Multiple genetic variants have been associated with adult obesity and a few with severe obesity in childhood; however, less progress has been made in establishing genetic influences on common early-onset obesity. We performed a North American, Australian and European collaborative meta-analysis of 14 studies consisting of 5,530 cases (≥95th percentile of body mass index (BMI)) and 8,318 controls (<50th percentile of BMI) of European ancestry. Taking forward the eight newly discovered signals yielding association with P < 5 × 10(-6) in nine independent data sets (2,818 cases and 4,083 controls), we observed two loci that yielded genome-wide significant combined P values near OLFM4 at 13q14 (rs9568856; P = 1.82 × 10(-9); odds ratio (OR) = 1.22) and within HOXB5 at 17q21 (rs9299; P = 3.54 × 10(-9); OR = 1.14). Both loci continued to show association when two extreme childhood obesity cohorts were included (2,214 cases and 2,674 controls). These two loci also yielded directionally consistent associations in a previous meta-analysis of adult BMI(1).

Background
Because students may purchase food and drinks in and around their schools, the school food environment may be important for obesity-related eating behaviors such as soft drink and snack consumption. However, research exploring the associations between school environments and specific eating behaviors is sparse.

Methods
Associations of the availability of canteen food and drinks, the presence of food stores around schools, and individual cognitions (attitudes, norms, modeling, perceived behavioral control, and intentions) with soft drink and snack consumption were examined in a cross-sectional study (2005–2006) among 1293 adolescents aged 12–15 years. Soft drink and snack consumption and related cognitions were assessed with self-administered questionnaires. The presence of food stores and the distance to the nearest food store were calculated within a 500-meter buffer around each school. Data on the availability of soft drinks and snacks in school canteens were gathered by observation. In 2007, multilevel regression models were run to analyze associations and mediation pathways between cognitions, environmental factors, and behaviors.

Results
Adolescents' attitudes, subjective norms, parental and peer modeling, and intentions were positively associated with soft drink and snack consumption. There was an inverse association between the distance to the nearest store and the number of small food stores with soft drink consumption. These effects were mediated partly by cognitions.

Conclusions
This study provided little evidence for associations of environmental factors in the school environment with soft drink and snack consumption. Individual cognitions appeared to be stronger correlates of intake than physical school-environmental factors. Longitudinal research is needed to confirm these findings.

Although the recent history of human colonisation and impact on Mauritius is well documented, virtually no records of the pre-human native ecosystem exist, making it difficult to assess the magnitude of the changes brought about by human settlement. Here, we describe a 4000-year-old fossil bed at Mare aux Songes (MAS) in south-eastern Mauritius that contains both macrofossils (vertebrate fauna, gastropods, insects and flora) and microfossils (diatoms, pollen, spores and phytoliths). With >250 bone fragments/m2 and comprising 50% of all known extinct and extant vertebrate species (ns = 44) of Mauritius, MAS may constitute the first Holocene vertebrate bone Concentration-Lagerstätte identified on an oceanic volcanic island. Fossil remains are dominated by extinct giant tortoises Cylindraspis spp. (63%), passerines (10%), small bats (7.8%) and dodo Raphus cucullatus (7.1%). Twelve radiocarbon ages [four of them duplicates] from bones and other material suggest that accumulation of fossils took place within several centuries. An exceptional combination of abiotic conditions led to preservation of bones, bone collagen, plant tissue and microfossils. Although bone collagen is well preserved, DNA from dodo and other Mauritian vertebrates has proved difficult. Our analysis suggests that from ca 4000 years ago (4 ka), rising sea levels created a freshwater lake at MAS, generating an oasis in an otherwise dry environment which attracted a diverse vertebrate fauna. Subsequent aridification in the south-west Indian Ocean region may have increased carcass accumulation during droughts, contributing to the exceptionally high fossil concentration. The abundance of floral and faunal remains in this Lagerstätte offers a unique opportunity to reconstruct a pre-human ecosystem on an oceanic island, providing a key foundation for assessing the vulnerability of island ecosystems to human impact.

Objective. To explore the association between home environmental variables and television (TV) time, and the mediating pathways underlying this association.

Methods. The current study used data from the longitudinal ENDORSE study. Self-reported data was available for 1 265 adolescents (mean age of 12–15 years at baseline) on home environment (availability of a TV in the bedroom, perceived parental modelling, family rules), potential mediators (intention, attitude, perceived behavioural control, subjective norm towards TV viewing) and TV viewing time. Mediation analyses were conducted using General Estimating Equations and mediation effects were calculated as the product-of-coefficients.

Results.
Significant overall positive associations were found for the presence of a TV in the bedroom and parental modelling with self-reported TV viewing. Controlling family rules showed an inverse association with reported TV time. Similarly, parental modelling and a TV in the bedroom were significantly positively associated with the Theory of Planned Behaviour variables and habit strength, while family rules showed an inverse association with these potential mediators. In turn, most potential mediators were positively associated with TV viewing. Intention, attitude and habit strength were the strongest mediators in all three associations explaining more than 55% of the overall association. Habit strength alone explained 38.2%–58.0% of the overall associations.

Conclusions. Home and family environmental predictors of TV time among adolescents may be strongly mediated by habit strength and other personal factors. Future intervention studies should explore if changes in home and family environments indeed lead to reductions in TV time through these mediators.

Objective. To examine associations between social ecological factors and Dutch adolescents’ TV viewing. Design. Cross-sectional examination of predictors of adolescents’ TV viewing.

Participants. A total of 338 adolescents, aged 14 years (55% boys).

Measurements. Adolescents self-reported their age, ethnicity and TV viewing (dichotomized at two hours/day) and responded to items from all three social ecological domains; individual (cognitions based on the Theory of Planned Behaviour and TV viewing habit strength, and other behaviours, such as computer use), social (parental rules about TV viewing and parental TV viewing behavior) and physical environmental factors (TV in bedroom, physical activity equipment available). Parents reported demographic factors (e.g., ethnicity, education level), and their own TV viewing (mins/day); adolescents’ weight status (not overweight vs. overweight/obese) was calculated from objective measures of height and weight. Logistic regression analyses examined associations between socio-ecological factors and adolescents’ TV viewing, and whether associations were moderated by adolescents’ sex, parents’ education and ethnicity.

Results. Compared with others, overweight/obese adolescents (odds ratio (OR)=3.0; p≤0.001), those with high computer use (OR=2.3; p≤0.0001), with high TV viewing habit strength (OR=1.3; p≤0.0001), and those whose parents had high levels of TV viewing (OR=2.4; p≤0.01) were more likely to exceed two hours of TV viewing per day. The association with habit strength was moderated by gender, and the association with parents’ TV viewing was moderated by parents’ education and ethnicity.

Conclusions. Interventions should target parents’ TV viewing behaviors and aim to amend habitual, ‘mindless’ TV viewing among adolescents.

Aicardi-Goutières syndrome (AGS) is a genetic encephalopathy whose clinical features mimic those of acquired in utero viral infection. AGS exhibits locus heterogeneity, with mutations identified in genes encoding the 3′→5′ exonuclease TREX1 and the three subunits of the RNASEH2 endonuclease complex. To define the molecular spectrum of AGS, we performed mutation screening in patients, from 127 pedigrees, with a clinical diagnosis of the disease. Biallelic mutations in TREX1, RNASEH2A, RNASEH2B, and RNASEH2C were observed in 31, 3, 47, and 18 families, respectively. In five families, we identified an RNASEH2A or RNASEH2B mutation on one allele only. In one child, the disease occurred because of a de novo heterozygous TREX1 mutation. In 22 families, no mutations were found. Null mutations were common in TREX1, although a specific missense mutation was observed frequently in patients from northern Europe. Almost all mutations in RNASEH2A, RNASEH2B, and RNASEH2C were missense. We identified an RNASEH2C founder mutation in 13 Pakistani families. We also collected clinical data from 123 mutation-positive patients. Two clinical presentations could be delineated: an early-onset neonatal form, highly reminiscent of congenital infection seen particularly with TREX1 mutations, and a later-onset presentation, sometimes occurring after several months of normal development and occasionally associated with remarkably preserved neurological function, most frequently due to RNASEH2B mutations. Mortality was correlated with genotype; 34.3% of patients with TREX1, RNASEH2A, and RNASEH2C mutations versus 8.0% RNASEH2B mutation-positive patients were known to have died (P = .001). Our analysis defines the phenotypic spectrum of AGS and suggests a coherent mutation-screening strategy in this heterogeneous disorder. We present measurements of Underlying Event observables in pp collisions at root s = 0 : 9 and 7 TeV. The analysis is performed as a function of the highest charged-particle transverse momentum p(T),L-T in the event. Different regions are defined with respect to the azimuthal direction of the leading (highest transverse momentum) track: Toward, Transverse and Away. The Toward and Away regions collect the fragmentation products of the hardest partonic interaction. The Transverse region is expected to be most sensitive to the Underlying Event activity. The study is performed with charged particles above three different p(T) thresholds: 0.15, 0.5 and 1.0 GeV/c. In the Transverse region we observe an increase in the multiplicity of a factor 2-3 between the lower and higher collision energies, depending on the track p(T) threshold considered. Data are compared to PYTHIA 6.4, PYTHIA 8.1 and PHOJET. On average, all models considered underestimate the multiplicity and summed p(T) in the Transverse region by about 10-30%.

The ALICE experiment at the LHC has studied J/psi production at mid-rapidity in pp collisions at root s = 7 TeV through its electron pair decay on a data sample corresponding to an integrated luminosity L-int = 5.6 nb(-1). The fraction of J/psi from the decay of long-lived beauty hadrons was determined for J/psi candidates with transverse momentum p(t) > 1,3 GeV/c and rapidity vertical bar y vertical bar < 0.9. The cross section for prompt J/psi mesons, i.e. directly produced J/psi and prompt decays of heavier charmonium states such as the psi(2S) and chi(c) resonances, is sigma(prompt J/psi) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 8.3 +/- 0.8(stat.) +/- 1.1 (syst.)(-1.4)(+1.5) (syst. pol.) mu b. The cross section for the production of b-hadrons decaying to J/psi with p(t) > 1.3 GeV/c and vertical bar y vertical bar < 0.9 is a sigma(J/psi <- hB) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 1.46 +/- 0.38 (stat.)(-0.32)(+0.26) (syst.) mu b. The results are compared to QCD model predictions. The shape of the p(t) and y distributions of b-quarks predicted by perturbative QCD model calculations are used to extrapolate the measured cross section to derive the b (b) over bar pair total cross section and d sigma/dy at mid-rapidity.

Coats plus is a highly pleiotropic disorder particularly affecting the eye, brain, bone and gastrointestinal tract. Here, we show that Coats plus results from mutations in CTC1, encoding conserved telomere maintenance component 1, a member of the mammalian homolog of the yeast heterotrimeric CST telomeric capping complex. Consistent with the observation of shortened telomeres in an Arabidopsis CTC1 mutant and the phenotypic overlap of Coats plus with the telomeric maintenance disorders comprising dyskeratosis congenita, we observed shortened telomeres in three individuals with Coats plus and an increase in spontaneous gamma H2AX-positive cells in cell lines derived from two affected individuals. CTC1 is also a subunit of the alpha-accessory factor (AAF) complex, stimulating the activity of DNA polymerase-alpha primase, the only enzyme known to initiate DNA replication in eukaryotic cells. Thus, CTC1 may have a function in DNA metabolism that is necessary for but not specific to telomeric integrity.

Ziel der Untersuchungen war, Pilze aus geschädigtem und ungeschädigtem Wurzelmaterial konventionell und ökologisch bewirtschafteter Weinbergsböden zu isolieren und diese auf ihre Durchsetzungsfähigkeit gegenüber den anderen Arten bzw. deren Pilzmetabolitsuspensionen unter unterschiedlichen Nahrungsbedingungen zu prüfen und eine eventuelle substratabhängige Verhaltensänderung bei den Spezies in Interaktion festzustellen. Zudem wurde in weiteren In-vitro- Versuchen das pathogene Potenzial der gefundenen Arten gegenüber Vitis spp. getestet. Hintergrund dieser Untersuchungen war die Hypothese, dass Absterbeerscheinungen in Rebanlagen nicht durch die Reblaus per se verursacht werden, sondern dass ein Zusammenhang zwischen der Bewirtschaftungsmethode und dem Schadbild in reblausbefallenen Rebanlagen besteht und dessen Entstehung auf pathogenkonduktive und –suppressive Eigenschaften des Bodens zurückgeführt werden kann. Aus rund 2400 Wurzelproben konnten insgesamt 49 Pilzarten isoliert und bestimmt und mehr als die Hälfte davon in Wurzeln beider Versuchsflächen nachgewiesen werden. Ein Großteil der Pilze wurde sowohl in geschädigten als auch in ungeschädigten Wurzelgeweben identifiziert. Darunter waren Arten, die in der Literatur als Parasiten und Saprobier beschrieben werden, aber auch Arten, die eine andere Lebensweise pflegen oder deren Lebensweise nicht bekannt ist. Mit Hilfe von Interaktionsversuchen auf unterschiedlichen Nährmedien (einem Voll- und einem Mangelmedium) konnte bei den untersuchten Arten teilweise starke substratabhängige Verhaltensänderung in Interaktion mit bestimmten Pilzkolonien festgestellt und auf die Verfügbarkeit von organischem Kohlenstoff zurückgeführt werden. Starke Konkurrenz um organischen Kohlenstoff und dadurch entstehende fungistatische und antibiotische Effekte können in diesem Zusammenhang pathogenkonduktive bzw. pathogensuppressive Bodeneigenschaften fördern oder hemmen. Weiterhin konnte gezeigt werden, dass alle 15 in vitro an Vitis spp. inokulierten Pilze (Absidia glauca, Acremonium kiliense, Aspergillus ustus, Cylindrocarpon magnusianum, Cylindrocarpon sp., Fusarium culmorum, F. detonianum, F. oxysporum, F. sacchari, F. semitectum, Gliocladium roseum, Leptosphaeria coniothyrium, Penicillium expansum, Trichoderma harzianum, T. pseudokoningii), unter denen sich auch als Saprobier bekannte Arten befanden (P. expansum, T. harzianum), selbst bei Verfügbarkeit organischer Kohlenstoffverbindungen im Substrat, gegenüber Vitis spp. ein fakultativ pathogenes Potenzial besitzen. Diese aus In-vitro-Interaktionsversuchen gewonnenen Erkenntnisse geben Hinweise darauf, welchen Einfluss die Bewirtschaftung, insbesondere die Versorgung der Weinbergsböden mit organischem Kohlenstoff, auf fakultativ pathogene Sekundärparasiten in Form von Bodenpilzen und folglich auf die Entwicklung von Schadbildern an durch die Reblaus prädispositionierten Rebpflanzen in vivo haben kann.

Die zwischen allen Objekten vorhandenen Wechselwirkungen können repulsiver und attraktiver Natur sein. Bei den attraktiven Kräften kommt der Bestimmung von Dispersionskräften eine besondere Bedeutung zu, da sie in allen kolloidalen Systemen vorhanden sind und entscheidenden Einfluss auf die Eigenschaften und Prozesse dieser Systeme nehmen. Eine der Möglichkeiten, Theorie und Experiment zu verbinden, ist die Beschreibung der London-Van der Waals-Wechselwirkung durch die Hamaker-Konstante, welche durch Berechnungen der Wechselwirkungsenergie zwischen Objekten erhalten werden kann. Für die Beschreibung von Oberflächenphänomenen wie Adhäsion, die in Termen der totalen potentiellen Energie zwischen Partikeln und Substrat beschrieben werden, benötigt man exakt bestimmte Hamaker-Konstanten. In der vorliegenden Arbeit wurde die asymmetrische Fluss Feld-Fluss Fraktionierung in Kombination mit einem auf dem Newton-Algorithmus basierenden Iterationsverfahren zur Bestimmung der effektiven Hamaker-Konstanten verschiedener Nanopartikeln sowie Polystyrollatex-Partikel in Toluol bzw. Wasser verwendet. Der Einfluss verschiedener Systemparameter und Partikeleigenschaften wurde im Rahmen der klassischen DLVO-Theorie untersucht.
