Sodium dodecyl sulfate (SDS) is commonly used to extract polyhedra from infected cells and diseased dead larval tissues. It was found, however, that 80% of Helicoverpa armigera nucleopolyhedrovirus (HaSNPV) polyhedra produced via cell culture were damaged after 30 min of 0.5% SDS treatment whereas only 20% of in vivo produced polyhedra were damaged by the same treatment. Transmission and scanning electron microscopy revealed that the damaged polyhedra had lost their polyhedron envelopes and virions were dislodged from the polyhedrin matrix, leaving empty spaces that were previously occupied by the occluded virions. Up to 20% in vitro produced polyhedra were resistant to SDS and remained intact, even after a 24 h exposure to SDS. This sensitivity to SDS was observed across a range of cell culture media, including serum supplemented media. Electron microscopy also revealed that the inferior polyhedron envelope of in vitro produced polyhedra is likely due to poor interaction between the polyhedron envelope, polyhedron envelope protein (PEP), and polyhedrin matrix. The PEP gene was cloned and sequenced and mutations in this gene were ruled out as an explanation. In vitro produced polyhedra that were passed through insect larva once were resistant to SDS, indicating that a critical component is lacking in insect cell culture medium used for producing HaSNPV or the cells growing in culture are inefficient in some ways in relation to production of polyhedra. Obstructive apnea (OA) can exert significant effects on renal sympathetic nerve activity (RSNA) and hemodynamic parameters. The present study focuses on the modulatory actions of RSNA on OA-induced sodium and water retention. The experiments were performed in renal-denervated rats (D; N = 9), which were compared to sham (S; N = 9) rats. Mean arterial pressure (MAP) and heart rate (HR) were assessed via an intrafemoral catheter. A catheter was inserted into the bladder for urinary measurements. OA episodes were induced via occlusion of the catheter inserted into the trachea. After an equilibration period, OA was induced for 20 s every 2 min and the changes in urine, MAP, HR and RSNA were recorded. Renal denervation did not alter resting MAP (S: 113 ± 4 vs D: 115 ± 4 mmHg) or HR (S: 340 ± 12 vs D: 368 ± 11 bpm). An OA episode resulted in decreased HR and MAP in both groups, but D rats showed exacerbated hypotension and attenuated bradycardia (S: -12 ± 1 mmHg and -16 ± 2 bpm vs D: -16 ± 1 mmHg and 9 ± 2 bpm; P < 0.01). The basal urinary parameters did not change during or after OA in S rats. However, D rats showed significant increases both during and after OA. Renal sympathetic nerve activity in S rats increased (34 ± 9%) during apnea episodes. These results indicate that renal denervation induces elevations of sodium content and urine volume and alters bradycardia and hypotension patterns during total OA in unconscious rats.

O cisto nasolabial é um cisto não odontogênico raro que se desenvolve na região inferior da asa nasal, com patogênese ainda incerta. Esta lesão, que possui crescimento lento e dimensões variáveis (1,5-3cm), caracteriza-se clinicamente por uma tumefação flutuante na região do sulco nasolabial ao redor da asa do nariz, causando uma elevação do lábio superior. Seu diagnóstico pode ser feito basicamente pelo quadro clínico e, se necessário, complementando-se com exames auxiliares por imagens. O presente trabalho relata o caso de uma paciente do sexo feminino de 48 anos, que se queixava da presença de uma massa consistente na região da asa esquerda do nariz e cujas características clínicas eram compatíveis com cisto nasolabial. As imagens de TC demonstraram uma formação expansiva com densidade de tecido mole, localizadas na região nasal esquerda medindo cerca de 1,2cm de diâmetro e apresentando contornos nítidos e bem definidos e densidade homogenia ao redor de 50 UH. Já as imagens de RM mostraram uma lesão de aspecto circular localizada em tecido mole, apresentando hiperintensidade nas imagens ponderadas em T1, T2 e no recurso de supressão da gordura, sendo a hipótese diagnóstico de cisto nasolabial, confirmado pelo exame histopatológico depois da cirurgia.

O objetivo foi avaliar a eficiência de trifloxysulfuron-sodium isolado, em mistura, ou em aplicação sequencial a outros herbicidas no controle de Cyperus rotundus L. na cultura da cana-de-açúcar. O estudo foi realizado em área de produção comercial do cultivar RB 72-454 na Usina Paraíso e em casa de vegetação da Universidade Estadual do Norte Fluminense Darcy Ribeiro, em Campos dos Goytacazes, Rio de Janeiro. As parcelas no campo foram submetidas a nove tratamentos: 2,4-D; trifloxysulfuron-sodium + ametryn; trifloxysulturon-sodium; 2,4-D + 2,4-D (20 dias após a primeira aplicação -DAP); 2,4-D + trifloxysulfuron-sodium + ametryn (20 DAP); 2,4-D + trifloxysulfuron-sodium (20 DAP); halosulfuron (20 DAP) e testemunha com e sem capina. Em casa de vegetação avaliou-se a viabilidade dos bulbos de C. rotundus em função dos tratamentos utilizados no experimento instalado em campo. Os resultados indicaram que o trifloxysulfuron-sodium isolado ou em mistura com ametryn, em aplicação sequencial ao 2,4-D, foi excelente no controle de C. rotundus. Esse herbicida e o halosulfuron, aplicados isoladamente, reduziram a capacidade de reinfestação da espécie daninha em razão da redução de sua propagação e do seu crescimento.
Hydraulic conductivity in response to exchangeable sodium percentage and solution salt concentration

Hydraulic conductivity is determined in laboratory assays to estimate the flow of water in saturated soils. However, the results of this analysis, when using distilled or deionized water, may not correspond to field conditions in soils with high concentrations of soluble salts. This study therefore set out to determine the hydraulic conductivity in laboratory conditions using solutions of different electrical conductivities in six soils representative of the State of Pernambuco, with the exchangeable sodium percentage adjusted in the range of 5-30%. The results showed an increase in hydraulic conductivity with both decreasing exchangeable sodium percentage and increasing electrical conductivity in the solution. The response to the treatments was more pronounced in soils with higher proportion of more active clays. Determination of hydraulic conductivity in laboratory is routinely performed with deionized or distilled water. However, in salt affected soils, these determinations should be carried out using solutions of electrical conductivity different from 0 dS m-1, with values close to those determined in the saturation extracts.

Actualmente a Tomografia Computorizada (TC) é um dos métodos de diagnóstico por imagem que tem uma maior contribuição para a dose de radiação X recebida pelos pacientes. Pretende-se com este estudo avaliar as doses praticadas em TC e contribuir para o estabelecimento de Níveis de Referência de Diagnóstico (NRD) na região da Grande Lisboa, Portugal. Foram efectuadas medições de dose em 5 equipamentos de TC multidetectores, considerando o abdómen como área anatómica de interesse. Recorreu-se a uma câmara de ionização e a um fantoma para obter o índice de dose de TC (CTDI) e o produto dose-comprimento (DLP), que permitem determinar os NRD. Estes valores foram comparados com os NRD propostos pela Guideline Europeia e com os estudos desenvolvidos em outros países, como o Reino Unido, Grécia e Taiwan. Os resultados revelaram que os valores de NRD obtidos neste estudo (16,7 mGy para o CTDIvol e 436,5 mGy·cm para o DLP) são discrepantes relativamente à Guideline Europeia (±50%), mas muito próximos relativamente aos NRD estabelecidos nos países considerados. Estes valores podem ser eventualmente explicados pelos equipamentos em análise e pela utilização de protocolos de exame adoptados pelos profissionais de Radiologia nas instituições analisadas. ABSTRACT - Nowadays Computed Tomography (CT) is one of the imaging techniques which have a large contribution to radiation dose received by patients. The purpose of this study is to evaluate CT doses and contribute to the establishment of Diagnostic Reference Levels (DRL) in Lisbon, Portugal. Dose measurements on 5 multidetector CT scanners have been performed, considering the abdomen as the anatomic region of interest. All measurements were performed using an ionization chamber and a phantom to obtain the index CT dose (CTDI) and the dose-length product (DLP), which are used to determine DRL. These values were compared not only with European reference dose values but also with DRL studies developed in other countries like United Kingdom, Greece and Taiwan. The results revealed that DRL values obtained in this study (CTDIvol is 16,7 mGy and DLP is 436,5 mGy·cm) have a higher discrepancy to European Guideline (±50%), while the DRL´s of other countries are nearest to values obtained in this study. Those differences may be eventually explained by the type of the evaluated equipments but also by the exam protocols used by the Radiology professionals on the analyzed institutions.

O aparecimento da Tomografia Computorizada (TC) Multicorte no início dos anos noventa veio revolucionar os estudos crânio-encefálicos, possibilitando a avaliação funcional da hemodinâmica cerebral através dos estudos de perfusão.Com a natural evolução dos equipamentos de TC e dos softwares comerciais de análise de imagem foi crescendo o campo de aplicabilidade da TC de perfusão, surgindo os sistemas de análise fisiológica, úteis no estudo da vasculatura tumoral. Na actualidade, a TC tem provas dadas no diagnóstico de patologias tumorais, mostrando-se útil na caracterização da lesão primária e das possíveis metástases associadas, na avaliação do risco e na monitorização da resposta terapêutica, revelando-se um excelente indicador da angiogénese tumoral. Os estudos de perfusão são obtidos através da monitorização do comportamento de um bólus de contraste iodado através da vasculatura tumoral e os dados e correntes do padrão de distribuição deste no tecido tumoral podem fornecer informações relativas às suas propriedades fisiológicas.

Introduction: The quantification of th e differential renal function in adults can be difficult due to many factors - on e of the se is the variances in kidney depth and the attenuation related with all the tissue s between the kidney and the camera. Some authors refer that t he lower attenuation i n p ediatric patients makes unnecessary the use of attenuation correction algorithms. This study will com pare the values of differential renal function obtained with and with out attenuation correction techniques . Material and Methods: Images from a group consisting of 15 individuals (aged 3 years +/ - 2) were used and two attenuation correction method s were applied – Tonnesen correction factors and the geometric mean method . The mean time of acquisition (time post 99m Tc - DMSA administration) was 3.5 hours +/ - 0.8h. Results: T he absence of any method of attenuation correction apparently seems to lead to consistent values that seem to correlate well with the ones obtained with the incorporation of methods of attenuation correction . The differences found between the values obtained with and without attenuation correction were not significant. Conclusion: T he decision of not doing any kind of attenuation correction method can apparently be justified by the minor differences verified on the relative kidney uptake values. Nevertheless, if it is recognized that there is a need for a really accurate value of the relative kidney uptake, then an attenuation correction method should be used.

Introduction: In the XXI Century ’s Society the scientific investigation process has been growing steadily , and the field of the pharmaceutical research is one of the most enthusiastic and relevant . Here, it is very important to correlate observed functional alterations with possibly modified drug bio distribution patterns . Cancer, inflammation and inf ection are processes that induce many molecular intermediates like cytokines, chemokines and other chemical complexes that can alter the pharmacokinetics of many drugs. One cause of such changes is thought to be the modulator action of these complexes in t he P - Glyco p rotein activity, because they can act like inducers/inhibitors of MDR - 1 expression. This protein results from the expression of MDR - 1 gene, and acts as an ATP energy - dependent efflux pump, with their substrates including many drugs , like antiretrovirals, anticancers, anti - infectives, immunosuppressants, steroids or opioids . Objectives: Because of the lack of methods to provide helpful information in the investigation of in vivo molecular changes in Pgp activity during infection/infl ammation processes, and its value in the explanation of the altered drug pharmacokinetic, this paper want to evaluate the potential utility of 99m Tc - Sestamibi scintigraphy during this kind of health sciences investigation. Although the a im is indeed to create a technique to the in vivo study of Pgp activity, this preliminary Project only reaches the in vitro study phase, assumed as the first step in a n evaluation period for a new tool development. Materials and Methods: For that reason , we are performing in vitro studies of influx and efflux of 99m Tc - Sestamibi ( that is a substrate of Pgp) in hepatocytes cell line (HepG2). We are interested in clarify the cellular behavior of this radiopharmaceutical in Lipopolysaccharide(LPS) stimulated cells ( well known in vitro model of inflammation) to possibly approve this methodology. To validate the results, the Pgp expression will be finally evaluated using Western Blot technique. Results: Up to this moment , we still don’t have the final results, but we have already enough data to let us believe that LPS stimulation induce a downregulation of MDR - 1, and consequently Pgp, which could conduce to a prolonged retention of 99m Tc - Sestamibi in the inflamed cells . Conclusions: If and when this methodology demonstrate the promising results we expect, one will be able to con clude that Nuclear Medicine is an important tool to help evidence based research also on this specific field .

GOAL: The manufacturing and distribution of strips of instant thin - layer chromatography with silica gel (ITLC - SG) (reference method) is currently discontinued so there is a need for an alternative method f or the determination of radiochemical purity (RCP) of 99m Tc - tetrofosmin. This study aims to compare five alternative methods proposed by the producer to determine the RCP of 99m Tc - tetrofosmin. METHODS: Nineteen vials of tetrofosmin were radiolabelled with 99m Tc and the percentages of the RCP were determined. Five different methods were compared with the standard RCP testing method (ITLC - SG, 2x20 cm): Whatman 3MM (1x10 cm) with acetone and dichloro - methane (method 1); Whatman 3MM (1x1 0 cm) with ethyl acetate (method 2); aluminum oxide - coated plastic thin - layer chromatography (TLC) plate (1x10 cm) and ethanol (method 3); Whatman 3MM (2x20 cm) with acetone and dichloro - methane (method 4); solid - phase extraction method C18 cartridge (meth od 5). RESULTS: The average values of RCP were 95,30% ± 1,28% (method 1), 93,95 ± 0,61% (method 2), 96,85% ± 0,93% (method 3), 92,94% ± 0,99% (method 4) and 96,25% ± 2,57% (method 5) (n=12 each), and 93,15% ± 1,13% for the standard method (n=19). There we re statistical significant differences in the values obtained for methods 1 (P=0,001), 3 (P=0,000) and 5 (P=0,004), and there were no statistical significant differences in the values obtained for methods 2 (P=0,113) and 4 (P=0,327). CONCLUSION: From the results obtained, methods 2 and 4 showed a higher correlation with the standard method. Unlike method 4, method 2 is less time - consuming than the reference method and can overcome the problems associated with the solvent toxicity. The remaining methods (1, 3 and 5) tended to overestimate RCP value compared to the standard method.

A delimitação precisa é crucial na RT. TC: informação anatómica, pobre na definição do envolvimento ganglionar; alguns tumores são praticamente invisíveis (p.e., esófago, fígado, baço e glândulas salivares), mapa de densidades electrónicas essencial para o cálculo. 18F-FDG PET: 1) mapeamento metabólico que permite alterar volumes alvo: BTV, elimina incertezas no GTV; 2) vantagens: estadiamento tumoral, predição da resposta tumoral, avaliação da resposta tumoral, detecção precoce da doença recorrente. PET/TC: maior precisão, menor variabilidade inter e intra-observador, menor erro no reposicionamento, adaptação à RT, ainda em Investigação Clínica.

Titanate nanotubes (TNT) with different sodium contents have been synthesised using a hydrothermal approach and a swift and highly controllable post-washing processes. The influence of the sodium/proton replacement on the structural and morphological characteristics of the prepared materials was analysed. Different optical behaviour was observed depending on the Na+/H+ samples’ content. A band gap energy of 3.27±0.03 eV was estimated for the material with higher sodium content while a value of 2.81±0.02 eV was inferred for the most protonated material, which therefore exhibits an absorption edge in the near visible region. The point of zero charge of the materials was determined and the influence of the sodium content on the adsorption of both cationic and anionic organic dyes was studied. The photocatalytic performance of the TNT samples was evaluated in the rhodamine 6G degradation process. Best photodegradation results were obtained when using the most protonated material as catalyst, although this material has shown the lowest R6G adsorption capability.

White mice were used to study the infectivity of the eggs of Lagochilascaris minor Leiper, 1909 after incubation in liquid media, with or without preservative substances. Potassium bichromate (K2Cr2O7) at 1% restrict hatching, while 1% formalin gave a greater larval yield. Incubation of eggs in distilled water, in Roux or Falcon flasks gave a good yield, whether the eggs were obtained from human feces or from experimentally infected cats. Treatment of eggs with Sodium hypochlorite (NaOCl) at 5.25% for 2 min prior to inoculation, produced a notable increment of the larval yield in the infections.
