Apis mellifera honeybees are social insects of economic importance, by providing honeybee products, and by the pollination of natural areas of vegetation or agricultural areas. The constant use of pesticides, including the thiamethoxam, which is an insecticide belonging to the class of neonicotinoids with neurotoxic action, is subjecting pollinators to situations of severe stress, which has been evidenced by the decrease in the density of honeybees in many parts of the world. By these considerations, the present study aimed to assess the acute toxicity of thiamethoxam for newly emerged workers of A. mellifera, and to investigate the effect of sublethal doses of this insecticide on the survival time and its cytotoxicity to the brain and midgut to the honeybees. It was established the value of the lethal concentration 50 (LC50) equal to 4.28 ng thiamethoxam/μL of food and from it, the sublethal concentrations of 0.428 ng thiamethoxam/μL (CL50/10) and 0.0428 ng thiamethoxam/μL (CL50/100), which were used in bioassays of intoxication of the honeybees. After the bioassays the bees were dissected and the brain and midguts were collected to analyze possible morphological (staining with Hematoxylin-Eosin) and histochemical alterations (Xylidine Ponceau technique, and Feulgen and PAS reactions) caused by exposure to thiamethoxam, and to calculate the lethal time (LT50) for the workers. The duration of the bioassays was 8 days after beginning of feeding. The results obtained showed that the thiamethoxam is toxic to newly-emerged workers of A. mellifera, causing changes in survival time of individuals. This study also shown that the thiamethoxam causes morphological and histochemical alterations on the midgut and brain of workers. These alterations may be reflected in physiological and behavioral changes that can modify the operation of the colony

The studies related to research on new antimicrobial products have received special attention from researchers, especially given the emergence of microbial strains resistant to conventional antimicrobials. Thus, the present study was aimed to test the antimicrobial action of hydro-alcoholic extracts of plants collected in Cerrado region of Botucatu, following the species: Achyrocline satureioides (Lam) DC (macela), Stryphnodendron adstringens (Mart) Coville (barbatimão), Miconia rubiginosa (Bonpl.) DC (quaresma-branca), Davilla elliptica A. St-Hil (lixinha), Siparuna guianensis (negramina) e Solanum lycocarpum A.St-Hil (lobeira). The plants were always collected in the morning, in areas near the town of Botucatu, and extracts were prepared using a solvent such as methanol 70% from materials dried (50°C) and ground into mill knives. The extraction was performed for 48 hours at refrigerator temperature, followed by filtration, removal of methanol solvent in a rotary evaporator, determination of the dry weight of the extracts (mg / mL) and phytochemical analysis of the same. The sensitivity tests for 10 S. aureus, 11 E. coli and 11 P. aeruginosa, isolated from human clinical cases were performed by diluting volumes of the extracts in Mueller Hinton Agar (MHA) and determination of minimum inhibitory concentration (MIC) (mg / mL). According to the results and statistical analysis, it was found that depending on the bacteria tested, and in descending order of antibacterial activity for S. aureus: Lixinha sheet > Barbatimão sheet > Quaresma-Branca > Macela > Lixinha fruit > Barbatimão shell > Lobeira > Negramina; E. coli: Lixinha sheet > Barbatimão sheet > Lixinha fruit = barbatimão peel > Quaresma-Branca > Macela = Lobeira > Negramina and P. aeruginosa: Lixinha leaf > Barbatimão bark > Barbatimão leaf > Lixinha fruit > Macela > Lobeira > Quaresma - Branca = Negramina... Coumarin is a natural active compound that can be found in many plants. The coumarins have many properties such as bronchodilator, anti-inflammatory, antioxidant, anticoagulant, antibiotics, immunomodulatory, antimicrobial and antiviral, thus, they are widely used in medical applications. More recently the coumarin derivatives have attracted the interest of many research groups in the field of new materials, for example the possibility of their use as sensitizers in dye-sensitized solar cells (DSSC) and lasers. The MCRs are defined as a process in which three or more reactants are combined in the same reaction pot, resulting in products with good structural complexity a single step, in addition to economy of atoms and selectivity and is a very important feature in modern synthetic methodology. In this work we investigated the use of niobium pentachloride as catalyst of the multicomponent reactions between phenolic derivatives, various aromatic aldehydes and β-diester derivatives in the synthesis of 4-aryl-3,4-dihydrocoumarin derivatives. The reactions were carried out at room temperature, under inert atmosphere (N2), using dichloromethane anhydrous (CH2 Cl2) as solvent, with a reaction time of most 120 hours. The products were isolated by column chromatography on silica gel and submitted to spectrometric and spectroscopic analysis. The results show that NbCl5 is an excellent agent for promoting the synthesis of 4-aryl-3,4-dihydrocoumarin derivatives through multicomponent reactions, obtaining yields varying from 45 to 95%

Objective: To compare the efficacy of the mouthwashes 0.12% chlorhexidine, Listerine, and 0.5% and 2% Melaleuca Alternifolia oil against the salivary levels of Streptococcus mutans and total microorganisms. Methods: This study was double-blind controlled and paired clinical assay. Twenty-six volunteers aged 21 to 35 years old were enrolled. At baseline, 1 mL of unstimulated saliva was collected from each subject, 1 and 15 min after mouthrinsing with the following solutions: sterile distilled water, 0.12% chlorhexidine digluconate, Listerine (©Johnson & Johnson do Brasil), 0.5% and 2% concentrations of Melaleuca Alternifolia (Sigma-Aldrich). The volunteers used all the evaluated mouthrinses with a 15-day interval between the solutions. Immediately after rinsing, saliva was collected and serial dilutions were performed, followed by plating in blood agar culture medium for growth of total microorganisms and SB-20 (Sucrose-Bacitracin agar) for growth of S. mutans, and incubation at 37 °C for 48 h in microaerophilia. After incubation, the number of colonies was counted and expressed as colony forming units (UFC/mL). Results: Chlorhexidine showed antimicrobial action by reducing total microorganisms and S. mutans, while the action of 0.5% Melaleuca Alternifolia was similar to that of distilled water. Listerine and 2% Melaleuca Alternifolia oil reduced total microbial counts by 11% and 9% respectively, and S. mutans by 20% and 11%. Conclusion: A single rinse with 0.12% chlorhexidine is effective in reducing the levels of total microorganisms and S. mutans present in saliva. Under the same testing conditions, Listerine and 0.5% and 2% Melaleuca Alternifolia oil presented lower efficacy than chlorhexidine.

The objective of this review was reporting an overview about the properties of two irrigating solutions, solution of sodium hypochlorite and chlorhexidine, used in the treatment of root canals. A solution must submit irrigating antimicrobial action, dissolve waste tissue, promote wetting to facilitate the instrumentation, and present biocompatibility with the adjacent tissues. The sodium hypochlorite used in different concentrations has been the solution of choice among professionals by presenting antimicrobial action and act as a solvent tissue. However, today, chlorhexidine has been widely used as an irrigating solution due to specific properties that enable its use, such as substantivity, antimicrobial effectiveness, and low toxicity. Thus the solution of chlorhexidine with different concentrations is presented as an alternative to irrigating solution for the treatment of root canals.

CR-LAAO is an l-amino acid oxidase from Calloselasma rhodostoma snake venom that has been broadly studied regarding its structural and biochemical characteristics, however, few studies have investigated its pharmacological effects. The present study aimed at the evaluation of the biotechnological potential of CR-LAAO by determining its bactericidal, antifungal, leishmanicidal and trypanocidal activity, as well as its cytotoxicity on human tumor and non-tumor cell lines. After 24h of preincubation, CR-LAAO showed bactericidal effects against both Staphylococcus aureus (MIC 0.78μg/mL) and Escherichia coli (MIC 31.25μg/mL) strains, inducing dismantle of bacterial cell walls. After 6h of preincubation with Candida albicans, CR-LAAO was able to inhibit 80% of the yeast growth, and it also showed cytotoxic activity on Leishmania species and Trypanosoma cruzi. Additionally, CR-LAAO showed high cytotoxicity on HepG2 and HL-60 tumor cells (IC50 10.78 and 1.7μg/mL), with lower effects on human mononuclear cells (PBMC). The cytotoxic effects of CR-LAAO were significantly inhibited in the presence of catalase, which suggests the involvement of hydrogen peroxide in its mechanisms of toxicity. Therefore, CR-LAAO showed promising pharmacological effects, and these results provide important information for the development of therapeutic strategies with directed action, such as more effective antimicrobial agents.

To assess the fluoride (F) content in commercially available milk formulae in Brazil and to estimate the F intake in children from this source in the first year of life. Samples of cow's milk (n = 51), infant formulae (n = 15), powdered milk (n = 13), and soy-based products (n = 4) purchased in Araçatuba (Brazil) had their F content measured using an ion-specific electrode, after hexamethyldisiloxane-facilitated diffusion. Powdered milk and infant formulae were reconstituted with deionized water, while ready-to-drink products were analyzed without any dilution. Using average infant body masses and suggested volumes of formula consumption for infants 1-12 months of age, possible F ingestion per body mass was estimated. Data were analyzed by descriptive analysis. Mean F content ranged from 0.02 to 2.52 mg/L in all samples. None of the cow's milk provided F intake higher than 0.07 mg/kg. However, two infant formulae, one powdered milk, and one soy-milk led to a daily F intake above the suggested threshold for fluorosis when reconstituted with deionized water. Assuming reconstitution of products with tap water at 0.7 ppm F, two infant formulae, five powdered milk, and four soymilks led to daily F intake ranging from 0.108 to 0.851 mg/kg. The results suggest that the consumption of some brands of infant formulae, powdered milk, and soy-based milk in the first year of age could increase the risk of dental fluorosis, reinforcing the need for periodic surveillance of the F content of foods and beverages typically consumed by young children.

Objective: The use of Er:YAG laser operating in the 3 μm range with adjustable power and pulses has become popular for dental and medical practice due to its high photoablative capacity, surgical precision and antimicrobial action. Background data: The existing fiberoptic tips irradiate lasers parallel to the long axes of the tooth limiting its efficiency in the root canal. Methods: We evaluated hollow fiberoptic tips obtained from silicate glass as a means of Er:YAG laser conduction in dental procedures. The fiber tips were molded from capillary tubes with different profiles so that their ends would have cylindric, conical or spherical shapes. The performance of the three fibers as a means of propagation of Er:YAG (λ = 2.94 μm) laser radiation was compared to that of a solid sapphire fiber at 10 Hz and 200 mJ and of 20 Hz and 500 mJ. The profiles of frontal and lateral burning were visualized on thermal paper. Results: Analysis of these profiles demonstrated that the sapphire tip and the hollow fiber of cylindric section did not differ significantly in the profiles of frontal burning, and no lateral burning was detected. The fibers of the conical and spherical sections, although presenting attenuation in the frontal output power, showed a larger burning area in the frontal profile, in addition to producing lateral burning. Conclusions: The results indicate that commercial hollow fiberoptics have advantages such as easy manufacture of the different tip shapes, great adaptability, low cost, and a low loss of transmission. © Mary Ann Liebert, Inc.
