Protein aggregation, linked to many of diseases, is initiated when monomers access rogue conformations that are poised to form amyloid fibrils. We show, using simulations of src SH3 domain, that mechanical force enhances the population of the aggregation-prone (N*) states, which are rarely populated under force free native conditions but are encoded in the spectrum of native fluctuations. The folding phase diagrams of SH3 as a function of denaturant concentration (C]), mechanical force (f), and temperature exhibit an apparent two-state behavior, without revealing the presence of the elusive N* states. Interestingly, the phase boundaries separating the folded and unfolded states at all C] and f fall on a master curve, which can be quantitatively described using an analogy to superconductors in a magnetic field. The free energy profiles as a function of the molecular extension (R), which are accessible in pulling experiments, (R), reveal the presence of a native-like N* with a disordered solvent-exposed amino-terminal beta-strand. The structure of the N* state is identical with that found in Fyn SH3 by NMR dispersion experiments. We show that the timescale for fibril formation can be estimated from the population of the N* state, determined by the free energy gap separating the native structure and the N* state, a finding that can be used to assess fibril forming tendencies of proteins. The structures of the N* state are used to show that oligomer formation and likely route to fibrils occur by a domain-swap mechanism in SH3 domain. (C) 2014 Elsevier Ltd. All rights reserved.

An elastic organic crystal, 2,6-dichlorobenzylidine-4-fluoro-3-nitroaniline (DFNA), which also shows thermosalient behavior, is studied. The presence of these two distinct properties in the same crystal is unusual and unprecedented because they follow respectively from isotropy and anisotropy in the crystal packing. Therefore, while both properties lead from the crystal structure, the mechanisms for bending and thermosalience are quite independent of one another. Crystals of the low-temperature (a) form of the title compound are bent easily without any signs of fracture with the application of deforming stress, and this bending is within the elastic limit. The crystal structure of the a-form was determined (P2(1)/c, Z = 4, a = 3.927(7) angstrom, b = 21.98(4) angstrom, c = 15.32(3) angstrom). There is an irreversible phase transition at 138 degrees C of this form to the high-temperature beta-form followed by melting at 140 degrees C. Variable-temperature X-ray powder diffraction was used to investigate the structural changes across the phase transition and, along with an FTIR study, establishes the structure of the beta-form. A possible rationale for strain build-up is given. Thermosalient behavior arises from anisotropic changes in the three unit cell parameters across the phase transition, notably an increase in the b axis parameter from 21.98 to 22.30 angstrom. A rationale is provided for the existence of both elasticity and thermosalience in the same crystal. FTIR studies across the phase transition reveal important mechanistic insights: (i) increased pi...pi repulsions along 100] lead to expansion along the a axis; (ii) change in alignment of C-Cl and NO2 groups result from density changes; and (iii) competition between short-range repulsive (pi...pi) interactions and long-range attractive dipolar interactions (C-Cl and NO2) could lie at the origin of the existence of two distinctive properties.

Collisions of a particle pair induced by optical tweezers have been employed to study colloidal stability. In order to deepen insights regarding the collision-sticking dynamics of a particle pair in the optical trap that were observed in experimental approaches at the particle level, the authors carry out a Brownian dynamics simulation. In the simulation, various contributing factors, including the Derjaguin-Landau-Verwey-Overbeek interaction of particles, hydrodynamic interactions, optical trapping forces on the two particles, and the Brownian motion, were all taken into account. The simulation reproduces the tendencies of the accumulated sticking probability during the trapping duration for the trapped particle pair described in our previous study and provides an explanation for why the two entangled particles in the trap experience two different statuses. To improve the cod stocks in the Baltic Sea, a number of regulations have recently been established by the International Baltic Sea Fisheries Commission (IBSFC) and the European Commission. According to these, fishermen are obliged to use nets with escape windows (BACOMA nets) with a mesh size of the escape window of 120 mm until end of September 2003. These nets however, retain only fish much larger than the legal minimum landing size would al-low. Due to the present stock structure only few of such large fish are however existent. As a consequence fishermen use a legal alternative net. This is a conventional trawl with a cod-end of 130 mm diamond-shaped meshes (IBSFC-rules of 1st April 2002), to be increased to 140 mm on 1st September 2003, according to the mentioned IBSFC-rule. Due legal alterations of the net by the fishermen (e.g. use of extra stiff net material) these nets have acquired extremely low selective properties, i. e. they catch very small fish and produce great amounts of discards. Due to the increase of the minimum landing size from 35 to 38 cm for cod in the Baltic, the amount of discards has even increased since the beginning of 2003. Experiments have now been carried out with the BACOMAnet on German and Swedish commercial and research vessels since arguments were brought forward that the BACOMA net was not yet sufficiently tested on commercial vessels. The results of all experiments conducted so far, are compiled and evaluated here. As a result of the Swedish, Danish and German initiative and research the European Commission reacted upon this in June 2003 and rejected the increase of the diamond-meshed non-BACOMA net from 130 mm to 140mm in September 2003. To protect the cod stocks in the Baltic Sea more effectively the use of traditional diamond meshed cod-ends with-out escape window are prohibited in community waters without derogation, becoming effective 1st of September 2003. To enable more effective and simplified control of the bottom trawl fishery in the Baltic Sea the principle of a ”One-Net-Rule“ is enforced. This is going to be the BACOMA net, with the meshes of the escape window being 110 mm for the time being. The description of the BACOMA net as given in the IBSFC-rules no.10 (revision of the 28th session, Berlin 2002) concentrates on the cod-end and the escape window but only to a less extent on the design and mesh-composition of the remaining parts of the net, such as belly and funnel and many details. Thus, the present description is not complete and leaves, according to fishermen, ample opportunity for manipulation. An initiative has been started in Germany with joint effort from scientists and the fishery to better describe the entire net and to produce a proposal for a more comprehensive description, leaving less space for manipulation. A proposal in this direction is given here and shall be seen as a starting point for a discussion and development towards an internationally uniform net, which is agreed amongst the fishery, scientists and politicians. The Baltic Sea fishery is invited to comment on this proposal, and recommendations for further improvement and specifications are welcomed. Once the design is agreed by the Baltic Fishermen Association, it shall be proposed to the IBSFC and European Commission via the Baltic Fishermen Association.

ENGLISH: Data from tagging experiments initiated during 1968-1974 in the eastern Pacific Ocean were used to study the migrations of yellowfin tuna in that area. The map method, the parallel-area method, and the Jones method were employed in the analyses. The map method gives a useful impression of the distances and directions traveled, but does not express these parameters in quantitative terms. The parallel-area method is particularly useful for determining whether or not there is net movement in particular directions, i.e. inshore-offshore, east-west, or north-south. The first of these is of particular interest, as the incidence of smaller fish is much higher in the catches made inshore than in those made offshore, and it is desirable to know whether this is due to relatively greater abundance or to relatively greater vulnerability of the smaller fish in the inshore areas. If the former were the case an offshore movement of the fish as they grew older would probably be detected. Such a movement was not detected, however, so it appears likely that the differences in the catches of smaller fish in the inshore and offshore areas are due mainly to differences in vulnerability. Few or no east-west or north-south tendencies in the movements of the fish were detected. The Jones method indicates that the movement is not random, but reveals no pronounced directional tendencies. SPANISH: Se emplearon los datos de los experimentos de marcado, iniciados en el Océano Pacífico oriental durante 1968-1974 para estudiar los desplazamientos del atún aleta amarilla en esa zona. En los análisis se emplearon los métodos cartográficos, de las zonas paralelas y de Jones. El método cartográfico ofrece una idea útil sobre la distancia y dirección de los desplazamientos, pero no expresa estos parámetros en términos cuantitativos. El método de las zonas paralelas es particularmente conveniente para determinar si existe o nó un desplazamiento neto en una dirección especial, es decir, hacia la costa-fuera de la costa, este-oeste o norte-sur. El primero de éstos tiene un interés especial, ya que la incidencia de peces más pequeños es muy superior en las capturas realizadas cerca de la costa que en las de mar afuera, y se desea conocer si ésto se debe a la abundancia relativamente superior o a las vulnerabilidad relativamente mayor de los pequeños peces en las zonas costeras. Si el caso fuera el primero, se podría descubrir probablemente un movimiento de los peces mar afuera a medida que crecen. Sin embargo, no se ha descubierto tal movimiento, así que es probable que las diferencias en las capturas de peces pequeños en las zonas costeras y mar afuera se deban principalmente a diferencias en la vulnerabilidad. Se descubrió poca o ninguna tendencia en los peces a desplazarse este-oeste o norte-sur. El método de Jones indica que el movimiento no es aleatorio, pero no revela una tendencia pronunciada a orientarse direccionalmente.

Abstract—Burst-and-coast is the most common locomotion type in freely routine swimming of koi carps (Cyprinus carpio koi), which consists of a burst phase and a coast phase in each cycle and mostly leads to a straight-line trajectory. Combining with the tracking experiment, the flow physics of koi carp’s burst-andcoast swimming is investigated using a novel integrated CFD method solving the body-fluid interaction problem. The dynamical equations of a deforming body are formulated. Following that, the loose-coupled equations of the body dynamics and the fluid dynamics are numerically solved with the integrated method. The two burst modes, MT (Multiple Tail-beat) and HT (Half Tail-beat), which have been reported by the experiments, are investigated by numerical simulations in this paper. The body kinematics is predicted and the flow physics is visualized, which are in good agreement with the corresponding experiments. Furthermore, the optimization on the energy cost and several critical control mechanisms in burst-and-coast swimming of koi carps are explored, by varying the parameters in its selfpropelled swimming. In this paper, energetics is measured by the two mechanical quantities, total output power CP and Froude efficiency Fr. Results and discussion show that from the standpoint of mechanical energy, burst-and-coast swimming does not actually save energy comparing with steady swimming at the same average speed, in that frequently changing of speed leads to decrease of efficiency.

The aim of this study is to analyze the gender segregation in undergraduate studies in the University from Basque Country (UPV/EHU). We use data from UPV/EHU for the period 2003-2013. We focus on the period from 2003 to 2013 to analyze the changes in the segregation over ten years. We analyze the tendencies of the men and the women inside undergraduate studies. Undergraduate studies are decomposed into five fields: Legal and social sciences, experimental sciences, engineering, arts and humanities, and health sciences. We draw segregation curves and compute the Gini segregation index within the Lorenz approach. Our results show that the gender segregation in undergraduate studies in the UPV/EHU has decreased from 2003 to 2013.

The muscles of the various regions and zones of the body of the two teleosts, A. dussumieri and O. striatus have been analyzed for lipid contents. There is a significant dorsum-ventral gradient in lipid concentration exhibited by both the fishes with higher lipid values in the ventral aspect of the body, especially the belly flaps. As regards the vertical series, both the fishes exhibit comparatively higher lipid contents at the dorsal aspects of the caudal region and at anterior portion of the dorsal fin area with lower lipid values at cephalic and middle portions of the body. The red muscle of Arius exhibits higher lipid content than the white muscle.

An experiment was undertaken in which silver bellies (Leiognathus splendens) of different quality were used to produce silages using different concentrations of hydrochloric acid and formic acid. The quality and storage life of the various preparations are reported. Silages which keep for at least 30 days can be produced from silver belly held for 3 or 12 hours at 28°C by: 1) reducing the pH by addition of hydrochloric acid; 2) adding 0.5% formic acid and reducing the pH to 3.5 with hydrochloric acid; or 3) adding 2.5% formic acid.

An investigation was undertaken on the production of dried products from silver belly (Leiognathus splendens) silage mixed with plant filter materials. Silages produced using hydrochloric acid and/or formic acid when mixed with rice bran or maize meal and dried, yielded powders having an acceptable appearance and a pleasant odour, which are suitable for use in compounded chicken feeds.

Economic aspects of producing liquid and dried silage from silver belly (Leiognathus splendens) in Sri Lanka are considered. A discounted cash flow analysis for the production of a dried fish silage/rice bran product suitable for use in compounded poultry feeds shows that the internal rate of return for a 10-year project would be between 34-77% and for a 5-year project between 26-73%. Thus it is concluded that the project would be extremely profitable.

Amphibian skin contains rich neuropeptides. In the present study, a novel neuromedin U (NmU) analog was isolated from skin secretions of Chinese red belly Load Bombina maxima. Being 17-amino acids long, its primary structure was established as DSSGIVGRPFFLFRPRN-NH2, in which the C-terminal 8-residue segment (FFLFRPRN) is the same as that of rat NmU, while the N-terminal part DSSGIVGRP shows a great sequence variation compared with those of NmU peptides from different resources. The peptide, named Bm-NmU-17, was found to elicit concentration-dependent contractile effects on smooth muscle of rat uterus horns. The cDNA Structure of the peptide, as obtained by a 3'-RACE strategy and subsequently cloning from a skin cDNA library, was found to contain a coding region of 438 nucleotides. The encoded precursor is composed of 145 amino acids with a single copy of Bm-NmU-17 located towards the C-terminus. The sequence of the peptide is preceded by a dibasic site (Lys-Arg) and followed by the sequence of Gly-Arg-Lys, providing the sites of cleavage and releasing of the mature peptide. Antimicrobial peptides secreted by the skin of many amphibians play an important role in innate immunity. From two skin cDNA libraries of two individuals of the Chinese red belly toad (Bombina maxima), we identified 56 different antimicrobial peptide cDNA sequences, each of which encodes a precursor peptide that can give rise to two kinds of antimicrobial peptides, maximin and maximin H. Among these cDNA, we found that the mean number of nucleotide substitution per non-synonymous site in both the maximin and maximin H domains significantly exceed the mean number of nucleotide substitution per synonymous site, whereas the same pattern was not observed in other structural regions, such as the signal and propiece peptide regions, suggesting that these antimicrobial peptide genes have been experiencing rapid diversification driven by Darwinian selection. We cloned and sequenced seven genes amplified from skin or liver genomic DNA. These genes have three exons and share the same gene structure, in which both maximin and maximin H are encoded by the third exon. This suggests that alternative splicing and somatic recombination are less likely to play a role in creating the diversity of maximins and maximin Hs. The gene trees based on different domain regions revealed that domain shuffling or gene conversion among these genes might have happened frequently.
