Endo-oligopeptidase A, EC 3.4.22.19, converts small enkephalin-containing peptides into the corresponding enkephalins in vitro. We investigated the presence of endooligopeptidase A in the retina and its possible colocalization with enkephalins in retinal neurons. The specific activity of endo-oligopeptidase A found in pigeon retinae (30.3 +/- 7.3 mU/mg, mean +/- standard deviation) was four times higher than in rabbit retinae (7.0 +/- 1.1 mU/mg). The enzyme activity was not modified by EDTA, but it was enhanced by dithiothreitol and inhibited by zinc and 5,5'-dithiobis(2-nitrobenzoic acid). Immunohistochemical experiments with a purified antiserum against rabbit endo-oligopeptidase A revealed labeled neurons in both the inner nuclear layer and the ganglion cell layer of pigeon and rabbit retinae. Double-labeling immunofluorescence experiments demonstrated that about 90% of neurons containing endo-oligopeptidase A-like immunoreactivity also contained [Leu5]-enkephalin-like immunoreactivity. These colocalization results may represent an important step toward the demonstration of the possible involvement of endo-oligopeptidase A in enkephalin generation in vivo.

Two assays were accomplished being, both using a randomized block design: Assay 1 the aimed was to evaluate the blood parameters of weaned recently pigs and receiving rations with and without probiotic. Twenty pigs with body weight of 6.99 +/- 0.42 kg were used and were collected samples of blood to the 0, 7, 14, 21 and 28 days after wean, for determinations of the bool parameters. Assay 2 the objective was to determine the rations digestibility for growth swine supplemented or not with probiotic in the rations. The rations and the ages of the animals influenced most of the variables of the blood parameters. The probiotic didn't alter the ration digestibility to exception for mineral matter. It is ended that the blood parameters were altered for the use of the probiotic and the age of the animals, both emphasized to challenges happened after wean and the mineral matter was better taken advantage with the use of the probiotic.

This study aims to evaluate structurally a heat exchanger that has been in operation since the mid-60's and was built COSAN for operation in one of its sugar production plants in São Paulo, this equipment as well as many similar devices are operation in plants throughout Brazil, and thus to be acquired by large corporations as in the case of COSAN, pass the scrutiny of inspection work, and work such as recalculations in the latter case in structural evaluations to verify the possible need for adjustments to project or operation and thus ensure the structural integrity of the equipment. To this were first done field survey using techniques of NDT and NDT's for determining the thickness of the main parts of the equipment, made the revaluation dimensional and existing project and considering the loads operating performing the characterization of the equipment as specified in the standard regulatory number 13 - NR13 and the other for this type of equipment and finally an analysis using a static approach, as an analysis tool using the finite element method

There are few reports in the literature of the absence of Wharton's Jelly. Here we report the seventh case in a primigravida, 22 years old, admitted after vaginal delivery of stillborn. The umbilical cord have a long segment with disruption of cord structures and the three blood vessels were completely separated from each other, with a minimum amount of Wharton's jelly remaining around each vessel. The absence of Wharton' jelly is associated with fetal distress, intrauterine growth restriction, and fetal death. Quantitative/qualitative studies of Wharton's jelly represent an open field of research for possible correlations with obstetric conditions and fetal deaths.

The production systems on substrates have been employed in most commercial cultivation of flowers and ornamental plants, including gerbera. However, its introduction as potted flower is recent in Brazil and many studies, especially those related to the determination of substrates physical and chemical properties, which contribute to production quality are still needed. The present study aimed to assess the influence of substrates characteristics for nutrient absorption and production of potted gerbera. The experiment was carried out in greenhouse with the experimental design in randomized blocks, factorial arrangement 5x2 (5 substrates and 2 cultivars) and 4 replications. Treatments consisted of substrates with different physical and chemical characteristics and gerbera cultivars (Cherry and Red). Plants grown on substrate with pH above 7.0 had a reduction in iron absorption, resulting in lower intensity of green coloration in the leaves. Plants grown on substrate with pH below 5.0 showed toxic levels of manganese and lower dry mass. The characteristics of the substrate, especially the pH, influence the nutrients absorption and the production of potted gerbera, altering the final plant quality.

The feeding diets were evaluated containing low and high levels of soybean oil for horses athletes subjected to two protocols of aerobic training on the response of basal blood biochemical parameters. Four horses were used in latin square design with treatments in a 2 x 2 factorial arrangement. Treatments consisted levels of 5 and 15% oil concentrates and two aerobic training, 40' and 60' minutes. Plasmatic parameters were monitored, triglyceride (TG), total cholesterol (TC), glucose (GLU) and lactate (LAC), during basal metabolism. The TG, TC, GLU and LAC from horses at rest were not affected (P> 0.05) neither of diet and physical activity, 0.21, 3.79, 4.18, 0.93 mmol L-1, respectively. It can be concluded that offer concentrate with high content of soybean oil to athletic horses in aerobic activities can be performed without altering the blood biochemical profile of basal metabolism.

Fertigation management of gerbera crop has been many times performed inadequately, and it has been worsened when mixtures of substrates with different physical and chemical characteristics are used. Aiming at evaluating the production and quality of potted gerbera in two substrates and different levels of fertigation, the experiment was conducted in the greenhouse of the DRN/Soil Science, FCA/UNESP, Botucatu (SP). A 5 x 2 factorial randomized block design (5 levels of fertigation and 2 substrates) was adopted with 4 replications. Levels of fertigation corresponded to maintenance of 100% available water (AW) in the substrate; 100 to 80% of AW; 100 to 60% of AW; 100 to 40% of AW and 100 to 20% of AW. The substrates were as follows: 1- mixed coconut fiber (50% pellet coconut fiber and 50% coir fiber), 2- 40% red soil, 40% decomposed pine bark, 10% composition 1 (40% decomposed pine bark, 30% vermiculite and 30% carbonized rice husk) and 10% composition 2 (75% decomposed pine bark and 25% needles of pine). Plants at the marketing stage were evaluated according to the number of leaves; diameter of leaf surface; leaf area; fresh and dry phytomass of leaves, inflorescence and total plant; inflorescence number and diameter, stem diameter, plant height, leaf area rate and electrical conductivity of the substrate solution. Plants of better quality were obtained when they were maintained in levels of 100% available water and the mixed coconut fiber was used as substrate.

The turtle retina has been extensively used for the study of chromatic processing mechanisms. Color opponency has been previously investigated with trichromatic paradigms, but behavioral studies show that the turtle has ail ultraviolet (UV) channel and a tetrachromatic visual system. Our laboratory has been working ill the characterization of neuronal responses in the retina of vertebrates using stimuli in the UV-visible range of the electromagnetic spectrum. In the present investigation, we recorded color-opponent responses from turtle amacrine and ganglion cells to UV and visible stimuli and extended our previous results that UV color-opponency is present at the level of the inner nuclear layer. We recorded from 181 neurons, 36 of which were spectrally opponent. Among these, there were 10 amacrine (5%), and 26 ganglion cells (15%). Morphological identification of color-opponent neurons was possible for two ganglion cell classes (G17 and G22) and two amacrine cell classes (A22 and A23b). There was a variety of cell response types and a potential for complex processing of chromatic stimuli, with intensity- and wavelength-dependent response components. Ten types of color opponency were found in ganglion cells and by adding previous results from our laboratory, 12 types of opponent responses have been found. The majority of the ganglion cells were R+UVBG- and RG+UVB-color-opponents but there were other less frequent types of chromatic opponency. This study confirms the participation of a UV channel in the processing of color opponency in the turtle inner retina and shows that the turtle visual system has the retinal mechanisms to allow many possible chromatic combinations.

Many animal species make use of ultraviolet (UV) light in a number of behaviors, such as feeding and mating. The goldfish (Carassius auratus) is among those with a UV photoreceptor and pronounced UV sensitivity. Little is known, however, about the retinal processing of this input. We addressed this issue by recording intracellularly from second-order neurons in the adult goldfish retina. In order to test whether cone-driven horizontal cells (HCs) receive UV cone inputs, we performed chromatic adaptation experiments with mono- and biphasic HCs. We found no functional evidence of a projection from the UV-sensitive cones to these neurons in adult animals. This suggests that goldfish UV receptors may contact preferentially triphasic HCs, which is at odds with the hypothesis that all cones contact all cone-driven HC types. However, we did find evidence of direct M-cone input to monophasic HCs, favoring the idea that cone-HC contacts are more promiscuous than originally proposed. Together, our results suggest that either UV cones have a more restricted set of post-synaptic partners than the other three cone types, or that the UV input to mono- and biphasic HCs is not very pronounced in adult animals.

Oxidative stress and mitochondrial impairment are essential in the ischemic stroke cascade and eventually lead to tissue injury. C-Phycocyanin (C-PC) has previously been shown to have strong antioxidant and neuroprotective actions. In the present study, we assessed the effects of C-PC on oxidative injury induced by tert-butylhydroperoxide (t-BOOH) in SH-SY5Y neuronal cells, on transient ischemia in rat retinas, and in the calcium/phosphate-induced impairment of isolated rat brain mitochondria (RBM). In SH-SY5Y cells, t-BOOH induced a significant reduction of cell viability as assessed by an MTT assay, and the reduction was effectively prevented by treatment with C-PC in the low micromolar concentration range. Transient ischemia in rat retinas was induced by increasing the intraocular pressure to 120 mmHg for 45 min, which was followed by 15 min of reperfusion. This event resulted in a cell density reduction to lower than 50% in the inner nuclear layer (INL), which was significantly prevented by the intraocular pre-treatment with C-PC for 15 min. In the RBM exposed to 3 mM phosphate and/or 100 mu M Ca2+, C-PC prevented in the low micromolar concentration range, the mitochondrial permeability transition as assessed by mitochondrial swelling, the membrane potential dissipation, the increase of reactive oxygen species levels and the release of the pro-apoptotic cytochrome c. In addition, C-PC displayed a strong inhibitory effect against an electrochemically-generated Fenton reaction. Therefore, C-PC is a potential neuroprotective agent against ischemic stroke, resulting in reduced neuronal oxidative injury and the protection of mitochondria from impairment. (C) 2012 Elsevier Inc. The vertebrate retina has a very high dynamic range. This is due to the concerted action of its diverse cell types. Ganglion cells, which are the output cells of the retina, have to preserve this high dynamic range to convey it to higher brain areas. Experimental evidence shows that the firing response of ganglion cells is strongly correlated with their total dendritic area and only weakly correlated with their dendritic branching complexity. On the other hand, theoretical studies with simple neuron models claim that active and large dendritic trees enhance the dynamic range of single neurons. Theoretical models also claim that electrical coupling between ganglion cells via gap junctions enhances their collective dynamic range. In this work we use morphologically reconstructed multi-compartmental ganglion cell models to perform two studies. In the first study we investigate the relationship between single ganglion cell dynamic range and number of dendritic branches/total dendritic area for both active and passive dendrites. Our results support the claim that large and active dendrites enhance the dynamic range of a single ganglion cell and show that total dendritic area has stronger correlation with dynamic range than with number of dendritic branches. In the second study we investigate the dynamic range of a square array of ganglion cells with passive or active dendritic trees coupled with each other via dendrodendritic gap junctions. Our results suggest that electrical coupling between active dendritic trees enhances the dynamic range of the ganglion cell array in comparison with both the uncoupled case and the coupled case with cells with passive dendrites. The results from our detailed computational modeling studies suggest that the key properties of the ganglion cells that endow them with a large dynamic range are large and active dendritic trees and electrical coupling via gap junctions.

The visual system is particularly sensitive to methylmercury (MeHg) exposure and, therefore, provides a useful model for investigating the fundamental mechanisms that direct toxic effects. During a period of 70 days, adult of a freshwater fish species Hoplias malabaricus were fed with fish prey previously labeled with two different doses of methylmercury (0.075 and 0.75 mu g g(-1)) to determine the mercury distribution and morphological changes in the retina. Mercury deposits were found in the photoreceptor layer, in the inner plexiform layer and in the outer plexiform layer, demonstrating a dose-dependent bioaccumulation. The ultrastructure analysis of retina revealed a cellular deterioration in the photoreceptor layer, morphological changes in the inner and outer segments of rods, structural changes in the plasma membrane of rods and double cones, changes in the process of removal of membranous discs and a structural discontinuity. These results lead to the conclusion that methylmercury is able to cross the blood-retina barrier, accumulate in the cells and layers of retina and induce changes in photoreceptors of H. malabaricus even under subchronic exposure. Objetivo: o objetivo deste trabalho foi estudar os aspectos morfológicos e ultra-estruturais na gênese de capilares sanguíneos em músculo esquelético do membro caudal de ratos submetidos à isquemia sob a ação da Prostaglandina E1 (PGE1), administrada por via intramuscular ou endovenosa. Métodos: foram utilizados 60 ratos (Rattus norvegicus albinus), linhagem Wistar-UEM, distribuídos aleatoriamente em três grupos de 20, redistribuídos igualmente em dois subgrupos, observados no 7o e 14o dias, sendo um grupo controle onde apenas foi provocada a isquemia no membro, outro com a isquemia e a injeção da PGE1 via intramuscular (IM), e outro com a isquemia e a injeção da PGE1 endovenosa (EV). Para análise dos resultados, foram realizadas a coloração com hematoxilina & eosina (HE), a imuno-histoquímica e a microscopia eletrônica de transmissão (MET). Resultados: constatou-se um aumento estatisticamente significante no número de capilares nos subgrupos com o uso da PGE1 IM e EV, através da contagem nos cortes corados com HE. Houve marcação de capilares e vasos de maior calibre nestes mesmos subgrupos, porém, esta reação não foi eficiente para a quantificação dos capilares. Na MET encontraram-se evidências de formação de novos capilares. Conclusões: a PGE1, administrada por via IM ou EV, promoveu, após 14 dias de observação, um aumento no número de capilares no músculo esquelético de ratos submetido à isquemia, identificáveis histologicamente com a coloração em HE. Na análise ultra-estrutural encontraram-se alterações que sugerem, nos animais sob a ação da PGE1, que a neoformação vascular possa ter ocorrido por angiogênese e vasculogênese. A imuno-coloração, apesar da marcação de capilares e vasos maiores, não permitiu estabelecer uma correlação com o aumento de vasos encontrados na coloração com HE.
