AbstractIn this thesis t-BOC and methacrylate functionalisedoligothiophenes with 3 5 thiophene units where synthesizedby Stille coupling. The material was structured by methodslike stamping and photolithography.The polymerizability of the methacrylates was proved by thepolymerization in solution with AIBN and subsequent analysisby gel permeation chromatography and differential scanningcalorimetry. The conductivities of the doped polymer filmswere in the range published for oligothiophenes.The optical and electrochemical properties of the compoundswere measured and compared to known compounds.The cleavage of the t-BOC groups was followed bythermogravimetry and infrared spectroscopy. The cleavagetemperature can be lowered by up to 100 °C by the use ofphoto acid generators. The cleavage is complete after 2hours annealing.The methacrylates were structured by microinjection mouldingin capillaries (MIMIC), stamping with soft silicone moulds,filling of patterned substrates and preperation of an opalreplica. By MIMIC line patterns with 5- 50 µm line widthswere obtained. The stamping succeeded in structures with 500nm line width only, on which liquid crystals wereorientated. This shows the possible application asorientation layers in LEDs with polarized emission. Withsilica opal templates the three-dimensional structuring ofthe oligomers succeeded.ZusammenfassungIn dieser Arbeit wurden neue funktionalisierteOligothiophene hergestellt und unter Anwendung verschiedenerVerfahren wie Stempeltechniken oder Photolithographiestrukturiert. Dazu wurden Oligothiophene mit drei bis fünfThiopheneinheiten durch Stille-Kupplung synthetisiert. Alsfunktionelle Gruppen wurden t-BOC-Ester undMethacrylsäureester eingeführt.Die Polymerisierbarkeit der Methacrylate wurde durch diePolymerisation mit AIBN in Lösung und anschließendeGelpermeationschromatographie und Differentialkalorimetrienachgewiesen. Die Leitfähigkeiten der dotierten Polymerfilmelagen im Bereich der für Oligothiophene bekannten Werte.Die optischen und elektrochemischen Eigenschaften derVerbindungen wurden untersucht und mit den Eigenschaftenbekannter Verbindungen verglichen. Die Abspaltung der t-BOC-Gruppen wurde thermogravimetrischund infrarotspektroskopisch verfolgt. Es wurde gezeigt, daßdie Abspaltungstemperatur durch den Zusatz einesPhotosäuregenerators um bis zu 100°C gesenkt wird und dieAbspaltung nach zweistündigem Tempern vollständig ist.Die methacrylatfunktionalisierten Verbindungen wurdenstrukturiert durch Micro Injection Moulding in Capillaries(MIMIC), Prägen mit weichen Silikonstempeln, Füllen vonstrukturierten Substraten und die Herstellung einerOpalreplika. Durch die Strukturierung mit MIMIC wurdenLinienstrukturen mit Linienbreiten von 5-50 µm erhalten.Durch Prägen wurden Strukturen mit Linienbreiten von nur 500nm erreicht, auf diesen gelang die Orientierung vonFlüssigkristallen. Dies zeigt die mögliche Anwendung alsOrientierungsschichten in Leuchtdioden mit polarisierterEmission. Durch die Verwendung von Siliziumoxidopalen alsTemplate gelang die dreidimensionale Strukturierung derOligomere.

In dieser Arbeit wurden neue, ionische Oligo- und Poly-thiopene synthetisiert. Diese Verbindungen wurden unterAnwendung der Multischichttechnik von Decher zwei- unddreidimensional strukturiert. Zur Synthese der Verbindungen wurde die Stille-Kupplung vonThiophenbromiden und -zinnorganylen eingesetzt. Als ionischeGruppen fanden Sulfonium- und Phosphoniumsalze Verwendung. Bei optischen Untersuchungen konnte die effektive Kon-jugations länge der Substanzen bestimmt werden. MittelsCyclovoltammetrie wurden ihre Redoxeigenschaften untersucht.Die Polymere wurden mittels GPC, DSC und TGA charak-terisiert. Die Charakterisierung des Polythiophen-Multischichtaufbausdurch UV-Spektroskopie bewies die Gleichförmigkeit desSchichtaufbaus. Der Schichtaufbau unter Verwendung vonBolaamphiphilen verlief ebenfalls erfolgreich. Elektrochemische Untersuchungen an den Multischichtenergaben ein reversibles Verhalten, ebenfalls durchgeführteImpedanz-Messungen bewiesen das reversible Verhalten derRedox-Reaktion. Polyelektrolytschichten wurden ebenfalls auf ionisch imMikrometerbereich vorstrukturierten Substraten aufgebaut, umeine dreidimensionale Struktur zu erhalten. Ionischfunktionalisierte Polymerlatices wurden ebenfalls alsTemplat zum Aufbau dreidimensionaler Strukturen eingesetzt. Es wurden Gelbildner auf Terthiophenbasis synthetisiert,deren Struktur durch Röntgenmessungen untersucht wurde. DerGelbildner konnte ab 4% Massenanteil organische Lösungs-mittel vergelen.

The capability of goats to maintain milk production during water deprivation is remarkable and not yet fully understood. The aim of the present study was to investigate whether intravenous infusions of hypertonic NaCl cause release of both vasopressin and oxytocin and whether the peptides, in combination with the hyperosmolality, affect milk flow and milk composition. Six Swedish domestic landrace goats in their first to third lactation were milked every 30 min during experiments. Hypertonic NaCl (HNaCl) or isotonic NaCl (IsoNaCl) were infused for 90 min. Goats were not allowed to drink during infusions. Plasma vasopressin concentration increased during HNaCl infusions, and did not change in response to IsoNaCl infusions. Plasma oxytocin concentration did not change during either infusion. Milk flow was maintained during the infusions. Milk fat concentration decreased in the three samples taken before onset of the infusions, but then increased gradually during HNaCl infusions, while it continued to fall during the IsoNaCl infusions. Milk osmolality followed the rise in plasma osmolality during the HNaCl infusions and did not change in IsoNaCl experiments. Milk lactose concentration increased throughout both series of experiments, the concentration being higher during HNaCl infusions. Milk protein concentration did not change during HNaCl infusions, but fell in the IsoNaCl experiments. It is concluded that the hyperosmolality in combination with elevated plasma vasopressin levels did not disturb the secretory activity of the mammary cells, but rather facilitated emptying of the alveolar milk. Such a mechanism may help to explain the sustained milk production in water deprived goats.

Three Clark faculty members are studying the development of the Worcester Biotechnology Cluster for any lessons it might hold for current efforts to catalyze the growth of a sustainable energy/green jobs cluster in Worcester or elsewhere. Mary Ellen Boyle (GSOM), Jennie Stephens (IDCE/ESP), and Jing Zhang (GSOM) have conducted extensive in-depth interviews and combed the literature of cluster development to produce several articles and working papers.

The first cases of early-onset progressive polyneuropathy appeared in the Alaskan Malamute population in Norway in the late 1970s. Affected dogs were of both sexes and were ambulatory paraparetic, progressing to non-ambulatory tetraparesis. On neurologic examination, affected dogs displayed predominantly laryngeal paresis, decreased postural reactions, decreased spinal reflexes and muscle atrophy. The disease was considered eradicated through breeding programmes but recently new cases have occurred in the Nordic countries and the USA. The N-myc downstream-regulated gene (NDRG1) is implicated in neuropathies with comparable symptoms or clinical signs both in humans and in Greyhound dogs. This gene was therefore considered a candidate gene for the polyneuropathy in Alaskan Malamutes. The coding sequence of the NDRG1 gene derived from one healthy and one affected Alaskan Malamute revealed a non-synonymous G>T mutation in exon 4 in the affected dog that causes a Gly98Val amino acid substitution. This substitution was categorized to be "probably damaging" to the protein function by PolyPhen2 (score: 1.000). Subsequently, 102 Alaskan Malamutes from the Nordic countries and the USA known to be either affected (n = 22), obligate carriers (n = 7) or healthy (n = 73) were genotyped for the SNP using TaqMan. All affected dogs had the T/T genotype, the obligate carriers had the G/T genotype and the healthy dogs had the G/G genotype except for 13 who had the G/T genotype. A protein alignment showed that residue 98 is conserved in mammals and also that the entire NDRG1 protein is highly conserved (94.7%) in mammals. We conclude that the G>T substitution is most likely the mutation that causes polyneuropathy in Alaskan Malamutes. Our characterization of a novel candidate causative mutation for polyneuropathy offers a new canine model that can provide further insight into pathobiology and therapy of human polyneuropathy. Furthermore, selection against this mutation can now be used to eliminate the disease in Alaskan Malamutes.

BACKGROUND Primary hyperoxaluria type 3 (PH3) is characterized by mutations in the 4-hydroxy-2-oxoglutarate aldolase (HOGA1) gene. PH3 patients are believed to present with a less severe phenotype than those with PH1 and PH2, but the clinical characteristics of PH3 patients have yet to be defined in sufficient detail. The aim of this study was to report our experience with PH3. METHODS Genetic analysis of HOGA1 was performed in patients with a high clinical suspicion of PH after the presence of mutations in the alanine-glyoxylate aminotransferase gene had been ruled out. Clinical, biochemical and genetic data of the seven patients identified with HOGA1 mutations were subsequently retrospectively reviewed. RESULTS Among the seven patients identified with HOGA1 mutations the median onset of clinical symptoms was 1.8 (range 0.4-9.8) years. Five patients initially presented with urolithiasis, and two other patients presented with urinary tract infection. All patients experienced persistent hyperoxaluria. Seven mutations were found in HOGA1, including two previously unreported ones, c.834 + 1G > T and c.3G > A. At last follow-up, two patients had impaired renal function based on estimated glomerular filtration rates (GFRs) of 77 and 83 mL/min per 1.73 m(2), respectively. CONCLUSIONS We found that the GFR was significantly impaired in two of our seven patients with PH3 diagnosed during childhood. This finding is in contrast to the early-impaired renal function in PH1 and PH2 and appears to refute to preliminary reassuring data on renal function in PH3.

El aborto está penalizado en nuestro país, con excepción del que se practica por razones terapéuticas. Sin embargo, la desproporción entre el número de abortos que se producen –el último estudio estimado de Delicia Ferrando habla de 410,000 al año1[2]-, aquellos que son denunciados y, los que terminan en procesos judiciales, revela que si bien la práctica del aborto es ilegal, la sociedad, con su silencio, parece ampararla y de esa manera, tolerar la muerte de miles de mujeres por esa causa. Desde el Derecho Penal y los estudios de criminología se puede afirmar que las leyes penalizadoras no han cumplido con su finalidad de prevención específica ni general2, es decir, la amenaza de una posible sanción no ha logrado inhibir la práctica del aborto, sino más bien, se ha convertido en una forma de ejercer violencia contra las mujeres en múltiples planos. Ello se manifiesta en el incremento de las muertes maternas y en los altos costos que la atención de abortos incompletos y de los problemas derivados de su práctica clandestina representan para los servicios de salud. En este sentido, es válido y pertinente considerar al aborto como un problema de salud pública.
