Weathering of mine tailings in Adak results in high As concentrations in surface and ground water, sediments, and soil. In spite of the oxic conditions, As-rich surface and ground, water samples indicate As(III) species predominantly (up to 83%). Several microorganisms were isolated from the enrichment cultures that were involved in As cycling. Amongst them was Arsenicicoccus bolidensis - a novel gram-positive, facultatively anaerobic, coccus-shaped actinomycete, which actively reduced As(V) to As(III) in aqueous media. A. bolidensis reduced 0.06-0.20 mM day(-1) As(V). As(V) reduction displays a direct correlation between the initial As(V) concentration, growth rate, and biomass yield. (c) 2006 Elsevier B.V. The aim of this study was to evaluate the effects of inulin as fat replacer on short dough biscuits and their corresponding doughs. A control formulation, with no replacement, and four formulations in which 10, 20, 30, and 40 % of shortening was replaced by inulin were studied. In the dough, shortening was observed surrounding flour components. At higher fat replacement levels, flour was more available for hydration leading to significant (P<0.05) harder doughs: from 2.76 (0.12)N in 10 % fat-replaced biscuits to 5.81 (1.56)N in 30 % fat-replaced ones. Biscuit structure was more continuous than dough structure. A continuous fat layer coated the matrix surface, where starch granules were embedded. In general, weight loss during baking and water activity decreased significantly (P<0.05) as fat replacement increased. Biscuit dimensions and aeration decreased when fat replacement increased, e.g., width gain was +1.20 mm in 10 fat-replaced biscuits and only +0.32 mm in 40 % fat-replaced ones. Panelist found biscuits with 20 % of fat replacement slightly harder than control biscuits. It can be concluded that shortening may be partially replaced, up to 20 %, with inulin. These low fat biscuits are similar than the control biscuits, and they can have additional health benefits derived from inulin presence.

This study investigated the stability of freeze dried and fluid bed dried alginate microcapsules coated with chitosan containing model probiotic bacteria, Lactobacillus plantarum, during storage for up to 45 days at different water activities (0.11, 0.23, 0.40 and 0.70) and temperatures (4, 30 and 37 °C). The loss in cell viability was around 0.8 log in the case of fluid bed drying and around 1.3 in the case of freeze drying, with the former method resulting in dried capsules of smaller size (~ 1 mm vs 1.3 mm), more irregular shape, and with a rougher surface. In both cases, the water activity and water content were less than 0.25 and 10% w/w, respectively, which favours high storage stability. The storage stability studies demonstrated that as the water activity and temperature decreased the survival of the dried encapsulated cells increased. Considerably better survival was observed for fluid bed dried encapsulated cells compared to freeze dried encapsulated cells and freeze dried free cells with 10% sucrose (control), and in some cases, e.g. at 4 and 30 °C at water activities of 0.11, 0.23 and 0.40, there was more than 1 log difference after 45 days, with concentrations higher than 108 CFU/g after 45 days of storage. The results indicate that fluid bed drying is an effective and efficient manufacturing method to produce probiotic containing capsules with enhanced storage stability.

The aim of this study was to assess in vitro the influence of Er:YAG laser irradiation distance on the shear strength of the bond between an adhesive restorative system and primary dentin. A total of 60 crowns of primary molars were embedded in acrylic resin and mechanically ground to expose a flat dentin surface and were randomly assigned to six groups (n = 10). The control group was etched with 37% phosphoric acid. The remaining five groups were irradiated (80 mJ, 2 Hz) at different irradiation distances (11, 12, 16, 17 and 20 mm), followed by acid etching. An adhesive agent (Single Bond) was applied to the bonding sites, and resin cylinders (Filtek Z250) were prepared. The shear bond strength tests were performed in a universal testing machine (0.5 mm/min). Data were submitted to statistical analysis using one-way ANOVA and the Kruskal-Wallis test (p < 0.05). The mean shear bond strengths were: 7.32 +/- 3.83, 5.07 +/- 2.62, 6.49 +/- 1.64, 7.71 +/- 0.66, 7.33 +/- 0.02, and 9.65 +/- 2.41 MPa in the control group and the groups irradiated at 11, 12, 16, 17, and 20 mm, respectively. The differences between the bond strengths in groups II and IV and between the bond strengths in groups II and VI were statistically significant (p < 0.05). Increasing the laser irradiation distance resulted in increasing shear strength of the bond to primary dentin.

The proliferation of mesangial cells (MC) in the presence of glutamine (0-20 mM) was determined in both low (5 mM) and high (25 mM) glucose-containing medium. Glutamine in a high glucose (HG) environment increased cell proliferation in a dose-dependent manner. Inhibition of glutamine:fructose 6-phosphate amidotransferase (GFAT) and of phosphodiesterase significantly reduced glutamine-induced proliferation. Supraphysiologic levels of glutamine increase MC proliferation in a HG milieu via GFAT and cAMP-dependent pathways, suggesting that glutamine could pose a risk for diabetic nephropathy.

Islet neogenesis associated protein (INGAP) increases islet mass and insulin secretion in neonatal and adult rat islets. lit the Present Study, we measured the short- and long-term effects of INGAP-PP (a pentadecapeptide having the 104-118 amino acid sequence of INGAP) upon islet protein expression and phosphorylation of components of the PI3K, MAPK and cholinergic pathways, and on insulin secretion. Short-term exposure of neonatal islets to INGAP-PP (90 s, 5, 15, and 30 min) significantly increased Akt1(-Ser473) and MAPK3/1(-Thr202/Tyr204) phosphorylation and INGAP-PP also acutely increased insulin secretion from islets perifused with 2 and 20 mM glucose. Islets cultured for 4 days in the presence of INGAP-PP showed an increased expression of Akt1, Frap1, and Mapk1 mRNAs as well as of the muscarinic M3 receptor subtype, and phospholipase C (PLC)-beta 2 proteins. These islets also showed increased Akt1 and MAPK3/1 protein phosphorylation. Brief exposure of INGAP-P-treated islets to carbachol (Cch) significantly increased P70S6K(-Thr389) and MAPK3/1 phosphorylation and these islets released more insulin when challenged with Cch that was prevented by the M3 receptor antagonist 4-DAMP in a concentration-dependent manner. In conclusion, these data indicate that short- and long-term exposure to INGAP-PP significantly affects the expression and the phosphorylation of proteins involved in islet PI3K and MAPK signaling pathways. The observations of INGAPP-PP-stimulated up-regulation of cholinergic M3 receptors and PLC-beta 2 proteins, enhanced P70S6K and MAIIK3/1 phosphorylation and Cch-induced insulin secretion suggest a participation of the cholinergic pathway in INGAP-PP-mediated effects.

At low ionic strength dimyristoylphosphatidylglycerol (DMPG) exhibits a broad phase transition region characterized by several superimposed calorimetric peaks. Peculiar properties, such as sample transparency, are observed only in the transition region. In this work we use differential scanning calorimetry (DSC), turbidity. and optical microscopy to study the narrowing of the transition region with the increase of ionic strength (0-500 mM NaCl). Upon addition of salt, the temperature extension of the transition region is reduced, and the number of calorimetric peaks decreases until a single cooperative event at T(m) = 23 degrees C is observed in the presence of 500 mM NaCl. The transition region is always coupled with a decrease in turbidity, but a transparent region is detected within the melting process only in the presence of up to 20 mM NaCl. The vanishing of the transparent region is associated with one of the calorimetric peaks. Optical microscopy of giant vesicles shows that bilayers first rupture when the transition region is reached and Subsequently lose optical contrast. Fluorescence microscopy reveals a blurry and undefined image in the transparent region, suggesting a different lipid self-assembly. Overall sample turbidity can be directly related to the bilayer optical contrast. Our observations are discussed in terms of the bilayer being perforated along the transition region. In the narrower temperature interval of the transparent region, dependent on the ionic strength, the perforation is extensive and the bilayer completely loses the optical contrast.

Electroactive nanostructured membranes have been produced by the layer-by-layer (LbL) technique, and used to make electrochemical enzyme biosensors for glucose by modification with cobalt hexacyanoferrate redox mediator and immobilisation of glucose oxidase enzyme. Indium tin oxide (ITO) glass electrodes were modified with up to three bilayers of polyamidoamine (PAMAM) dendrimers containing gold nanoparticles and poly(vinylsulfonate) (PVS). The gold nanoparticles were covered with cobalt hexacyanoferrate that functioned as a redox mediator, allowing the modified electrode to be used to detect H(2)O(2), the product of the oxidase enzymatic reaction, at 0.0 V vs. SCE. Enzyme was then immobilised by cross-linking with glutaraldehyde. Several parameters for optimisation of the glucose biosensor were investigated, including the number of deposited bilayers, the enzyme immobilisation protocol and the concentrations of immobilised enzyme and of the protein that was crosslinked with PAMAM. The latter was used to provide glucose oxidase with a friendly environment, in order to preserve its bioactivity. The optimised biosensor, with three bilayers, has high sensitivity and operational stability, with a detection limit of 6.1 mu M and an apparent Michaelis-Menten constant of 0.20 mM. CaNb(2)O(6) single crystal fibers were grown by the laser-heated pedestal growth technique, directly from the starting reagents. Optically transparent fibers were obtained in the form of rods with elliptical cross-section, free from cracks, impurities, and secondary phases, with an average diameter of 0.4 mm and about 20 mm of length. The fibers grew within the orthorhombic Pbcn columbite structure, with the growth axis nearly parallel to the crystallographic a-direction. The parameters b and c were parallel to the shorter and larger ellipsis axes. A special setup using a microscope was developed to obtain the far-infrared reflectivity spectra of these micrometer-sized fibers, allowing the identification and assignment of 34 of the 38 polar phonons foreseen for the material. From these phonons, the intrinsic dielectric constant ( of 185 THz) could be estimated, showing the potential of the material for applications in microwave circuitry. These results, along with previous polarized Raman data (Cryst. Growth Des. 2010, 10, 1569), allow us to present a comprehensive set of optical phonon modes and to discuss the potential use of designed CaNb(2)O(6) microcrystals in compact optical devices.

Electrochemical impedance spectroscopy (EIS) in pH 6.9 phosphate buffer solution was used to investigate each step of the procedure employed to modify a screen-printed electrode (SPE). The SPE was modified with self-assembled monolayers (SAMs) of cystamine (CYS, deposited from 20 mM solution), followed by glutaraldehyde (GA, 0.3 M solution). The Trypanosoma cruzi antigen was immobilized using different deposition times. The influence of incubation time (2-18 h) of protein was also investigated. The topography of modified electrode with this protein was investigated by atomic force microscopy (AFM). Interpretation of impedance data was based on physical and chemical adsorption, and degradation of the layer at high and meddle frequencies, and charge transfer reaction involving mainly the reduction of oxygen at low frequencies. EIS studies on modified electrodes with Tc85 protein immobilized for different incubation times indicated that the optimum incubation time was 6-8 h. It was demonstrated that EIS is a good technique to evaluate the different steps and the integrity of the surface modifications, and to optimize the incubation time of protein in the development of biosensors. A acidemia glutárica tipo I (AG I) é um erro inato do metabolismo de herança autossômica recessiva que se caracteriza bioquimicamente pela deficiência da atividade da enzima glutaril-CoA desidrogenase da rota de degradação dos aminoácidos lisina, hidroxilisina e triptofânio. O bloqueio da rota na conversão de glutaril-CoA à crotonil-CoA e resultante da deficiência enzimática leva ao acúmulo tecidual e aumento da concentração nos líquidos biológicos predominantemente dos ácidos glutárico e 3-hidroxiglutárico e, em alguns casos, do ácido glutacônico. Os pacientes afetados apresentam sintomas neurológicos especialmente após crises encefalopáticas, comuns no primeiro ano de vida, em que ocorre necrose bilateral dos gânglios da base, degeneração do globo pálido e da substância branca. Tendo em vista que a patogênese do dano cerebral na AG I é pouco conhecida e que as lesões do SNC dos pacientes muito se assemelham às lesões devido à excitotoxicidade, no presente estudo investigamos a ação dos ácidos glutárico e 3-hidroxiglutárico sobre alguns parâmetros neuroquímicos envolvendo o sistema glutamatérgico em córtex cerebral e astrócitos cultivados de córtex cerebral de ratos. Alguns desses parâmetros já haviam sido testados previamente com o ácido glutárico. Observamos que o ácido 3-hidroxiglutárico, em concentrações de 10 µM a 1 mM, não alterou a captação de L-[3H]glutamato por preparações sinaptossomais. Verificamos também que os ácidos glutárico e 3- hidroxiglutárico, nas mesmas concentrações, não alteraram a liberação do L- [3H]glutamato por estas estruturas em condições basais ou estimuladas (despolarizadas) por potássio (20 mM e 40 mM). Já nos ensaios de união do L- [3H] glutamato a membranas plasmáticas, o ácido 3-hidroxiglutárico inibiu a união do neurotransmissor em meio de incubação sem sódio e cloreto (sítios receptores), quando em baixas concentrações (1 µM a 100 µM), enquanto não alterou esta união em altas concentrações (500 µM a 2 mM). Na presença de alto sódio (sítios transportadores), ambos os ácidos inibiram a união de L- [3H]glutamato em todas as concentrações testadas (10 µM a 1 mM). Outros experimentos demonstraram que o ácido 3-hidroxiglutárico não alterou a captação vesicular de L-[3H]glutamato. Avaliamos também a influência dos ácidos glutárico e 3-hidroxiglutárico sobre a captação de L-[3H]glutamato por astrócitos cultivados de córtex cerebral. Inicialmente verificamos a viabilidade destas células na presença dos ácidos pelo teste do MTT que reflete a atividade das desidrogenases mitocondriais e também pela liberação de lactato desidrogenase para o meio de incubação. Na concentração de 1 mM , os ácidos glutárico e 3- hidroxiglutárico não se mostraram citotóxicos para estas células. Por outro lado, quando incubados por 60 minutos, nas concentrações de 50 µM e 1mM, aumentaram significativamente a captação de L-[3H]glutamato pelos astrócitos.

A adsorção em leito fixo de carvão ativado granular é uma das mais importantes alternativas para a remoção de compostos orgânicos voláteis em efluentes aquosos, em concentrações baixas. O objetivo deste trabalho é colocar em funcionamento uma unidade piloto de adsorção e desenvolver uma metodologia para avaliar o comportamento de adsorventes nesta planta como a usada no projeto de unidades de adsorção de leito fixo. Esta planta piloto possui uma coluna de 20 mm de diâmetro interno, encamisada. Ela é equipada com espectrofotômetro UV on-line, que permite a medida da concentração de componentes orgânicos voláteis na corrente de efluente. Também, possui módulos para a aquisição de dados de temperatura, vazão e pressão diferencial. Nessa planta, foram realizados experimentos, à temperatura ambiente, para aquisição de curvas de ruptura experimentais da adsorção de tolueno de uma corrente aquosa, em um leito fixo de carvão ativado. O leito era composto por um carvão ativado granular (8 x 12 mesh). Diferentes alturas de leito (5 e 10 cm) e vazões ( 0,8 e 1,6 L.h-1) foram testados, mantendo a concentração de tolueno na alimentação em aproximadamente 50 ppm. Concomitantemente, foram realizados testes para a caracterização do adsorvente, obtendo-se 15,4 % de conteúdo de umidade e 8,7 % de cinzas, massa específica bulk de 0,5 g.mL-1, massa específica real de 1,72 g.mL-1 e 681 m2.g-1 de área superficial A estimação da difusividade mássica e do coeficiente de transferência de massa referente à fase sólida foram obtidos através de dados de testes cinéticos, obtendo-se, respectivamente, os valores médios de 6,1 cm2.h-1 e 0,0162 cm.h-1. As alturas das zonas de transferência de massa obtidas para a vazão de 0,8 L.h-1 aumentaram com o aumento da altura do leito, indicando comportamento de padrões proporcionais. Além disso, analisaram-se dados preliminares de pressão diferencial no leito, que indicam que ela aumentou enquanto havia adsorção. A metodologia desenvolvida poderá ser aplicada para estudar a adsorção com outros pares adsorvente/adsorvato.

A resistência aos antibióticos dos patógenos mais comuns do trato respiratório está aumentando mundialmente. Recentemente, Streptococcus pneumoniae resistente à penicilina tem sido isolado em diversos países, e a freqüência dessas cepas tem elevado de modo alarmante. O aumento da resistência, com conseqüentes implicações terapêuticas, tem levado a uma reavaliação do uso dos antibióticos ß-lactâmicos para o tratamento de infecções pneumocócicas. No presente trabalho, um total de 107 amostras de Streptococcus pneumoniae, obtidas de materiais provenientes de pacientes adultos ambulatoriais e hospitalizados, em dois centros médicos de duas cidades do Rio Grande do Sul (Porto Alegre e Caxias do Sul), os quais apresentavam quadro clínico-radiológico de infecção pulmonar, foram analisadas com o objetivo de estudar-se a resistência do germe à penicilina. As amostras constituídas de escarro (80,4%), lavado brônquico (13,5%) e aspirado traqueal (6,6%) foram coletadas no período compreendido entre Julho de 1998 e Julho de 1999. O material foi semeado em meio de Agar sangue e as colônias suspeitas de Streptococcus pneumoniae foram transferidas para meio de Mueller-Hinton para teste de optoquina e de sensibilidade à penicilina com discos de oxacilina. Um halo de inibição da oxacilina menor do que 20 mm indicava a realização de teste para determinação da concentração inibitória mínima (MIC) com E-test. Um total de nove cepas foi identificado como tendo resistência intermediária à penicilina (MIC 0,1-1,0μg/ml) e nenhuma cepa resistente (resistência elevada: MIC > 2,0 μg/ml) foi identificada. Uma monitorização local das cepas quanto à resistência antimicrobiana é de grande importância para os clínicos no manejo de infecções pneumocócicas.

Nanoemulsões catiônicas têm sido recentemente consideradas como potenciais sistemas de liberação de oligonucleotídeos. O objetivo do presente trabalho foi avaliar a influência da adição de quantidades crescentes dos lipídeos catiônicos estearilamina (EA), oleilamina (OA) ou DOTAP (DT) sobre propriedades físico-químicas de nanoemulsões como sistemas de liberação de um oligonucleotídeo modelo (pdT16). As formulações foram preparadas pelo procedimento de emulsificação espontânea. As nanoemulsões constituídas de triglicerídeos de cadeia média, lecitina de gema de ovo, glicerol, quantidades crescentes dos lipídeos catiônicos (até 20 mM) e água foram caracterizadas em termos de diâmetro médio, pH, potencial zeta e viscosidade. Baseado nos resultados obtidos, as formulações contendo EA, OA e DT na concentração de 2 mM foram selecionadas para a continuidade do trabalho, uma vez que apresentam diâmetro de gotícula e potencial zeta de cerca de 250 nm e 40-50 mV, respectivamente. A taxa de associação do pdT16 às nanoemulsões foi determinada, indiretamente, pelo doseamento deste, na fase aquosa externa após separação em membranas de ultrafiltração/centrifugação, por meio de espectrofotometria no UV. A maior taxa de associação do pdT16 foi detectada para a formulação contendo o lipídeo catiônico DT (até ~70 mg/g de fase interna). Evidências adicionais da associação do pdT16 com as nanoemulsões foram detectadas pelo aumento do diâmetro de gotícula, inversão do potencial zeta e morfologia das gotículas. Em uma última etapa, um estudo preliminar da liberação do pdT16 foi realizado especialmente para a formulação contendo o lipídeo catiônico DT. O fator de diluição e relação de cargas [+/-] foram identificados como os principais parâmetros que influenciam a liberação do pdT16 a partir das nanoemulsões. Em conclusão, o conjunto dos resultados obtidos demonstra a influência da concentração e da natureza do lipídeo catiônico empregado sobre as propriedades físico-químicas de nanoemulsões catiônicas como sistemas de liberação de oligonucleotídeos.

The Cruzeta impoundment, situated in the city of Cruzeta, hinterland of the state of the Rio Grande do Norte state has significant importance to the municipality as it represents the only source of supplying water to the region. It was hypothesized that the regional consequence of the global warming and the warming of surface water could substantially contribute the significant growth of the aquatic macrophytes in the years 2008-2009. The growth of these vegetation believed to be improved the degree of water transparency and as a consequence of the improved growth of phytoplankton species and chlorophyll biomass. At the same time the aquatic macrophyte could interact and compete potentially for the dissolved inorganic nutrients resources and the phytoplankton community. This study presents a comparison of years 2004-2005 when it did not have the expressive presence of the aquatic macrophyte community or restricted to the littoral region. In contrast, the years 2008-2009 showed a significant growth of the aquatic macrophyte in the Cruzeta/RN impoundments. The present study is an attempt to elucidate the significant presence of the aquatic macrophyte, Eichhornia crassipes, Ceratophyllum submersum, Nymphea sp and Pistia sp, and its interference on the ecology of phytoplankton. The samplings had been carried out from September of 2008 to April of 2009 and consistently between 10:00 h and 12:00 h with the aid of Van Dorn bottle and the plankton net of mesh size 20 Qm. The collections were made in three depths ie., surface, mid-column and bottom. The Physical-Chemical parameters such as pH, temperature, electrical conductivity and dissolved oxygen had been analyzed in situ. The samples for analysis of nutrients and chlorophyll were kept under refrigeration for posterior analysis in the laboratory. Phytoplankton samples were preserved in Lugol-iodine and kept for sedimentation for quali-quantitative analysis of phytoplankton. Enumeration of cells, colonies and filaments was done with the aid of Sedgwick-Rafter counting chamber and expressed as numbers/ml. Chlorophyll a was analyzed as a functional component of phytoplankton biomass and extracted with cold 90% acetone. The results indicate that the chlorophyll concentration varied between 5,65-8,08 Qg.L-1 for the dry period and 5,09-6,23 Qg.L-1 for the rainy period and showed considerable reduction when compared to the values to the 2004-2005 study period. The temperature was always presented higher in relation to the 2004-2005 study. Phytoplankton species showed a relative abundance of the Cyanophyceae for both the period of dry and rainy. The predominance species are filamentous Leptolymbya geophila Borzi (Planctolyngbya sp), Anabaena plankctônica Brunnthaler, Oscillatória limosa Ag. and Cylindrospermopsis raciborskii (Wolosz). The concentration of the nutrients such as nitrate and orto-phosfato had always presented higher values during the rainy period and the ammoniacal nitrogen retained moderate values in the dry period and a slight increase in rainy season. The main conclusions are the reduction of the concentration of chlorophyll, diversity of phytoplankton, and the increase in temperature and transparency of the water during the period of the study
