A mancha de alternaria, causada por Alternaria alternata f. sp. citri, afeta tangelos Minneola, tangerinas Dancy, tangores Murcote e, menos freqüentemente, tangelos Orlando, tangerinas Novas, Lees e Sunburst. Esta doença causa desfolha grave, queda de frutos e manchas nas frutas. O objetivo deste trabalho foi estabelecer o melhor fungicida e a melhor dose para o controle da mancha marrom de alternaria. O delineamento experimental foi de parcelas subdivididas em blocos, com 10 tratamentos principais e 3 doses (subparcelas), com 5 repetições. Foram feitas 5 aplicações, com intervalo de 15 dias. Os tratamentos foram: azoxystrobin, pyraclostrobin, trifloxystrobin, trifloxystrobin + tebuconazole (2 aplicações) seguido de 3 aplicações de mancozeb, difenoconazole, trifloxystrobin + propiconazole, iprodione, trifloxystrobin + propineb (2 aplicações) seguido de 3 aplicações de oxicloreto de cobre, oxicloreto de cobre + óleo e testemunha. Simultaneamente foram feitas avaliações de incidência e número de lesões por folha. Ao surgimento dos frutos foram avaliadas a incidência em frutos e a produtividade em Kg/ha. Todos os tratamentos foram superiores à testemunha quanto a produtividade. Entre os produtos utilizados o tratamento com trifloxystrobin + propiconazole foi rentável comparando-se custo e produtividade.

A exploração da atividade biológica de compostos secundários presentes nas tinturas ou em óleos essenciais de plantas podem representar, ao lado da indução de resistência, mais uma forma potencial de controle de doenças em plantas cultivadas. O presente trabalho objetivou avaliar o potencial de tinturas de Lippia alba, Lippia sidoides, Mikania glomerata, Equisetum sp. e Hedera helix e óleos essenciais de Rosmarinus officinalis e Cinnamomum zeylanicum nas atividades in vitro, in vivo e na produção de proteínas na indução de resistência, em plantas de feijão vagem cultivar Bragança. Os resultados obtidos demonstraram que as tinturas de L. alba e L. sidoides e os óleos essenciais (R. officinalis e C. zeylanicum) apresentaram atividade in vitro aos isolados de Xanthomonas axonopodis pv. phaseoli. Todas as tinturas ensaiadas apresentaram menores valores do progresso da doença (AACPD), em relação à testemunha, merecendo destaque a tintura de L. alba, que estavam correlacionadas com os maiores teores de polifenoloxidase, peroxidase e proteínas solúveis totais, evidenciando uma possível indução de resistência. Os óleos essenciais não apresentaram diferença na AACPD e nem na indução de proteínas.

Conduziu-se simultaneamente dois experimentos, em casa de vegetação, com o objetivo de avaliar a resistência de híbridos de brócolis 'tipo cabeça única' (AF 649, Titleist, Centenário, Green Power, BR068, Magestic Crown, Marathon, Laguna, Legacy, Green Parasol, Packman e Mônaco) à podridão negra, causada por Xanthomonas campestris pv. campestris. Foram utilizados os métodos de inoculação no ápice das folhas por cortes com tesoura embebida na suspensão bacteriana e por ferimento provocado no caule com palito de dente umedecido na suspensão bacteriana. A inoculação foi realizada aos 25 dias após transplante (6 a 8 folhas definitivas). Avaliou-se no experimento de inoculação com tesoura, as áreas abaixo da curva de progresso da doença nas folhas inoculadas. No experimento de inoculação por palito de dente, avaliou-se a proporção de altura necrosada do caule, aos 26 dias após inoculação. Verificou-se que, em ambos os experimentos, o híbrido BRO68 apresentou-se mais suscetível à podridão negra e os híbridos Marathon, Legacy e Green Power foram os que apresentaram os maiores níveis de resistência à podridão negra.

Sintomas do cancro bacteriano da videira na variedade Red Globe foram observados em agosto de 2009 em pomar de Tupi Paulista, Estado de São Paulo, Brasil, e o agente causal Xanthomonas campestris pv. viticola foi identificado por meio de testes patológicos e moleculares. O procedimento de erradicação foi adotado e aproximadamente 4.700 plantas foram destruídas. Um levantamento realizado nas regiões produtoras do Estado de São Paulo não encontrou nenhum outro pomar contaminado, e essa espécie bacteriana é considerada ausente neste estado.

The genome of the bacterium Xylella fastidiosa contains four ORFs (XF2721, XF2725, XF2739 and XF0295) related to the restriction modification type I system, ordinarily named R-M. This system belongs to the DNA immigration control region (ICR). Each CIRF is related to different operon structures, which are homologues among themselves and with subunit Hsd R from the endonuclease coding genes. In addition, these ORFs are highly homologous to genes in Pseudomonas aeruginosa, Methylococcus capsulatus str. Bath, Legionella pneumophila, Helicobacter pylori, Xanthomonas oryzae pv. Oryzae and Silicibacter pomeroyi, as well as to genes from X. fastidiosa strains that infect grapevine, almond and oleander plants. This study was carried out on R-M ORFs from forty-three X. fastidiosa strains isolated from citrus, coffee, grapevine, periwinkle, almond and plum trees, in order to assess the genetic diversity of these loci through PCR-RFLP. PCR-RFLP analysis of the four ORFs related to the R-M system from these strains enabled the detection of haplotypes for these loci. When the haplotypes were defined, wide genetic diversity and a large range of similar strains originating from different hosts were observed. This analysis also provided information indicating differences in population genetic structures, which led to detection of different levels of gene transfer among the groups of strains. Assays were done under greenhouse conditions in order to evaluate the effect of pyraclostrobin (0.0375, 0.0750 and 0.150 mL.L-1) and acibenzolar-S-methyl (ASM) (0.025 g.L-1) in common bacterial blight on leaves of snap beans cultivar Braganca. These chemicals were sprayed at three different times: five days before; five days before + five days after; and five days after leaf inoculation with an isolate of Xanthomonas axonopodis pv. phaseoli. They were determinate the levels of polyphenoloxidase, peroxidase and total soluble proteins on inoculated and non-inoculated leaves of snap beans sprayed with pyraclostrobin (0.075 g.L-1) and ASM (0.025 g.L-1). All concentration of pyraclostrobin and ASM reduced the area under the disease progress curve (AUDPC) on leaves of snap beans, and the least AUDPC value was observed when this products were sprayed five days before + five days after inoculation. Higher levels of polyphenoloxidase, peroxidase and the total soluble proteins were observed on leaves sprayed with pyraclostrobin or ASM.

Com o propósito de avaliar o comportamento de seis cultivares de couve-flor de verão, quanto à produção e resistência à podridão negra (Xanthomonas campestris pv. campestris), instalou-se um experimento em campo, em Ipameri-GO, com o delineamento experimental em blocos completos casualizados com seis tratamentos e quatro repetições. Cada parcela experimental, de 10m², constou de 20 plantas distribuídas no espaçamento de 1,0m entre linhas e 0,5m entre plantas. Foram avaliadas as cultivares: híbridos 'Cindy', 'Sarah AF-1169', 'Sharon', 'Verona', 'Lisvera' e a variedade Piracicaba precoce. Os híbridos Verona e Sharon mostraram-se promissores para cultivo nas condições edafoclimáticas em que foram avaliados, com produtividades de 34,17 e 30,64t ha-1, respectivamente, e com moderados níveis de resistência à podridão negra. A variedade Piracicaba precoce apresentou produção inferior às dos melhores híbridos avaliados e moderada resistência à podridão negra. O híbrido Sarah não é recomendado para plantio na região de Ipameri-GO por apresentar baixa produção e baixa resistência à podridão negra.

Estudou-se o efeito das pulverizações de azocyclotin por turboatomizadores Jacto Arbus 2000/Export e Arbus 2000/850, sob diferentes condições operacionais, no controle do ácaro purpúreo, Panonychus citri (McGregor), e da leprose, Brevipalpus phoenicis (Geijskes); e de Arbus 2000/Export contra o ácaro da falsa ferrugem, Phyllocoptruta oleivora (Ashmead). Foram instalados três experimentos em pomares de citros (var. Valência) no período de 1994/95. O acaricida azocyclotin na dose 25 g i.a. / 100 litros d'água foi eficiente por 52 dias no controle de B. phoenicis e por 28 dias contra P. oleivora. Variações nas condições operacionais dos turboatomizadores não influenciaram no controle dos ácaros pragas. O produto azocyclotin pulverizado a 75% da dosagem comercial com 35 bicos do tipo JA-2 (Jacto), a 3,6 km/h e na pressão de 2070 kPa proporcionaram níveis de controle equivalentes aos obtidos com 100% da dosagem comercialmente recomendada desse acaricida, até 16 dias, quando aplicado com o turboatomizador Jacto Arbus 2000/Export no controle de P. citri.

The huanglongbing (HLB) disease of citrus trees, caused by Candidatus Liberibacter asiaticus and Ca. Liberibacter americanus, was first reported in Brazil in March, 2004. The presence of the disease has caused serious concerns among growers. Pruning experiments were conducted to determine if removal of symptomatic branches or the entire canopy (decapitation) would eliminate infected tissues and save HLB-affected trees. Pruning was done in five blocks on a total of 592 3- to 16 year-old 'Valencia', 'Hamlin' or 'Pera' sweet orange trees showing no symptoms or with two levels of symptom severity. Ten decapitated trees per block were caged and all trees were treated with insecticides to control the psyllid vector, Diaphorina citri. Mottled leaves reappeared on most symptomatic (69.2%) as well on some asymptomatic (7.6%) pruned trees, regardless of age, variety, and pruning procedure. Presence of the pathogen (Ca. Liberibacter americanus) in all symptomatic trees was confirmed by PCR. In general, the greater the symptom severity before pruning the lower the percentage of trees that remained asymptomatic after pruning.

In greenhouse trials, copper hydroxide, pyraclostrobin, and famoxadone were applied to actively crowing young citrus seedlings to determine the duration of protection of young leaves provided by these fungicides against melanose, caused by Diaporthe citri, citrus scab, caused by Elsinoe fawcettii, and Alternaria brown spot, caused by Alternaria alternata. Fungicides were applied to different sets of potted plants of grapefruit for control of melanose, of rough lemon for control of scab, and of Dancy tangerine for control of Afternaria brown spot 1 to 6 days prior to inoculation. as well as on the day of inoculation. Leaf area of treated shoots was estimated on the day of fungicide application and the day of inoculation and disease severity evaluated subsequently. In most cases. copper hydroxide and famoxadone provided at least 50% control of all three diseases for only about 2 days after application. Generally, there was little or no disease control when the products were applied 4 or more days before inoculation. In contrast, pyraclostrobin usually provided a high level of control of all three diseases when applied up to 5 days prior to inoculation. The level of disease control decreased as the interval between a fungicide application and inoculation increased and the relationship between disease control and leaf expansion best fit a quadratic equation. Effective disease control was observed with copper hydroxide and famoxadone until leaf area had increased by 100 to 200%, whereas control with pyraclostrobin was observed up to 400 to 500% increase in leaf area. In postinoculation tests with scab and melanose, pyraclostrobin provided high levels of disease control (>75%) when applied up to 2 days after inoculation. whereas copper hydroxide and famoxadone had minimal postinoculation activity. Applications of pyraclostrobin to the spring flush growth of citrus trees are much more likely to provide control of melanose, scab, and Alternaria brown spot than those of famoxadone or copper hydroxide.

Symptoms of huanglongbing (HLB) were reported in São Paulo State (SPS), Brazil, in March 2004. In Asia, HLB is caused by 'Candidatus Liberibacterasiaticus'and in Africa by 'Candidatus Liberibacter africanus'. Detection of the liberibacters is based on PCR amplification of their 16S rRNA gene with specific primers. Leaves with blotchy mottle symptoms characteristic of HLB were sampled in several farms of SIPS and tested for the presence of liberibacters. 'Ca. L. asiaticus' was detected in a small number of samples but most samples gave negative PCR results. Therefore, a new HLB pathogen was suspected. Evidence for an SPS-HLB bacterium in symptomatic leaves was obtained by PCR amplification with universal primers for prokaryotic 16S rRNA gene sequences. The amplified 16S rRNA gene was cloned and sequenced. Sequence analysis and phylogeny studies showed that the 16S rRNA gene possessed the oligonucleotide signatures and the secondary loop structure characteristic of the alpha-Proteobacteria, including the liberibacters. The 16S rRNA gene sequence phylogenetic tree showed that the SPS-HLB bacterium clustered within the a-Proteobacteria, the liberibacters being its closest relatives. For these reasons, the SPS-HLB bacterium is considered a member of the genus 'Ca. Liberibacter'. However, while the 16S rRNA gene sequences of 'Ca. L. asiaticus' and 'Ca. L. africanus' had 98-4% similarity, the 16S rRNA gene sequence of the SPS-HLB liberibacter had only 96(.)0% similarity with the 16S rRNA gene sequences of 'Ca. L. asiaticus'or'Ca. L. africanus'. This lower similarity was reflected in the phylogenetic tree, where the SPS-HLB liberibacter did not cluster within the 'Ca. L asiaticus'/'Ca. L. africanus group', but as a separate branch. Within the genus 'Candidatus Liberibacter' and for a given species, the 16S/23S intergenic region does not vary greatly. The intergenic regions of three strains of 'Ca. L. asiaticus', from India, the People's Republic of China and Japan, were found to have identical or almost identical sequences. In contrast, the intergenic regions of the SPS-HLB liberibacter, 'Ca. L. asiaticus' and 'Ca. L. africanus' had quite different sequences, with similarity between 66(.)0 and 79(.)5%. These results confirm that the SPS-HLB liberibacter is a novel species for which the name 'Candidatus Liberibacter americanus' is proposed. Like the African and the Asian liberibacters, the 'American' liberibacter is restricted to the sieve tubes of the citrus host. The liberibacter could also be detected by PCR amplification of the 16S rRNA gene in Diaphorina citri, the psyllid vector of 'Ca. L. asiaticus', suggesting that this psyllid is also a vector of 'Ca. L. americanus' in SPS. 'Ca. L. americanus' was detected in 216 of 218 symptomatic leaf samples from 47 farms in 35 municipalities, while 'Ca. L. asiaticus' was detected in only 4 of the 218 samples, indicating that 'Ca. L. americanus' is the major cause of HLB in SIPS.

A novel L-amino acid oxidase (LAO) (Casca LAO) from Crotalus durissus cascavella venom was purified to a high degree of molecular homogeneity using a combination of molecular exclusion and ion-exchange chromatography system. The purified monomer of LAO presented a molecular mass of 68 kDa and pI estimated in 5.43, which were determined by two-dimensional electrophoresis. The 71st N-terminal amino acid sequence of the LAO from Crotalus durissus cascavella presented a high amino acid sequence similarities with other LAOs from Colloselasma rhosostoma, Crotalus adamanteus, Agkistrodon h. blomhoffi, Agkistrodon h. halys and Trimeresurus stejnegeri. LAO displayed a Michaelis-Menten behavior with a kilometer of 46.7 mu M and an optimum pH for enzymatic activity of 6.5. Casca LAO induced a dose-dependent platelet aggregation, which was abolished by catalase and inhibited by indomethacin and aspirin. These results suggest that the production of H2O2 is involved in subsequent activation of inflammatory enzymes, such as thromboxane. Casca LAO also inhibited the bacterial Growth of Gram-negative (Xanthomonas axonopodis pv passiflorae) and Gram-positive (S. mutans) strains. Electron microscopy assessments of both bacterial strains suggest that the hydrogen peroxide produced by LAO induce bacterial membrane rupture and consequently loss of cytoplasmatic content. This LAO exhibited a high antileishmanic activity against the promastigote of Leishmania amazonensis in vitro, its activity was dependent on the production of hydrogen peroxide, and the 50% inhibitory concentration was estimated in 2.39 mu g/ml. The objective of this research was to evaluate de reaction of leaves and pods of five cultivars of bean (Phaseolus vulgaris L.) plants to twenty strains of Xanthomonas campestris pv. phaseoli. The strains were inoculated onto leaves in a greenhouse, and onto pods in a growth chamber. The results obtained were analyzed and the strains classified into three groups: low, medium, and high virulence. Most of the strains showed high virulence on leaves of Carioca and Rio Negro cultivars, as opposed to only low to medium virulence on leaves of IAPAR 14, IAPAR 16, and G. N. Nebraska # 1 sel. 27 cultivars. There were, however, individual strains powerful enough to overcome the leaf resistance of IAPAR 14, IAPAR 16, and G. N. Nebraska # 1 sel. 27. With regard to pods, most strains showed high virulence on all bean cultivars, with exception of IAPAR 14 where virulence was at medium level. A correlation between leaf and pod symptoms was found to exist in Carioca, Rio Negro, and IAPAR 14 cultivars. No such correlation was observed in IAPAR 16 and G. N. Nebraska # 1 sel. 27. Comparing strains producing melanine in vitro with those not producing this pigment, no difference was observed with regard to virulence.
