The aim of this study was to analyze the effect of flushing on the reproductive performance of Morada Nova (MN) and Santa Inês (SI) ewes submitted to fixed time artificial insemination (TAI). Twenty seven SI and 24 MN supplemented with concentrate (1% of live weight, on average), for 75 days during the breeding season. After 30 days of supplementation, ewes were synchronized with the aid of a hormonal protocol (HP) based on progesterone, eCG and cloprostenol. The estrus observation was conducted at 12, 24, 36 and 48 h after the end of HP with the aid of two ruffians. TAI was done 55 h after the end of HP. From 20 to 45 days after the beginning of the HP ewes were exposed to rams (natural breeding). The pregnancy diagnosis was evaluated 70 days after TAI. We analyzed the weight, body condition score, estrus rate, pregnancy rate and prolificacy testing the effects of race, week of supplementation and body condition score class. The weight and body conditions of ewes varied according to the week of supplementation, with higher values in the first two weeks following TAI. The estrus rate was 88.2% and 43.2% of the ewes showed estrus up to 24 hours of the end of the HP. The pregnancy rate per TAI was 31.3% and the pregnancy rate after natural breeding was 50.0%. It was observed that body condition score classes interfered in pregnancy rates. There was a higher percentage of multiple births by pregnancy by TAI than by natural breeding. It was concluded that the flushing resulted in weight gain and better body conditions ensuring the standardization of the herd for breeding season, which therefore improved reproductive performance. The HP used advanced the onset of estrus and increased prolificacy, but was inefficient in the synchronization of woolless sheep.

The purpose of this work was to validate isolation methods for sheep mesenchymal stem cells (MSC) from different sources and to explore the hypothesis that MSC exhibit markers of the same phenotype independent from tissue source. Cells derived from ovine bone marrow, synovial membrane and adipose tissue were characterized using the following markers: CD44, CD45, CD11b and MHC-I. The isolated MSC were cultivated, went through osteogenic, chondrogenic and adipogenic differentiation, and were characterized by flow cytometry using mouse anti-ovine CD44, CD45 and MHC-I monoclonal antibody (mAb), and mouse anti-bovine CD11b mAb. Ovine MSC from all three sources differentiated under chondorgenic, osteogenic and adipogenic conditions. Also, MSC from the three tissues were found to express CD44 and MHC-I but lack of CD11b and CD45. The results obtained revealed that our isolation methods for the different tissues tested are valid and that MSC from the three sources studied have same immunophenotic characteristics. Streptococcosis is one of the major causes of mortality in tilapia's creation in Brazil, inducing great economic losses. As soon, the study objectived to determinate the frequency of isolation and identification the Streptococcus agalactiae in organs different of Oreochromis niloticus naturally infected, derived from eight fish farms in the northern region of the state of Parana, that presented clinical signs characteristics of streptococcal disease. However, blood samples and fragments (kidney, liver, spleen, heart and brain) were collected. These all samples were plated on solid medium of brain and heart infusion (BHI) added 5% ovine blood and incubated at 29 degrees C for 7 days in aerophilic conditions. Behind, the bacterial growth and from the macro and microscopic features, colonies compatibles with Streptococcus sp. gender, were selected. The species were identified by PCR reaction and confirmed by sequencing of 16S rDNA gene. The results exhibited that in tilapia of Nile infected with S. agalactiae the isolation is more common in brain, kidney and liver in descending order.

Eight reproductive rams with no prior reproductive disease were distributed into three groups of infection with T. gondii: GI, 3 rams, 2.0 x 10(5) P strain oocysts; GII, 3 rams, 1.0 x 10(6) RH strain tachyzoites; GIII, 2 control rams. Clinical parameters were measured and serological evaluations (IIF) were performed. Presence of the parasite in the semen was investigated by PCR and bioassay techniques. The rams presented clinical alterations (hyperthermia and apathy) related to toxoplasmosis in both groups infected with Toxoplasma gondii. All the inoculated rams responded to antigenic stimulus, producing antibodies against T. gondii from postinoculation day 5 onwards. In ovine groups I and II, the greatest titers observed were 1 : 4096 and 1 : 8192, respectively. In semen samples collected from these two groups, the presence of T. gondii was detected by bioassay and PCR. This coccidian was isolated (bioassay and PCR) in tissue pools (testicles, epididymis, seminal vesicle, and prostrate) from two rams infected presenting oocysts and in one presenting tachyzoites.
