Amphiphysin is a protein enriched at mammalian synapses thought to function as a clathrin accessory factor in synaptic vesicle endocytosis. Here we examine the involvement of amphiphysin in synaptic vesicle recycling at the giant synapse in the lamprey. We show that amphiphysin resides in the synaptic vesicle cluster at rest and relocates to sites of endocytosis during synaptic activity. It accumulates at coated pits where its SH3 domain, but not its central clathrin/AP-2-binding (CLAP) region, is accessible for antibody binding. Microinjection of antibodies specifically directed against the CLAP region inhibited recycling of synaptic vesicles and caused accumulation of clathrin-coated intermediates with distorted morphology, including flat patches of coated presynaptic membrane. Our data provide evidence for an activity-dependent redistribution of amphiphysin in intact nerve terminals and show that amphiphysin is a component of presynaptic clathrin-coated intermediates formed during synaptic vesicle recycling.

During migratory journeys, birds may become displaced from their normal migratory route. Experimental evidence has shown that adult birds can correct for such displacements and return to their goal. However, the nature of the cues used by migratory birds to perform long distance navigation is still debated. In this experiment we subjected adult lesser black-backed gulls migrating from their Finnish/Russian breeding grounds (from >60°N) to Africa (to < 5°N) to sensory manipulation, to determine the sensory systems required for navigation. We translocated birds westward (1080 km) or eastward (885 km) to simulate natural navigational challenges. When translocated westwards and outside their migratory corridor birds with olfactory nerve section kept a clear directional preference (southerly) but were unable to compensate for the displacement, while intact birds and gulls with the ophthalmic branch of the trigeminal nerve sectioned oriented towards their population-specific migratory corridor. Thus, air-borne olfactory information seems to be important for migrating gulls to navigate successfully in some circumstances.

Illegal, Unreported and Unregulated fishing has had a major role in the overexploitation of global fish populations. In response, international regulations have been imposed and many fisheries have been 'eco-certified' by consumer organizations, but methods for independent control of catch certificates and eco-labels are urgently needed. Here we show that, by using gene-associated single nucleotide polymorphisms, individual marine fish can be assigned back to population of origin with unprecedented high levels of precision. By applying high differentiation single nucleotide polymorphism assays, in four commercial marine fish, on a pan-European scale, we find 93-100% of individuals could be correctly assigned to origin in policy-driven case studies. We show how case-targeted single nucleotide polymorphism assays can be created and forensically validated, using a centrally maintained and publicly available database. Our results demonstrate how application of gene-associated markers will likely revolutionize origin assignment and become highly valuable tools for fighting illegal fishing and mislabelling worldwide.

Purpose: To investigate the clinical implications of a variable relative biological effectiveness (RBE) on proton dose fractionation. Using acute exposures, the current clinical adoption of a generic, constant cell killing RBE has been shown to underestimate the effect of the sharp increase in linear energy transfer (LET) in the distal regions of the spread-out Bragg peak (SOBP). However, experimental data for the impact of dose fractionation in such scenarios are still limited.

Methods and Materials: Human fibroblasts (AG01522) at 4 key depth positions on a clinical SOBP of maximum energy 219.65 MeV were subjected to various fractionation regimens with an interfraction period of 24 hours at Proton Therapy Center in Prague, Czech Republic. Cell killing RBE variations were measured using standard clonogenic assays and were further validated using Monte Carlo simulations and parameterized using a linear quadratic formalism.

Results: Significant variations in the cell killing RBE for fractionated exposures along the proton dose profile were observed. RBE increased sharply toward the distal position, corresponding to a reduction in cell sparing effectiveness of fractionated proton exposures at higher LET. The effect was more pronounced at smaller doses per fraction. Experimental survival fractions were adequately predicted using a linear quadratic formalism assuming full repair between fractions. Data were also used to validate a parameterized variable RBE model based on linear α parameter response with LET that showed considerable deviations from clinically predicted isoeffective fractionation regimens.

Conclusions: The RBE-weighted absorbed dose calculated using the clinically adopted generic RBE of 1.1 significantly underestimates the biological effective dose from variable RBE, particularly in fractionation regimens with low doses per fraction. Coupled with an increase in effective range in fractionated exposures, our study provides an RBE dataset that can be used by the modeling community for the optimization of fractionated proton therapy.

Concetualmente ancorado em referenciais teóricos que demonstram o papel fundamental da narrativa e a ubiquidade dos produtos dos esquemas narrativos nos vários domínios da vida humana, bem como a importância do desenvolvimento de competências narrativas adequadas enquanto processo determinante no sentido da construção da literacia, este trabalho procurou compreender os processos de estruturação e a estrutura de histórias contadas por crianças do 1º Ciclo do Ensino Básico (CEB), numa abordagem morfológica suportada pela teoria e modelo de Vladimir Propp, no sentido da concetualização e prototipagem de uma aplicação multimédia de suporte ao conto de histórias. Os estudos empíricos conduzidos, de natureza comparativo-exploratória, permitiram que as crianças assumissem o papel de contadoras de histórias e informantes, fornecendo-nos o corpus e conteúdo para análises morfológicas e de conteúdo, sobretudo qualitativas e com objetivos fundamentalmente descritivos. Os resultados do estudo suportaram o delineamento de linhas orientadoras para o desenvolvimento do modelo concetual e requisitos funcionais de uma aplicação multimédia de suporte ao conto de histórias, de natureza educativa, que veio a designar-se por Curucucu©, Conto eu ou contas tu?. O processo de prototipagem da aplicação multimédia implementou uma abordagem iterativa e centrada no aluno para efeitos de concetualização, desenho, implementação e testes da ferramenta e tendo como preocupação central o desenvolvimento de interfaces adequadas, quer às crianças, quer aos objetivos e contextos de utilização, integrando crianças e professores do 1.º CEB, bem como outros informantes (na qualidade de peritos) em fases específicas do processo. Para além das limitações encontradas no estudo, avançam-se propostas de investigação futura.

Summary form only given, as follows. In Vol. 12, no. 3 (Summer 2007), page 9, bottom of the left column, in 'Computer Architecture and Amdahl??s Law' by Gene Amdahl, the claim about invalidating Amdahl??s Law in 1988 came from a team at Sandia National Laboratories, and not Los Alamos. The correct text should read: "Several years later I was informed of a proof that Amdahl's Law was invalidated by someone at Sandia National Laboratories, where a number of computers interconnected as an Ncube by communication lines, but with each computer also connected to I/O devices for loading the operating system, initial data, and results." On page 20 of the same issue, in the second sentence of the diagram explanation note by Justin Rattner, the percentage figures for the sequential and the system coordination parts of the workload were interchanged. The correct version of this sentence should read: "assuming a fixed sized problem, Amdahl speculated that most programs would require at least 10% of the computation to be sequential (only one instruction executing at a time), with overhead due to interprocessor coordination averaging 25%."

In this paper we present a concept of an agent-based strategy to allocate services on a Cloud system without overloading nodes and maintaining the system stability with minimum cost. To provide a base for our research we specify an abstract model of cloud resources utilization, including multiple types of resources as well as considerations for the service migration costs. We also present an early version of simulation environment and a prototype of agent-based load balancer implemented in functional language Scala and Akka framework.
