The small G protein Ras has been implicated in hypertrophy of cardiac myocytes. We therefore examined the activation (GTP loading) of Ras by the following hypertrophic agonists: phorbol 12-myristate 13-acetate (PMA), endothelin-1 (ET-1), and phenylephrine (PE). All three increased Ras.GTP loading by 10-15-fold (maximal in 1-2 min), as did bradykinin. Other G protein-coupled receptor agonists (e.g. angiotensin II, carbachol, isoproterenol) were less effective. Activation of Ras by PMA, ET-1, or PE was reduced by inhibition of protein kinase C (PKC), and that induced by ET-1 or PE was partly sensitive to pertussis toxin. 8-(4-Chlorophenylthio)-cAMP (CPT-cAMP) did not inhibit Ras.GTP loading by PMA, ET-1, or PE. The association of Ras with c-Raf protein was increased by PMA, ET-1, or PE, and this was inhibited by CPT-cAMP. However, only PMA and ET-1 increased Ras-associated mitogen-activated protein kinase kinase 1-activating activity, and this was decreased by PKC inhibition, pertussis toxin, and CPT-cAMP. PMA caused the rapid appearance of phosphorylated (activated) extracellular signal-regulated kinase in the nucleus, which was inhibited by a microinjected neutralizing anti-Ras antibody. We conclude that PKC- and Gi-dependent mechanisms mediate the activation of Ras in myocytes and that Ras activation is required for stimulation of extracellular signal-regulated kinase by PMA.

Estrogen is an important steroid hormone that mediates most of its effects on regulation of gene expression by binding to intracellular receptors. The consensus estrogen response element (ERE) is a 13 bp palindromic inverted repeat with a three nucleotide spacer. However, several reports suggest that many estrogen target genes are regulated by diverse elements, such as imperfect EREs and ERE half sites (ERE 1/2),which are either the proximal or the distal half of the palindrome. To gain more insight into ERE half site-mediated gene regulation, we used a region from the estrogen-regulated chicken riboflavin carrier protein (RCP) gene promoter that contains ERE half sites. Using moxestrol, an analogue of estrogen and transient transfection of deletion and mutation containing RCP promoter/reporter constructs in chicken hepatoma (LMH2A) cells, we identified an estrogen response unit (ERU) composed of two consensus ERE 1/2 sites and one non-consensus ERE 1/2 site. Mutation of any of these sites within this ERU abolishes moxestrol response. Further, the ERU is able to confer moxestrol responsiveness to a heterologous promoter. Interestingly, RCP promoter is regulated by moxestrol in estrogen responsive human MCF-7 cells, but not in other cell lines such as NIH3T3 and HepG2 despite estrogen receptor-alpha (ER-�) co transfection. Electrophoretic mobility shift assays (EMSAs) with promoter regions encompassing the half sites and nuclear extracts from LMH2A cells show the presence of a moxestrol-induced complex that is abolished by a polyclonal anti-ER� antibody. Surprisingly, estrogen receptor cannot bind to these promoter elements in isolation. Thus, there appears to be a definite requirement for some other factor(s) in addition to estrogen receptor, for the generation of a suitable response of this promoter to estrogen. Our studies therefore suggest a novel mechanism of gene regulation by estrogen, involving ERE half sites without direct binding of ER to the cognate elements.

The aim of the study was to verify whether post-menopausal hormone replacement therapy (HRT) modifies autoantibody titers against oxidized low-density lipoprotein (LDL) (anti-LDLoxi), against epitopes of oxidized apolipoprotein B100 and common carotid intima-media thickness (IMT) in these women. Sixty-eight women in pre-menopause (PMW) and 216 in post-menopause (POMW) were recruited; eighty-three had undergone HRT for at least 12 months, where 48 received conjugated estrogens alone (EHRT) and 35 received conjugated estrogen and medroxyprogesterone acetate (CHRT). ELISA was used to determine autoantibodies. Lipoprotein lipase (LPL), hepatic lipase (HL), cholesterol ester transfer protein (CETP) and phospholipid transfer protein (PLTP) activities were assayed by radiometric methods. IMT was measured using Doppler ultrasound. Anti-oxidized LDL and anti-D antibodies increased by 40% (p <= 0.003) and 42% (p <= 0.006), respectively, with menopause. There was a surprising and significant 7% reduction in anti-D2 antibody titers with HRT (p <= 0.050), indicating a positive effect of treatment on the immune response to oxidized LDL. Combined HRT decreased activities of HL and LPL. HRT did not change common carotid IMT, which was increased by 32% as expected after menopause (p <= 0.030). This study describes, for the first time, the protective effect of HRT on decreasing autoantibody titers against oxidized apolipoprotein B in LDL.

Endostatin (ES) is a potent inhibitor of angiogenesis and tumor growth. Continuous ES delivery of ES improves the efficacy and potency of the antitumoral therapy. The TheraCyte (R) system is a polytetrafluoroethylene (PTFE) semipermeable membrane macroencapsulation system for implantation of genetically engineered cells specially designed for the in vivo delivery of therapeutic proteins, such as ES, which circumvents the problem of limited half-life and variation in circulating levels. In order to enable neovascularization at the tissues adjacent to the devices prior to ES secretion by the cells inside them, we designed a scheme in which empty TheraCyte (R) devices were preimplanted SC into immunodeficient mice. Only after healing (17 days later) were Chinese hamster ovary cells expressing ES injected into the preimplanted devices. In another model for device implantation, the cells expressing ES where loaded into the immunoisolation devices prior to implantation into the animals, and the TheraCyte (R) were then immediately implanted SC into the mice. Throughout the 2-month study, constant high ES levels of up to 3.7 mu g/ml were detected in the plasma of the mice preimplanted with the devices, while lower but also constant levels of ES (up to 2.1 mu g/ml plasma) were detected in the mice that had received devices preloaded with the ES-expressing cells. Immunohistochemistry using anti-ES antibody showed reaction within the device and outside it, demonstrating that ES, secreted by the confined recombinant cells, permeated through the membrane and reached the surrounding tissues.

Background: Hepatocyte growth factor (HGF) is overexpressed after acute kidney injury (AKI). The aim of this study was to evaluate the role of endogenous HGF in the progression of the inflammatory response in glycerol-induced AKI (Gly-AKI) in rats. Methods: Renal and systemic HGF expressions were evaluated during the development of Gly-AKI. Subsequently, the blockade of endogenous HGF was analyzed in rats treated with anti-HGF antibody concomitant to glycerol injection. Apoptosis, cell infiltration and chemokine and cytokine profiles were investigated. Results: We detected an early peak of renal and plasma HGF protein expressions 3 h after glycerol injection. The pharmacological blockade of the endogenous HGF exacerbated the renal impairment, the tubular apoptosis, the renal expression of monocyte chemoattractant protein-1 and the macrophage, CD43+, CD4+ and CD8+ T lymphocytes renal infiltration. The analysis of mRNA expressions of Th1 (t-bet, TNF-alpha, IL-1 beta) and Th2 (gata-3, IL-4, IL-10) cytokines showed a Th1-polarized response in Gly-AKI rats that was aggravated with the anti-HGF treatment. Conclusion: Endogenous HGF attenuates the renal inflammatory response, leukocyte infiltration and Th1 polarization after glycerol injection. The control of cellular immune response may partly explain the protective effect of endogenous HGF in the development of Gly-AKI. Copyright (C) 2008 S. Karger AG, Basel

This study was undertaken to evaluate the prevalence of GB virus C (GBV-C) viraemia and anti-E2 antibody, and to assess the effect of co-infection with GBV-C and HIV during a 10-year follow-up of a cohort of 248 HIV-infected women. Laboratory variables (mean and median CD4 counts, and HIV and GBV-C viral loads) and clinical parameters were investigated. At baseline, 115 women had past exposure to GBV-C: 57 (23%) were GBV-C RNA positive and 58 (23%) were anti-E2 positive. There was no statistical difference between the groups (GBV-C RNA + /anti-E2 -, GBV-C RNA - /anti-E2 + and GBV-C RNA - /anti-E2 -) regarding baseline CD4 counts or HIV viral loads (P = 0.360 and 0.713, respectively). Relative risk of death for the GBV-C RNA + /anti-E2 - group was 63% lower than that for the GBV-C RNA - /anti-E2 - group. Multivariate analysis demonstrated that only HIV loads >= 100,000 copies/mL and AIDS-defining illness during follow-up were associated with shorter survival after AIDS development. It is likely that antiretroviral therapy (ART) use in our cohort blurred a putative protective effect related to the presence of GBV-C RNA.

Introdução A mastocitose abrange um grupo heterogêneo de condições crônicas caracterizado pela proliferação excessiva de mastócitos nos tecidos. Os sinais e sintomas clínicos são decorrentes da distribuição anatômica dos mastócitos e do efeito funcional dos mediadores produzidos e liberados por estas células. Na infância, a doença é considerada uma condição benigna na maioria dos casos, cujo comprometimento característico é o cutâneo. As mais freqüentes manifestações na pele são os mastocitomas e a urticária pigmentosa. Lesões cutâneas bolhosas podem manifestar-se e acompanhar todas as formas de mastocitose e quando esta apresentação é a predominante, é denominada de mastocitose bolhosa. O diagnóstico de mastocitose é suspeitado clinicamente e confirmado pela histologia. A demonstração do aumento do número de mastócitos nas lesões cutâneas características se constitui no principal critério diagnóstico. Contudo, este método tem dificuldades técnicas que impedem a adequada reprodutibilidade dos achados, dificultando a elucidação de casos duvidosos e retardando seu tratamento. Considerando as propriedades imunológicas e a importância clínica dos mastócitos reveste-se de maior importância compreender o papel destas células nas doenças, sendo indispensável identificá-las e enumerá-las com acurácia nos tecidos. Objetivos Quantificar o número de mastócitos marcados com anticorpo monoclonal antitriptase, através de técnica imuno-histoquímica e análise de imagem, em biópsias cutâneas de crianças, com diagnóstico clínico de mastocitose. Descrever os achados histológicos; quantificar o número de mastócitos marcados com o anticorpo antitriptase entre as diferentes expressões clínicas da mastocitose cutânea; comparar o número de mastócitos entre os casos de mastocitose cutânea e mastocitose associada à sintomas sistêmicos e correlacionar as contagens de mastócitos entre os dois diferentes métodos (coloração por Giemsa com contagem manual e marcação com anticorpo antitriptase e análise digital). Material e Método Foram incluídas no estudo biópsias cutâneas de crianças de 0 a 14 anos, com diagnóstico clínico e histológico de mastocitose. Os casos foram classificados de acordo com a apresentação clínica cutânea em mastocitoma, urticária pigmentosa ou mastocitose bolhosa e assinalada a presença de sintomas sistêmicos associados. Os fragmentos de pele fixados em formalina e emblocados em parafina foram cortados e utilizados para diagnóstico histopatológico convencional, corados com hematoxilina-eosina e Giemsa, e para análise imuno-histoquímica com estreptavidina peroxidase marcados com anticorpo antitriptase. A densidade de mastócitos (número de células por área) foi realizada por um único observador na técnica histológica e através de um sistema de análise de imagem de vídeo no método imuno-histoquímico. Resultados Foram avaliados 33 casos de mastocitose, sendo 21 do sexo masculino. Dez casos (30,3%) apresentavam mastocitoma, 21 (63,6%) urticária pigmentosa e 2 (6,1%) mastocitose bolhosa. Todos os casos da amostra foram classificados como tendo mastocitose incipiente e em 6 (18,8%) pacientes pôde ser identificada a associação com sintomas sistêmicos. Prurido foi o sintoma mais freqüente, sendo relatado em 21 casos. Em 21 dos 33 casos foi identificada a infiltração de mastócitos na derme havendo predominância pela região perivascular (p=0,001, teste exato de Fisher). Não houve diferenças significativas entre a presença de infiltrado mastocitário e as várias formas cutâneas de mastocitose ou a mastocitose sistêmica. A presença de eosinófilos foi identificada em 15 casos (45,5%) e em 10 casos associadamente ao infiltrado perivascular de mastócitos. A densidade de mastócitos na técnica histológica, incluindo-se todos os casos, foi 50,00 células/mm2. Não houve diferença significativa das contagens entre os pacientes com mastocitoma e aqueles com urticária pigmentosa, assim como entre os pacientes com e sem sintomas sistêmicos associados aos cutâneos. A densidade de mastócitos encontrada com a técnica imuno-histoquímica e contagem por análise de imagem foi 158,85 células/mm2. Não houve diferença significativa das contagens entre os pacientes com mastocitoma e aqueles com urticária pigmentosa, assim como entre aqueles com e sem sintomas sistêmicos. Comparando-se a contagem dos mastócitos por área (densidade) entre a histologia e a imuno-histoquímica houve uma diferença significativa (p=0,0001 teste não-paramétrico de Wilcoxon). A média da diferença entre as contagens foi 199,98 células/mm2 (±365,31 DP). Também não houve semelhança, entre os dois métodos, nos grupos mastocitoma e urticária pigmentosa (p=0,005 e p=0,01, respectivamente, teste não-paramétrico de Wilcoxon). Puderam ser identificados 518% a mais de mastócitos com a técnica imunohistoquímica quando comparada com a histológica. Conclusões O presente estudo permite concluir que: 1) a localização preferencial da infiltração de mastócitos é dérmica e perivascular, não sendo possível identificar diferenças histológicas entre casos de urticária pigmentosa e mastocitoma; 2) o número de mastócitos marcados com o anticorpo monoclonal antitriptase e contados com análise digital de imagem, em biópsia de pele de crianças com diagnóstico clínico de mastocitose, foi 159 células por milímetro quadrado; 3) a densidade de mastócitos, foi semelhante entre os casos de urticária pigmentosa e mastocitoma e entre os casos com e sem sintomas sistêmicos associados nas duas diferentes técnicas empregadas; 4) o número de mastócitos por milímetro quadrado com a técnica imuno-histoquímica e a contagem através de análise de imagem foi significativamente maior quando comparada com a coloração através de Giemsa e a contagem manual, com uma diferença média entre os dois métodos de 200 células por milímetro quadrado; 5) a densidade de mastócitos com a técnica imunohistoquímica foi significativamente maior tanto nos casos com urticária pigmentosa quanto nos com mastocitoma, quando comparada com a técnica empregada rotineiramente e 6) com a técnica imuno-histoquímica e a contagem através de análise de imagem foi possível identificar 518% a mais de mastócitos quando comparada com a técnica histológica.

Visceral leishmaniasis hás adapted in the past 20 years to periurban and urban areas, and in Natal, Rio Grande do Norte, it became endemic. Thid study aimed to evaluate the environmental and social aspectsof Leishmania chagasi infection and its epidemiologic transmission chain in an urban, periurban and rural area of Parnamirim-RN. A study with three sections was conducted: Section 1: Sectional study of the human and canine infection by L. chagasi and its environmental and social determinants. Section 2: Observational longitudinal cohort to evaluate the dynamics of the canine infection. Section 3: Longitudinal study to evaluate the behavior of Lu. Longipalpis vector and the seasonal factors related to its dynamics. To include in the study the hauses were randomly selected and georreferenciated. Montenegro skin test was done in the human population and blood samples were collected for anti-Leishmania antibody detection. The canine population was examinated for L. chagasi infection by RIFI, ELISA and ELISA for rK39. An entomologic surveillance was monthly done with CDC light trapsin 10 houses of each locality. Quantitative and qualitative analyses was done using STATISC 6.0. Probality and prediction maps were done using ArcGis 9.0 model. In the human population L. chagasi infection was associated with the area of the hause, age, sex, population densyti, vegetation, kind of the floor of thr hause, water and resides destiny. In the canine population L. chagasi infection was associated with the breed, size, time of living in the hause, presence of dogs in the neighborhood, presence of horses and donkeys in the neighborhood, vegetation, kind of the floor and walls of the hause. The human infection was associated with canine infction only when analyzed taking into account the locality. In the prospective study, serum conversion and antibody lost observed in 30,8% and 22% of the animals examined, respectively. The human infection rate by L. chagasi was 24,6%, by the presence of anti-Leishmania antibody and 38,6% by the Montenegro skin test. The canine infection rate 32,5% by the presence of anti-Leishmania antibody. The vector Lu longipalpis showed an atypical behavior. These results indicate that environmental and social factors are important variables associated with L. chagasi infection in humans and canines, with punctual association of thr last two. Control measures of the infection on the studied points are necessary, in the aim to reduce the endemic focus of the disease in the study area. 1. The relationship between repeated thermal treatments and hepatic synthesis of Hsp 70 was studied in broiler chickens.2. Sixty broilers were submitted to 5 different treatments (12 birds each) from day 1 to day 42. Four groups were kept in a thermoneutral environment and subjected to 0, 1, 2 and 3 heat stress episodes at 35 degrees C for 4 h per week (TN-0, TN-1, TN-2 and TN-3, respectively). The last group (HT-35) was reared at a room temperature of 35 degrees C.3. From 39 to 42 old, the birds experienced acute heat stress at 41 degrees C. Resistance to heat stress was evaluated by the time taken for rectal temperature to increase by 3 degrees C above the pre-treatment value. Livers were collected (before and after heat stress) and Hsp70 was determined using Western Blot analysis with monoclonal anti-Hsp70 antibody.4. Resistance to heat stress and concentration of Hsp70 were higher in those birds subjected to more heat stress episodes during the experimental period (TN-3) and HT-35. A positive correlation was observed between Hsp70 concentration and the time taken for a 3 degrees C increase in rectal temperature (r=0.42; P<0.01).5. Exposing birds to episodes of heat stress (35 degrees C) during rearing may improve their resistance to acute heat stress, but the previous thermal history did not seem to influence the hepatocyte Hsp70 content after exposure to more severe heat stress (41 degrees C).

Humoral immune response of water buffalo naturally infected with Toxocara vitulorum was monitored using three different antigens of this parasite in serum and colostrum of buffalo cows and calves. Soluble extract (Ex) and excretory/secretory (ES) larval antigens and perienteric fluid antigen (Pe) of adult T vitulorum were used to measure the antibody levels by an indirect ELISA. Serum of 7-12 buffalo cows for the first 365 days and colostrum of the same number of buffalo cows for the first 60 days of parturition, and serum of 8-10 buffalo calves for the first 365 days afterbirth were assayed. The ELISA detected antibodies against all three T vitulorum antigens in the colostrum and serum of 100% of buffalo cows and calves examined. The highest antibody levels against Ex, ES and Pe antigens were detected in the buffalo cow sera during the perinatal period and were maintained at high levels through 300 days after parturition. on the other hand, colostrum antibody concentrations of all three antigens were highest on the first day post-parturition, but decreased sharply during the first 15 days. Concomitantly to the monitoring of immune response, the parasitic status of the calves was also evaluated. In calves, antibodies passively acquired were at the highest concentrations 24 h after birth and remained at high levels until 45 days coincidentally with the peak of T vitulorum infection. The rejection of the worms by the calves occurred simultaneously with the decline of antibody levels, which reached their lowest levels between 76 and 150 days. Thereafter, probably because of the presence of adults/larvae stimulation, the calves acquired active immunity and the antibodies started to increase slightly in the serum and plateaued between the days 211 and 365. All three antigens were detected by the serum antibodies of buffalo calves; however, the concentration of anti-Pe antibody was higher than anti-EX and anti-ES, particularly after 90 days of age. By conclusion, the buffalo cows develop immunity and keep high levels of antibodies against T vitulorum-Ex, ES and Pe antigens and these antibodies are transferred to their calves through the colostrum. This passively acquired immunity does not protect the calves against the acquisition of the infection, but these antibodies, passively or actively acquired, may have an important role during worm rejection by the calves and prevention of intestinal reinfection. Foram utilizadas 99 vacas prenhes distribuídas em oito grupos que receberam os seguintes tratamentos: grupo I, com 29 vacas não vacinadas e seus bezerros que não receberam probiótico, ficando como controle; grupo II, com 10 vacas vacinadas e seus bezerros que não receberam probiótico; grupos III, IV e V, com 10 animais cada, vacas vacinadas e seus bezerros que receberam probiótico durante 5, 15 e 30 dias, respectivamente; os grupos VI, VII e VIII, com 10 animais cada, vacas não vacinadas e seus bezerros que receberam probiótico durante 5, 15 e 30 dias, respectivamente. Cada animal dos grupos vacinados recebeu duas doses vacinais contendo os pili K99 e A14 de Escherichia coli na dose de 5,0ml por via subcutânea. O probiótico contendo Ruminobacter amylophilum, Ruminobacter succinogenes, Succinovibrio dextrinosolvens, Bacillus cereus, Lactobacillus acidophilus e Streptococcus faecium, na dose de 3,0× 10(8) células vivas (UFC) de cada amostra em 250ml de leite, era adiministrado por via oral. Os animais foram observados diariamente e foram determinados os títulos de anticorpos anti-K99 e anti-A14 no soro e no colostro. Anotaram-se os pesos dos bezerros ao nascimento e aos 30 dias. Os resultados mostraram que a associação de vacina com probiótico administrado por 15 e 30 dias foram os tratamentos mais eficientes no controle da diarréia e ganho de peso.

Este estudo objetivou caracterizar a resposta imune celular no sistema nervoso central (SNC) de eqüinos com infecção crônica experimental por Trypanosoma evansi. Para este propósito, foram utilizados os métodos histoquímicos (HE) e imunoistoquímicos do complexo avidina-biotina peroxidase (ABC). O fenótipo do infiltrado celular foi caracterizado com o auxílio de anticorpos anti - CD3, para linfócitos T e antiBLA36 para linfócitos B. Os macrófagos foram marcados com anticorpo antiantígenos da linhagem mielóide/histiócitos (Clone Mac387). A lesão no sistema nervoso central (SNC) dos eqüinos infectados com T. evansi foi caracterizada como meningoencefalite e meningomielite não supurativa. A gravidade das lesões variou em diferentes segmentos do SNC, refletindo distribuição irregular das alterações vasculares. A distribuição de células T e B e antígenos do complexo maior de histocompatibilidade classe II foram avaliados dentro do SNC de eqüinos cronicamente infectados com T. evansi. O infiltrado perivascular e meníngeo eram constituídos predominantemente por células T e B. Macrófagos foram raramente visualizados. T.evansi não foi identificado no parênquima do SNC dos eqüinos.
