Latin-American Society of Forensic Genetics (SLAGF) Interlaboratory Quality Control Exercise (2010-2011) included the analysis of three bloodstain samples in FTA Classic Card (three persons, biologically unrelated) and one theoretical exercise. There were 56 participating laboratories from 13 Latin-American countries that belong to society, were reported 70 STRs, including autosomal and sex chromosome markers with consensus in 53 STRs with a rate in reporting errors of 2.3%. Fifty-six laboratories reported results in theoretical exercise with mistakes in calculation of IP for each marker. It is necessary to hold meetings to discuss the results of this exercise to reach conclusions and recommendations on all aspects of DNA forensics analysis and paternity test, to improve results and quality in the results of each laboratory. © 2011 Elsevier B.V.

The use and inadequate exploitation of natural resources is restricting the occurrence of aroeira (Myracrodruon urundeuva F.F. & M.F. Allemão), which now is on the FAO list of endangered species. This exploitation causes a decrease in the genetic base of M. urundeuva populations, which makes it difficult to find genotypes with stability and adaptability to different growing conditions. This study aimed at estimating the genetic variation and productivity, stability and adaptability of progenies of a M. urundeuva natural population, from the Ecological Station of Paulo de Faria-SP, under different planting systems. DBH (diameter at breast height) was evaluated in four progeny tests of M. urundeuva: i) planted with Anandenanthera falcata and Guazuma ulmifolia (TP-AMA); ii) single (TP-ASO); iii) planted with annual crops (TP-SAF) and iv) planted with Corymbia citriodora (TP-EUCA), installed in Selvíria-MS. The experimental design consisted of complete randomized blocks with three replications and a variable number of plants per plot in each of the four planting systems. From the joint analysis of the planting systems studied, it was found that: i) there were variations among planting systems particularly in TP-SAF; ii) only in TP-EUCA it was possible to detect variations among the progenies; iii) the effects of the genotype x environment interaction were not significant. Thereby, the harmonic mean of genotypic values (MHVG), the relative performance of genotypic values from the mean of each site (PRVG) and the harmonic mean of the relative performance of genotypic values (MHPRVG) for DBH showed, respectively: progenies with greater stability, adaptability, and stability and simultaneous adaptability within different planting systems. The use of these selection criteria provided a more refined selection of the best progenies of M. urundeuva under the different planting systems studied.

A malária é uma doença parasitária causada por protozoários do gênero Plasmodium que completam seu ciclo de desenvolvimento alternando entre hospedeiros humanos e mosquitos do gênero Anopheles. No contexto mundial, constitui um grave problema de saúde pública que afeta principalmente os países em desenvolvimento de clima tropical e subtropical. No Brasil, acredita-se que 99,5% dos casos registrados de malária encontram-se na Amazônia Legal. Muito do sucesso deste agravo nesta região deve-se a fatores biológicos e ambientais que favorecem níveis altos de vetores, além de fatores sociais que comprometem os esforços para controlar a doença. Assim, o objetivo deste trabalho foi estudar o perfil epidemiológico da malária no Pará, durante uma série histórica (1999 – 2003), analisando a influência de variáveis ambientais e socioeconômicas sobre a prevalência dos casos. Para tal foram calculados os índices parasitários anuais (IPA) de cada município e, através de um SIG, estes dados foram georreferenciados e estudados de forma temporal e espacial. Dados sobre o desmatamento no Estado foram analisados, através de uma regressão por permutação, para tentar explicar a variação temporal da malária. Para o estudo espacial foi testada (regressão múltipla) a influência das variáveis: temperatura, pluviosidade, altitude, educação, longevidade e renda, sobre a prevalência da malária. No estudo temporal a malária apresentou uma tendência decrescente no Estado, entretanto, apenas 31 municípios apresentaram a mesma tendência, não houve tendência crescente, os 112 municípios restantes apresentaram tendência estável. Além disso, muitos municípios alternaram aumento e diminuição dos casos ao longo da série, indicando uma boa ação de controle, mas uma fraca atuação da vigilância. Neste contexto o desmatamento parece influenciar a série temporal da malária, obteve-se resultados significativos em dois (2001 e 2002) dos três anos estudados. No estudo espacial o modelo final adotado, apesar de uma baixo poder explicativo (R²=0.31), apresentou três variáveis significativas: número de meses secos, renda e educação. No entanto, o resultado das duas primeiras não se apresenta de uma forma direta, sendo reflexo de outras atividades. Apesar da escala adotada e de problemas na agregação dos dados (só estão disponíveis por município), este trabalho apresenta resultados relevantes que podem auxiliar os gestores da saúde (ou de endemias) a direcionar ações de controle para as áreas apontadas como críticas, atuando nos fatores de maior significância, obtendo assim melhor aproveitamento dos recursos humanos e materiais disponíveis.

The desert locust (Schistocerca gregaria) has been used as material for numerous cytogenetic studies. Its genome size is estimated to be 8.55 Gb of DNA comprised in 11 autosomes and the X chromosome. Its X0/XX sex chromosome determinism therefore results in females having 24 chromosomes whereas males have 23. Surprisingly, little is known about the DNA content of this locust's huge chromosomes. Here, we use the Feulgen Image Analysis Densitometry and C-banding techniques to respectively estimate the DNA quantity and heterochromatin content of each chromosome. We also identify three satellite DNAs using both restriction endonucleases and next-generation sequencing. We then use fluorescent in situ hybridization to determine the chromosomal location of these satellite DNAs as well as that of six tandem repeat DNA gene families. The combination of the results obtained in this work allows distinguishing between the different chromosomes not only by size, but also by the kind of repetitive DNAs that they contain. The recent publication of the draft genome of the migratory locust (Locusta migratoria), the largest animal genome hitherto sequenced, invites for sequencing even larger genomes. S. gregaria is a pest that causes high economic losses. It is thus among the primary candidates for genome sequencing. But this species genome is about 50 % larger than that of L. migratoria, and although next-generation sequencing currently allows sequencing large genomes, sequencing it would mean a greater challenge. The chromosome sizes and markers provided here should not only help planning the sequencing project and guide the assembly but would also facilitate assigning assembled linkage groups to actual chromosomes.

Haplotypes formed by polymorphisms (T-786C, rs2070744; a variable number of tandem repeats in intron 4, and Glu298Asp, rs1799983) of the eNOS gene were associated previously with gestational hypertension (GH) and preeclampsia (PE). However, no study has explored the Tag SNPs rs743506 and rs7830 in these disorders. The aim of the current study was to compare the distribution of the genotypes and haplotypes formed by the five eNOS polymorphisms mentioned among healthy pregnant (HP, n = 122), GH (n = 138), and PE (n = 157). The haplotype formed by "C b G G C" was more frequent in HP compared to GH and PE (p = 0.0071), which is supported by previous findings that demonstrated the association of the combination "C b G" with a higher level of nitrite (NO marker). Our results suggest a protective effect of the haplotype "C b G G C" against the development of hypertensive disorders of pregnancy.
