The proper management of agricultural crop residues could produce benefits in a warmer, more drought-prone world. Field experiments were conducted in sugarcane production areas in the Southern Brazil to assess the influence of crop residues on the soil surface in short-term CO2 emissions. The study was carried out over a period of 50 days after establishing 6 plots with and without crop residues applied to the soil surface. The effects of sugarcane residues on CO2 emissions were immediate; the emissions from residue-covered plots with equivalent densities of 3 (D50) and 6 (D100) t ha-1 (dry mass) were less than those from non-covered plots (D0). Additionally, the covered fields had lower soil temperatures and higher soil moisture for most of the studied days, especially during the periods of drought. Total emissions were as high as 553.62 ± 47.20 g CO2 m-2, and as low as 384.69 ± 31.69 g CO2 m-2 in non-covered (D0) and covered plot with an equivalent density of 3 t ha-1 (D50), respectively. Our results indicate a significant reduction in CO2 emissions, indicating conservation of soil carbon over the short-term period following the application of sugarcane residues to the soil surface.

Laparoscopy is not widely used as a tool to perform assisted reproduction techniques in South American cervids; thus, scarce information in literature is available regarding its effects and appropriate anesthetic protocols to perform it. This study evaluated the effect of laparoscopy on heart rate (HR), respiration rate (RR), saturation of oxyhemoglobin (SpO2) and rectal temperature (RT) of six female brown brocket deer (Mazama gouazoubira) anesthetized with ketamine (5mg/kg), xylazine (0.3mg/kg), midazolam (0.5mg/kg) combination i.v. and isoflurane. Twelve laparoscopies were performed and each animal was used twice with a 40-day interval. After anesthetized, the animals were placed in dorsal recumbency to perform laparoscopy procedure using abdominal CO2 insufflations (14.2 ± 2.39mmHg; M ± SE). The main events of the laparoscopy procedure were divided into three periods: animal without (P1) and with abdominal insufflation (P2) and abdominal insufflation with the hips raised at 45º (P3). As a control, the animals were anesthetized again 40 days after the last laparoscopy, and were maintained in a dorsal recumbency for the same average duration of the previous anesthesia and no laparoscopy procedure was conducted. The period of anesthesia for the controls was also divided into P1, P2, and P3 considering the average duration of these periods in previous laparoscopies performed. Data were analyzed through the (ANOVA) variance analysis followed by Tukey test and values at P<0.05 were considered significant. No significant differences were observed in the parameters evaluated at P1, P2 and P3 between the animals submitted to laparoscopy and control. However, the RR mean between P1 (38.8 ± 4.42) and P3 (32.7 ± 4.81); and the RT mean between the P1 (38.2ºC ± 0.17), P2 (37.6ºC ± 0.19) and P3 (37.0ºC ± 0.21) varied significantly, independent of the laparoscopy. These data indicated that laparoscopy didn't cause any significant alterations in the cardiorespiratory parameters evaluated, even though the anesthetic protocol used can cause a reduction in the RT contributing to development of hypothermia during anesthesia.

Water table draw-down is thought to increase peat decomposition and, therefore, DOC release. However, several studies have shown lower DOC concentrations during droughts relative to ‘normal’ periods with high water table. We carried out controlled incubation experiments at 10°C on 10x10 cm peat soil cores collected from six UK sites across a sulphur deposition gradient. Our aim was to quantify the balance between microbial consumption and chemical precipitation of DOC due to episodic acidification driven by sulphur redox reactions by comparing changes in soil water chemistry to microbial activity (i.e. soil respiration and trace gas fluxes). During dry periods, all sites showed a concurrent increase in SO4 and soil respiration and a decline in DOC. However, the magnitude of change in both DOC and SO4 varied considerably between sites according to historical sulphur deposition loads and the variation in acid/base chemistry.

The effect of episodic drought on dissolved organic carbon (DOC) dynamics in peatlands has been the subject of considerable debate, as decomposition and DOC production is thought to increase under aerobic conditions, yet decreased DOC concentrations have been observed during drought periods. Decreased DOC solubility due to drought-induced acidification driven by sulphur (S) redox reactions has been proposed as a causal mechanism; however evidence is based on a limited number of studies carried out at a few sites. To test this hypothesis on a range of different peats, we carried out controlled drought simulation experiments on peat cores collected from six sites across Great Britain. Our data show a concurrent increase in sulphate (SO4) and a decrease in DOC across all sites during simulated water table draw-down, although the magnitude of the relationship between SO4 and DOC differed between sites. Instead, we found a consistent relationship across all sites between DOC decrease and acidification measured by the pore water acid neutralising capacity (ANC). ANC provided a more consistent measure of drought-induced acidification than SO4 alone because it accounts for differences in base cation and acid anions concentrations between sites. Rewetting resulted in rapid DOC increases without a concurrent increase in soil respiration, suggesting DOC changes were primarily controlled by soil acidity not soil biota. These results highlight the need for an integrated analysis of hydrologically driven chemical and biological processes in peatlands to improve our understanding and ability to predict the interaction between atmospheric pollution and changing climatic conditions from plot to regional and global scales.

Climacteric and non-climacteric fruits have traditionally been viewed as representing two distinct programmes of ripening associated with differential respiration and ethylene hormone effects. In climacteric fruits, such as tomato and banana, the ripening process is marked by increased respiration and is induced and co-ordinated by ethylene, while in non-climacteric fruits, such as strawberry and grape, it is controlled by an ethylene-independent process with little change in respiration rate. The two contrasting mechanisms, however, both lead to texture, colour, and flavour changes that probably reflect some common programmes of regulatory control. It has been shown that a SEPALLATA(SEP)4-like gene is necessary for normal ripening in tomato. It has been demonstrated here that silencing a fruit-related SEP1/2-like (FaMADS9) gene in strawberry leads to the inhibition of normal development and ripening in the petal, achene, and receptacle tissues. In addition, analysis of transcriptome profiles reveals pleiotropic effects of FaMADS9 on fruit development and ripening-related gene expression. It is concluded that SEP genes play a central role in the developmental regulation of ripening in both climacteric and non-climacteric fruits. These findings provide important information to extend the molecular control of ripening in a non-climacteric fruit beyond the limited genetic and cultural options currently available.

We used a light-use efficiency model of photosynthesis coupled with a dynamic carbon allocation and tree-growth model to simulate annual growth of the gymnosperm Callitris columellaris in the semi-arid Great Western Woodlands, Western Australia, over the past 100 years. Parameter values were derived from independent observations except for sapwood specific respiration rate, fine-root turnover time, fine-root specific respiration rate and the ratio of fine-root mass to foliage area, which were estimated by Bayesian optimization. The model reproduced the general pattern of interannual variability in radial growth (tree-ring width), including the response to the shift in precipitation regimes that occurred in the 1960s. Simulated and observed responses to climate were consistent. Both showed a significant positive response of tree-ring width to total photosynthetically active radiation received and to the ratio of modeled actual to equilibrium evapotranspiration, and a significant negative response to vapour pressure deficit. However, the simulations showed an enhancement of radial growth in response to increasing atmospheric CO2 concentration (ppm) ([CO2]) during recent decades that is not present in the observations. The discrepancy disappeared when the model was recalibrated on successive 30-year windows. Then the ratio of fine-root mass to foliage area increases by 14% (from 0.127 to 0.144 kg C m-2) as [CO2] increased while the other three estimated parameters remained constant. The absence of a signal of increasing [CO2] has been noted in many tree-ring records, despite the enhancement of photosynthetic rates and water-use efficiency resulting from increasing [CO2]. Our simulations suggest that this behaviour could be explained as a consequence of a shift towards below-ground carbon allocation.

Paracoccidioides brasiliensis is a thermo-dimorphic fungus that is the causative agent of paracoccidioidomyicosis (PCM), a human systemic granulomatous mycosis found in Latin America. Dimorphic transition from mycelium to yeast is required for establishing pathogenicity. Dimorphism is marked by changes in mitochondrial physiology, including modulation of respiration rate. In this work, we present the identification of three P. brasiliensis nuclear genes PbCOX9, PbCOX12, and PbCOX16 that code for structural sub-units and a putative assembly facilitator (PbCOX16) of the mitochondrial cytochrome c oxidase (COX), the terminal enzyme complex of the respiratory chain. We measured their expression pattern during the dimorphic transition from mycelium to yeast and back by real-time reverse transcription quantitative polymerase chain reaction (real-time RT-qPCR). Our results show that messages from these genes increase during the mycelium to yeast transition and decrease during the opposite conversion. This result supports active mitochondrial participation in the transition. Heterologous complementation of the corresponding Saccharomyces cerevisiae null mutant with the PbCOX9 gene was successfully obtained. (C) 2008 The British Mycological Society. O nitrogênio e potássio são os nutrientes requeridos em maiores quantidades pelas gramas e no Brasil não se tem informação da quantidade a ser aplicada para se obter a formação de tapete em menor tempo possível. Dois experimentos foram instalados em vasos em casa de vegetação, com o objetivo de avaliar o efeito de doses de nitrogênio e de potássio na produção de tapetes de grama esmeralda (Zoysia japonica). O delineamento utilizado para cada experimento foi fatorial com doses de N ou K e épocas de avaliação. Foram aplicadas quatro doses de nitrogênio (0, 200, 400 e 600 kg ha-1) e quatro doses de potássio (0, 100, 200, e 300 kg ha-1). As doses de nitrogênio e potássio foram aplicadas parceladamente em cobertura. O aumento das doses de N influenciou a taxa de cobertura do solo pela grama (TCS) permitindo a formação do tapete com a dose de 408 kg ha-1 de N aos 198 dias após a colheita do tapete anterior, tempo menor quando comparado com as demais doses. A concentração de N na folha e da cor verde da grama foram influenciadas pelas doses de N podendo ser utilizadas para auxiliar na recomendação das doses de N. O aumento das doses de K não influenciou na TCS pela grama, sendo o teor no solo (1,4 mmol c dm-3) suficiente para a produção dos tapetes de grama esmeralda.

Among the toxic elements, Cd has received considerable attention in view of its association with a number of human health problems. The objectives of this study were to evaluate the Cd availability and accumulation in soil, transfer rate and toxicity in lettuce and rice plants grown in a Cd-contaminated Typic Hapludox. Two simultaneous greenhouse experiments with lettuce and rice test plants were conducted in a randomized complete block design with four replications. The treatments consisted of four Cd rates (CdCl2), 0.0; 1.3; 3.0 and 6.0 mg kg(-1), based on the guidelines recommended by the Environmental Agency of the State of São Paulo, Brazil (Cetesb). Higher Cd rates increased extractable Cd (using Mehlich-3, Mehlich-1 and DTPA chemical extractants) and decreased lettuce and rice dry matter yields. However, no visual toxicity symptoms were observed in plants. Mehlich-1, Mehlich-3 and DTPA extractants were effective in predicting soil Cd availability as well as the Cd concentration and accumulation in plant parts. Cadmium concentration in rice remained below the threshold for human consumption established by Brazilian legislation. on the other hand, lettuce Cd concentration in edible parts exceeded the acceptable limit.

A aplicação de fertilizantes fosfatados por meio de fertirrigação com sistemas de irrigação localizada pode causar obstrução de emissores. Para evitar esse problema, pode ser utilizado o ácido fosfórico como fonte de fósforo às plantas. Porém, têm sido pouco investigados os efeitos da irrigação relacionados às perdas de CO2 do solo para a atmosfera, em conseqüência da decomposição do carbono orgânico e da infiltração de água no solo. Neste trabalho, investigou-se, no período de um mês, o efeito da fertirrigação com ácido fosfórico nas taxas de emissão de CO2 de um latossolo desprovido de vegetação, na Área Experimental de Irrigação da UNESP, Câmpus de Jaboticabal - SP. Utilizou-se de um sistema de irrigação por gotejamento, com delineamento experimental em blocos casualizados, constando de cinco repetições e cinco tratamentos (0; 30; 60; 90 e 120 kg ha-1de P2O5), aplicados via fertirrigação com ácido fosfórico. Verificou-se que as taxas de emissão de CO2 aumentaram significativamente após as fertirrigações, porém não houve efeito da dose do ácido fosfórico sobre as taxas. A umidade do solo mostrou-se um fator importante na relação entre as variações das taxas de emissão e a temperatura do solo ao longo do período estudado.

O presente trabalho teve como objetivo avaliar a qualidade pós-colheita de jabuticabas submetidas a diferentes temperaturas de armazenamento refrigerado (AR). Após a colheita, os frutos fisiologicamente maduros foram acondicionados em bandejas de poliestireno expandido (EPS), revestidas por filme plástico de polietileno de baixa densidade (PEBD) e armazenados sob refrigeração a 0; 3; 6; 9 e 12 ± 1ºC e U.R. 87 ± 2%, sendo avaliados a cada 5 dias. Os frutos foram avaliados quanto à perda de massa, atividade respiratória, pH, acidez titulável, sólidos solúveis, ácido ascórbico, textura, pectina total e solúvel, atividade da enzima polifenoloxidase (PFO), compostos fenólicos e atividade antioxidante. Para frutos refrigerados a 9 e 12ºC, o pico respiratório atrasou em relação aos demais tratamentos, além de apresentarem as menores taxas respiratórias. O teor de sólidos solúveis aumentou com o tempo de armazenamento para todas as temperaturas, contudo, em 9 e 12ºC, esse aumento foi em menor proporção. A firmeza e o teor de ácido ascórbico também foram superiores nos frutos armazenados a 9 e 12ºC, enquanto os contéudos de pectina solúvel foram menores. Observou-se a diminuição da atividade da enzima PFO ao longo dos 30 dias do AR, independentemente da temperatura utilizada; entretanto, os menores valores foram encontrados nos frutos mantidos a 9 e 12ºC. Os frutos armazenados a 12ºC apresentaram os maiores conteúdos de compostos fenólicos totais e a maior atividade antioxidante ao final do experimento. Nesse sentido, a temperatura de 12ºC foi a mais efetiva na manutenção da qualidade pós-colheita das jabuticabas.

O N é o nutriente que proporciona as maiores respostas no crescimento das gramas, e a adubação nitrogenada adequada pode proporcionar a formação do tapete com boa qualidade em menor tempo. Neste trabalho, objetivou-se avaliar a influência da adubação nitrogenada na produção e qualidade de tapetes de grama-bermuda. O experimento foi instalado e conduzido em área de produção comercial de grama, localizada na cidade de Capela do Alto, SP. A grama utilizada foi a Cynodon dactylon (Pers) L., conhecida como grama-bermuda. O delineamento experimental utilizado foi o de blocos casualizados, com cinco tratamentos e quatro repetições. Os tratamentos foram constituídos por cinco doses de N: 0, 150, 300, 450 e 600 kg ha-1. O aumento das doses de N aumentou a taxa de cobertura do solo pela gramabermuda, reduzindo o tempo para formação do tapete. O máximo acúmulo de matéria seca de rizomas + estolões + raízes foi proporcionado pela dose de 354 kg ha-1 de N, e a resistência dos tapetes, pela dose de 365 kg ha-1 de N. Doses de N entre 354 e 365 kg ha-1, aumentaram a resistência dos tapetes e, com isso, a capacidade deles serem manuseados após a colheita, podendo promover assim maior rendimento da área.

Avaliaram-se os efeitos da irradiação ultravioleta (254nm, UV-C) na conservação de pêssegos cultivar Douradão, aos 5; 10; 15 e 20 dias de armazenamento com o uso da refrigeração (0ºc e 75-85% de uR). os tratamentos foram: controle (0 minuto de exposição - 0 kj. m -2); T1 (5 minutos -1,85 kj. m -2); T2 (10 minutos - 3,71 kj. m -2); (15 minutos - 5,55 kj. m -2), e T4 (20 minutos - 7,52 kj. m -2). o delineamento experimental foi um esquema inteiramente casualizado, com 5 tempos de armazenamento: (0; 5; 10; 15 e 20 dias), com 3 repetições e 2 frutos por unidade experimental. os parâmetros avaliados foram: sólidos solúveis (SS); acidez titulável (AT); relação SS/AT Ratio; pH; firmeza da polpa, respiração e vida útil. Os pêssegos não apresentaram alteração do pH e da At durante o tempo de armazenamento.os melhores valores de SS e firmeza da polpa foram obtidos com o tratamento T4. A taxa respiratória foi menor em T4. As alterações de SS, firmeza da polpa e respiração apresentadas pelos frutos do tratamento T4 foram menores que aquelas dos demais tratamentos, demonstrando a eficácia do uso da irradiação ultravioleta.
