L’oestradiol joue un rôle important dans la reproduction en général, particulièrement dans la croissance folliculaire chez la vache. La production de l’œstradiol nécessite l’expression du gène CYP19A1 suite à la stimulation des cellules de granulosa par l’hormone folliculostimulante (FSH) ou le facteur de croissance insulinique de type 1 (IGF-1). Chez la vache, il existe six promoteurs (1.1 ; 1.2 ; 1.3 ; 1.4 ; 1.5 et 2) qui dirigent la transcription du gène CYP19A1 dans les cellules de la granulosa. Le principal promoteur qui dirige la transcription au niveau de l’ovaire (cellules de granulosa) est le promoteur 2 (P2). Cependant, l’effet de la FSH et de l’IGF-1 sur l’activation de ces promoteurs d’aromatase demeure mal connu. De plus, la demi-vie du transcrit CYP19A1 est très courte avec une région 3’UTR relativement longue. L’analyse de la séquence 3’UTR montre la présence des motifs ARE (séquence riche en AU), des études antérieur montrent que ces séquences impliquent dans la régulation de la stabilité ou la dégradation de l’ARNm, ce qui est fort probable que la courte demi-vie de l’ARNm CYP19A1 est sous le contrôle post-transcriptionel. L’objectif de la thèse visait à étudier la régulation de l’expression du gène CYP19A1 chez la vache. Il y a deux thèmes soit étude de la régulation transcriptionnelle ciblant le promoteur et soit étude de la régulation post-transcriptionnelle impliquant la région 3’non traduite (3’UTR). Le premier objectif vise à étudier la régulation transcriptionnelle du gène CYP19A1. Nous avons étudié l'activité du promoteur ovarien bovin dans deux modèles de cellules de la granulosa, les cellules lutéinisées et nonlutéinisées in vitro, suite à une stimulation des cellules par la FSH ou IGF-1. Nous avons également évalué la voie de signalisation impliquée dans la régulation des différents promoteurs en utilisant un RT-PCR et un gène rapporteur (les différents promoteurs d’aromatase ont été insérés dans le vecteur pGL3promoter en amont du gène exprimant la luciférase). Les résultats de RT-PCR démontrent que la FSH et l’IGF-1 augmentent les concentrations d’ARNm provenant des deux promoteurs 2 et 1.1 dans les cellules de la granulosa non lutéinisées. Des expériences subséquentes ont montré que la FSH stimule le promoteur 2 via la voie PKA tandis que l'IGF-1 stimule le promoteur 2 via la voie PKC. La FSH et l’IGF-1 stimulent l’expression du promoteur 1.1 via la voie PI3K. L’analyse de l’activité luciférase démontre que dans les cellules de granulosa lutéinisées, la FSH stimule le promoteur 1.1 de façon dose dépendante et ne semble y avoir aucun effet significatif sur le promoteur 2. Nous avons donc comparé l’activité du promoteur PII/P2 humain, du rat, de la chèvre et de la vache dans les cellules de granulosa bovine lutéinisées. Le résultat le plus significatif est que le promoteur 2 bovine (et caprine) dépend de plusieurs facteurs de transcription (NR5A2, FOXL2) comparé au promoteur PII humain et celui du promoteur proximal du rat qui dépendent principalement de l'AMPc. En effet, nos résultats ont démontré une expression raisonnablement robuste du P2 bovine lorsque les cellules sont traitées à la forskoline, NR5A2 et FOXL2. Le facteur FOXL2 semble déterminer l'activité du promoteur 2 chez le ruminant. Le deuxième objectif vise à étudier la régulation post-transcriptionnelle du gène CYP19A1. Pour ce faire, nous avons déterminé la séquence minimale de l'ARNm CYP19A1 requise pour la régulation de sa demi-vie. Différents séquences de la région 3’UTR ont été insérés dans le vecteur pGL3promoter en aval du gène exprimant la luciférase ou soit dans le vecteur pGEMTeasy. Le vecteur pGL3promoter a été transfecté dans les cellules de granulosa lutéinisées pour évaluer l'impact de la séquence 3'UTR sur l'expression du gène rapporteur de la luciférase, alors que le vecteur pGEMTeasy a été utilisé pour la transcription in vitro afin de générer de l’ARNm. Ce dernier sera utilisé en réaction croisée au UV avec des extraits protéiques pour démontrer l’association du complexe ARNm/protéine. L’analyse de l’activité luciférase a permis d’identifier une séquence de 200 pb située entre 926 et 1134 pb de la région 3'UTR de l’ARNm CYP19A1 qui a réduit significativement l’activité de la luciférase. Selon les analyses de la réaction croisée au UV, une ou plusieurs protéines de 66 et 80 kDA se lient spécifiquement à la séquence de 200 pb qui réduit l’activité de luciférase. Cette protéine s'exprime dans les cellules de granulosa, mais n’a pas été détectée dans d'autres tissus comme le foie et le cœur. Par ailleurs, l’utilisation du gène rapporteur sensible à la FSH a suscité l’intérêt d'une compagnie pharmaceutique qui vend de l’equine chorionic gonadotropin (eCG) pour lui permettre de distinguer facilement l’eCG ayant une forte activité FSH et donc, avoir un produit commercial plus efficace et de meilleure qualité. Dans cette étude, nous avons développé un système de bioessai à la FSH basé sur la transfection des cellules avec un récepteur à la FSH et un gène rapporteur colorimètrique qui permet d’estimer l’activité de la FSH dans le sérum de la jument et qui pourrait être applicable au niveau de la ferme/industrie.

La infertilidad afecta en la actualidad a aproximadamente 1 de cada 7 parejas a nivel mundial. La falla ovárica prematura (FOP) es una condición común en la población femenina, afectando al 1% de mujeres menores de 40 años. La etiología de la FOP es idiopática entre el 50% y el 80% de los casos, lo que sugiere causas genéticas, epigenéticas y ambientales aún desconocidas. A pesar de los avances en las técnicas de cartografía genética y de sistematización de la técnica de Sanger, pocos genes etiológicos de FOP fueron identificados en los últimos 20 años. Este fracaso relativo se asoció principalmente a que cientos de genes, que abarcan grandes regiones del genoma, son candidatos pero la técnica de secuenciación directa sólo permite el análisis de unas 700bp en cada reacción. En el presente trabajo se empleó la secuenciación de siguiente generación (NGS) para la búsqueda de mutaciones en 70 genes candidatos que potencialmente contribuyen con el desarrollo de la patología. Se identificaron mutaciones en 3 de 12 pacientes. La paciente POF-7 presentaba una mutación no sinónima en el gen ADAMTS19 (c.2828C>T, p.Thr943Ile). La proteína ADAMTS19 se clasifica dentro de la familia ADAMTS como huérfana ya que no se ha identificado su sustrato. Mediante el sistema de doble hibrido en levaduras se buscó identificar las potenciales proteínas que interactúan con ADAMTS19. Permitió identificar, a partir de las versiones murinas, la interacción de Adamts19 y Col6a2. Para comprobar la interacción entre las proteínas ADAMTS19 y COL6A2 humanas se empleó el sistema de doble hibrido en células eucariotas. Los hallazgos no permitieron replicar los resultados obtenidos previamente. En síntesis de identificó una mutación potencialmente causal de FOP en un gen nuevo y una muy probable interacción entre ADAMTS19 y COL6A2.

The mechanisms whereby the high variation in numbers of morphologically healthy oocytes and follicles in ovaries (ovarian reserve) may have an impact onovarian function, oocyte quality, and fertility are poorly understood. The objective was to determine whether previously validated biomarkers for follicular differentiation and function, as well as oocyte quality differed between cattle with low versus a high antral follicle count (AFC). Ovaries were removed (n = 5 per group) near the beginning of the nonovulatory follicular wave, before follicles could be identified via ultrasonography as being dominant, from heifers with high versus a low AFC. The F1, F2, and F3 follicles were dissected and diameters determined. Follicular fluid and thecal, granulosal, and cumulus cells and the oocyte were isolated and subjected to biomarker analyses. Although the size and numerous biomarkers of differentiation, such as mRNAs for the gonadotropin receptors, were similar, intrafollicular concentrations of estradiol and the abundance of mRNAs for CYP19A1 in granulosal cells and ESR1, ESR2, and CTSB in cumulus cells were greater, whereas mRNAs for AMH in granulosal cells and TBC1D1 in thecal cells were lower for animals with low versus a high AFC during follicle waves. Hence, variation in the ovarian reserve may have an impact on follicular function and oocyte quality via alterations in intrafollicular estradiol production and expression of key genes involved in follicle-stimulating hormone action (AMH) and estradiol (CYP19A1) production by granulosal cells, function and survival of thecal cells (TBC1D1), responsiveness of cumulus cells to estradiol (ESR1, ESR2), and cumulus cell determinants of oocyte quality (CTSB).

Members of the transforming growth factor-beta (TGF-beta) superfamily have wide-ranging influences on many tissue and organ systems including the ovary. Two recently discovered TGF-beta superfamily members, growth/differentiation factor-9 (GDF-9) and bone morphogenetic protein-15 (BMP-15; also designated as GDF-9B) are expressed in an oocyte-specific manner from a very early stage and play a key role in promoting follicle growth beyond the primary stage. Follicle growth to the small antral stage does not require gonadotrophins but appears to be driven by local autocrine/paracrine signals from both somatic cell types (granulosa and theca) and from the oocyte. TGF-beta superfamily members expressed by follicular cells and implicated in this phase of follicle development include TGF-beta, activin, GDF-9/9B and several BMPs. Acquisition of follicle-stimulating hormone (FSH) responsiveness is a pre-requisite for growth beyond the small antral stage and evidence indicates an autocrine role for granulosa-derived activin in promoting granulosa cell proliferation, FSH receptor expression and aromatase activity. Indeed, some of the effects of FSH on granulosa cells may be mediated by endogenous activin. At the same time, activin may act on theca cells to attenuate luteinizing hormone (LH)-dependent androgen production in small to medium-size antral follicles. Dominant follicle selection appears to depend on differential FSH sensitivity amongst a growing cohort of small antral follicles. Activin may contribute to this selection process by sensitizing those follicles with the highest "activin tone" to FSH. Production of inhibin, like oestradiol, increases in selected dominant follicles, in an FSH- and insulin-like growth factor-dependent manner and may exert a paracrine action on theca cells to upregulate LH-induced secretion of androgen, an essential requirement for further oestradiol secretion by the pre-ovulatory follicle. Like activin, BMP-4 and -7 (mostly from theca), and BMP-6 (mostly from oocyte), can enhance oestradiol and inhibin secretion by bovine granulosa cells while suppressing progesterone secretion; this suggests a functional role in delaying follicle luteinization and/or atresia. Follistatin, on the other hand, may favor luteinization and/or atresia by bio-neutralizing intrafollicular activin and BMPs. Activin receptors are expressed by the oocyte and activin may have a further intrafollicular role in the terminal stages of follicle differentiation to promote oocyte maturation and developmental competence. In a reciprocal manner, oocyte-derived GDF-9/9B may act on the surrounding cumulus granulosa cells to attenuate oestradiol output and promote progesterone and hyaluronic acid production, mucification and cumulus expansion.(C) 2003 Elsevier Science B.V. In vivo and in vitro assays were performed with S91 murine melanoma cells aiming to investigate the effects of testosterone and photoperiod on tumor growth and melanogenesis (tyrosinase activity). In vivo assays were performed by inducing melanoma tumors in castrated mice receiving increasing concentrations of testosterone and submitted to varying photoperiod regimens. The results demonstrated that the increase of melanin content was higher in animals submitted to the longest days, thus demonstrating the importance of photoperiod length in melanin synthesis. Increase in tumor growth and protein content was observed in testosterone-treated animals submitted to 12L:12D; in testosterone-treated animals submitted to 4L:20D and 20L:4D tumor growth was significantly smaller. In S91 cultured cells, testosterone increased cell proliferation and reduced tyrosinase activity in a dose-dependent manner. Radioactive binding assays demonstrated that the hormone was acting through low affinity testosterone receptors, since the presence of aromatase inhibitor did not affect the binding assay in a statistically significant way, and all the in vitro experiments were performed in the presence of the inhibitor. Our in vivo data added to the in vitro results corroborate the hypothesis that S91 melanoma cells directly respond to testosterone and that this effect is modulated by light.

Objective: To investigate whether submicroscopic copy number variants (CNVs) on the X chromosome can be identified in women with primary ovarian insufficiency (POI), defined as spontaneous secondary amenorrhea before 40 years of age accompanied by follicle-stimulating hormone levels above 40 IU/L on at least two occasions. Design: Analysis of intensity data of single nucleotide polymorphism (SNP) probes generated by genomewide Illumina 370k CNV BeadChips, followed by the validation of identified loci using a custom designed ultra-high-density comparative genomic hybridization array containing 48,325 probes evenly distributed over the X chromosome. Setting: Multicenter genetic cohort study in the Netherlands. Patient(s): 108 Dutch Caucasian women with POI, 97 of whom passed quality control, who had a normal karyogram and absent fragile X premutation, and 235 healthy Dutch Caucasian women as controls. Intervention(s): None. Main Outcome Measure(s): Amount and locus of X chromosomal microdeletions or duplications. Result(s): Intensity differences between SNP probes identify microdeletions and duplications. The initial analysis identified an overrepresentation of deletions in POI patients. Moreover, CNVs in two genes on the Xq21.3 locus (i.e., PCDH11X and TGIF2LX) were statistically significantly associated with the POI phenotype. Mean size of identified CNVs was 262 kb. However, in the validation study the identified putative Xq21.3 deletions samples did not show deviations in intensities in consecutive probes. Conclusion(s): X chromosomal submicroscopic CNVs do not play a major role in Caucasian POI patients. We provide guidelines on how submicroscopic cytogenetic POI research should be conducted. Besides the effects on peripheral energy homeostasis, insulin also has an important role in ovarian function. Obesity has a negative effect on fertility, and may play a role in the development of the polycystic ovary syndrome in susceptible women. Since insulin resistance in the ovary could contribute to the impairment of reproductive function in obese women, we evaluated insulin signaling in the ovary of high-fat diet-induced obese rats. Female Wistar rats were submitted to a high-fat diet for 120 or 180 days, and the insulin signaling pathway in the ovary was evaluated by immunoprecipitation and immunoblotting. At the end of the diet period, we observed insulin resistance, hyperinsulinemia, an increase in progesterone serum levels, an extended estrus cycle, and altered ovarian morphology in obese female rats. Moreover, in female obese rats treated for 120 days with the high-fat diet, the increase in progesterone levels occurred together with enhancement of LH levels. The ovary from high-fat-fed female rats showed a reduction in the insulin receptor substrate/phosphatidylinositol 3-kinase/AKT intracellular pathway, associated with an increase in FOXO3a, IL1B, and TNF alpha protein expression. Objective: To evaluate the effects of soy isoflavone supplementation on profile lipid and endogenous hormone levels. Methods: In this double-blind, placebo-controlled Study, 47 post menopausal women 47-66 v of age received 40 mg of isoflavone (n = 25) or 40 mg of casein placebo (11 = 22). Cardiovascular risk factors were assessed by evaluating lipid profile at baseline and after 6 mo of treatment. To examine the effects of this regime on endogenous hormone levels, follicle-stimulating hormone and beta-estradiol were measured. Urinary isoflavone concentrations (genistein and daidzein) were measured as markers of both compliance and absorption using high performance liquid chromatography. Baseline characteristics were compared by the unpaired Student`s t-test. Within-group changes were determined by paired Student`s t-test and comparison between the isoflavone and casein placebo groups were determined by analysis of variance. Results: Lipid levels (low-density lipoprotein and total cholesterol) similarly decreased in both,groups. High-density lipoprotein increased significantly in both groups and cannot thus be attributable to treatment: the reason for Such variation is unknown and can be attributed to chance or to bias (even that of a real placebo effect in both groups or perhaps in spontaneous changes in exercise and dietary habits of patients after their inclusion). Furthermore, in both groups very low-density lipoprotein and triacylglycerol levels increased in a non-significant manner. Conclusion: The results of the present Study do not support any biologically significant estrogenic effects of isoflavone on the parameters assessed. Further research will he necessary to definitively assess the safety and efficacy of isoflavone. (C) 2008 Elsevier Inc. All rights reserved.

Introduction: Hypoestrogenism is the main characteristic of female aging. It promotes significant changes in body composition, both in fat mass as in lean body mass, leading to a decrease in muscle strength and physical performance. Objective: The aim of this study was to test whether menopausal status and hormone levels are associated with muscular strength and physical performance in middle-aged women. Methods: In a cross-sectional study it was collected sociodemographic data, gynecological history, anthropometric and biochemical measures in women aged 40 to 65 years in Parnamirim-RN. The menopause status (pre, peri and post menopause) was determined by menstrual history. All women underwent three dimensions of physical performance assessment: handgrip dynamometry, gait speed and chair stands test - Short Physical Performance Battery (SPPB). Categorical data were presented as absolute and relative frequencies. Quantitative data were showed as mean and standard deviation and the normality of distribution was verified with Kolmogorov-Smirnov (KS) test. Biochemical measures of estradiol and follicle-stimulating hormone (FSH) were transformed to log10. ANOVA with Tukey post-test for comparison of variables between the groups pre, peri and post-menopausal was performed and then multiple linear regression analyzes. Results: Two hundred and seventy eight women aged 50.2 (±5.58) years composed this study, being 50 women in premenopausal status (18%), 122 in perimenopausal (43.9%), and 106 postmenopausal stage (38.1%). The groups were different in age (p=0.001), marital relationship duration (p <0.001), number of pregnancies (p=0.001) and parity (p=0.001). Differences in biochemical measures were observed among the groups: estradiol (p<0.001), FSH (p<0.001), total cholesterol (p=0.001). There were no differences in gait velocity between menopausal status. Values in mean of grip strength decreased by postmenopausal women to perimenopausal and premenopausal ones (24.5 ± 5.1, 25.6 ± 5.4, 26.9 ± 4.9 for post-stage, pre and peri menopausas, respectively, p = 0.02) and the performance of chair stands test was better in premenopausal women compared with that in peri and postmenopausal status (p = 0.02). In multiple linear regression for muscle strength, the variables that remained were: age, estradiol and somatic symptoms measured by Menopause Rating Scale-MRS (R2=0.15). While for the xiv chair-stands test the predictors were number of births and FSH values (R2=0.04). Conclusion: There is a relationship between the stages of menopause and muscle performance in measures of grip strength and sit-up test and these are influenced by the fall of estrogens levels. Data suggest that the decrease in muscle strength and physical performance already appear in the transition to menopause stage, pointing to the need for more research in this area and appropriate preventive interventions

In Experiments 1, 2, and 3, we evaluated the effects of temporary weaning (TW), equine chorionic gonadotropin (eCG), and follicle-stimulating hormone (FSH) treatments on results of it fixed-time artificial insemination (TAI) protocol in postpartum Bos indicus cows. In Experiment 1, treatment with 400 IU eCG or with TW for 48 h consistently improved pregnancy rates (PRs) at TAI, but, in Experiment 2, FSH treatment was less effective than eCG or TW. In Experiment 3, the inclusion of eCG treatment in cows subjected to TW did not improve PRs. We concluded that TW or 400 IU eCG should be included in the TAI protocol in postpartum Bos indicus cows to enhance fertility. In Experiment 4, we used records front heifers and cows treated with the proposed protocol during the 2006-2007 (n = 27,195) and 2007-2008 (n = 36,838) breeding seasons from multiple locations in Brazil to evaluate factors potentially affecting PRs. Overall PR at TAI was 49.6% (31,786 of 64,033). Pregnancy rate differed (P < 0.01) among farm within location (results ranging between 26.8% and 68.0%; P < 0.01), cow group within farm, by breed (Bos indicus, 48.3% [26,123 of 54,145]; Bos taurus, 61.7% [3652 of 5922]; and crossbred Bos indicus x Bos taurus, 50.7% [2011 of 3966]), category (nulliparous, 39.6% [2095 of 52901] suckled primiparous, 45.2% [3924 of 8677]; suckled multiparous, 51.8% [24,245 of 46,767]; and nonsuckled multiparous, 46.1% [1522 of 3299]), body condition score at TAI (<= 2.5 43.0% [3409 of 7923]; 3.0, 49.6% [18,958 of 38,229]; and >3.5, 52.7% [9419 of 17,881]). Days postpartum at beginning, of protocol did not affect PR (30 to 60 d, 47.6% [4228 of 8881]; 61 to 90 d, 51.7% 116,325 to 31,5721; and 91 to 150 d, 50.8% [7616 to 14,991]; P > 0.1). Pregnancy rate was also consistently affected (P < 0.01) by sire (results ranging from 7.2% to 77.3%) and artificial insemination technician (results ranging from 15.1% to 81.8%). Sibutramine is a drug globally used for the treatment of obesity. The aim of this study was to investigate male reproductive disorders caused by sibutramine in adult rats. Wistar rats were treated for 28 consecutive days (gavage) with 10 mg/kg of sibutramine. Control animals received only vehicle (dimethylsulfoxide and saline). The rats were sacrificed for evaluation of body and reproductive organ weights, sperm parameters, hormone levels (luteinizing hormone, follicle-stimulating hormone, and testosterone), testicular and epididymal histopathology, sexual behavior, fertility and in vitro contractility of the epididymal duct. Sibutramine decreased (P < .05) weights of the epididymis and ventral prostate, but not of other reproductive organs. The sperm number and transit time in the epididymal cauda were decreased (P < .001), but the daily sperm production was not altered. Moreover, morphology and sperm motility, histopathology of the testes and epididymis, sexual behavior, fertility, and serum hormone levels were not altered by the treatment. Sibutramine increased the potency of norepinephrine and, per se, increased the mechanical activity of the epididymal duct in vitro. Thus, although sibutramine in these experimental conditions did not interfere with the reproductive process of rats, it provoked acceleration of the sperm transit time and a decrease in the sperm reserves in the epididymal cauda. This alteration is probably related to the sympathomimetic effect of this drug, as shown by the in vitro assays. In humans, use of this drug might present a threat for male fertility because sperm reserves in men are naturally lower than those in rats.

Exposure to ultraviolet (UV) radiation induces generation of reactive oxygen species, production of proinflammatory cytokines and melanocyte-stimulating hormone (MSH) as well as increase in tyrosinase activity. The potential photoprotective effects of Coccoloba uvifera extract (CUE) were evaluated in UV-stimulated melanocytes.Human epidermal melanocytes were used as an in vitro model to evaluate the effects of CUE on the production interleukin-1 alpha (IL-1 alpha), tumor necrosis factor alpha (TNF-alpha), and alpha-MSH under basal and UV-stimulated conditions. Antioxidant and anti-tyrosinase activities were also evaluated in membrane lipid peroxidation and mushroom tyrosinase assay, respectively.Coccoloba uvifera L. showed antioxidant and anti-tyrosinase activities and also inhibited the production of IL-1 alpha, TNF-alpha and alpha-MSH in melanocytes subjected to UV radiation (P < 0.01). Moreover, CUE inhibited the activity of tyrosine kinase in cell culture under basal and UV radiation conditions (P < 0.001), corroborating the findings of the mushroom tyrosinase assay.This study supports the photoprotective potential of CUE.
