La saprolégniose est une maladie fongique causée par le champignon aquatique Saprolegnia sp. qui affecte les poissons sauvages et ceux provenant des piscicultures. L’apparition de touffes cotonneuses semblables à de la ouate de couleur blanche à grise est souvent la première indication de l’infection. Ce saprophyte ubiquitaire se nourrit habituellement des œufs de poissons morts, mais peut se propager rapidement aux œufs sains causant la mort de ces derniers. La saprolégniose est souvent une infection secondaire, mais des souches virulentes peuvent facilement se développer sur les salmonidés ayant subi un stress ou une mauvaise manipulation. De grandes pertes économiques associées à la saprolégniose sont rapportées chaque année à travers le monde surtout dans l’industrie de la pisciculture. Jusqu’en 2002, le contrôle de la saprolégniose pouvait se faire par l’utilisation du vert de malachite, un colorant organique ayant une grande activité antifongique. Malheureusement, cette molécule a été bannie à cause de ses propriétés cancérigènes. Aucun composé aussi efficace n’est actuellement disponible pour traiter les infections de la saprolégniose. Des molécules ou extraits naturels ayant un potentiel antifongique ont donc été testés à l’aide de deux techniques (par graines de chanvre et par cylindre d’agar). Les molécules d’un extrait de propolis (cire de ruches d’abeilles) démontrant de l’activité anti-Saprolegnia ont été identifiées. De plus, une bactérie, Pseudomonas aeruginosa, pouvant être retrouvée dans le même environnement que Saprolegnia sp. a démontré un effet antagoniste au champignon. Une molécule de signalisation intercellulaire produite par P. aeruginosa, 4-hydroxy-2-heptylquinoline (HHQ), a été identifiée comme responsable de l’effet antagoniste contre Saprolegnia sp.

Biodegradable films based on cassava starch and with addition of natural antimicrobial ingredients were prepared using the casting technique. The tensile properties tensile strength (TS) [MPa] and percent elongation (E) at break [%] and the water vapor transmission (WVT) of the biodegradable films were evaluated and compared with the control (without antimicrobial ingredients). The evaluation of the Colony Forming Units per gram [CFU/g] of pan bread slices packed with the best biodegradable films, in terms of packaging performance, was also determined. The addition onto the matrix of only clove and cinnamon powders could reduce the films WVT when compared to the control, however TS and E were lower than the control and the effect of cinnamon was milder regarding this property. Since water activity of the pan bread slices packed with the biodegradable films increased considerably during the storage period, the antimicrobial effect could not be clearly determined. Em quatro experimentos foram avaliados como agentes antibacterianos os produtos própolis em solução alcoólica e álcool etílico, adicionados às rações artificialmente contaminadas com os respectivos sorotipos: Salmonella typhimurium Nalr - Specr, (resistentes ao ácido Nalidíxico e a Spectinomicina) nos três primeiros experimentos e Salmonella agona Nalr - Specr, Salmonella infantis Nalr - Specr e Salmonella enteritidis Nalr - Specr no quarto experimento. As rações foram fornecidas a grupos de 10-16 pintos de corte de um dia. em todos os experimentos os produtos testados foram adicionados na base de 2% da ração. Quando se utilizou solução hidroalcóolica de própolis (exp. 1), seguidas 120 horas após o desafio, detectou-se a presença da bactéria nos cecos. No segundo experimento, testou-se a solução de própolis e seu diluente, o álcool etílico; seguidas 96 horas após o desafio, não foi observada a presença da bactéria nos cecos (< 2,0 log10). Avaliou-se, no terceiro experimento, a ação da solução de própolis e do álcool etílico no tempo, adicionados na ração 14 dias e 28 dias antes do fornecimento às aves. Após 72 horas do desafio, a leitura nas placas acusou a presença da bactéria nos cecos. Dentro deste último período, também se avaliou a ação da própolis em pó (extrato seco) e esse mesmo extrato em uma solução aquosa, adicionados à ração 48 horas antes do fornecimento às aves sendo que os resultados confirmaram a presença da bactéria nos cecos. No quarto experimento avaliou-se somente o álcool etílico nas rações artificialmente contaminadas com os sorotipos S. agona, S. enteritidis e S. infantis, registrando-se contagem zero (<2,0 log10) apenas com o último sorotipo. Os resultados obtidos permitem concluir que o tratamento com a solução de própolis apresentou ação sobre a S. typhimurium somente quando em solução alcóolica, dentro de um período de 48 horas, indicando que o efeito bactericida se deveu ao álcool etílico presente na solução. A ação do tratamento com o álcool etílico sobre os demais sorotipos demonstrou resultado parcial sendo observado efeito bactericida nos sorotipos S. typhimurium e S. enteritidis artificialmente inoculados na ração.

Currently, a wide range of research involving natural products is focused on the discovery of new drugs in many different therapeutic areas. A great number of the synthetic compounds on the market were derived from natural products, especially plants. Nemorosone is the major constituent of the floral resin of Clusia rosea Jacq., Clusiaceae, and in Cuban propolis. In vitro studies have shown cytotoxic activity in this substance against various tumor cell lines, including those resistant to various cytotoxic drugs, whereas it has low cytotoxicity to non-tumoral cells. Therefore, in order to characterize the biological activity of nemorosone, a substance with potential antitumor activity, and in view of preclinical testing of the toxicity of drug candidate compounds, the main aim of this study was to determine the mutagenic and antimutagenic activity of nemorosone by the Ames test, using the strains TA97a, TA98, TA100 and TA102 of Salmonella typhimurium. Secondly, to characterize the estrogenic activity in an experimental recombinant yeast model (Recombinant Yeast Assay) mutagenic activity was observed at in any of the concentrations in any of the test strains. To evaluate the antimutagenic potential, direct and indirect mutagenic agents were used: 4 nitro-o-phenylenediamine (NPD), mitomycin C (MMC) and aflatoxin B1 (AFL). Nemorosone showed moderate antimutagenic activity (inhibition level 31%), in strain TA100 in the presence of AFL, and strong antimutagenic activity in TA102 against MMC (inhibition level 53%). Estrogenic activity was observed, with an EEq of 0.41±0.16 nM at various tested concentrations.

Avaliou-se a atividade antiinflamatória do extrato etanólico de própolis - EEP, . sobre o edema desencadeado por carragenina, dextrana e histamina. O EEP apresentou dose eficaz (DE50) de 650 mg/kg (v.o), inibindo significativamente o processo inflamatório desencadeado pela carragenina, mas não inibiu o produzido por dextrana. O EEP antagonizou ainda o efeito edematogênico produzido por histamina. Nas úlceras produzidas por estresse, o EPP inibiu de forma significativa a geração dos diversos tipos classificados. em todos os parâmetros analisados no estudo da toxicidade em fase de tratamento subcrônico , (hematológicos, bioquímicos e histopatológicos), o grupo tratado com o EEP não apresentou diferença significativa em relação ao grupo controle. Desta forma, sugere-se que na dose de 650 mg/kg (dose eficaz) não existe a presença de efeitos tóxicos que possam comprometer a utilização deste extrato.

Since it is not always possible to reduce human exposure to mutagens, attempts have been directed to identify potential antimutagens and anticarcinogens for use in protecting the population against environmental disease. The purpose of this paper is to provide the reader with information about the antimutagenic and anticarcinogenic potentials of some dietary constituents and foods widely consumed in Brazil, and to reinforce diet as a key factor in determining genomic stability and preventing human diseases. In this report, we have summarized data that show interactive effects between some dietary components and specific chemical mutagens or carcinogens using in vitro and in vivo short- or medium-term assays. The summary indicates that certain dietary compounds may be useful agents for disease prevention. (C) 2003 Elsevier B.V. All rights reserved.

The aim of this work was to evaluate the quality of Hass avocado submitted to application of propolis and vegetable wax. The selected fruits were submitted to the treatments (1) without wax and without propolis, (2) with vegetable wax, (3) propolis alcoholic extract 100%, (4) wax and etanol 70%, (5) propolis alcoholic extract 2% and wax, (6) propolis alcoholic extract 4% and wax, (7) propolis alcoholic extract 6% and wax, (8) propolis alcoholic extract 8% and wax, and (9) propolis alcoholic extract 10% and wax. The fruits were maintained under refrigeration (10 +/- 1 degrees C and 90 +/- 5% relative humidity), being appraised every three days for 15 days and more 3 days of commercialization simulation at room temperature (23 +/- 1 degrees C and 18% relative humidity). The evaluation parameters were the weight loss, respiratory activity, pH, firmness, tritable acidity (AT), soluble solids (SS), and ratio. The results of weigth loss, respiratory activity and firmness were the most significant to show the difference among the treatments. 'Hass' avocado fruits submitted the application of propolis associated the vegetable wax present smaller weight loss, CO2 production and firmness in relation to the control fruits. The fruits of the treatment with propolis alcoholic extract 2% and wax presented the most promising results.

OBJECTIVE: The aim of this study was to evaluate histomorphometrically the effect of alveolex (Propolis 10%) on the repair of bone cavities in the calvaria of rats. MATERIALS and METHODS: A 5 mm diameter bone defect was made in the calvaria of male Wistar rats using the drill-type trephine. The defects were filled with rhBMP-21Alveolex, rhBMP-2, Alveolex, or coagulum. Twenty-eight animals with seven subjects on each were sacrificed 30 days after surgery and samples were fixed and embedded in paraffin. Histological sections stained by HE (hematoxylin and eosin) were obtained from the calvaria bone defect and analyzed by a differential point-counting method. RESULTS: Group I and II, rhBMP-21Alveolex and rhBMP-2, respectively, presented higher levels of newly formed bone than other groups (P < 0.001). There were not significant differences between groups I and II (P > 0.05). In addition, there was not significant difference between groups III and IV, Control-Coagulum and Alveolex, respectively (P > 0.05). CONCLUSION: Alveolex has increased the bone repair in calvaria defects of rats when associated to rhBMP-2, however without significant differences for rhBMP-2 isolated group; Alveolex isolated group showed the lowest levels of newly formed bone with no significant differences to coagulum group (control). The aim of this in vitro study was to determine the maximum inhibitory dilution (MID) of four cetylpyridinium chloride (CPC)based mouthwashes: CPC+Propolis, CPC+Malva, CPC+Eucaliptol+Jua+Roma+Propolis (Natural Honey (R)) and CPC (Cepacol (R)), against 28 Staphylococcus aureus field strains, using the agar dilution method. Decimal dilutions ranging from 1/10 to 1/655,360 were prepared and added to Mueller Hinton Agar. Strains were inoculated using Steers multipoint inoculator. The inocula were seeded onto the surface of the culture medium in Petri dishes containing different dilutions of the mouthwashes. The dishes were incubated at 37 degrees C for 24 h. For readings, the MID was considered as the maximum dilution of mouthwash still capable of inhibiting microbial growth. The obtained data showed that CPC+Propolis had antimicrobial activity against 27 strains at 1/320 dilution and against all 28 strains at 1/160 dilution, CPC+Malva inhibited the growth of all 28 strains at 1/320 dilution, CPC+Eucaliptol+Jua+Roma+Propolis inhibited the growth of 2 strains at 1/640 dilution and all 28 strains at 1/320 dilution, and Cepacol (R) showed antimicrobial activity against 3 strains at 1/320 dilution and against all 28 strains at 1/160 dilution. Data were submitted to Kruskal-Wallis test, showing that the MID of Cepacol (R) was lower than that determined for the other products (p<0.05). In conclusion, CPC-mouthwashes showed antimicrobial activity against S. aureus and the addition of other substances to CPC improved its antimicrobial effect.

Baccharis dracunculifolia D.C. (Asteraceae), a shrub which grows wild in Brazil, is the main botanical source of Brazilian green propolis. Since Brazilian propolis shows an immunomodulatory activity, the goal of this work was to evaluate the action of B. dracunculifolia extracts and some of its isolated compounds on reactive oxygen intermediate (H2O2) production by macrophages obtained from male BALB/c mice. The results showed that the leaf (Bd-L) (25, 50, and 100 mu g mL(-1)), leaf rinse (Bd-LR) (25 mu g mL(-1)), and the root (Bd-R) (25 mu g mL(-1)) extracts enhanced H2O2 release by macrophages. A phytochemical study of the root and leaves of B. dracunculifolia was carried out. The chromatographic fractionation of Bd-R, using several techniques, afforded the isolation of baccharis oxide (1), friedelanol (2), viscidone (11), 11-hydroxy-10,11-dihydro-euparin (12), and 6-hydroxy-tremetona (13), while Bd-LR gave the following isolated compounds: baccharis oxide (1), friedelanol (2), isosakuranetin (3), aromadendrin-4'-methyl ether (4), dihydrocumaric acid (5), baccharin (6), hautriwaic acid lactone (7), hautriwaic acid acetate (8), drupanin (9), and cumaric acid (10). Among the isolated compounds, baccharis oxide (1) and friedelanol (2) increased H2O2 production at a concentration of 1001,M. This is the first time that the presence of compounds 7, 8, 12, and 13 in B. dracunculifolia has been reported. Based on these results it is suggested that the crude extracts and some isolated compounds from B. dracunculifolia display an immunomodulatory action.

As propriedades biológicas da própolis de Apis mellifera são amplamente relatadas sendo comuns variações nas mesmas em função da região onde foram produzidas. A ação antimicrobiana de própolis obtidas em três regiões do Brasil (Botucatu-SP, Mossoró-RN e Urubici-SC) foi investigada sobre linhagens isoladas de infecções clínicas humanas (Staphylococcus aureus, Escherichia coli, Enterococcus sp, Pseudomonas aeruginosa e Candida albicans). Foram preparados extratos alcoólicos de própolis (EAP) e determinada a Concentração Inibitória Mínima (CIM) seguida do cálculo da CIM90%. A própolis de Botucatu foi a mais eficiente sobre S. aureus (0,3%v/v), Enterococcus sp (1,1%v/v) e C. albicans (2,1% v/v). Para E. coli, a própolis eficiente foi de Urubici (7,0%v/v) e para P. aeruginosa a de Mossoró (5,3%v/v). Os resultados mostram maior sensibilidade das bactérias Gram positivas e levedura em relação às Gram negativas. É possível concluir que, para os microrganismos testados e amostras de própolis testadas, há diferenças na atividade antimicrobiana em função do local de produção e que isso se explica pela diferença de composição química da própolis.

Os autores propõem procedimentos para estabelecer o controle de qualidade da própolis. Para tanto, foram adaptadas e executadas técnicas farmacopéicas e/ou desenvolvidas técnicas novas. Foram estabelecidos perfis dos teores de flavonóides, de ceras, de substâncias voláteis totais (através da perda por dessecação) e de cinzas totais. Os valores obtidos para as amostras de própolis estudadas, coletadas em um apiário-escola, foram comparados com os de própolis adquiridas no mercado. Foi estabelecido, ainda, o perfil destes parâmetros para amostras de própolis coletadas nos diferentes locais da colméla, no periodo de três anos, com coletas de inverno e verão. Os teores de flavonóides variaram entre 2,05 - 5,52%.
