Para testar o efeito da temperatura e do tempo de embebição nos valores de condutividade elétrica e verificar sua aplicabilidade para determinar a qualidade fisiológica de sementes de Dalbergia nigra (jacarandá-da-bahia), foram utilizadas sementes colhidos em três anos (lote I _ 1998; lote II _ 1997 e lote III - 1994). Inicialmente determinou-se o teor de água e depois conduziu-se os testes de germinação em laboratório e viveiro, utilizando-se quatro repetições de 25 sementes. Para estudar a condutividade elétrica (CE) foram utilizadas quatro repetições de 50 sementes. Cada subamostra foi colocada em recipiente contendo 75ml de água deionizada, embebidas por 6, 12, 18, 24, 30 e 36 horas, a 20, 25 e 30ºC. Sementes do lote II apresentaram maiores valores na primeira contagem da germinação, diferindo significativamente dos demais lotes. O lote III apresentou qualidade inferior. Com o aumento do período e da temperatura de embebição ocorreu aumento nos valores de CE das sementes dos três lotes. O lote III mostrou qualidade fisiológica inferior a dos lotes I e II, que apresentaram valores mais elevados de CE. Não houve diferenciação entre os lotes I e II. Concluiu-se que o teste de CE foi eficiente para diferenciar os lotes de sementes de Dalbergia nigra, com alto grau de associação com o teste de germinação.

O presente trabalho teve como objetivos verificar o efeito do número de sementes e do volume de água utilizada no teste de condutividade elétrica (CE) para avaliar o vigor de três lotes de sementes de Dalbergia nigra (jacarandá-da-bahia) e correlacionar esses resultados com os dados de germinação em laboratório e em viveiro. Os testes de germinação em laboratório e viveiro foram conduzidos com quatro repetições de 25 sementes. O teste de CE foi realizado com 25, 50 e 75 sementes embebidas a 75, 100 e 125ml de água, por diferentes períodos. A porcentagem de germinação e de plântulas normais em laboratório, indicaram o lote III como de qualidade inferior aos lotes I e II. A primeira contagem da germinação e o índice de velocidade de germinação em laboratório e a emergência, índice de velocidade de emergência e porcentagem de plântulas normais em viveiro identificaram o lote II como superior ao lote I e o III como inferior. A CE diminui com o aumento do volume de água e aumentou com o período de embebição. A diferenciação dos lotes foi mais eficiente, quando se utilizou 75ml de água deionizada e amostras de 50 sementes com pelo menos 36 horas de embebição, com valores de CE menores nos lotes I e II do que no lote II. Os coeficientes de correlação simples entre a CE e as demais características avaliadas, em laboratório e viveiro, foram elevados e significativos, evidenciando alta associação entre os mesmos. Assim, pode-se recomendar que o teste de CE seja conduzido a 25ºC, com amostras de 50 sementes embebidas em 75ml de água deionizada, por períodos iguais ou superiores a 36 horas de embebição, para determinar a qualidade fisiológica de lotes de sementes de jacarandá-da-bahia.

Licania rigida Benth., Licania tomentosa (Benth.) Fritsch, and Couepia impressa Prance (Chrysobalanaceae family) plants have long been used medicinally by the people from Northeastern Brazil. Crude extracts and infusions of these plants have been applied in the treatment of several conditions such as diabetes and rheumatism, degenerative diseases with involvement of reactive oxygen species (ROS). The aim of this study was to evaluate the aqueous, ethanolic, and hydroethanolic leaves extracts antioxidant capacity of these species, using several in vitro assay systems (reducing power, DPPH● scavenging, the β-carotene linoleate model system and lipid peroxidation inhibition in rat brain homogenate, using thiobarbituric acid reactive substances - TBARS). The oral acute toxicity of aqueous extracts was also evaluated in vivo. Results revealed that these extracts possess a potent reducing power and DPPH scavenging ability, as well as the ability to prevent TBARS formation in rat brain homogenate in a concentration-dependent manner. Regarding in vivo oral acute toxicity of the aqueous species extracts, no toxic effects were observed upon evaluating physiological, hematological and biochemical parameters. The presence of high levels of phenolics and flavonoids was determined mainly in the ethanol extract. However, the C. impressa hydroethanolic extract, fractionated with hexane, chloroform and ethyl acetate for analysis by NMR 1H, showed more efficient results than the reference antioxidant Carduus marianus. The classes of organics compounds were determined were phenolics in the fraction of ethyl acetate and terpenes in chloroform and hexane fractions. The ethil acetate fraction had the highest content of flavonoids and increased scavenging capacity of DPPH●, possibly by the presence of phenolic compounds. Therefore, a detailed investigation of the phytochemical composition and in vivo study of the C. impressa hydroethanolic extract is suggested to characterize the active compounds of the species

The Caatinga is the predominant vegetation type in semi-arid region of Brazil, where many inhabitants depend on hunting and gathering for survival, obtaining resources for: food and feed, folk medicine, timber production, etc. It‟s the dry ecosystem with highest population density in the world. The early stages of development are the most critical during the life cycle of a flowering plant and they‟re primordial to its establishment in environments exposed to water stress. Information about adjustments to the growth of the species, correlated with their studies of distribution in Seridó oriental potiguar, are an important ecological and economic standpoint, because they provide subsidies for the development of cultivation techniques, to programs of sustainable use and recovery of degraded areas. This thesis aimed to study the initial growth and foliar morphology in plants like Enterolobium contortisiliquum (Vell.) Morong. (tamboril) and Erythrina velutina Mart. ex Benth (mulungu), species of occurrence in the Caatinga, under water stress. After sowing and emergency, the seedlings were exposed to three water regimes: 450 (control), 225 (moderate stress) and 112.5 (severe stress) mm of water slide for 40 days. Seeding occurred in bags of 5 kg and after the establishment of seedlings thinning was carried out leaving a plantlet per bag. At the beginning the waterings occurred daily with distilled water, passing to be on alternate days after thinning. Twenty and forty days after the thinning seedlings collections were held to be done analysis of growth and biomass partition. When compared to the control group, the treatments with water stress showed reduction in the growth of the aerial part, growth of the greater root, number of leaves and leaflets, dry leaf area and total phytomass in both species, but in general, this effect was most marked for E. velutina. Regarding the partition of biomass, there were few changes throughout the experiment. Morphological changes in the leaves as a function of stress were not significant, however, there was a trend, in both species, to produce narrower leaves, that facilitate heat loss to the environment. It has not been possible to establish a positive relationship between inhibition of growth and distribution of species, whereas E. velutina is a species of most common occurrence in Seridó oriental potiguar. In this way, other aspects should be taken into account when studying the adaptation of species the dry environments, such as salinity, presence of heavy metals, wind speed, etc

O desempenho germinativo de sementes de Dalbergia nigra (jacarandá-da-bahia) foi avaliado em laboratório e viveiro, com o objetivo de verificar a eficiência do teste de germinação em laboratório em predizer o comportamento de lotes de sementes em condições de viveiro. Para tanto sementes pertencentes a três lotes (anos de colheita: lote I - 1998, lote II - 1997 e lote III - 1994) foram utilizados para determinar os dados biométricos, teor de água, germinação, índice de velocidade e primeira contagem da germinação e porcentagem de plântulas normais em laboratório e emergência, índice de velocidade de emergência e porcentagem de plântulas normais em viveiro. Sementes pertencentes ao lote III apresentaram qualidade inferior aos lotes I e II, tanto em laboratório como em viveiro. Os altos valores do coeficiente de correlação simples entre as características avaliadas nas duas condições, evidenciam a eficiência do teste de germinação em laboratório em predizer o desempenho germinativo de sementes de jacarandá-da-bahia em viveiro.

O angico-de-bezerro (Piptadenia moniliformis Benth.) é uma espécie arbórea melífera, de crescimento rápido, característica da caatinga do nordeste brasileiro, onde é muito abundante e com dispersão contígua e irregular. Seus ramos finos, junto com as folhas, servem como alimento para animais. Como suas sementes apresentam dormência por impermeabilidade do tegumento à água, objetivou-se neste trabalho avaliar a eficiência de tratamentos pré-germinativos para superar a dormência de suas sementes visando maximizar e uniformizar o processo de germinação. Foram estudados 28 tratamentos pré-germinativos: imersão em água a 70, 80, 90 ºC e fervente por 1, 2, 3, 4 e 5 minutos e imersão em ácido sulfúrico concentrado por 1, 5, 10, 15, 20, 25 e 30 minutos, além da testemunha. A semeadura foi realizada sobre papel em temperatura alternada de 20-30 ºC, utilizando-se quatro repetições de 25 sementes. Foram avaliadas as porcentagens de germinação das sementes considerando a protrusão da raiz primária, de plântulas normais e de sementes duras e o índice de velocidade de germinação. O delineamento experimental utilizado foi o inteiramente casualizado e as médias comparadas pelo teste de Scott-Knott a 5% de probabilidade. Os tratamentos de imersão em água, independentemente da temperatura e do período de imersão, aumentaram a porcentagem de germinação das sementes, comparativamente ao tratamento controle, mas foram menos eficientes que os tratamentos em ácido sulfúrico nos maiores períodos de imersão. Desta forma, a imersão por 20, 25 e 30 minutos em ácido sulfúrico é recomendada para a superação da dormência de sementes de Piptadenia moniliformis Benth.

Seeds of Caesalpinea ferrea Mart. ex Tu. var. leiostachya Benth. present low and irregular germination due to dormancy caused by the impermeability of the tegument. With the purpose to determine an efficient method to accelerate and uniformize seed germination, they were submitted to different immersion periods (0, 5, 10, 15, 20, 25 and 30 minutes) in sulfuric acid (95-98%), influence emergency and vigor (first count, speed index, medium time and relative frequency of emergency, length and dry matter of the seedlings). The experiment was carried out under greenhouse conditions with an experimental design of completely randomized, with seven treatments and four replications. The pre-conditioning of the seeds with immersion in concentrated sulfuric acid was efficient to overcoming seed dormancy, by increasing the percentage and speed emergency, the first count of emergency, length and dry matter of seedlings and reduction in the medium time for emergency. The efficiency of this chemical treatment with concentrated sulfuric depends on the immersion time, and 10 to 20 minutes were the most adequate to provide larger emergency uniformity percentages and vigor.

In germination tests, substrate and ideal temperature are important for providing favorable conditions for the germination of seeds and the eventual development of seedlings. This research aimed at evaluating the effects of temperature and substrate on the germination of Piptadenia moniliformis Benth seeds. Two trials were done. In the first, seeds were submitted to germination under constant temperatures of 5 to 40 degrees C (increases of 5 degrees C) and alternated temperatures of 20-25, 25-30, 30-35, 20-30, 25-35 and 20-35 degrees C on paper. In the second trial, temperatures of 25, 20-30 e 20-35 degrees C were used in the substrates on (OP) and between paper (BP), on (OS) and between sand (BS), on (0V) and between vermiculite (BV) and paper roll (PR). The following traits were evaluated: percentage of seeds with protrusion of primary root and normal seedlings; speed germination index; length of primary root and of the hypocotyls and the dry mass of seedlings. A completely randomized design was used with 14 treatments. In the first experiment and a factorial scheme of 7 x 3 (7 substrates and 3 temperatures) was used in the second with four replicates of 25 seeds each. In the first experiment the constant temperatures of 25 e 30 degrees C and the alternated of 20-30 e 20-35 degrees C provided best results for the germination of seeds. In the second, the temperature of 25 C associated with the substrates BP, BS and BV surpassed the other treatments proving adequate for carrying out the germination tests of Piptadenia moniliformis Benth seeds.

Stem bark of the two species Stryphnodendron polyphyllum Mart. and Stryphnodendron obovatum Benth., Leguminosae, was investigated for wound healing, antibacterial and antioxidant activity. These plants contain 12 and 19% tannins in their stem bark, respectively, and are widely used in traditional medicine in Brazil. The total content of phenolics of the crude extract (CE) of Stryphnodendron obovatum was 76.95 +/- 2.98% (CV = 3.87%) and of the ethyl-acetate fraction (EAF) was 89.13 +/- 0.34% (CV = 0.38%); whereas in Stryphnodendron polyphyllum the CE phenolics content was 51.62 +/- 1.53% (CV = 2.96%) and the EAF phenolics content was 59.00 +/- 1.91% (CV = 3.24%). The tannin content of CE from Stryphnodendron obovatum [36.58 +/- 0.35% (CV = 0.98%)] was about 11% higher than in CE from Stryphnodendron polyphyllum [25.43 +/- 0.96% (CV = 3.77%)]. The difference between the species was even greater in the EAF: in Stryphnodendron obovatum the EAF phenolics content was 55.01 +/- 0.36% (CV = 0.65%), whereas in Stryphnodendron polyphyllum the content was 36.16 +/- 0.42% (CV = 1.16%). The healing effect of ointments containing 2.5% crude lyophilised extract (PCE) and 2.5% ethyl-acetate lyophilised fraction (PEA) of the stem bark of Stryphnodendron polyphyllum and Stryphnodendron obovatum was studied in cutaneous wounds of Wistar((R)) rats after 4, 7 and 10 days of treatment. Epithelial cell proliferation in the area of re-epithelialisation of the wounds was evaluated by counting the metaphases blocked by vincristine sulfate. With PCE an increase in epidermal growth was observed after 4 and 7 days of treatment with Stryphnodendron polyphyllum, and after 7 and 10 days of treatment with Stryphnodendron obovatum. Wounds treated with PEA of Stryphnodendron obovatum showed increased epidermal growth only 4 days after the treatment, for Stryphnodendron polyphyllum, epidermal growth was observed after 4 and 7 days of treatment. Both the CE and the EAF fractions of Stryphnodendron polyphyllum and Stryphnodendron obovatum showed antibacterial activity against Staphylococcus aureus with MIC values of 125 and 250 mu g/ml, respectively. Gram-negative bacteria tested were not inhibited by extracts and fractions at concentrations > 1000 mu g/ml. The antioxidant activity through reduction of the DPPH radical in TLC, confirmed the anti-radical properties of these extracts in both species. CE and EAF of both species showed a radical scavenging activity (RSA) and protected DPPH from discolouration, already at 0.032 mu g/ml. The extract from Stryphnodendron polyphyllum were more effective than those Stryphnodendron obovatum, although the former had a lower tannin content. (c) 2005 Elsevier B.V.. All rights reserved.

Considerando-se a qualidade dos fitoterápicos, é importante salientar que a preocupação com esta questão inclui rigoroso acompanhamento das diferentes etapas de desenvolvimento e produção, desde a coleta do vegetal até o produto final. O controle de qualidade de drogas vegetais e seus extratos é essencial quando utilizados como matéria-prima para o desenvolvimento de fitoterápicos. Neste trabalho foram utilizadas diversas técnicas (farmacopeicas e não farmacopeicas) visando estabelecer parâmetros de qualidade dos frutos de Dimorphandra mollis Benth. (Leguminosae), popularmente conhecida como faveiro. Os resultados obtidos determinaram as características físico-químicas da droga vegetal e mostraram que o pó dos frutos e o extrato etanólico 70% de D. mollis apresentam um teor de flavonóides de 10,25% e 17,21%, respectivamente. A análise fitoquímica preliminar dos frutos caracterizou a presença de taninos, saponinas e flavonóides. Na análise microbiológica do extrato não houve crescimento de patógenos entre os testes realizados.

In this present work an ethnographic research was performed with 84 native medicinal specimens from the Litoral Norte Riograndense, from which two plants Cleome spinosa Jacq e Pavonia varians Moric were submitted to ethnobotanic, phytochemistry and pharmacologic investigations. Additionally, a phytopharmacological research of the medicinal specimen Croton cajucara Benth ( native plant of the Amazon region of Brazil) was improved. The obtained phytochemical results of the C. spinosa and P. varians showed the presence of flavonoids constituents, among other components. The two flavonoids (2S)-5-hydroxy-7,4 -dimethoxy-flavanone and 5,4 -dihydroxy-3,7,3 -trimethoxy-flavone were isolated from C. spinosa. The antioxidant activity of the hydroalcoholic extracts of C. spinosa and P. varians solubilized in the microemulsion systems SME-1 and SME-4, was evaluated in the DPPHmethod. The used SME systems [obtained with Tween 80: Span 20 (3:1) and isopropyl myristate (IPM)] improved the dissolution of those tested polar extracts, with higher efficacy to the SME-1 system (in which ethanol was included as cosurfactant). The CE50 values evidenced for P. varians were 114 [g/mL (SME-1) and 246 [g/mL (SME-4); for C. spinosa it was 224 [g/mL (SME-1) and 248 [g/mL (SME-4), being the system SME-1 more effective for both tested extracts. The hydroalcoholic extracts of P. varians (HAE-PV) was also submitted to pharmacological screening for antinociceptive activity in animal models. The oral administration of this extract (100, 300 and 1000 mg/kg) inhibited the acetic acid-induced writhing in mice. The higher inhibition (74%) was evidenced to the 1000 mg/kg administered dose. Its effect on the central nervous system (CNS) was investigated by tail flick and formalin-method and reveled that it has negligible antinociceptive action on the CNS. After taking consideration of HAE-PV interaction, Pavonia varians Moric could be used as a potent analgesic agent in case of peripheral algesia, without affecting the CNS. The phytochemical study of the stem bark of Croton cajucara Benth lead to the isolation of 19-nor-clerodanetype diterpenes, as well as to the separation of its fixed oil FO-CC. This non polar oil material reveled to be rich in sesquiterpenes and 19-nor-clerodanes components. The biologic effect of OF-CC was evaluated in the development in vitro of the fungis phytopatogens such as Fusarium oxysporum, Rhizoctonia solani and Sclerotium rolfsii. Significant inhibitory effect of the tested fungis (at 0,2 mg.mL-1 dosage) were comproved. A Mass Spectrometry study of clerodane-type diterpenes was developed in order to identify characteristic fragments on mass spectrometra of both clerodane and 19-nor-clerodane presenting an α,β-insaturated carbonyl moiety at ring A of the decalin-system. For that study, mass spectroscopy data were analysed for 19-nor-clerodanes [trans-dehydrocrotonin (DCTN), trans-crotonin (CTN), cis-cajucarin B (c-CJC-B), and cajucarinolide (CJCR)] and for clerodanes [isosacacarin (ISCR) and transcajucarin A (t-CJC-A)] obtained from the stem bark of C. cajucara, and also clerodane-type from other species. The trans-junction of the enone-system clerodanes was clear correlated with the presence of the characteristic ions at m/z 95, 121 e 205. Meanwhile, the characteristics ions at m/z 122 e 124 were correlated to cis-junction. The trans-junction of the enone-system 19-nor-clerodanes showed characteristics ions at m/z 161, 134 e 121. This study could be successful employed for identification of clerodane constituents from other specimens without any additional spectroscopic analyses, as well as a previously phytochemical analyzes in clerodane project search
