Molecular neurobiology has provided an explanation of mechanisms supporting mental functions as learning, memory, emotion and consciousness. However, an explanatory gap remains between two levels of description: molecular mechanisms determining cellular and tissue functions, and cognitive functions. In this paper we review molecular and cellular mechanisms that determine brain activity, and then hypothetize about their relation with cognition and consciousness. The brain is conceived of as a dynamic system that exchanges information with the whole body and the environment. Three explanatory hypotheses are presented, stating that: a) brain tissue function is coordinated by macromolecules controlling ion movements, b) structured (amplitude, frequency and phase-modulated) local field potentials generated by organized ionic movement embody cognitive information patterns, and c) conscious episodes are constructed by a large-scale mechanism that uses oscillatory synchrony to integrate local field patterns. © by São Paulo State University.

Neuropsychiatric syndromes are highly prevalent in Alzheimer's disease (AD), but their neurobiology is not completely understood. New methods in functional magnetic resonance imaging, such as intrinsic functional connectivity or resting-state analysis, may help to clarify this issue. Using such approaches, alterations in the default-mode and salience networks (SNs) have been described in Alzheimer's, although their relationship with specific symptoms remains unclear. We therefore carried out resting-state functional connectivity analysis with 20 patients with mild to moderate AD, and correlated their scores on neuropsychiatric inventory syndromes (apathy, hyperactivity, affective syndrome, and psychosis) with maps of connectivity in the default mode network and SN. In addition, we compared network connectivity in these patients with that in 17 healthy elderly control subjects. All analyses were controlled for gray matter density and other potential confounds. Alzheimer's patients showed increased functional connectivity within the SN compared with controls (right anterior cingulate cortex and left medial frontal gyrus), along with reduced functional connectivity in the default-mode network (bilateral precuneus). A correlation between increased connectivity in anterior cingulate cortex and right insula areas of the SN and hyperactivity syndrome (agitation, irritability, aberrant motor behavior, euphoria, and disinhibition) was found. These findings demonstrate an association between specific network changes in AD and particular neuropsychiatric symptom types. This underlines the potential clinical significance of resting state alterations in future diagnosis and therapy. © 2013 Wiley Periodicals, Inc.

The bed nucleus of the stria terminalis (BNST) is a limbic structure that has a direct influence on the autonomic, neuroendocrine, and behavioral responses to stress. It was recently reported that reversible inactivation of synaptic transmission within this structure causes antidepressant-like effects, indicating that activation of the BNST during stressful situations would facilitate the development of behavioral changes related to the neurobiology of depression. Moreover, noradrenergic neurotransmission is abundant in the BNST and has an important role in the regulation of emotional processes related to the stress response. Thus, this study aimed to test the hypothesis that activation of adrenoceptors within the BNST facilitates the development of behavioral consequences of stress. To investigate this hypothesis, male Wistar rats were stressed (forced swimming, 15 min) and 24 h later received intra-BNST injections of vehicle, WB4101, RX821002, CGP20712, or ICI118,551, which are selective α1, α2, β1, and β2 adrenoceptor antagonists, respectively, 10 min before a 5-min forced swimming test. It was observed that administration of WB4101 (10 and 15 nmol), CGP20712 (5 and 10 nmol), or ICI118,551 (5 nmol) into the BNST reduced the immobility time of rats subjected to forced swimming test, indicating an antidepressant-like effect. These findings suggest that activation of α1, β1, and β2 adrenoceptors in the BNST could be involved in the development of the behavioral consequences of stress. © 2013 Wolters Kluwer Health | Lippincott Williams & Wilkins.

The medullary raphé is an important component of the central respiratory network, playing a key role in CO2 central chemoreception. However, its participation in hypoxic ventilatory responses is less understood. In the present study, we assessed the role of nucleus raphé obscurus (ROb), and specifically 5-HT neurons confined in the ROb, on ventilatory and thermoregulatory responses to hypoxia. Chemical lesions of the ROb were performed using either ibotenic acid (non-specific lesion; control animals received PBS) or anti-SERT-SAP (5-HT specific lesion; control animals received IgG-SAP). Ventilation (VE; whole body plethysmograph) and body temperature (Tb; data loggers) were measured during normoxia (21% O2, N2 balance) and hypoxia exposure (7% O2, N2 balance, 1h) in conscious adult rats. Ibotenic acid or anti-SERT-SAP-induced lesions did not affect baseline values of VE and Tb. Similarly, both lesion procedures did not alter the ventilatory or thermoregulatory responses to hypoxia. Although evidence in the literature suggests a role of the rostral medullary raphé in hypoxic ventilatory responses, under the present experimental conditions our data indicate that caudal medullary raphé (ROb) and its 5-HT neurons neither participate in the tonic maintenance of breathing nor in the ventilatory and thermal responses to hypoxia. © 2013 Elsevier B.V.

O reconhecimento dos limites entre o envelhecimento normal e o patológico é essencial para a adoção de políticas de saúde baseadas em evidências para o grupo etário acima de 65 anos. Este estudo é parte de um esforço sistemático que o grupo de pesquisa do Laboratório de Investigações em Neurodegeneração e Infecção da Universidade Federal do Pará tem feito para fornecer informações translacionais sobre a neurobiologia do envelhecimento normal e alterada. A meta principal em longo prazo é permitir políticas públicas para o envelhecimento saudável na Região Amazônica. No presente trabalho investigamos os efeitos do envelhecimento sobre os desempenhos em testes neuropsicológicos selecionados para avaliar aprendizagem, memória e alterações de linguagem. 29 adultos jovens (29,9 ± 1,06 anos) e 31 idosos (74,1 ± 1,15 anos) saudáveis foram submetidos aos testes e os resultados de seus desempenhos foram comparados por testes paramétricos e estatística multivariada. Uma anamnese e uma variedade de testes cognitivos, incluindo Mini Exame do Estado Mental, tarefas visuo-espaciais de aprendizagem e de memória da bateria de testes neuropsicológicos automatizados - CANTAB e testes de linguagem incluindo fluência verbal semântica e fonológica, teste de nomeação de Boston reduzido, performance narrativa utilizando a figura “o roubo dos biscoitos” e alguns testes da Bateria Montreal de Comunicação - MAC. O programa BioEstat versão 5.0 e o pacote estatístico SPSS foram utilizados para a análise. O teste paramétrico t de Student ou o não paramétrico de Mann-Whitney foram aplicados para detectar diferenças significativas (fixadas em valores de p <0,05). Os grupos foram pareados por escolaridade e incluiu homens e mulheres. A análise das subcategorias que compõem o MEEM mostrou diferença significativa apenas na recuperação da memória de evocação de lista de palavras, em que o grupo de idosos apresentou pior desempenho em comparação com o grupo de jovens. Foram encontradas diferenças estatisticamente significativas entre os desempenhos de idosos e jovens adultos nos seguintes testes de linguagem: 1) Nomeação de Boston, 2) Testes de Narrativa, 3) Metáforas; 4) Prosódia Emocional e Linguística. Em comparação com os adultos jovens, indivíduos idosos apresentaram pior desempenho em 10 medidas diferentes nos testes de memória visuo-espacial e de aprendizagem do CANTAB. Distâncias Euclidianas e análise discriminante obtidas a partir do CANTAB e dos testes de linguagem demonstraram que os primeiros distinguem os grupos com maior resolução. Os efeitos do envelhecimento sobre o desempenho nos testes neuropsicológicos selecionados revelam que a Bateria CANTAB, empregada para testar a memória visuo-espacial, é mais sensível e discrimina melhor a formação de subgrupos tanto no grupo de adultos jovens quanto no grupo de idosos. Por essa razão sugerimos que a aplicação em larga escala de testes selecionados da bateria CANTAB, tanto em estudos transversais como em longitudinais, vai aumentar nossa capacidade de resolução na distinção dos limites entre o envelhecimento normal e o patológico.

A análise da morfologia celular é um aspecto crucial da neurobiologia, pois a relação entre forma e função pode definir os processos fisiológicos na saúde e na doença. Um dos principais métodos para avaliar a morfologia celular é por meio da fotooxidação de marcadores fluorescentes intracelulares, dentre os quais se tem o percloreto de 1,1’-dioctadecil-3,3,3’,3’-tetrametil-indocarbocianina (DiI), uma carbocianina lipofílica que pode marcar células vivas ou fixadas. O DiI foi escolhido para este trabalho devido, dentre outros fatores, à sua importante utilização para estudos de morfologia celular. Como modelo para avaliar a qualidade da fotooxidação do aparelho construído para essa finalidade, elegeu-se as células horizontais da retina humana com o intuito de prosseguir com estudo morfométrico posterior dessas células, tendo em vista a escassez de trabalhos com essa abordagem em retina humana. Assim, este estudo teve como objetivo avaliar a qualidade do método de fotooxidação do DiI através de LED e utilizando como modelo células horizontais da retina humana. O material foi obtido do Banco de Olhos do Hospital Ophir Loyola e, em sequência dissecado, marcado com cristais de DiI e fotooxidado com o aparelho de LED. As imagens que resultaram do novo método de iluminação para fotooxidação de traçador fluorescente apresentou elevada qualidade de detalhes da morfologia neuronal, similares aos resultados obtidos em reações de fotoconversão convencional com microscópio, o que permite concluir que o aparelho mantém a eficiência da fotooxidação por revelar detalhes finos da morfologia celular, inclusive com as vantagens de processar áreas maiores de tecido e considerável redução de custo por dispensar o emprego de microscópio para o processo.

A number of speech disorders including stuttering have been shown to have important genetic contributions, as indicated by high heritability estimates from twin and other studies. We studied the potential contribution to stuttering from variants in the FOXP2 gene, which have previously been associated with developmental verbal dyspraxia, and from variants in the CNTNAP2 gene, which have been associated with specific language impairment (SLI). DNA sequence analysis of these two genes in a group of 602 unrelated cases, all with familial persistent developmental stuttering, revealed no excess of potentially deleterious coding sequence variants in the cases compared to a matched group of 487 well characterized neurologically normal controls. This was compared to the distribution of variants in the GNPTAB, GNPTG, and NAGPA genes which have previously been associated with persistent stuttering. Using an expanded subject data set, we again found that NAGPA showed significantly different mutation frequencies in North Americans of European descent (p = 0.0091) and a significant difference existed in the mutation frequency of GNPTAB in Brazilians (p = 0.00050). No significant differences in mutation frequency in the FOXP2 and CNTNAP2 genes were observed between cases and controls. To examine the pattern of expression of these five genes in the human brain, real time quantitative reverse transcription PCR was performed on RNA purified from 27 different human brain regions. The expression patterns of FOXP2 and CNTNAP2 were generally different from those of GNPTAB, GNPTG and NAPGA in terms of relatively lower expression in the cerebellum. This study provides an improved estimate of the contribution of mutations in GNPTAB, GNPTG and NAGPA to persistent stuttering, and suggests that variants in FOXP2 and CNTNAP2 are not involved in the genesis of familial persistent stuttering. This, together with the different brain expression patterns of GNPTAB, GNPTG, and NAGPA compared to that of FOXP2 and CNTNAP2, suggests that the genetic neuropathological origins of stuttering differ from those of verbal dyspraxia and SLI. In the LTP (Long-Term Potentiation) model of learning and memory formation, elaborated in the context of molecular neurobiology, the opening of NMDA (N-Metyl-D-Aspartate) channels to entry of calcium ions into the post-synaptic neuron depends on two excitatory events: the repeated activation of the post-synaptic neuron by only one pre-synaptic neuron (Alternative 1), or its concomitant activation by two or more pre-synaptic neurons (Alternative 2). With the purpose of testing these alternatives, in the context of Cognitive Psychology, we presented to 73 university students a sequence of slides, with the duration of 6 seconds each, containing sentences (one for each slide) considered as being relevant or irrelevant for the subjects. Relevant sentences (R1) were presented only one time, while irrelevant ones were divided in three groups: the first one with sentences presented only one time (I1), the second with sentences presented three times (I3) and the third with sentences presented five times (I5). We conjectured that relevant sentences presented only one time would mobilize two or more brain excitatory pathways (corresponding to Alternative 2 above), while repeated irrelevant sentences would progressively activate the same sensory pathway. After the presentation of the sentences, the subjects answered a written questionnaire with questions about each presented sentence. The results indicate a prevalence of correct answers to R1 over I1, I3 and I5, suggesting that the relevance factor has greater weight than repetition in the induction of declarative memories
