Adding Zn improves crop growth, increases seed yield and also positively affects nutritional quality. After Zn fertilization, there is normally a period of several years in which residual effects provide an adequate supply of Zn to successive crops. Immediately after the application of Zn sources water-soluble Zn slowly but continually decreases. Various factors, including time and moisture conditions, affect the aging process and modify the solubility of the metal in soil and therefore its availability. In previous experiments, we studied the residual effect of synthetic chelates, obtained that the amounts of potentially available Zn decreased in the second cropping year due to aging processes. The present study was undertaken to verify variations in the residual effects of applying four different synthetic Zn sources

ABSTRACT This thesis aims to demonstrate that the underground cellars represent a model of efficient construction and environmentally sustainable for aging and storing wines. During the production, aging and storing wine, temperature and relative humidity are crucial to the final product quality. The appearance of defects in wine as a result of high temperatures during the aging process is something known since the last century, so that winemakers seek to achieve the parameters considered suitable for proper production and aging of wine using the most wineries to artificial air conditioning. The underground cellars are historical buildings in La Rioja and all the peoples of the Designation of Origin Rioja and the aim of this thesis is to demonstrate that naturally meet the conditions for aging wine due to its construction design and without the use of other energy to get it. Furthermore, the presence of these buildings in the Rioja Denomination of Origin represents the origin of the fame of its wines and is a cultural heritage today untapped. To demonstrate the importance of these areas of underground cellars, germ of the Designation of Origin Rioja, they have been inventoried all the districts of municipalities benefiting from this. This will have visited all municipalities and have photographed their cellars. In visits made during cataloging it has been observed that constitute today a forgotten heritage that has evolved to picnic or homes, sometimes even abandoned or demolished. The conservation of the buildings has been more careful in the subzone of Rioja Alta, where you can still find wine cellars where a particular way, something exceptional to find in La Rioja Baja is made. It has also been observed that the building materials used in the drafts La Rioja Alta usually carved stone or masonry, while in La Rioja Baja is most uses of river stone or masonry. This work is reflected in 2159 cataloging records located underground cellars. In order to demonstrate that they meet the conditions suitable for breeding they were installed sensors that recorded the temperature and relative humidity in the underground cellars and commercial wineries which differ both in the building design and the solutions adopted by sought to gather these parameters. The results obtained in the underground cellars show that these traditional buildings have fewer annual fluctuations of temperature and relative humidity of the aging rooms commercial wineries, which implies a more constructive solution suitable for the breeding and conservation of wine. To corroborate the results obtained on domestic conditions, has made a sensory analysis of wines stored and raised underground cellars compared with stored without being subjected to control these parameters, as can occur in domestic conditions, once marketed as winemaker loses control over them. The results show that the wines stored in private homes have differences with those stored in underground cellars. These results are consistent with data obtained from sensors placed in these underground cellars, where the maximum annual temperature is below 20 ° C, considered as maximum allowable threshold for raising by various authors.

ABSTRACT The preservation of bibliographic and documentary heritage is one of the biggest challenges that libraries and archives around the world have to face. The search for solutions to the problem of degraded paper has historically been focused from two predominants lines of work: the conservation of these documents by the neutralization of acids in them with alkaline agents, and their restoration by lining them with, basically, cellulose from vegetal sources. However, the possibility of strengthening the damaged cellulose has not been successfully explored, and the problem still persists. Until today, the development of biotechnology-based treatments in documentary heritage conservation has been scarce, although the ability of certain bacteria to produce cellulose takes to propose its use in the field of conservation and restoration of paper. The bacterial cellulose (BC) is chemically identical to the plant cellulose, but its macroscopic organization is different. Its unique properties (high degree of crystallinity, durability, strength and biocompatibility), makes it an excellent resource in different fields. The use of high-quality BC generated by Gluconacetobacter sucrofermentans CECT 7291 to restore damaged documents and to consolidate those that may be at risk of degradation, has been studied in this thesis, trying to prevent the document destruction, and to get reinforced papers with good mechanical, optical and structural properties. Protocols that allow the implementation of the BC as a reinforcing material were also developed. First of all, in order to select the culture medium that provides a cellulose suitable for its use in restoration, it has been evaluated the effect that the carbon and nitrogen sources from the culture medium have on the generated BC, keeping the temperature and the initial pH of the medium as fixed parameters, and performing the culture without shaking. The effect of the addition of 1% ethanol to the culture medium on BC properties was also evaluated. The cellulose layers were collected at four different times, characterizing in all of them the culture medium (pH and carbon source consumption), and the BC sheets (pH, dry weight and optical and mechanical properties). The best combination of carbon and nitrogen sources proved to be fructose plus yeast extract and corn steep liquor, with or without ethanol, which provided a good balance between the cellulose production and the consumption of carbon source, and generating BC sheets homogeneous and resistant. The addition of ethanol to the culture medium increased productivity but caused a noticeable decrement in pH. The BC layers generated with these optimized culture media, have been characterized in terms of tear and burst index, optical properties, scanning electron microscopy (SEM), X-ray diffraction, infrared Fourier transform spectroscopy (FTIR), polymerization degree, static and dynamic contact angles, and mercury intrusion porosimetry. Moreover it must be kept in mind that the restored materials should be stable over time. Therefore, the same characterization was performed after subjecting the layers of BC to an accelerated aging process. The results showed that the BC sheets obtained have a high crystallinity index, low internal porosity, good mechanical properties, and high stability over time. To develop working protocols to use this optimized BC in paper restoration, the first step was to select the samples to restore. Three types of model papers, made from mechanical pulp, chemical pulp and filter paper (before and after an accelerated aging process), and three old books purchased in the second hand market, were chosen. These specimens to be restored were also characterized in terms of its mechanical and physicochemical properties. The first protocol of restoration with BC to be evaluated is called linning. It consists on applying a reinforcing material to the document using an adhesive. The BC produced in the optimized culture medium with 1% ethanol was selected. An alkali purification method (1 hour at 90 °C in 1% NaOH) was applied, and wheat starch was selected as adhesive. The linning process was also carried out with Japanese paper (JP), a material commonly used in conservation, in order to compare both materials. It was concluded that there are no significant differences in the characteristics studied of the two types of reinforcing materials. The reinforced materials were characterized before and after undergoing to an accelerated aging. Papers lined with BC showed more marked differences in the optical properties that papers restored with JP. However, the text was more readable when BC was the reinforcing material. Wettability decreased with both types of reinforcement, although in the papers linned with BC it happened more marked and independently of the sample to restore. This is due to the closed structure of BC, which also leads to a decrement in air permeance. This study suggests that BC improves the deteriorated paper quality, without altering the information on it, and that this improvement is maintained over time. Therefore, the BC may be used as reinforcing material for linning, being more suitable than the JP to restore certain types of papers. The other restoration method to be evaluated was the in situ generation of BC over the paper to restore. For this purpose the culture medium without ethanol was selected, as the pH decrement caused by his presence would damage the document to restore. As purification method a heat treatment (24 hours at 65 °C) was chosen, less aggressive to the material to restore than the alkaline treatment. It was decided to apply the culture medium with the bacteria onto the material to restore with a brush. The reinforced material was characterized before and after an accelerated aging process. It was concluded that there was no substantial change in any characteristic, except for air permeance, which decreases very sharply after the generation of BC, getting a substantially air impermeable material. In general, it can be concluded that the ability of BC produced by Gluconacetobacter sucrofermentans CECT 7291 for its use as a reinforcing material in the restoration of paper documentary heritage, has been demonstrated. Also, two restoration methods, one ex situ and another in situ have been developed.

A permanent line of mouse embryo fibroblasts was treated with concentrations of the anticancer drug methotrexate (MTX) that left 20–50% surviving colonies. The surviving population initially multiplied at a much slower rate than controls after subculture in the absence of the drug, and required 9–12 days of serial subculture, with selective growth of the faster growing cells, to approximate the control rate. To determine the distribution of growth rates of cells in the original posttreatment populations, many single cells were isolated in multiwell plates immediately after the treatment period, and the resulting clones were serially subcultured. Most of the control clones underwent about 2 population doublings per day (PD/D). Almost all the survivors of MTX treatment multiplied at heterogeneously reduced rates, ranging from 0.6 PD/D to as high as control rates for a very few clones. They maintained the reduced rates through many subcultivations. The heritability of the reduced growth rates indicates that most cells that retain proliferative capacity after treatment with MTX carry random genetic damage that is perpetuated through many divisions of their progeny. Similar results have been described for cells that survive x-irradiation, and suggest random genetic damage is a common occurrence among cells in rapidly growing tissues that survive cytotoxic treatment. It also occurs in serial subcultures of cells that had been held under the constraint of confluence for extended periods, which suggests that the accumulation of random genetic damage to somatic cells during aging of mammals underlies the reduction of growth rate and function of the cells that characterizes the aging process.

Werner syndrome (WS) is a human progeroid syndrome characterized by the early onset of a large number of clinical features associated with the normal aging process. The complex molecular and cellular phenotypes of WS involve characteristic features of genomic instability and accelerated replicative senescence. The gene involved (WRN) was recently cloned, and its gene product (WRNp) was biochemically characterized as a helicase. Helicases play important roles in a variety of DNA transactions, including DNA replication, transcription, repair, and recombination. We have assessed the role of the WRN gene in transcription by analyzing the efficiency of basal transcription in WS lymphoblastoid cell lines that carry homozygous WRN mutations. Transcription was measured in permeabilized cells by [3H]UTP incorporation and in vitro by using a plasmid template containing the RNA polymerase II (RNA pol II)–dependent adenovirus major late promoter. With both of these approaches, we find that the transcription efficiency in different WS cell lines is reduced to 40–60% of the transcription in cells from normal individuals. This defect can be complemented by the addition of normal cell extracts to the chromatin of WS cells. Addition of purified wild-type WRNp but not mutated WRNp to the in vitro transcription assay markedly stimulates RNA pol II–dependent transcription carried out by nuclear extracts. A nonhelicase domain (a direct repeat of 27 amino acids) also appears to have a role in transcription enhancement, as revealed by a yeast hybrid–protein reporter assay. This is further supported by the lack of stimulation of transcription when mutant WRNp lacking this domain was added to the in vitro assay. We have thus used several approaches to show a role for WRNp in RNA pol II transcription, possibly as a transcriptional activator. A deficit in either global or regional transcription in WS cells may be a primary molecular defect responsible for the WS clinical phenotype.

The yeast Saccharomyces cerevisiae has a limited life-span, which is measured by the number of divisions that individual cells complete. Among the many changes that occur as yeasts age are alterations in chromatin-dependent transcriptional silencing. We have genetically manipulated histone deacetylases to modify chromatin, and we have examined the effect on yeast longevity. Deletion of the histone deacetylase gene RPD3 extended life-span. Its effects on chromatin functional state were evidenced by enhanced silencing at the three known heterochromatic regions of the genome, the silent mating type (HM), subtelomeric, and rDNA loci, which occurred even in the absence of SIR3. Similarly, the effect of the rpd3Δ on life-span did not depend on an intact Sir silencing complex. In fact, deletion of SIR3 itself had little effect on life-span, although it markedly accelerated the increase in cell generation time that is observed during yeast aging. Deletion of HDA1, another histone deacetylase gene, did not result in life-span extension, unless it was combined with deletion of SIR3. The hda1Δ sir3Δ resulted in an increase in silencing, but only at the rDNA locus. Deletion of RPD3 suppressed the loss of silencing in rDNA in a sir2 mutant; however, the silencing did not reach the level found in the rpd3Δ single mutant, and RPD3 deletion did not overcome the life-span shortening seen in the sir2 mutant. Deletion of both RPD3 and HDA1 caused a decrease in life-span, which resulted from a substantial increase in initial mortality of the population. The expression of both of these genes declines with age, providing one possible explanation for the increase in mortality during the life-span. Our results are consistent with the loss of rDNA silencing leading to aging in yeast. The functions of RPD3 and HDA1 do not overlap entirely. RPD3 exerts its effect on chromatin at additional sites in the genome, raising the possibility that events at loci other than rDNA play a role in the aging process.

A comparison was made of the competence for neoplastic transformation in three different sublines of NIH 3T3 cells and multiple clonal derivatives of each. Over 90% of the neoplastic foci produced by an uncloned transformed (t-SA′) subline on a confluent background of nontransformed cells were of the dense, multilayered type, but about half of the t-SA′ clones produced only light foci in assays without background. This asymmetry apparently arose from the failure of the light focus formers to register on a background of nontransformed cells. Comparison was made of the capacity for confluence-mediated transformation between uncloned parental cultures and their clonal derivatives by using two nontransformed sublines, one of which was highly sensitive and the other relatively refractory to confluence-mediated transformation. Transformation was more frequent in the clones than in the uncloned parental cultures for both sublines. This was dramatically so in the refractory subline, where the uncloned culture showed no overt sign of transformation in serially repeated assays but increasing numbers of its clones exhibited progressive transformation. The reason for the greater susceptibility of the pure clones is apparently the suppression of transformation among the diverse membership that makes up the uncloned parental culture. Progressive selection toward increasing degrees of transformation in confluent cultures plays a major role in the development of dense focus formers, but direct induction by the constraint of confluence may contribute by heritably damaging cells. In view of our finding of increased susceptibility to transformation in clonal versus uncloned populations, expansion of some clones at the expense of others during the aging process would contribute to the marked increase of cancer with age.

To determine whether pathogenic mutations in mtDNA are involved in phenotypic expression of Alzheimer’s disease (AD), the transfer of mtDNA from elderly patients with AD into mtDNA-less (ρ0) HeLa cells was carried out by fusion of platelets or synaptosomal fractions of autopsied brain tissues with ρ0 HeLa cells. The results showed that mtDNA in postmortem brain tissue survives for a long time without degradation and could be rescued in ρ0 HeLa cells. Next, the cybrid clones repopulated with exogenously imported mtDNA from patients with AD were used for examination of respiratory enzyme activity and transfer of mtDNA with the pathogenic mutations that induce mitochondrial dysfunction. The presence of the mutated mtDNA was restricted to brain tissues and their cybrid clones that formed with synaptosomes as mtDNA donors, whereas no cybrid clones that isolated with platelets as mtDNA donors had detectable mutated mtDNA. However, biochemical analyses showed that all cybrid clones with mtDNA imported from platelets or brain tissues of patients with AD restored mitochondrial respiration activity to almost the same levels as those of cybrid clones with mtDNA from age-matched normal controls, suggesting functional integrity of mtDNA in both platelets and brain tissues of elderly patients with AD. These observations warrant the reassessment of the conventional concept that the accumulation of pathogenic mutations in mtDNA throughout the aging process is responsible for the decrease of mitochondrial respiration capacity with age and with the development of age-associated neurodegenerative diseases.

To determine the importance of mitochondrial reactive oxygen species toxicity in aging and senescence, we analyzed changes in mitochondrial function with age in mice with partial or complete deficiencies in the mitochondrial antioxidant enzyme manganese superoxide dismutase (MnSOD). Liver mitochondria from homozygous mutant mice, with a complete deficiency in MnSOD, exhibited substantial respiration inhibition and marked sensitization of the mitochondrial permeability transition pore. Mitochondria from heterozygous mice, with a partial deficiency in MnSOD, showed evidence of increased proton leak, inhibition of respiration, and early and rapid accumulation of mitochondrial oxidative damage. Furthermore, chronic oxidative stress in the heterozygous mice resulted in an increased sensitization of the mitochondrial permeability transition pore and the premature induction of apoptosis, which presumably eliminates the cells with damaged mitochondria. Mice with normal MnSOD levels show the same age-related mitochondrial decline as the heterozygotes but occurring later in life. The premature decline in mitochondrial function in the heterozygote was associated with the compensatory up-regulation of oxidative phosphorylation enzyme activity. Thus mitochondrial reactive oxygen species production, oxidative stress, functional decline, and the initiation of apoptosis appear to be central components of the aging process.

A fundamental question in the basic biology of aging is whether there is a universal aging process. If indeed such a process exists, one would expect that it develops at a higher rate in short- versus long-lived species. We have quantitated pentosidine, a marker of glycoxidative stress in skin collagen from eight mammalian species as a function of age. A curvilinear increase was modeled for all species, and the rate of increase correlated inversely with maximum life-span. Dietary restriction, a potent intervention associated with increased life-span, markedly inhibited glycoxidation rate in the rodent. On the assumption that collagen turnover rate is primarily influenced by the crosslinking due to glycoxidation, these results suggest that there is a progressive age-related deterioration of the process that controls the collagen glycoxidation rate. Thus, the ability to withstand damage due to glycoxidation and the Maillard reaction may be under genetic control.

Prolonged incubation of NIH 3T3 cells under the growth constraint of confluence results in the death of some cells in a manner suggestive of apoptosis. Successive rounds of prolonged incubation at confluence of the surviving cells produce increasing neoplastic transformation in the form of increments in saturation density and transformed focus formation. Cells from the postconfluent cultures are given a recovery period of various lengths to remove the direct inhibitory effect of confluence before their growth properties are studied. It is found that with each round of confluence the exponential growth rate of the cells at low densities gets lower and the size of isolated colonies of the same cells shows a similar progressive reduction. The decreased growth rate of cells from the third round of confluence persists for > 60 generations of growth at low density. The proportion of colonies containing giant cells is much higher after a 2-day recovery from confluence than after a 7-day recovery. Retardation of growth at low density and increased saturation density appear to be two sides of the same coin: both occur in the entire population of cells and precede the formation of transformed foci. We propose that the slowdown in growth and the formation of giant cells result from heritable damage to the cells, which in turn drives their transformation. Similar results have been reported for the survivors of x-irradiation and of treatment with chemical carcinogens and are associated with the aging process in animals. We suggest that these changes result from free radical damage to membrane lipids with particular damage to lysosomes. Proteases and nucleases would then be released to progressively modify the growth behavior and genetic stability of the cells toward autonomous proliferation.
