In vivo production of viral biopesticides is the major source of viral insecticides currently in the marketplace. However, this system presents limitations during production scale-up. For the Spodoptera frugiperda nucleopolyhedrovirus (SfMNPV), the insect used for replication has cannibalistic characteristics, thus production is even more difficult. Insect cells are commonly used for in vitro baculovirus production. Most of these cell lines are derived from Lepidoptera species. The Sf21 cell line is derived from Spodoptera frugiperda caterpillar ovarian tissue, and its clonal isolate Sf9 has been used for biopesticide production due to its ease of growth in suspension cultures. In this work, the in vitro production capabilities of a Brazilian SfMNPV isolate obtained from cornfields was evaluated. Comparison of polyhedra production was carried out using both Sf21 and Sf9 cells, based on volumetric and specific yields. Both cell lines were cultivated in Hyclone medium supplemented with different fetal bovine serum concentrations (2,5 and 5%). The best results were obtained using Sf9 cells supplemented with 5% serum. These results were further confirmed quantitively through kinetic parameter estimation for both cells lines and different serum concentrations. After seven successive passages, this system still presented high specific polyhedra production

This paper describes a geostatistical method, known as factorial kriging analysis, which is well suited for analyzing multivariate spatial information. The method involves multivariate variogram modeling, principal component analysis, and cokriging. It uses several separate correlation structures, each corresponding to a specific spatial scale, and yields a set of regionalized factors summarizing the main features of the data for each spatial scale. This method is applied to an area of high manganese-ore mining activity in Amapa State, North Brazil. Two scales of spatial variation (0.33 and 2.0 km) are identified and interpreted. The results indicate that, for the short-range structure, manganese, arsenic, iron, and cadmium are associated with human activities due to the mining work, while for the long-range structure, the high aluminum, selenium, copper, and lead concentrations, seem to be related to the natural environment. At each scale, the correlation structure is analyzed, and regionalized factors are estimated by cokriging and then mapped.

Viable cells immobilized in inert supports are currently studied for a wide range of bioprocesses. The intrinsic advantages of such systems over suspended cultures incite new research, including studies on fundamental aspects as well as on the industrial viability of these non-conventional processes. In aerobic culture of filamentous fungi, scale-up is hindered by oxygen mass transfer limitation through the support material and bioprocess kinetics must be studied together with mass transfer limitation. In this work, experimental and simulated data of cephalosporin C production were compared. Concentrations in the bulk fermentation medium and cellular mass profiles inside the bioparticles are focused. Immobilized cells were used in a tower bioreactor, operated in fed-batch mode. To describe the radial variation of oxygen concentration within the pellet, a dead core model was used. Despite the extremely low sugar concentrations, bioreaction rates in the pellets were limited by the dissolved oxygen concentration. Cell growth occurs only in the outer layers, a result also confirmed by scanning electron microscopy. (C) 2001 Elsevier B.V. Ltd. All rights reserved.

The use of sewage sludge in agricultural land as a means of sludge disposal and recycling has been shown to be economical and suitable because of the presence of nutrients such as nitrogen and phosphorus. However, municipal sludges often contain high quantities of toxic metals and other compounds that must be removed for its safe use in agricultural soils. The biological leaching of metals from sewage sludges has been shown to be a promising technique for metal detoxifying in such complex matrix. The process efficiency is dependent on several physico-chemical parameters, such as total solids concentration, metal forms, pH-ORP, and temperature. Scale-up of the process has not yet been defined and is still pursuing the correct operational design. Current research involving the bioleaching of metals from sewage sludge and its application to land, which affects soil physical properties, are presented and discussed.

This article extends results contained in Buzzi et al. (2006) [4], Llibre et al. (2007, 2008) [12,13] concerning the dynamics of non-smooth systems. In those papers a piecewise C-k discontinuous vector field Z on R-n is considered when the discontinuities are concentrated on a codimension one submanifold. In this paper our aim is to study the dynamics of a discontinuous system when its discontinuity set belongs to a general class of algebraic sets. In order to do this we first consider F :U -> R a polynomial function defined on the open subset U subset of R-n. The set F-1 (0) divides U into subdomains U-1, U-2,...,U-k, with border F-1(0). These subdomains provide a Whitney stratification on U. We consider Z(i) :U-i -> R-n smooth vector fields and we get Z = (Z(1),...., Z(k)) a discontinuous vector field with discontinuities in F-1(0). Our approach combines several techniques such as epsilon-regularization process, blowing-up method and singular perturbation theory. Recall that an approximation of a discontinuous vector field Z by a one parameter family of continuous vector fields is called an epsilon-regularization of Z (see Sotomayor and Teixeira, 1996 [18]; Llibre and Teixeira, 1997 [15]). Systems as discussed in this paper turn out to be relevant for problems in control theory (Minorsky, 1969 [16]), in systems with hysteresis (Seidman, 2006 [17]) and in mechanical systems with impacts (di Bernardo et al., 2008 [5]). Biopulping is a technology which application can be advantageous to mechanical or chemical pulping. It presents benefits such as the creation of stronger pulp, as well as energy or chemicals savings. This paper gives an overview of the recent efforts to develop biopulping processes in Brazil as well as provides critical information on biopulping development worldwide. Eucalyptus grandis wood chips have been biotreated by Ceriporiopsis subvermispora in a 50-ton biopulping pilot-plant and used to produce TMP and CTMP pulps on a mill scale, Up to 18% and 27% energy savings have been observed for producing 450-470 CSFreeness TMP and CTMP pulps. Despite darker bio-TMP pulps are produced, one-stage bleaching with 5% H2O2 was sufficient to improve brightness values to 70% and 72% for bio-TMP and control pulps, respectively. Understanding biopulping mechanisms is also relevant because more resistant and competitive fungal species could be selected with basis on a function-directed screen-ing project. As far as the chemical changes induced by the fungus in wood are concerned, recent efforts have pointed out for two different types of wood transformations. One of them involves intense lignin depolymerization in short biotreatment periods, while the other indicates that esterification reactions of oxalate secreted by the Jungas on the polysaccharides chains increase the water saturation point of the fibers. Both transformations are expected to affect the fiber-fiber bonding and, consequently, the physical resistance of wood.

This paper proposes to use a state-space technique to represent a frequency dependent line for simulating electromagnetic transients directly in time domain. The distributed nature of the line is represented by a multiple 1t section network made up of the lumped parameters and the frequency dependence of the per unit longitudinal parameters is matched by using a rational function. The rational function is represented by its equivalent circuit with passive elements. This passive circuit is then inserted in each 1t circuit of the cascade that represents the line. Because the system is very sparse, it is possible to use a sparsity technique to store only nonzero elements of this matrix for saving space and running time. The model was used to simulate the energization process of a 10 km length single-phase line. ©2008 IEEE.

A extração com fluido supercrítico de materiais líquidos e sólidos despertou o interesse para aplicações industriais nas últimas décadas, mais particularmente sob o conceito de química verde e biorrefinarias, portanto é fundamental que se faça uma modelagem desse processo a fim de otimizar as condições operacionais e simular o processo. O objetivo geral deste trabalho consiste na determinação de parâmetros de transferência de massa do processo de extração supercrítica de matriz sólida, empregando o dióxido de carbono como solvente, a partir de dados cinéticos de extração e na avaliação sistemática de cinco modelos matemáticos para descrever as cinéticas de extração dos óleos da polpa e da casca do buriti, do óleo de açaí de da oleoresina de cúrcuma, medidas no Laboratório de Extração Supercrítica, da Faculdade de Engenharia Química (UFPA), a fim de contribuir para o estudo de ampliação de escala e análise de custo de produção. Foram avaliados os modelos de Tan e Liou, Goto et al. (1993), Martinez et al. (2003), Esquível et al. (1999), e Sovová (1994). A modelagem das cinéticas de extração foi realizada utilizando aplicativos computacionais desenvolvidos e validados neste trabalho a partir de diferentes dados experimentais publicados na literatura. Diante de 40 cinéticas medidas com diferentes equipamentos de extração, configurações de leito, tipos de matérias primas, preparo dos materiais, pressão e temperatura e outros parâmetros de processo (com destaque ao rendimento global e a vazão de solvente), foi construído um panorama dos resultados acerca da capacidade dos modelos de transferência de massa em descrever as mais diferentes curvas globais de extração. De forma geral, os modelos de Goto et al. (1993) e Sovová (1994) apresentaram as melhores previsões aos dados experimentais das matérias primas tratadas neste trabalho com menores valores de quadrado, erros relativo, faixa de erro e desvios padrão e valores de R2 próximos da unidade.

O objetivo deste trabalho foi avaliar a maturação oocitária in vitro (MIV) na espécie C. apella, relacionando com a expansão das células do cumulus e promovendo a produção in vitro de embriões (PIVE), por meio da ativação partenogenética e fecundação in vitro (FIV). Os oócitos puncionados dos folículos antrais medindo 2-9 mm de diâmetro foram classificados em desnudos (OD), com poucas células do cumulus (PCC) e complexo cumulus-oophorus intacto (CCO intacto). A expansão das células do cumulus foi analisada nos tempos de 0, 36 e 40 h de MIV e classificada em cinco categorias de expansão. A competência meiótica nos oócitos foi verificada pela extrusão do 1º corpúsculo polar (CP) após 40 h de MIV. Os oócitos foram divididos em 3 grupos para PIVE: grupo controle (FIV), ativação partenogenética utilizando 5 μM de ionomicina em associação com 2 mM de 6-DMAP ou em associação com 50 μM de roscovitina. Para a FIV, os espermatozóides obtidos do coágulo seminal, foram diluídos em água de coco em pó (ACP-118®) e submetidos ao resfriamento para serem levados posteriormente ao cultivo in vitro com os oócitos. O aumento no tempo da MIV e a presença de várias células do cumulus associadas aos oócitos proporcionaram maior expansão das células do cumulus, pois somente os CCO intactos alcançaram a expansão total em 40 h de MIV (p < 0,005). A presença das células do cumulus nos oócitos (PCC e CCO intactos) maturados in vitro por 40 h promoveu a competência meiótica (metáfase II) significamente mais elevada que os OD (p < 0,005). Após 6 h de resfriamento em ACP-118® e separação pelo método do swin-up, os espermatozóides utilizados na FIV possuíam 80% de motilidade e vigor 4. O tratamento com ionomicina/roscovitina promoveu a extrusão do 2º CP e formação pronuclear e com ionomicina/6-DMAP formou pronúcleos, sem extrusão do 2º CP. O protocolo utilizando a ionomicina/6-DMAP e da FIV originou as primeiras divisões embrionárias, observada pela taxa de clivagem. Os resultados encontrados sugerem que em C. apella a presença e a expansão total das células do cumulus estão relacionadas com a competência oocitária. A utilização dos oócitos maturados in vitro e dos espermatozóides diluídos e resfriados em ACP-118® promoveu a PIVE por diferentes métodos nesta espécie.

In this work the separation of multicomponent mixtures in counter-current columns with supercritical carbon dioxide has been investigated using a process design methodology. First the separation task must be defined, then phase equilibria experiments are carried out, and the data obtained are correlated with thermodynamic models or empirical functions. Mutual solubilities, Ki-values, and separation factors aij are determined. Based on this data possible operating conditions for further extraction experiments can be determined. Separation analysis using graphical methods are performed to optimize the process parameters. Hydrodynamic experiments are carried out to determine the flow capacity diagram. Extraction experiments in laboratory scale are planned and carried out in order to determine HETP values, to validate the simulation results, and to provide new materials for additional phase equilibria experiments, needed to determine the dependence of separation factors on concetration. Numerical simulation of the separation process and auxiliary systems is carried out to optimize the number of stages, solvent-to-feed ratio, product purity, yield, and energy consumption. Scale-up and cost analysis close the process design. The separation of palmitic acid and (oleic+linoleic) acids from PFAD-Palm Fatty Acids Distillates was used as a case study.
