An oxovanadium(IV) vitamin-B6 Schiff base complex, viz. VO(HL)( acdppz)] Cl, having (acridinyl) dipyridophenazine (acdppz) shows specific localization to endoplasmic reticulum (ER) and remarkable apoptotic photocytotoxicity in visible light (400-700 nm) in HeLa and MCF-7 cancer cells (IC50 < 0.6 mu M) while being non-toxic in the dark and to MCF-10A normal cells (IC50 > 40 mu M).

Motivated by several recent experimental observations that vitamin-D could interact with antigen presenting cells (APCs) and T-lymphocyte cells (T-cells) to promote and to regulate different stages of immune response, we developed a coarse grained but general kinetic model in an attempt to capture the role of vitamin-D in immunomodulatory responses. Our kinetic model, developed using the ideas of chemical network theory, leads to a system of nine coupled equations that we solve both by direct and by stochastic (Gillespie) methods. Both the analyses consistently provide detail information on the dependence of immune response to the variation of critical rate parameters. We find that although vitamin-D plays a negligible role in the initial immune response, it exerts a profound influence in the long term, especially in helping the system to achieve a new, stable steady state. The study explores the role of vitamin-D in preserving an observed bistability in the phase diagram (spanned by system parameters) of immune regulation, thus allowing the response to tolerate a wide range of pathogenic stimulation which could help in resisting autoimmune diseases. We also study how vitamin-D affects the time dependent population of dendritic cells that connect between innate and adaptive immune responses. Variations in dose dependent response of anti-inflammatory and pro-inflammatory T-cell populations to vitamin-D correlate well with recent experimental results. Our kinetic model allows for an estimation of the range of optimum level of vitamin-D required for smooth functioning of the immune system and for control of both hyper-regulation and inflammation. Most importantly, the present study reveals that an overdose or toxic level of vitamin-D or any steroid analogue could give rise to too large a tolerant response, leading to an inefficacy in adaptive immune function.

Tuberculosis is continuing as a problem of mankind. With evolution, MDR and XDR forms of tuberculosis have emerged from drug sensitive strain. MDR and XDR strains are resistant to most of the antibiotics, making the management more difficult. BCG vaccine is not providing complete protection against tuberculosis. Therefore new infections are spreading at a tremendous rate. At the present moment there is experimental evidence to believe that Vitamin A and Vitamin D has anti-mycobacterial property. It is in this context, we have hypothesized a host based approach using the above vitamins that can cause possible prevention and cure of tuberculosis with minimal chance of resistance or toxicity. Iron(III)-Schiff base complexes, namely, Fe(tsc-py)(2)](NO3) (1), Fe(tsc-acpy)(2)](NO3) (2) and Fe(tsc-VB6)(2)](NO3) (3), where tsc-py, tsc-acpy and tsc-VB6 are the respective Schiff bases derived from thiosemicarbazide (tsc) and pyridine-2-aldehyde (tsc-py), 2-acetyl pyridine (tsc-acpy) and vitamin B-6 (pyridoxal, tsc-VB6), have been prepared, structurally characterized and their photocytotoxicity studied in cancer HeLa cells. The single crystal X-ray structures of the complexes 1 and 2 show a distorted octahedral geometry formed by the FeN4S2 core. The low-spin and 1 : 1 electrolytic complexes display a broad absorption band in the visible region. Complexes 1 and 2, without any VB6 moiety are not cytotoxic under light or dark conditions. Complex 3 is significantly photocytotoxic under visible light of 400-700 nm giving an IC50 value of 22.5 mu M in HeLa cells with no dark toxicity (IC50 > 100 mu M). The photo-induced cell death is attributable to apoptotic pathways involving photo-assisted generation of intracellular ROS. The observed photocytotoxicity of complex 3 could be the result of its better photosensitizing property combined with its enhanced uptake into cancer cells via a VB6 transporting membrane carrier (VTC) mediated diffusion pathway due to the presence of the VB6 moiety compared to the two non-vitamin B-6 analogues, complexes 1 and 2.

Complexes 1a, as the perchlorate salt of 1, and 2a, as the hexafluorophosphate salt of 2, were structurally characterized. Vitamin-B6 transporting membrane carrier (VTC) mediated entry into tumour cells in preference to the normal ones seems to be responsible for the higher cellular uptake of the complexes into HeLa and MCF-7 cells over MCF-10A cells. Complexes 3 and 4 having acdppz as the photosensitizer exhibit remarkable photocytotoxicity in these cancer cells giving IC50 of < 0.9 mu M. The complexes remain non-toxic in the dark. The complexes show photo-induced apoptotic cell death via singlet oxygen (O-1(2)) generation. Fluorescence microscopy reveals specific localization of complex 4 to endoplasmic reticulum (ER) and generation of O-1(2) possibly leads to apoptotic cell death by triggering ER stress response (ERSR).

Vitamin-B6 (VB6) Schiff base (H2L) copper(II) complexes of pyridyl bases, viz. Cu(bpy)(L)] (1), Cu(phen)(L)] (2) and Cu(dppz)(L)] (3), where bpy is 2,2'-bipyridine, phen is 1,10-phenanthroline and dppz is dipyrido3,2-a:2',3'c]phenazine are synthesized, characterized and their phto-induced anticancer activity studied. The non-electrolytic one electron paramagnetic complexes exhibit a d-d band near 700 nm in DMF. The dppz complex intercalatively binds to calf-thymus DNA with binding constant (K-b) values of similar to 10(6) M-1. This complex exhibits low chemical nuclease activity but excellent DNA photocleavage activity when irradiated with red light of 705 nm forming (OH)-O-center dot radical. It displays remarkable photocytotoxicity in human cervical cancer cells (HeLa) giving IC50 value of 0.9 mu M in visible light (400-700 nm) while being less toxic in darkness (IC50 : 23 mu M). The cellular uptake of the complexes seems to be via VB6 transporting membrane carrier mediated diffusion pathway. Photo-induced cell death follows apoptotic pathway involving photo-generated intracellular reactive oxygen species.

El objetivo del experimento fue evaluar el efecto del Hidroxicolecalciferol [HyD (25 –OH- D3)] en pollos de engorde, atraves de porcentajes de ceniza, calcio, fosforo y diagnostico de síndrome de hueso negro, se evaluaron dos tratamientos ( HyD y Testigo) con seis repeticiones para cada uno en dos tiempos a los 21 y 35 días de edad en análisis de ceniza, calcio y fosforo para lo cual se extrajo una tibia por pollo, dichos análisis resultaron con diferencias no significativas en ambas edades, las evaluaciones dieron como resultado que de los 21 día a los 35 días disminuyen su valor, ceniza baja de 43.8% a 36.8% en el testigo y de 42.4% a 37.7% en HyD, calcio de 15.6% a 13.4% para el testigo y de 16% a 14.5% para HyD de igual manera para los porcentaje de fosforo de 21 a 35 días con 7.6% a 6.5% para testigo y 7.5% a 6.7% para HyD. A los 35 días los resultados son mayores en el grupo HyD, 37.7% HyD,36.8% testigo en ceniza, calcio 14.5% HyD, testigo 13.4% y fosforo 6.7% HyD , 6.5% para testigo, las diferencias de 21 a 35 días son notorias y conservan la parte proporcional en que los porcentajes estan en ceniza calcio y fosforo, pero estas disminuyen para una misma variable de los 21 días de edad a los 35, sin encontrar diferencia significativas. A los 35 días se realizo análisis de síndrome de hueso negro con un total de 22 repeticiones por tratamiento, la extracción de dicha muestra (tibia) se realizo en planta de proceso, el análisis determino diferencias entre las aves muestreadas dando los mejores resultados aquellas que fueron alimentadas con Hidroxicolecalciferol [HyD (25 –OH- D3)], se observo en los resultados que en el grupo con HyD alcanzo un 91% de individuos sanos superando significativamente al testigo que solo llego a un 77% de individuos sanos, empleando un grado de libertad y 0.05 de significancia, lo que indica diminución de la presencia del síndrome de hueso negro, producto del HyD, empleando la línea genética Cobb 500, y alimentando los pollos del día cero al día 21 con diferencias de tratamiento e igual alimento del día 22 al día 35.

[EN]Forty feedlot steers were fed a barleygrain-based finishing diet typical for western Canada, with two levels of supplementary vitamin E (468 or 1068 IU head_1 d_1) and the effect on backfat trans-18:1 isomeric profile was determined. Feeding 1068 IU vitamin E reduced the total trans-18:1 content in backfat (P<0.01), as well as the percentage of trans 10-18:1 (P<0.001), which are related to an increased risk for cardiovascular diseases. On the other hand, trans 11-18:1 (vaccenic acid) the precursor for cis 9,trans 11- 18:2 (rumenic acid), which have several purported health benefits, increased (P<0.01). Vitamin E could, therefore, be used to decrease trans-18:1 in beef and improve its isomeric profile.

A feeding trial was conducted to study the role of vitamin E in growth of Catla catla fry. Newly hatched larvae of Catla were fed with synthetic diet, supplemented with graded levels of vitamin E α0, 50, 100, 150, 200, 250 mg/Kg of diet. The spawn were fed with five times of their body weight for 30 days. Observation was made on the basis of survival, growth, daily weight gain and food conversion ratio. The significant weight gain and highest survival could be achieved by the diet supplemented with 150 mg of vitamin E per kg of the diet. The weight gain per day in 0, 50, 100, 150, 200 and 250 mg vitamin F/kg supplemented diet were 4.0, 5.2, 6.5, 7.8, 6.8 and 6.3 mg, while survival was 50, 51.8, 52.4, 52.8, 52.2 and 52% respectively.

Increase in body protein concentration was noted in the fry of rohu fed vitamin E at 0.0 mg, 25 mg, 50 mg, 75 mg 100 mg, 150 mg, 200 mg, 300 mg, and 400 mg/kg of formulated diet. The protein content of fry was found to be significantly different between the different levels of vitamin E. It was found to be maximum (18.7%) in the case of vitamin E at 75 mg/kg diet followed by vitamin E at 100 mg/kg diet. On increasing of vitamin E above 100 mg per kilogram diet there was further decline in the protein content of the fry. The protein increase, from the initial value (12.5%), were recorded to be 14.4%, control; 21.6%, 25 mg level; 31.2%, 50 mg level; 49.6%, 75 mg level; 45.6%, 100 mg level; 14.4%, 150 mg level; 28% 200 mg level; 29.6%, 300 mg level and 13.6% for 400 mg level in different experimental groups.

Effects of three different doses of vitamin D sub(3) on molting, growth, and calcium and phosphate composition of tissue and molt during the grow-out of the giant freshwater prawn Macrobrachium rosenbergii (average weight 10.56 ± 0.20 g), obtained from a grow-out pond, were studied. Intramuscular doses of vitamin D sub(3) (100, 500 and 2000 IU/kg body weight) were given on the 1st, 3rd, 5th, 7th, 9th, 11th, 13th, 15th, 20th, 25th and 30th days. All the experimental animals were fed with a basal diet containing fish meal, shrimp meal, wheat flour, groundnut de-oiled cake, soybean meal and wheat bran at 3% of the body weight. The numbers of molts were recorded as 20±0.50, 29±1.16, 51±1.87, and 30±1.60 in control, 100, 500 and 2000 IU/kg body weight physiological doses, respectively. Maximum growth was recorded in prawns given 500 IU/ kg dose. Survival was between 58.33 ± 9.13 and 77.77 ± 8.61%. The ash content and calcium level increased significantly (p<0.05) and recorded the highest values in 500 IU/kg physiological dose. However, the inorganic phosphate (P sub(i)) content recorded the highest values in tissue in 2000 IU/kg dose (p<0.05, F = 50.60613). There is no significant difference in calcium contents (p>0.05) in both tissue and molt at 500 and 2000 IU/kg doses. It was found that a higher physiological dose (2000 IU/kg) of vitamin D sub(3) increased the rate of mortality. Results have shown that vitamin D sub(3) has a positive impact on the growth and survival of M. rosenbergii and it interferes with the metabolism of Ca and P sub(i), in tissue, and alters molting frequency. Results on physiological dose suggest an alternative and effective dietary supplementation method of vitamin D sub(3) in the grow-out phase of M. rosenbergii.

Experiments on the study of different dietary levels of vitamin E on the growth and breeding performance of Heteropneustes fossilis brood fish were carried out in two phases. The first phase consisted of studying its ovarian development and the second phase on breeding performance. Sixty female fishes were stocked in twelve experimental chambers of a raceway. The effects of four dietary vitamin E levels viz. 0 (served as control), 50, 100 and 200 mg/kg feed, on the somatic growth, ovarian development of brood fish and on their breeding performance were studied. Each treatment had three replications. It was observed that body growth in terms of length and weight was best with 0 mg vitamin E/kg feed and 200 mg vitamin E/kg of feed gave poorest result. The gonado-somatic index and fecundity, however, was highest in the fish fed with 100 mg vitamin E/kg of feed. In case of breeding performance such as ovulation rate, fertilization rate, hatching rate and survival rate, the best result was obtained with 200 mg vitamin E/kg of feed. The overall result of this experiment indicates that 200 mg vitamin E/kg of feed is the best vitamin E dose for H fossilis brood and vitamin E content has a positive impact on ovarian development.
