In the State of S. Paulo, Brazil, squash plants (Cucurbita spp.) are attacked by the Javanese nematode (Meloidogyne javanica) and by M. incognita acrita. Squash belongs to that group of plants in which the root-knot nematodes break through the root surface, so that the egg-producing females protrude from the root, showing yellowish or brow nish egg masses attached to them. Washed roots show numerous small dark spots, each corresponding to an ootheca, which is adhering to a mature female. A curious abnormal female of M. i. acrita was obtained from a sample of squash roots. This female's body showed two globular parts, separated by a deep constriction. The convoluted ovaries were found to fill both portions of the body.

Young coffee plants (Coffea arabica L., var. Mundo Novo) were grown in nutrient solution purified from micronutrients contaminants by the method of MUNNS & JOHNSON (1960). All plants, except those in the control treatment, wer given all macronutrients and all micronutrients except one which was omitted in order to induce its shortage. Symptoms of deficiency were obtained for all known micronutrients but chlorine. Measurements, observations and chemical analysis of leaves allowed the following main conclusions to be drawn. 1. The relative influence of micronutrients in growth-measured by the fresh weight of the entire plant - was as follows: -Fe -Zn -Cu -Mo -Mn complete = -B = -CI. that is: the omission of iron from the nutrient solution caused the severest reduction in growth; lack of B and Cl had no effect. 2. Symptoms of deficiency of B, Fe, Mn, and Zn were found to be in good agreement with those in the literature. Effects of Cu and Mo shortage, however, had not been described so far: In the case of the Cu-deficient plants, the younger leaves were distorted, having an "S" shape, due probably to lack of growth of the veins; they lost their green color and developed rather large, necrotic patches near the margins. When molybdenum was omitted from the nutrient solution yellow spots develop near the margen of subterminal (fully mature) leaves; they became necrotic; there was a characteristic downward curling of the leaf blade along the mid rib so that the opposite edges touched each other underneath. 3. The levels of micronutrients found in normal and deficient leaves are given in Table 4. It is hoped that those values will serve as a basis of judgement of micronutrient contents found in leaves of field grown plants.

The Drosophila peruensis species group was recently proposed and includes four taxa: D. atalaia Vilela & Sene, 1982, D. boraceia Vilela & Val, 2004, D. pauliceia Ratcov & Vilela, 2007, and D. peruensis Wheeler, 1959. All these species have most of setae or setulae of mesonotum arinsing from dark spots, wings with crossveins darker (except in D. atalaia) and hypandrium squared-shaped mostly fused to gonopods. Here, we describe two new species, Drosophila itacorubi sp. nov. and Drosophila paraitacorubi sp. nov., belonging to this species group. The male genitalia of these species are figured. An identification key to the D. peruensis species group is provided.

ABSTRACT Cladodes illigeri (Kirby, 1818) is redescribed, and can be distinguished by the following features: color pattern overall black, paired spots and elytral margins pale yellow; pygidium bisinuate, posterior angles rounded, as long as median margin; and aedeagus with phallus 1/3 shorter than the parameres, which are sinuated apically. Cladodes lamellicornis (Motschulsky, 1854) is proposed as a junior synomym of C. illigeri. New records from the Atlantic Rainforest and illustrations for structural features are provided.

Hyla claresignata Lutz & Lutz, 1939, is a large species apparently not closely allied to the other known Brazilian hylas. It is characterized by the very small tympanum; the head is short and the snout rounded; the legs are long, the hands and feet unusually large, the latter extensively webbbed. The specific name is derived from the insular, irregular, or roughly triangular, dark spots, with a light halo, found mostly in the dorso-lateral region and on the legs. It belongs to the rain-forest fauna of the Marítime Range. The adult is a bromeliad-dweller and the tadpole rhyacophilous. DESCRIPTION. Vomerine teeth in two separate, oblique, groups, behind the large choanae, parallel to the posterior half of their inner border. Tongue entire, short, very broad and hardly free behind. Snout short, rounded, with distinct canthus rostralis and gradually sloping loreal region. Eye very large and prominent, its horizontal diameter almost equal to the distance between its anterior corner and the tip of the snout. Tympanum very small, less than one third of the diameter of the eye, but distinct, partly covered by a short, heavy ridge. Lateral fingers less than one third webbed; fourth finger slightly longer than the second, just reaching the base of the disk of the third; subarticular tubercles well developed; an angular pollex rudiment, more noticeable in the males. Toes almost completely webbed, the edge of the web inserted at the base of the disk on the third and the fifth; an inner metatarsal tubercle. Skin smooth above, granular beneath, on the throat minutely so. No dermal appendage on the hell. Habit robust, head broader than long, body rather heavy, slightly narrowed in the postaxillary region. Legs long, the tibiotarsal articulation reaching beyond the tip of the snout when adpressed. Type (female): 61 mm. (Fig. 1.) DIAGNOSIS of TADPOLE (by G. Orton). "A large specialized, mountain-stream tadpole, with wide head an elongated, flattened snout, greatly enlarged lips and high tooth formula. Eyes dorsal. Spiracle sinistral, projecting, situated far back on side. Anus dextral. Tooth formula 8/12 to 9/14 in fully grown larvae. Tail with a prominent, vertical dark band across musculature and fins; a second concentration of dark pigment near tip of tail, may or may not form a similar but narrower band. In this paper, based on the study of the materials of the collections belonging to Dr. C. A. CAMPOS SEABRA and the author's own, the author gives the additional characters of the rare species Volxemia dianella Lam., Sthelenus braconimus Bates and Sth. ichneumoneus Buq., the redescription of the genus Xenocrasis bates, the descriptions of allotypes of the males of Brasilianus testaceicornis melz., Xenocrasis badeni Bates and Oncideres castanea Dill. & Dill., and the key for the determination of the species Colobothea Serv. with 6 (8) pale spots on the elytra. All these species are from Brazil.

Estudi elaborat a partir d’una estada al Paul Scherrer Institut del Maig a l’Octubre del 2006 amb l’ajuda i supervisió dels Dr. Konstantins Jefimovs i Dr. Christian David. Focalitzar raigs X tous és una necessitat essencial per al microanàlisis, la microscopia, i fer imatges en moltes Instal·lacions de Radiació Sincrotró. Les Lents Zonals de Fresnel (FZP, de la denominació anglesa “Fresnel Zone Plates”) han demostrat donar uns punts focals amb una resolució espacial destacada i una baixa il·luminació de fons. Tanmateix, la fabricació de FZP és complexa i no totalment reproduïble. A més a més, el temps de vida de les FZP és força curt, ja que estant situades sobre membranes de nitrur de silici molt fines i altament absorbents. Per tant, hem fet esforços per implementar FZP de silici, que s’espera que siguin més resistents. L’element està fet d’una oblia de cristall de silici poc absorbent, i no presenta cap interfase entre materials. Així doncs, aquestes lents són especialment adequades per a aguantar les extremes càrregues de radiació de les fonts de raigs X més brillants. Particularment, això és molt important per a les aplicacions a les pròximes generacions de fonts de raigs X, com els Làsers d’Electrons Lliures (FEL, de la denominació anglesa “Free Electron Laser”). El silici també garanteix que no hi hagi cap banda d’absorció en el rang d’energies de la finestra de l’aigua (200-520 eV), fent aquestes lents ideals per a fer imatges de mostres biològiques. En aquest informe, hi ha una descripció detallada de tots els passos involucrats en la fabricació de les Lents Zonals de Fresnel de silici. En resum, les estructures de FZP es modelen sobre una resina utilitzant litografia per feix d’electrons i llavors el patró es transmet al silici mitjançant un gravat d’ions reactius (RIE, de la denominació anglesa ‘Reactive Ion Etching’) utilitzant una fina (20 nm) màscara de Crintermitja. Les membranes de silici es poden aprimar després de la fabricació de les estructures per a garantir una transmissió suficient fins i tot a baixes energies. Aquest informe també inclou l’anàlisi i la discussió d’alguns experiments preliminars per avaluar el rendiment de les Si FZPs fets a la línia de llum PolLux del Swiss Ligth Source amb l’ajuda dels Dr. Jörg Raabe i Dr. George Tzvetkov.

The males of the sandfly Lutzomyia longipalpis occur in two forms, one which bears a single pair of pale spots on tergite 4 and another in which an additional pair of spots characterizes tergite 3. In crosses between laboratory reared stocks of the two forms originating from allopatric and sympatric sites in Brazil nearly all males of one form fail to inseminate females of the other. In addition, insemination failure between some allopatric populaytions of Lu. longipalpis with similar tergal spot patterns is recorded, indicating the existence of additional forms in an apparent species complex. The possibility that Lu. longipalpis sensu latu represents more than a single taxon is discussed and the relevance of these findings to future epidemiological studies on kala-azar is considered.

The males of the sandfly Lutzomyia longipalpis occur in two forms, one which bears a single pair of pale spots on tergite 4 and another in which an additional pair of spots characterizes tergite 3. Almost complete reproductive isolation between sumpatric populations of the two forms and between allopatric populations of the one-spot form has been reported (Ward et al., 1983). Micromorphological differences in cuticular structures on the spots have also been observed and may be sites of pheromone release (Lane & Ward, 1984). Detais of the known distribution of the two are based on the examination of new specimens captured in Brazil during 1982, as well as museum collections and specimens from other laboratories. Males with a single pair of pale spots have been found from Mexico to southern Brazil, whilst the two-spot form is found only from the state of Maranhão in North Brazil to Minas Gerais and the border with Paraguay. In the eastern area of Brazil the two forms occur sympatrically in some locations and separately in others.

Ornament expression fluctuates with age in many organisms. Whether these changes are adaptively plastic is poorly known. In order to understand the ultimate function of melanin-based ornaments, we studied their within-individual fluctuations and their covariation with fitness-related traits. In barn owls (Tyto alba), individuals vary from reddish-brown pheomelanic to white and from immaculate to marked with black eumelanic spots, males being less reddish and less spotted than females. During the first molt, both sexes became less pheomelanic, females displayed larger spots and males fewer spots, but the extent of these changes was not associated with reproduction. At subsequent molts, intra-individual changes in melanin-based traits covaried with simultaneous reproduction changes. Adult females bred earlier in the season and laid larger eggs when they became scattered with larger spots, while adults of both sexes produced larger broods when they became whiter. These results suggest that the production of melanin pigments and fitness-related life history traits are concomitantly regulated in a sex-specific way.

BACKGROUND: Intra-specific variation in melanocyte pigmentation, common in the animal kingdom, has caught the eye of naturalists and biologists for centuries. In vertebrates, dark, eumelanin pigmentation is often genetically determined and associated with various behavioral and physiological traits, suggesting that the genes involved in melanism have far reaching pleiotropic effects. The mechanisms linking these traits remain poorly understood, and the potential involvement of developmental processes occurring in the brain early in life has not been investigated. We examined the ontogeny of rapid eye movement (REM) sleep, a state involved in brain development, in a wild population of barn owls (Tyto alba) exhibiting inter-individual variation in melanism and covarying traits. In addition to sleep, we measured melanistic feather spots and the expression of a gene in the feather follicles implicated in melanism (PCSK2). RESULTS: As in mammals, REM sleep declined with age across a period of brain development in owlets. In addition, inter-individual variation in REM sleep around this developmental trajectory was predicted by variation in PCSK2 expression in the feather follicles, with individuals expressing higher levels exhibiting a more precocial pattern characterized by less REM sleep. Finally, PCSK2 expression was positively correlated with feather spotting. CONCLUSIONS: We demonstrate that the pace of brain development, as reflected in age-related changes in REM sleep, covaries with the peripheral activation of the melanocortin system. Given its role in brain development, variation in nestling REM sleep may lead to variation in adult brain organization, and thereby contribute to the behavioral and physiological differences observed between adults expressing different degrees of melanism.

The hypothesis that extravagant ornaments signal parasite resistance has received support in several species for ornamented males but more rarely for ornamented females. However, recent theories have proposed that females should often be under sexual selection, and therefore females may signal the heritable capacity to resist parasites. We investigated this hypothesis in the socially monogamous barn owl, Tyto alba, in which females exhibit on average more and larger black spots on the plumage than males, and in which males were suggested to choose a mate with respect to female plumage spottiness. We hypothesized that the proportion of the plumage surface covered by black spots signals parasite resistance. In line with this hypothesis, we found that the ectoparasitic fly, Carnus hemapterus, was less abundant on young raised by more heavily spotted females and those flies were less fecund. In an experiment, where entire clutches were cross-fostered between nests, we found that the fecundity of the flies collected on nestlings was negatively correlated with the genetic mother's plumage spottiness. These results suggest that the ability to resist parasites covaries with the extent of female plumage spottiness. Among females collected dead along roads, those with a lot of black spots had a small bursa of Fabricius. Given that parasites bigger the development of this immune organ, this observation further suggests that more spotted females are usually less parasitized. The same analyses performed on male plumage spottiness all provided non-significant results. To our knowledge, this study is the first one showing that a heritable secondary sexual characteristics displayed by females reflects parasite resistance.

Die Differenzierung von Tinten erweist sich oft als wichtig in der Echtheitsprüfung von Dokumenten. Sie wird üblicherweise durch optische Vergleiche und Dünnschicht Chromatographie durchgeführt (TLC). Laser Desorption Ionisation Massenspektrometrie (LDI-MS) ist auch als nützlich gefunden worden und besonders leistungsfähig um Farbstoffe aus Kugelschreibertinte zu analysieren. Diese analytische Methode ist mit Hochleistungs Dünnschichtchromatografie TLC (HPTLC) verglichen worden, mit dem Ziel deren Tinten-Differenzierungskapazität zu testen. Tinteneinträge von 31 blauen Kugelschreibern sind analysiert worden und gemäß deren Farbstoffzusammensetzung klassifiziert worden. Typische Farbstoffe sind durch beide Methoden identifiziert worden und mehrere sind in vielen Tinten-Zusammensetzungen gefunden worden. LDI-MS ist leistungsfähiger als HPTLC um Tinten zu differenzieren, weil es Informationen über Farbstoffstrukturen (Molekular Gewicht) enthält und eine präzise relative Quantifizierung (Signalfläche) erlaubt. Dazu ist für LDI-MS Proben die Vorbereitung minimal und die Analysezeit kurz im Vergleich zu HPTLC mehr komplexen Schritte, wie Extraktionen, Spots Applikationen und Lösungsmittelelution. Allerdings sind mit LDI-MS zwei Analysen nötig um kationische und anionische Farbstoffe zu analysieren, während mit HPTLC nur eine Analyse nötig ist.

Metalworking fluid-associated hypersensitivity pneumonitis (MWF-HP) is a pulmonary disease caused by inhaling microorganisms present in the metalworking fluids used in the industrial sector. Mycobacterium immunogenum is the main etiological agent. Among the clinical, radiological and biological tools used for diagnosis, serological tests are important. The aim of this study was to identify immunogenic proteins in M. immunogenum and to use recombinant antigens for serological diagnosis of MWF-HP. Immunogenic proteins were detected by two-dimensional Western blot and candidate proteins were identified by mass spectrometry. Recombinant antigens were expressed in Escherichia coli and tested by enzyme-linked immunosorbent assay (ELISA) with the sera of 14 subjects with MWF-HP and 12 asymptomatic controls exposed to M. immunogenum. From the 350 spots visualized by two-dimensional gel electrophoresis with M. immunogenum extract, 6 immunogenic proteins were selected to be expressed as recombinant antigens. Acyl-CoA dehydrogenase antigen allowed for the best discrimination of MWF-HP cases against controls with an area under the receiver operating characteristics (ROC) curve of 0.930 (95% CI=0.820-1), a sensitivity of 100% and a specificity of 83% for the optimum threshold. Other recombinant antigens correspond to acyl-CoA dehydrogenase FadE, cytosol aminopeptidase, dihydrolipoyl dehydrogenase, serine hydroxymethyltransferase and superoxide dismutase. This is the first time that recombinant antigens have been used for the serodiagnosis of hypersensitivity pneumonitis. The availability of recombinant antigens makes it possible to develop standardized serological tests which in turn could simplify diagnosis, thus making it less invasive.

Trade-offs between the benefits of current reproduction and the costs to future reproduction and survival are widely recognized. However, such trade-offs might only be detected when resources become limited to the point where investment in one activity jeopardizes investment in others. The resolution of the trade-off between reproduction and self-maintenance is mediated by hormones such as glucocorticoids which direct behaviour and physiology towards self-maintenance under stressful situations. We investigated this trade-off in male and female barn owls in relation to the degree of heritable melanin-based coloration, a trait that reflects the ability to cope with various sources of stress in nestlings. We increased circulating corticosterone in breeding adults by implanting a corticosterone-releasing-pellet, using birds implanted with a placebo-pellet as controls. In males, elevated corticosterone reduced the activity (i.e. reduced home-range size and distance covered within the home-range) independently of coloration, while we could not detect any effect on hunting efficiency. The effect of experimentally elevated corticosterone on female behaviour was correlated with their melanin-based coloration. Corticosterone (cort-) induced an increase in brooding behaviour in small-spotted females, while this hormone had no detectable effect in large-spotted females. Cort-females with small eumelanic spots showed the normal body-mass loss during the early nestling period, while large spotted cort-females did not lose body mass. This indicates that corticosterone induced a shift towards self-maintenance in males independently on their plumage, whereas in females this shift was observed only in large-spotted females.
