1 The Early Holocene sediment of a lake at tree line (Gouillé Rion, 2343 m a.s.l.) in the Swiss Central Alps was sampled for plant macrofossils. Thin (0.5 cm) slices, representing time intervals of c. 50 years each from 11 800 to 7800 cal. year bp, were analysed and the data compared with independent palaeoclimatic proxies to study vegetational responses to environmental change. 2 Alpine plant communities (e.g. with Salix herbacea) were established at 11 600–11 500 cal. year bp, when oxygen-isotope records showed that temperatures increased by c. 3–4 °C within decades. Larix decidua trees reached the site at c. 11 350 cal. year bp, probably in response to further warming by 1–2 °C. Forests dominated by L. decidua persisted until 9600 cal. year bp, when Pinus cembra became more important. 3 The dominance of Larix decidua for two millennia is explained by dry summer conditions, and possibly low winter temperatures, which favoured it over the late-successional Pinus cembra. Environmental conditions were a result of variations in the earth's orbit, leading to a maximum of summer and a minimum of winter solar radiation. Other heliophilous and drought-adapted species, such as Dryas octopetala and Juniperus nana, could persist in the open L. decidua forests, but were out-competed when the shade-tolerant P. cembra expanded. 4 The relative importance of Larix decidua decreased during periods of diminished solar radiation at 11 100, 10 100 and 9400 cal. year bp. Stable concentrations of L. decidua indicate that these percentage oscillations were caused by temporary increases of Pinus cembra, Dryas octopetala and Juniperus nana that can be explained by increases in moisture and/or decreases in summer temperature. 5 The final collapse of Larix decidua at 8400 cal. year bp was possibly related to abrupt climatic cooling as a consequence of a large meltwater input to the North Atlantic. Similarly, the temporary exclusion of Pinus cembra from tree line at 10 600–10 200 cal. year bp may be related to slowing down of thermohaline circulation at 10 700–10 300 cal. year bp. 6 Our results show that tree line vegetation was in dynamic equilibrium with climate, even during periods of extraordinarily rapid climatic change. They also imply that forecasted global warming may trigger rapid upslope movements of the tree line of up to 800 m within a few decades or centuries at most, probably inducing large-scale displacements of plant species as well as irrecoverable biodiversity losses.

Three well-dated pollen diagrams from 1985 m, 2050 m, and at the tree line at 2150 m asl show the vegetational succession in the central Altai Mountains since 16 cal ka BP. Pioneer vegetation after deglaciation was recorded first at the lowest site. Subsequently, dense dry steppe vegetation developed coincident with the change from silt to organic sediments at the two lower sites, but silt lasted longer at the highest site, indicating the persistence of bare ground there. Forests of Pinus sibirica, Pinus sylvestris, Picea obovata, Larix sibirica, Abies sibirica, and Betula pendula started to develop about 12 cal ka BP with the change to a warmer and wetter climate at the beginning of the Holocene. Results indicate that the timberline did not rise above the highest site. Mesophilous dark-coniferous forests were fully developed by 9.5 cal ka BP. The role of Abies and Picea decreased by about 7.5 cal ka BP suggesting cooler climate, after which the forests changed little until today. The vegetational development in this portion of the central Altai Mountains is compatible with that described in neighbouring areas of the Altai, southern Siberia, Mongolia, and Kazakhstan.

Background: Bernese mountain dogs are reported to have a shorter life expectancy than other breeds. A Major reason for this has been assigned to a high tumour prevalence, especially of histiocytic sarcoma. The efforts made by the breeding clubs to improve the longevity with the help of genetic tests and breeding value estimations are impeded by insufficiently reliable diagnoses regarding the cause of death. The current standard for post mortem examination in animals is performance of an autopsy. In human forensic medicine, imaging modalities, such as computed tomography and magnetic resonance imaging, are used with increasing frequency as a complement to autopsy. The present study investigates, whether post mortem computed tomography in combination with core needle biopsy is able to provide a definitive diagnosis of histiocytic sarcoma. For this purpose we have analysed the results of post mortem computed tomography and core needle biopsy in eleven Bernese mountain dogs. In the subsequent autopsy, every dog had a definitive diagnosis of histiocytic sarcoma, based on immunohistochemistry. Results: Computed tomography revealed space-occupying lesions in all dogs. Lesion detection by post mortem computed tomography was similar to lesion detection in autopsy for lung tissue (9 cases in computed tomography / 8 cases in autopsy), thoracic lymph nodes (9/8), spleen (6/7), kidney (2/2) and bone (3/3). Hepatic nodules, however, were difficult to detect with our scanning protocol (2/7). Histology of the core needle biopsies provided definitive diagnoses of histiocytic sarcoma in ten dogs, including confirmation by immunohistochemistry in six dogs. The biopsy samples of the remaining dog did not contain any identifiable neoplastic cells. Autolysis was the main reason for uncertain histological diagnoses. Conclusions: Post mortem computed tomography is a fast and effective method for the detection of lesions suspicious for histiocytic sarcoma in pulmonary, thoracic lymphatic, splenic, osseous and renal tissue. Optimization of the procedure regarding the scanning protocol and tissue sample size and number will improve the accuracy of the method. BACKGROUND Recombinant bone morphogenetic protein two (rhBMP2) has been utilised for a variety of clinical applications in orthopaedic surgery and dental procedures. Despite its widespread use, concerns have been raised regarding its short half-life and transient bioactivity in vivo. Recent investigation aimed at developing rhBMP2 synthesized from a shorter polypeptide chain (108 amino acids) has been undertaken. METHODS The osteopromotive properties of BMP2 were investigated on cell behaviour. Five concentrations of rhBMP2_108 including 10, 50, 100, 200 and 500 ng/ml were compared to a commercially available rhBMP2 (100 ng/ml). Each of the working concentrations of rhBMP2_108 were investigated on MC3T3-E1 osteoblasts for their ability to induce osteoblast recruitment, proliferation and differentiation as assessed by alkaline phosphatase (ALP) staining, alizarin red staining, and real-time PCR for genes encoding ALP, osteocalcin (OCN), collagen-1 (COL-1) and Runx2. RESULTS The results demonstrate that all concentrations of rhBMP2_108 significantly improved cell recruitment and proliferation of osteoblasts at 5 days post seeding. Furthermore, rhBMP2_108 had the most pronounced effects on osteoblast differentiation. It was found that rhBMP2_108 had over a four fold significant increase in ALP activity at seven and 14 days post-seeding and the concentrations ranging from 50 to 200 ng/ml demonstrated the most pronounced effects. Analysis of real-time PCR for genes encoding ALP, OCN, COL-1 and Runx2 further confirmed dose-dependant increases at 14 days post-seeding. Furthermore, alizarin red staining demonstrated a concentration dependant increase in staining at 14 days. CONCLUSION The results from the present study demonstrate that this shorter polypeptide chain of rhBMP2_108 is equally as bioactive as commercially available rhBMP2 for the recruitment of progenitor cells by facilitating their differentiation towards the osteoblast lineage. Future in vivo study are necessary to investigate its bioactivity.

There is a great similarity between pollen types which occur in the early Holocene NE Tibetan pollen spectra and those which are commonly considered to be typical for the Würm Late Glacial period in Central Europe and for the Würm Pleniglacial period in Southern Europe. Evidently, this similarity is due to a remarkable general conformity of plant taxa growing in cold-arid regions of the northern hemisphere. The improvement of the climate and the retreat of the glaciers that commenced at the end of the Würm period had already terminated definitely before 9500 BP. In addition, the climatic situation as well as the vegetation belts must have remained rather constant during the following 3000 yr, i.e. through most parts of the climatic optimum of the Holocene.

En el contexto de la posible habilitación de una ruta entre Chile y Argentina a través del paso fronterizo Las Leñas en Chile central, alternativo al Cristo Redentor, se procedió a analizar las vulnerabilidades y los impactos provocados por la geodinámica en áreas montañosas. Se evaluaron procesos erosivos en los ambientes de baja, media y alta montaña en el valle del río Cachapoal. La alta energía de estos paisajes es causante de la generación y el aporte de sedimentos mediante erosión, la que es activada por las precipitaciones intensas y la acción periglacial. Las diferencias erosivas se entienden por la presencia de distintas formas del paisaje; así, los depósitos basales en alta montaña están asociados a movimientos del terreno en masa, y la erosión lineal se desarrolla sobre las terrazas fluviales ubicadas en fondo de valle, las que se encuentran estabilizadas por la mayor cobertura vegetal.

Bulk sediment chemistry from three Chilean continental margin Ocean Drilling Program sites constrains regional continental erosion over the past 30,000 years. Sediments from thirteen rivers that drain the (mostly igneous) Andes and the (mostly metamorphic) Coast Range, along with existing rock chemistry datasets, define terrestrial provenance for the continental margin sediments. Andean river sediments have high Mg/Al relative to Coast-Range river sediments. Near 36°S, marine sediments have high-Mg/Al (i.e. more Andean) sources during the last glacial period, and lower-Mg/Al (less Andean) sources during the Holocene. Near 41°S a Ti-rich source, likely from coast-range igneous intrusions, is prevalent during Holocene time, whereas high-Mg/Al Andean sources are more prevalent during the last glacial period. We infer that there is a dominant ice-sheet control of sediment sources. At 36°S, Andean-sourced sediment decreased as Andean mountain glaciers retreated after ~17.6 ka, coincident with local oceanic warming and southward retreat of the Patagonian Forest and, by inference, westerly winds. At 41°S Andean sediment dominance peaks and then rapidly declines at ~19 ka, coincident with local oceanic warming and the earliest deglacial sea-level rise. We hypothesize that this decreased flux of Andean material in the south is related to rapid retreat of the marine-based portion of the Patagonian Ice Sheet in response to global sea-level rise, as the resulting flooding of the southern portion of the Central Valley created a sink for Andean sediments in this region. Reversal of the decreasing deglacial Mg/Al trend at 41°S from 14.5 to 13.0 ka is consistent with a brief re-advance of the Patagonian ice sheet coincident with the Antarctic Cold Reversal.

We present new major and trace element and O-Sr-Nd-isotope data for igneous rocks from the western Mediterranean Alborán Sea, collected during the METEOR 51/1 cruise, and for high-grade schists and gneisses from the continental Alborán basement, drilled during the Ocean Drilling Programme (ODP Leg 161, Site 976). The geochemical data allow a detailed examination of crustal and mantle processes involved in the petrogenesis of the lavas and for the first time reveal a zonation of the Miocene Alborán Sea volcanism: (1) a keel-shaped area of LREE-depleted (mainly tholeiitic series) lavas in the central Alborán Sea, generated by high degrees of partial melting of a depleted mantle source and involving hydrous fluids from subducted marine sediments, that is surrounded by (2) a horseshoe-shaped zone with LREE-enriched (mainly calc-alkaline series) lavas subparallel to the arcuate Betic-Gibraltar-Rif mountain belt. We propose that the geochemical zonation of the Miocene Alborán Basin volcanism results from eastward subduction of Tethys oceanic lithosphere coupled with increasing lithospheric thickness between the central Alborán Sea and the continental margins of Iberia and Africa.

During Integrated Ocean Drilling Program Expedition 302 (Arctic Coring Expedition (ACEX)) a more than 200 m thick sequence of Paleogene organic carbon (OC)-rich (black shale type) sediments was drilled. Here we present new biomarker data determined in ACEX sediment samples to decipher processes controlling OC accumulation and their paleoenvironmental significance during periods of Paleogene global warmth and proposed increased freshwater discharge in the early Cenozoic. Specific source-related biomarkers including n-alkanes, fatty acids, isoprenoids, carotenoids, hopanes/hopenes, hopanoic acids, aromatic terpenoids, and long-chain alkenones show a high variability of components, derived from marine and terrestrial origin. The distribution of hopanoic acid isomers is dominated by compounds with the biological 17beta(H), 21beta(H) configuration indicating a low level of maturity. On the basis of the biomarker data the terrestrial OC supply was significantly enriched during the late Paleocene and part of the earliest Eocene, whereas increased aquatic contributions and euxinic conditions of variable intensity were determined for the Paleocene-Eocene thermal maximum and Eocene thermal maximum 2 events as well as the middle Eocene time interval. Furthermore, samples from the middle Eocene are characterized by the occurrence of long-chain alkenones, high proportions of lycopane, and high ratios (>0.6) of (n-C35 + lycopane)/n-C31. The occurrence of C37-alkenenones, which were first determined toward the end of the Azolla freshwater event, indicates that the OC becomes more marine in origin during the middle Eocene. Preliminary UK'37- based sea surface temperature (SST) values display a longterm temperature decrease of about 15C during the time interval 49-44.5 Ma (25° to 10°C), coinciding with the global benthic d18O cooling trend after the early Eocene climatic optimum. At about 46 Ma, parallel with onset of ice-rafted debris, SST (interpreted as summer temperatures) decreased to values <15°C. For the late early Miocene a SST of 11°-15°C was determined. Most of the middle Eocene ACEX sediments are characterized by a smooth short-chain n-alkane distribution, which may point to natural oil-type hydrocarbons from leakage of petroleum reservoirs or erosion of related source rocks and redeposition.
