O trabalho de investigação apresentado nesta dissertação foi desenvolvido tendo como objectivo a síntese e funcionalização de meso-triarilcorróis para utilização como quimiossensores. Este trabalho encontra-se apresentado ao longo de cinco capítulos. No primeiro capítulo são apresentadas as características gerais, as metodologias de síntese e de funcionalização de macrociclos de tipo corrólico, e descrevemse algumas aplicações em que têm sido utilizados. São ainda abordadas algumas das propriedades e características dos quimiossensores e os mecanismos de deteção de diversos analítos. No segundo capítulo, após uma pequena introdução às reações de Wittig e de Diels-Alder, escolhidas para a funcionalização do macrociclo corrólico, descreve-se o estudo efectuado para a obtenção do complexo de gálio(III) do 3- vinil-5,10,15-tris(pentafluorofenil)corrol e o seu comportamento como dieno, em reações de Diels-Alder na presença dos dienófilos 1,4-benzoquinona e 1,4- naftoquinona. Desses estudos resultaram dois aductos cuja habilidade sensorial, bem como a dos seus precursores, foi estudada, em solução, na presença de aniões esféricos (F-, Br-, Cl-), lineares (CN-) e volumosos (CH3COO-, H2PO4 -). Dos macrociclos estudados verificou-se que o corrol base-livre 5,10,15-tris(pentafluorofenil)corrol apresenta uma elevada sensibilidade para o anião fluoreto (F-), e que a coordenação do núcleo corrólico com gálio(III) diminui a afinidade para este anião. Em geral, todos os compostos mostraram afinidade para o anião cianeto (CN-) mesmo quando em suportes poliméricos. O gel de poliacrilamida revelou-se muito promissor na determinação de CN- em amostras de água. No terceiro capítulo é avaliada a reatividade do complexo de gálio(III) do 3- vinil-5,10,15-tris(pentafluorofenil)corrol ainda como dieno mas agora na presença de um dienófilo linear, o acetilenodicarboxilato de dimetilo. Desse estudo resultaram dois novos derivados corrólicos. A habilidade sensorial dos mesmos perante os aniões fluoreto, cianeto, acetato, e fosfato foi avaliada por espectroscopia de absorção e emissão tendo um dos aductos mostrado ser colorimétrico para o anião cianeto. No quarto capítulo descreve-se a síntese e caracterização de dois conjugados do tipo corrol-cumarina, resultantes de reações de Hetero-Diels-Alder entre o 3-vinil-5,10,15-tris(pentafluorofenil)corrolatogálio(III)(piridina) e orto-quinonasmetídeos gerados in situ a partir de reacções de Knoevenagel entre cumarinas e paraformaldeído. Realizaram-se estudos de afinidade sensorial para aniões e catiões com estes macrociclos, bem como com conjugados porfirinacumarina análogos. A inserção de uma unidade cumarina conferiu uma excepcional solubilidade tendo os novos derivados apresentado solubilidade em etanol. No quinto e último capítulo desta dissertação é avaliada a capacidade sensorial do 5,10,15-tris(pentafluorofenil)corrol e da sua espécie monoaniónica, para os catiões metálicos Na+, Ca2+, Cu2+, Cd2+, Pb2+, Hg2+, Ag+, Al3+, Zn2+, Ni2+, Cr3+, Ga3+, Fe3+ em tolueno e acetonitrilo. Os macrociclos corrólicos mostraram ser selectivos e colorimétricos para o catião Hg2+. Neste trabalho descreve-se ainda a síntese do derivado -iminocorrol, que após funcionalização com o 3-isocianatopropiltrimetoxisilano originou um derivado do tipo alcoxisilano, que foi, posteriormente, ancorado a nanopartículas comerciais de sílica. As novas nanopartículas ancoradas com o alcoxisilano corrol foram estudadas na presença de Cu2+, Hg2+ e Ag+. Na presença do catião Ag+ assistiu-se a uma mudança de cor, de verde para amarelo.

Photodynamic inactivation (PDI) is defined as the process of cell destruction by oxidative stress resulting from the interaction between light and a photosensitizer (PS), in the presence of molecular oxygen. PDI of bacteria has been extensively studied in recent years, proving to be a promising alternative to conventional antimicrobial agents for the treatment of superficial and localized infections. Moreover, the applicability of PDI goes far beyond the clinical field, as its potential use in water disinfection, using PS immobilized on solid supports, is currently under study. The aim of the first part of this work was to study the oxidative modifications in phospholipids, nucleic acids and proteins of Escherichia coli and Staphylococcus warneri, subjected to photodynamic treatment with cationic porphyrins. The aims of the second part of the work were to study the efficiency of PDI in aquaculture water and the influence of different physicalchemical parameters in this process, using the Gram-negative bioluminescent bacterium Vibrio fischeri, and to evaluate the possibility of recycling cationic PS immobilized on magnetic nanoparticles. To study the oxidative changes in membrane phospholipids, a lipidomic approach has been used, combining chromatographic techniques and mass spectrometry. The FOX2 assay was used to determine the concentration of lipid hydroperoxides generated after treatment. The oxidative modifications in the proteins were analyzed by one-dimensional polyacrylamide gel electrophoresis (SDS-PAGE). Changes in the intracellular nucleic acids were analyzed by agarose gel electrophoresis and the concentration of doublestranded DNA was determined by fluorimetry. The oxidative changes of bacterial PDI at the molecular level were analyzed by infrared spectroscopy. In laboratory tests, bacteria (108 CFU mL-1) were irradiated with white light (4.0 mW cm-2) after incubation with the PS (Tri-Py+-Me-PF or Tetra-Py+-Me) at concentrations of 0.5 and 5.0 μM for S. warneri and E. coli, respectively. Bacteria were irradiated with different light doses (up to 9.6 J cm-2 for S. warneri and up to 64.8 J cm-2 for E. coli) and the changes were evaluated throughout the irradiation time. In the study of phospholipids, only the porphyrin Tri-Py+-Me-PF and a light dose of 64.8 J cm-2 were tested. The efficiency of PDI in aquaculture has been evaluated in two different conditions: in buffer solution, varying temperature, pH, salinity and oxygen concentration, and in aquaculture water samples, to reproduce the conditions of PDI in situ. The kinetics of the process was determined in realtime during the experiments by measuring the bioluminescence of V. fischeri (107 CFU mL-1, corresponding to a level of bioluminescence of 105 relative light units). A concentration of 5.0 μM of Tri-Py+-Me-PF was used in the experiments with buffer solution, and 10 to 50 μM in the experiments with aquaculture water. Artificial white light (4.0 mW cm-2) and solar irradiation (40 mW cm-2) were used as light sources.

Expanded porphyrins are synthetic analogues of porphyrins, differing from the last ones and other naturally occurring tetrapyrrolic macrocycles by containing a larger central core, with a minimum of 17 atoms, while retaining the extended conjugation features that are a tremendous feature of these biological pigments. The core expansion results in various systems with novel spectral and electronic features, often uniques. Most of these systems can also coordinate cations and/or anions, and in some cases they can bind more than one of these species. In many cases, these molecules display structural features, such as non-planar structures, that have no antecedents in the chemistry of porphyrins or related macrocyclic compounds. This work will discuss several synthetic approaches for the synthesis of expanded porphyrins, namely the construction of new building blocks by Michael addition, as well as potential synthetic routes towards expanded porphyrins. The synthesis of smaller oligopyrrolic compounds namely, bipyrroles and dipyrromethanes, not only were developed for the synthesis of expanded porphyrins as they were also used in Knoevenagel condensations furnishing chromogenic compounds able to recognize different anions in solution. Also, an approach to the synthesis of novel expanded porphyrins namely sapphyrins has been done by aza-Michael additions. Several synthetic routes towards the synthesis of pyridyl and pyridinium N-Fused pentaphyrins and hexaphyrins have been explored in order to achieve compounds with potential applications in catalysis and PDI, respectively. Studies on the synthesis of compounds with potential anion binding properties, led to the structural characterization and NMR anion binding studies of [28]hexaphyrins functionalized with several diamines in the para position of their pentafluorophenyl groups. These compounds allow NH hydrogen bond interactions with various anions. All synthesized compounds were fully characterized by modern spectroscopic techniques.

Staphylococcus aureus are Gram-positive bacteria who integrate the human microbiota. Nevertheless, these bacteria can be pathogenic to the humans. Due to the increasing occurrence of antibiotic-resistant S. aureus new approaches to control this pathogen are necessary. The antimicrobial photodynamic inactivation process (PDI) is based in the combined use of a light source, an oxidizing agent like oxygen and an intermediary agent (a photosensitizer). These three components interact to form cytotoxic reactive oxygen species that irreversibly damage vital constituents of the microbial cells and ultimately lead to cell death. In fact, PDI is being shown to be a promising alternative to the antibiotic approach in the inactivation of pathogenic microorganisms. However, information on effects of photosensitization on particular virulence factors is strikingly scarce. The objective of this work was to evaluate the effect of PDI on virulence factors of S. aureus. For this, as photosensitizer the 5,10,15,20-tetrakis(1-methylpyridinium-4-yl)porphyrin tetra-iodide (Tetra-Py+-Me) and six strains of S. aureus (one reference strain, one strain with 1 enterotoxin, two strains with 3 enterotoxins and two strains resistant to methicillin, MRSA – one with 5 enterotoxins and the other without enterotoxins) were used. The effect of photosensitization on catalase activity, beta hemolysis, lipases, thermonuclease, enterotoxins, coagulase production and resistance to methicillin was assessed. The results indicate that the expression of some virulence factors in the cells subjected to this therapy is affected. Additionally the susceptibility of the strains to PDI did not decrease upon successive treatments.

Cationic porphyrins have been widely used as photosensitizers (PSs) in the inactivation of microorganisms, both in biofilms and in planktonic forms. However, the application of curcumin, a natural PS, in the inactivation of biofilms, is poorly studied. The objectives of this study were (1) to evaluate and compare the efficiency of a cationic porphyrin tetra (Tetra-Py+-Me) and curcumin in the photodynamic inactivation of biofilms of Pseudomonas spp and the corresponding planktonic form; (2) to evaluate the effect of these PSs in cell adhesion and biofilm maturation. In eradication assays, biofilms of Pseudomonas spp adherent to silicone tubes were subjected to irradiation with white light (180 J cm-2) in presence of different concentrations (5 and 10 μM) of PS. In colonization experiments, solid supports were immersed in cell suspensions, PS was added and the mixture experimental setup was irradiated (864 J cm-2) during the adhesion phase. After transference solid supports to new PS-containing medium, irradiation (2592 J cm-2) was resumed during biofilm maturation. The assays of inactivation of planktonic cells were conducted in cell suspensions added of PS concentrations equivalent to those used in experiments with biofilms. The inactivation of planktonic cells and biofilms (eradication and colonization assays) was assessed by quantification of viable cells after plating in solid medium, at the beginning and at the end of the experiments. The results show that porphyrin Tetra-Py+-Me effectively inactivated planktonic cells (3.7 and 3.0 log) and biofilms of Pseudomonas spp (3.2 and 3.6 log). In colonization assays, the adhesion of cells was attenuated in 2.2 log, and during the maturation phase, a 5.2 log reduction in the concentration of viable cells was observed. Curcumin failed to cause significant inactivation in planktonic cells (0.7 and 0.9 log) and for that reason it was not tested in biofilm eradication assays. In colonization assays, curcumin did not affect the adhesion of cells to the solid support and caused a very modest reduction (1.0 log) in the concentration of viable cells during the maturation phase. The results confirm that the photodynamic inactivation is a promising strategy to control installed biofilms and in preventing colonization. Curcumin, however, does not represent an advantageous alternative to porphyrins in the case of biofilms of Pseudomonas spp.

Sulfadimethoxine (SDM) is one of the drugs, often used in the aquaculture sector to prevent the spread of disease in freshwater fish aquaculture. Its spread through the soil and surface water can contribute to an increase in bacterial resistance. It is therefore important to control this product in the environment. This work proposes a simple and low-cost potentiometric device to monitor the levels of SDM in aquaculture waters, thus avoiding its unnecessary release throughout the environment. The device combines a micropipette tip with a PVC membrane selective to SDM, prepared from an appropriate cocktail, and an inner reference solution. The membrane includes 1% of a porphyrin derivative acting as ionophore and a small amount of a lipophilic cationic additive (corresponding to 0.2% in molar ratio). The composition of the inner solution was optimized with regard to the kind and/or concentration of primary ion, chelating agent and/or a specific interfering charged species, in different concentration ranges. Electrodes constructed with inner reference solutions of 1 × 10−8 mol/L SDM and 1 × 10−4 mol/L chromate ion showed the best analytical features. Near-Nernstian response was obtained with slopes of −54.1 mV/decade, an extraordinary detection limit of 7.5 ng/mL (2.4 × 10−8 mol/L) when compared with other electrodes of the same type. The reproducibility, stability and response time are good and even better than those obtained by liquid contact ISEs. Recovery values of 98.9% were obtained from the analysis of aquaculture water samples.

Solid-contact sensors for the selective screening of sulfadiazine (SDZ) in aquaculture waters are reported. Sensor surfaces were made from PVC membranes doped with tetraphenylporphyrin-manganese(III) chloride, α-cyclodextrin, β-cyclodextrin, or γ-cyclodextrin ionophores that were dispersed in plasticizer. Some membranes also presented a positive or a negatively charged additive. Phorphyrin-based sensors relied on a charged carrier mechanism. They exhibited a near-Nernstian response with slopes of 52 mV decade−1 and detection limits of 3.91 × 10−5 mol L−1. The addition of cationic lipophilic compounds to the membrane originated Nernstian behaviours, with slopes ranging 59.7–62.0 mV decade−1 and wider linear ranges. Cyclodextrin-based sensors acted as neutral carriers. In general, sensors with positively charged additives showed an improved potentiometric performance when compared to those without additive. Some SDZ selective membranes displayed higher slopes and extended linear concentration ranges with an increasing amount of additive (always <100% ionophore). The sensors were independent from the pH of test solutions within 2–7. The sensors displayed fast response, always <15 s. In general, a good discriminating ability was found in real sample environment. The sensors were successfully applied to the fast screening of SDZ in real waters samples from aquaculture fish farms. The method offered the advantages of simplicity, accuracy, and automation feasibility. The sensing membrane may contribute to the development of small devices allowing in locus measurements of sulfadiazine or parent-drugs.

Catalase dismutes H20 2 to O2 and H20. In successive twoelectron reactions H20 2 induces both oxidation and reduction at the heme group. In the first step the protoheme prosthetic group of beef liver catalase forms compound I, in which the heme has been oxidized from Fe3+ to Fe4+=0 and a porphyrin radical has been created. Compound II is formed by the oneelectron reduction of comp I. It retains Fe4+=0 but lacks the porphyrin radical and is catalytically inert. Molecular structures are available for Escherichia coli Hydroperoxidase II, Micrococcus Iysodeiktus, Penicillium vitale and beef liver enzymes, which contain different hemes and heme pockets. In the present work, the pockets and substrate access channels of protoheme (beef liver & Micrococcus) and heme d (HPII of E. coli and Penicillium) catalases have been analysed using Quanta™ and CharmMTM molecular modeling packages on the Silicon Graphics Iris Indigo 2 computer. Experimental studies have been carried out with two catalases, HPII (and its mutants) and beef liver. Fluoride and formate' are inhibitors of both enzymes, and their binding is modulated by the heme and by distal residues N201 & H128. Both HPII and beef liver enzymes form compound I with H202 or peracetate. The reduction of beef liver enzyme compound I to II and the decay of compound II are accelerated by fluoride. The decay of compound II is also accelerated by formate, and this reagent acts as a 2-electron donor towards compound I of both enzymes. It is concluded that heme d enzymes (Penicillium and HPII of E. coli) are formed by autocatalytic transformation of protoheme in a modified pocket which contains a characteristic serine residue as well as a partially occluded heme channel. They are less active than protoheme enzymes but also do not form the inactive compound II species. Binding of peroxide as well as fluoride and formate is prevented by mutation of H128 and modulated by mutation of N201.

There is considerable interest in intramolecular energy transfer, especially in complexes which absorb visible light, because it is crucial to the better understanding of photoharvesting systems in photosynthetic organisms and for utilizing solar energy as well. Porphyrin dimers represent one of the best systems for the exploration of light-induced intramolecular energy transfer. Many kinds of porphyrins and porphyrin dimers have been studied over the past decade, however little attention has been paid to the influence of paramagnetic metals on the behavior of their excited states. In this thesis, Electron Paramagnetic Resonance Spectroscopy (EPR) is used to study such compounds. After light irradiation, porphyrins easily produce a variety of excited states, which are spin polarized and can be detected by the time-resolved (TR) EPR technique. The spin polarized results for vanadyl porphyrins, their electrostatically-coupled dimers, a covalently-linked copper porphyrin-free base porphyrin dimer, and free base porphyrins are presented in this thesis. From these results we can conclude that the spin polarization patterns of vanadyl porphyrins come primarily from the trip-quartet state generated by intersystem crossing (lSC) from the excited sing-doublet state through the trip-doublet state. The spin polarization pattern of electrostatically-coupled vanadyl porphyrin-free base porphyrin dimer is produced by the triplet state of the free base porphyrin half which is coupled to the unpaired electron on the vanadyl ion.

Two classes of building blocks have been prepared and characterized and their coordination chemistry explored working towards the preparation of new molecule-based magnetic materials. In the first project, the amine functionality of 3,3'-diamino-2,2'- bipyridine was exploited for the preparation of a new family of ligands (H2L 1)-(H2L 4). The molecular structures of three ligands have been fully characterized by X-ray crystallography. [molecular structure diagram will not copy here, but is available in full pdf.] The coordination chemistry of these ligands with divalent first row transition metal ions was investigated. For ligand (H2L1), the molecular structures of four coordination complexes with stoichiometries [Zn2(Ll)(OAc)(MeO)]2 (I), [Cu2(L1)(OAc)2 (II), [Li(L1)]3 (III), and [Ni(L1)]3 (IV) were determined by X-ray crystallography. For ligand (H2L2), a Cu(II) complex of stoichiometry [Cu3(L2)(OAc)3MeO] (V) was determined by X-ray crystallography. The magnetic properties of complexes (II), (III), and (V) have been fully elucidated. In project two, synthetic strategies for the preparation of porphyrin molecules bearing triol substituents is presented. Following this approach, three new porphyrin derivatives have been prepared and characterized [Zn(HPTPP-CH2C(CH20H)3)] (VI), [P(TPP)(OCH2C(CH2)H)3)2]+CL- (VII), and [P(OEP)(C6H5)(OCH2C(CH2OH)3)]+Cl- (VIII). Attempts to exchange the labile methoxide bridges of a tetraironIIl single molecule magnet of stoichiometry [Fe4(OMe)6(dpm)6] (Hdpm = dipivaloylmethane) with the triol appended porphyrins will be discussed. ABSTRACT Photosystem II (PSII) of oxygenic photosynthesis has the unique ability to photochemically oxidize water, extracting electrons from water to result in the evolution of oxygen gas while depositing these electrons to the rest of the photosynthetic machinery which in turn reduces CO2 to carbohydrate molecules acting as fuel for the cell. Unfortunately, native PSII is unstable and not suitable to be used in industrial applications. Consequently, there is a need to reverse-engineer the water oxidation photochemical reactions of PSII using solution-stable proteins. But what does it take to reverse-engineer PSII’s reactions? PSII has the pigment with the highest oxidation potential in nature known as P680. The high oxidation of P680 is in fact the driving force for water oxidation. P680 is made up of a chlorophyll a dimer embedded inside the relatively hydrophobic transmembrane environment of PSII. In this thesis, the electrostatic factors contributing to the high oxidation potential of P680 are described. PSII oxidizes water in a specialized metal cluster known as the Oxygen Evolving Complex (OEC). The pathways that water can take to enter the relatively hydrophobic region of PSII are described as well. A previous attempt to reverse engineer PSII’s reactions using the protein scaffold of E. coli’s Bacterioferritin (BFR) existed. The oxidation potential of the pigment used for the BFR ‘reaction centre’ was measured and the protein effects calculated in a similar fashion to how P680 potentials were calculated in PSII. The BFR-RC’s pigment oxidation potential was found to be 0.57 V, too low to oxidize water or tyrosine like PSII. We suggest that the observed tyrosine oxidation in BRF-RC could be driven by the ZnCe6 di-cation. In order to increase the efficiency of iii tyrosine oxidation, and ultimately oxidize water, the first potential of ZnCe6 would have to attain a value in excess of 0.8 V. The results were used to develop a second generation of BFR-RC using a high oxidation pigment. The hypervalent phosphorous porphyrin forms a radical pair that can be observed using Transient Electron Paramagnetic Resonance (TR-EPR). Finally, the results from this thesis are discussed in light of the development of solar fuel producing systems.

Les complications vasculaires telles que l’augmentation de la contractilité et la prolifération cellulaire sont les complications les plus communes observées dans le diabète et l’hyperglycémie chronique est un facteur important dans ces processus. La voie de signalisation de Gαq joue un rôle important dans la régulation du tonus vasculaire et l’altération de celle-ci peut contribuer aux complications vasculaires observées dans les cas de diabète et d’hyperglycémie. Il a été observé que les taux et l’activité des protéines kinase C (PKC) et du diacylglycérol (DAG) sont augmentés dans ces conditions. Cependant, aucune étude n’a démontré l’implication de Gαq/11 et des PLCβ, molécules de signalisation en amont de PKC/DAG. Plusieurs études révèlent que l’augmentation des taux et de l’activité des PKC et du DAG induite par l’hyperglycémie dans des cellules du muscle lisse vasculaire (CMLV) est attribuée à l’augmentation du stress oxydatif. De plus, les niveaux de certains peptides vasoactifs, tels que l’angiotensine II et l’endothéline-1, augmentés dans les conditions de diabète/d’hyperglycémie, peuvent contribuer à l’augmentation du stress oxydatif observée. Le travail présenté dans cette thèse avait pour but d’examiner les effets de l’hyperglycémie sur les niveaux d’expression protéique de Gαq/11 et de ses molécules associées, ainsi que d’étudier le mécanisme moléculaire par lequel l’hyperglycémie module la voie de signalisation de Gαq dans les CMLV. Dans la première étude, nous avons examiné si l’hyperglycémie pouvait moduler l’expression des protéines Gαq, Gα11, PLCβ1 et PLCβ2. Le prétraitement des CMLV A10 avec 26 mM de glucose durant 72 heures augmente l’expression des protéines Gαq, Gα11, PLCβ-1 et PLCβ-2 en comparaison avec les CMLV témoins. Le traitement avec des antagonistes aux récepteurs AT1 de l’Ang II, et ETA/ETB de l’ET-1, atténue la hausse de Gαq, de Gα11, de PLCβ1 et de PLCβ2 induite par l’hyperglycémie. De plus, la formation d’IP3 stimulée par l’ET-1 était plus élevée dans les CMLV exposée à 26 mM de glucose. Le traitement des CMLV A10 avec l’Ang II et l’ET-1 augmente également les niveaux d’expression des protéines Gα q/11 et PLCβ. Cette augmentation de l’expression est restaurée au niveau des CMLV témoins par les antagonistes des récepteurs AT1, ETA et ETB. Ces résultats suggèrent que l’augmentation de l’expression des protéines Gαq/11 et PLCβ dans les CMLV induite par l’hyperglycémie est attribuée à l’activation des récepteurs AT1, ETA et ETB. Dans la seconde étude, nous avons examiné l’implication du stress oxydatif dans l’augmentation des niveaux d’expression des protéines Gαq/11 et PLCβ et de leur signalisation induite par l’hyperglycémie. Nous avons également déterminé le mécanisme responsable de l’augmentation du stress oxydatif induite par l’hyperglycémie. L’augmentation de l’expression des protéines Gαq/11 et PLCβ des CMLV A10 exposées à 26 mM de glucose est revenue au niveau basal après un traitement avec l’antioxydant diphenyleneiodonium (DPI), et la catalase, un chélateur du peroxyde d’hydrogène, mais pas par le 111Mn-tetralis(benzoic acid porphyrin) (MnTBAP) ni par l’acide urique, des chélateurs du peroxynitrite. De plus, l’augmentation de la formation d’IP3 stimulée par l’ET-1 dans les CMLV exposées à 26 mM de glucose est revenue au niveau basal après un traitement avec le DPI et la catalase. Ces résultats suggèrent que l’augmentation du stress oxydatif induite par l’hyperglycémie contribue à l’augmentation de l’expression des protéines Gαq/11 et les molécules associées à la voie de signalisation de Gq. De plus, l’augmentation de la production d’anion superoxyde (O2-), de l’activité de la NADPH oxydase et de l’expression des protéines p22(phox) et p47(phox) induite par l’hyperglycémie est revenue à un niveau basal après un traitement avec les antagonistes des récepteurs AT1, ETA et ETB. Ces résultats suggèrent que l’hyperglycémie augmente les niveaux endogènes de l’Ang II et de l’ET-1, ce qui augmente le stress oxydatif par la formation d’O2- et de H2O2 et peut contribuer à l’augmentation des niveaux de Gq/11α et de leurs molécules de signalisation. Puisqu’il a été observé que l’hyperglycémie transactive les récepteurs aux facteurs de croissance tels que le récepteur au facteur de croissance épidermique (EGF-R) et le récepteur au facteur de croissance dérivé des plaquettes (PDGF-R), nous avons entrepris d’examiner, dans la troisième étude, l’implication d’EGF-R et de PDGF-R dans l’augmentation des niveaux de Gαq/11, de PLCβ et de leur signalisation induite par l’hyperglycémie. L’augmentation des niveaux d’expression des protéines Gαq, Gα11, PLCβ-1 et PLCβ-2 induite par l’hyperglycémie est revenue au niveau basal après un traitement avec les inhibiteurs d’EGF-R (AG1478) et de PDGF-R (AG1295) et par l’inhibiteur de c-Src, PP2. L’augmentation de la phosphorylation d’EGF-R et de PDGF-R induite par l’hyperglycémie a été abolie par AG1478, AG1295 et PP2. De plus, l’augmentation des niveaux de Gαq/11, et de PLCβ induite par l’hyperglycémie est atténuée par l’inhibiteur des MAPK, le PD98059, et par l’inhibiteur d’AKT, le wortmannin. L’augmentation de la phosphorylation d’ERK et d’AKT était également atténuée par AG1478 et AG1295. Ces résultats suggèrent que la transactivation des récepteurs aux facteurs de croissance induite par c-Src peut contribuer à l’augmentation des niveaux de Gα q/11/PLC et de leur signalisation par la voie des MAPK/PI3K. En conclusion, les études présentées dans cette thèse indiquent que l’hyperglycémie augmente les niveaux de Gαq/11 et de PLCβ. Nous avons émis des évidences qui démontrent que l’augmentation endogène de l’Ang II et de l’ET-1 par l’hyperglycémie peut contribuer à l’augmentation de la production d’O2- et de H2O2 résultant ainsi en une augmentation du stress oxydatif qui pourrait être responsable de l’augmentation de Gαq/11/PLC et de leur signalisation dans les conditions d’hyperglycémie. Finalement, nous avons démontré que la transactivation des récepteurs aux facteurs de croissance induite par l’hyperglycémie peut être responsable de l’augmentation de Gαq/11/PLC et les molécules associées à la voie de signalisation de Gq dans les cas de diabète et d’hyperglycémie.

La vectorisation des médicaments est une approche très prometteuse tant sur le plan médical qu’économique pour la livraison des substances actives ayant une faible biodisponibilité. Dans ce contexte, les polymères en étoile et les dendrimères, macromolécules symétriques et branchées, semblent être les solutions de vectorisation les plus attrayantes. En effet, ces structures peuvent combiner efficacement une stabilité élevée dans les milieux biologiques à une capacité d’encapsulation des principes actifs. Grâce à leur architecture bien définie, ils permettent d’atteindre un très haut niveau de reproductibilité de résultats, tout en évitant le problème de polydispersité. Bien que des nombreuses structures dendritiques aient été proposées ces dernières années, il est cependant à noter que la conception de nouveaux nanovecteurs dendritiques efficaces est toujours d’actualité. Ceci s’explique par des nombreuses raisons telles que celles liées à la biocompatibilité, l’efficacité d’encapsulation des agents thérapeutiques, ainsi que par des raisons économiques. Dans ce projet, de nouvelles macromolécules branchées biocompatibles ont été conçues, synthétisées et évaluées. Pour augmenter leur efficacité en tant qu’agents d’encapsulations des principes actifs hydrophobes, les structures de ces macromolécules incluent un coeur central hydrophobe à base de porphyrine, décanediol ou trioléine modifié et, également, une couche externe hydrophile à base d’acide succinique et de polyéthylène glycol. Le choix des éléments structuraux de futures dendrimères a été basé sur les données de biocompatibilité, les résultats de nos travaux de synthèse préliminaires, ainsi que les résultats de simulation in silico réalisée par une méthode de mécanique moléculaire. Ces travaux ont permis de choisir des composés les plus prometteurs pour former efficacement et d’une manière bien contrôlable des macromolécules polyesters. Ils ont aussi permis d’évaluer au préalable la capacité de futurs dendrimères de capter une molécule médicamenteuse (itraconazole). Durant cette étape, plusieurs nouveaux composés intermédiaires ont été obtenus. L’optimisation des conditions menant à des rendements réactionnels élevés a été réalisée. En se basant sur les travaux préliminaires, l’assemblage de nouveaux dendrimères de première et de deuxième génération a été effectué, en utilisant les approches de synthèse divergente et convergente. La structure de nouveaux composés a été prouvée par les techniques RMN du proton et du carbone 13C, spectroscopie FTIR, UV-Vis, analyse élémentaire, spectrométrie de masse et GPC. La biocompatibilité de produits a été évaluée par les tests de cytotoxicité avec le MTT sur les macrophages murins RAW-262.7. La capacité d’encapsuler les principes actifs hydrophobes a été étudiée par les tests avec l’itraconazole, un antifongique puissant mais peu biodisponible. La taille de nanoparticules formées dans les solutions aqueuses a été mesurée par la technique DLS. Ces mesures ont montré que toutes les structures dendritiques ont tendance à former des micelles, ce qui exclue leurs applications en tant que nanocapsules unimoléculaires. L’activité antifongique des formulations d’itraconazole encapsulé avec les dendrimères a été étudiée sur une espèce d’un champignon pathogène Candida albicans. Ces tests ont permis de conclure que pour assurer l’efficacité du traitement, un meilleur contrôle sur le relargage du principe actif était nécessaire.

A metalloporphyrin incorporated carbon paste sensor has been developed for the determination of metronidazole benzoate (MTZB). Zn(II) complex of 5,10,15,20-tetrakis (3-methoxy-4-hydroxy phenyl) porphyrin (TMHPP) was used as the active material. The MTZB gave a well-defined reduction peak at - 0.713V in 0.1 mol l -1 phosphate buffer solution of pH around 7. Compared with bare carbon paste electrode (CPE), the TMHPP Zn(II) modified electrode significantly enhanced the reduction peak current of MTZB as well as lowered its reduction potential. Under optimum conditions the reduction peak current was proportional to MTZB concentration over the range 1×10-3 mol1-1 to 1×10-5mol1-1. The detection limit was found to be 4.36×10-6mol1-1 . This sensor has been successfully applied for the determination of MTZB in pharmaceutical formulations and urine samples.

Present thesis has discussed the design and synthesis of polymers suitable for nonlinear optics. Most of the molecules that were studied have shown good nonlinear optical activity. The second order nonlinear optical activity of the polymers was measured experimentally by Kurtz and Perry powder technique. The thesis comprises of eight chapters.The theory of NLO phenomenon and a review about the various nonlinear optical polymers has been discussed in chapter 1. The review has provided a survey of NLO active polymeric materials with a general introduction, which included the principles and the origin of nonlinear optics, and has given emphasis to polymeric materials for nonlinear optics, including guest-host systems, side chain polymers, main chain polymers, crosslinked polymers, chiral polymers etc.Chapter 2 has discussed the stability of the metal incorporated tetrapyrrole molecules, porphyrin, chlorin and bacteriochlorin.Chapter 3 has provided the NLO properties of certain organic molecules by computational tools. The chapter is divided into four parts. The first part has described the nonlinear optical properties of chromophore (D-n-A) and bichromophore (D-n-A-A-n-D) systems, which were separated by methylene spacer, by making use of DPT and semiempirical calculations.Chapter 4: A series of polyurethanes was prepared from cardanol, a renewable resource and a waste of the cashew industry by previously designed bifunctional and multifunctional polymers using quantum theoretical approach.Chapter 5: A series of chiral polyurethanes with main chain bis azo diol groups in the polymer backbone was designed and NLO activity was predicted by ZlNDO/ CV methods.In Chapter 7, polyurethanes were first designed by computational methods and the NLO properties were predicted by correction vector method. The designed bifunctional and multifunctional polyurethanes were synthesized by varying the chiral-achiral diol compositions
