Stoichiometry and thermodynamics of the interaction between the C-terminus of human 90 kDa heat shock protein Hsp90 and the mitochondrial translocase of outer membrane Tom70

A large majority of the 1000-1500 proteins in the mitochondria are encoded by the nuclear genome, and therefore, they are translated in the cytosol in the form and contain signals to enable the import of proteins into the organelle. The TOM complex is the major translocase of the outer membrane responsible for preprotein translocation. It consists of a general import pore complex and two membrane import receptors, Tom20 and Tom70. Tom70 contains a characteristic TPR domain, which is a docking site for the Hsp70 and Hsp90 chaperones. These chaperones are involved in protecting cytosolic preproteins from aggregation and then in delivering them to the TOM complex. Although highly significant, many aspects of the interaction between Tom70 and Hsp90 are still uncertain. Thus, we used biophysical tools to study the interaction between the C-terminal domain of Hsp90 (C-Hsp90), which contains the EEVD motif that binds to TPR domains, and the cytosolic fragment of Tom70. The results indicate a stoichiometry of binding of one monomer of Tom70 per dimer of C-Hsp90 with a K(D) of 360 30 nM, and the stoichiometry and thermodynamic parameters obtained suggested that Tom70 presents a different mechanism of interaction with Hsp90 when compared with other TPR proteins investigated. Fast Acquisition of (13)C NMR Spectra using the Steady-state Free Precession Sequence

In this paper, we show that the steady-state free precession sequence can be used to acquire (13)C high-resolution nuclear magnetic resonance spectra and applied to qualitative analysis. The analysis of brucine sample using this sequence with 60 degrees flip angle and time interval between pulses equal to 300 ms (acquisition time, 299.7 ms; recycle delay, 300 ms) resulted in spectrum with twofold enhancement in signal-to-noise ratio, when compared to standard (13)C sequence. This gain was better when a much shorter time interval between pulses (100 ms) was applied. The result obtained was more than fivefold enhancement in signal-to-noise ratio, equivalent to more than 20-fold reduction in total data recording time. However, this short time interval between pulses produces a spectrum with severe phase and truncation anomalies. We demonstrated that these anomalies can be minimized by applying an appropriate apodization function and plotting the spectrum in the magnitude mode.

Under många år har ”invandringsfrågan” varit en mycket debatterad fråga. Detta märks inte minst i alla de insändare som publicerats i Dalarnas morgontidningar. Integration och ekonomi är exempel på ämnen som nämns i dessa insändare då diskursen invandring diskuteras. En betydande faktor i debatten har bland annat varit Sverigedemokraternas valframgångar. Med denna undersökning vill vi försöka kartlägga hur diskursen invandring framställs i insändare från Dala-demokraten. Detta har gjorts med en innehålls- och diskursanalys. Analysen har utgått från insändare mellan tidsperioden augusti-april 2002/2003 och 2014/2015, vilket motsvarar tiden kring de båda riksdagsvalen.Analysen har genererat fyra teman (mångkultur och svenskhet, integrationsfrågan och invandring, demokrati och yttrandefrihet och ekonomi och välfärd) som diskursen invandring kretsar kring. Utifrån dessa teman har vår analys visat att Sverigedemokraternas valframgångar tenderar att delvis influera diskursen invandring som insändarskribenterna förmedlar. Diskursen invandring har gått från att kretsa kring integrationsproblematik 2002/2003 till ekonomisk problematik 2014/2015. Analysen har även visat på ett ”vi och dem”-tänk som genomsyrar diskursen invandring under bägge tidsperioder. Detta sker både genom fördomsfulla framställningar av andra människor och i uppdelningen av vilka som ”bör” få ta del av Sveriges gemensamma medel.

Qu est-ce que c est?: uma HQ autobiográfica de Fábio Moon e Gabriel Bá.

Partindo da ideia de que as histórias em quadrinhos, apesar de serem consideradas um gênero híbrido, possuem também uma linguagem autônoma, este trabalho busca investigar por meio de análise de conteúdo uma história em quadrinhos (HQ)autobiográfica dos gêmeos paulistas Fábio Moon e Gabriel Bá, Qu’est-ce que c’est?, publicada pela primeira vez em inglês na coletânea Autobiographix (Dark Horse Books, 2003) e republicada em português no livro Crítica (Devir, 2004). Serão analisados recursos estéticos e narrativos, tentando descobrir também quais são as particularidades de uma HQ autobiográfica. A história narra a saga dos irmãos durante uma viagem a Paris, onde foram visitar museus e colher referências para seu trabalho. No metrô, eles se depararam com uma gangue da periferia.

Influência do vírus da hepatite G (GBV-C) na resposta imune frente à infecção por Leishmania chagasi

GB virus type C (GBV-C) appears to promote a Th1 response and is associated with prolonged survival in HIV-infected people. L. chagasi causes a spectrum of illness that varies from severe visceral leishmaniasis, a disease that in the majority of cases is fatal if not treated, to self resolution of infection and development of positive DTH response that is protective against symptomatic disease. To determine if GBV-C viremia might influence the outcome of Leishmania infection, we characterized GBV-C status in a cohort of subjects residing in a L. chagasi endemic area in Brazil. GBV-C viremia was more prevalent in blood donors from urban than in periurban regions of Natal, Brazil (16% and 7.5% respectively). Evidence of prior GBV-C (anti-E2 antibodies) was detected in 24% and 12%of these groups respectively. Anti-E2 increased with age (p= 0.0121). No difference in GBV-C viremia was found in the DTH+ and VL groups (p= 0.269); however, subjects with visceral leishmaniasis were more likely to have anti-E2 than DTH+ subjects (p=0.0012), and DTH induration was smaller in subjects with E2 antibodies (4.5 mm) compared those without (7.12 mm) (p= 0.002). Furthermore, the size of the Leishmania DTH response was greater in GBV-C viremica subjects (6.8 mm) compared to non-viremic subjects (3.3 mm; p= 0.0054). There findings suggest that GBV-C virus may promote a type 1 immune response that could influence the outcome of Leishmania infection

Purificação e caracterização parcial de uma Fucana C de Dictyota mentrualis e estudo do seu efeito antiinflamatório e nociceptivo

In recent years, sulfated polysaccharides from marine algae have emerged as an important class of natural biopolymers with potential application in human and veterinary health care, while taking advantage of the absence of potential risk of contamination by animal viruses. Among these, fucans isolated from the cell walls of marine brown alga have been study due to their anticoagulant, antithrombotic, anti-inflammatory and antiviral activities. These biological effects of fucans have been found to depend on the degree of sulfation and molecular size of the polysaccharide chains. In the present study, we examined structural features of a fucan extracted from brown alga Dictyota menstrualis and its effect on the leukocyte migration to the peritoneum. The sulfated polysaccharides were extracted from the brown seaweed by proteolytic digestion, followed by sequential acetone precipitation producing 5 fractions. Gel lectrophoresis using 0.05 M 1,3-diaminopropane-acetate buffer, pH 9.0, stained with 0.1% toluidine blue, showed the presence of sulfated polysaccharides in all fractions. The chemical analyses demonstrated that all fractions are composed mainly of fucose, xylose, galactose, uronic acid, and sulfate. Electrophoresis in agarose gel in three different buffers demonstrated that the fraction 2.0v have only one population of fucan. This compound was purify by exclusion molecular. It has shown composition of fucose, xilose, sulfate and uronic acid in molar ration of 1.0: 1.7: 1.1: 0.5 respectively. The effect of this heterofucan on the leukocyte migration was observed 6h after zymozan (mg/g) administration into the peritoneum. The heterofucan showed higher antimigratory activity, it decrease the migration of leukocyte in 83.77% to peritoneum. The results suggest that this fucan is a new antimigratory compound with potential pharmacological appications
