Seed of 15 species of Brassicaceae were stored hermetically in a genebank (at -5 degrees C to -10 degrees C with c. 3% moisture content) for 40 years. Samples were withdrawn at intervals for germination tests. Many accessions showed an increase in ability to germinate over this period. due to loss in dormancy. Nevertheless, some dormancy remained after 40 years' storage and was broken by pre-applied gibberellic acid. The poorest seed survival occurred in Hormatophylla spinosa. Even in this accession the ability to germinate declined by only 7% between 1966 and 2006. Comparison of seeds from 1966 stored for 40 years with those collected anew in 2006 from the original sampling sites, where possible, showed few differences, other than a tendency (7 of 9 accessions) for the latter to show greater dormancy. These results for hermetic storage at sub-zero temperatures and low moisture contents confirm that long-term seed storage can provide a successful technology for ex situ plant biodiversity conservation.

The development of global orientation and morphological features in linear polyethylene crystallizing from a sheared melt are studied using in-situ time-resolving wide angle X-ray scattering (WAXS) and ex-situ transmission electron microscopy. It is found that samples subjected to a shear rate above a critical value of ~1s-1 result in macroscopically oriented structures in the crystallized sample. This critical shear rate appears to be independent of the differences in molecular weight distribution of the samples studied although the morphologies which develop are sensitive to quite small differences in molecular weight distributions. The presence of shish kebabs in the morphology is shown to differ markedly according to variations in the upper molecular weight fraction of the molecular weight distribution, even though the resulting global orientation does not. The WAXS also reveals that areas which evidence no row nucleated structures still realize high degrees of molecular orientation. It is proposed that the formation of shish kebab or lamellar morphologies in these samples is dependent on the critical density of contiguous elongated crystallization nuclei rather than any specific global criteria.

Europe has the greatest concentration of botanic gardens in the world, they cultivate extensive collections of plants that include samples of European threatened plant species. This study looks at the effectiveness of these collections in supporting species conservation. A three part study is presented: (1) the results of a survey and assessment of threatened plants in botanic gardens, as defined by the Bern Convention; (2) case studies illustrating current issues in the ex situ management of European threatened plant species; and (3) presentation of policy recommendations on further improving botanic garden contributions to European plant conservation. The survey indicated that of 119 European botanic gardens in 29 European countries, 105 are cultivating 308 of the 573 threatened plant species listed by the Bern Convention. The survey identified 25 botanic gardens in 14 countries undertaking 51 conservation projects focused on 27 Bern listed species. In particular this survey has established that the majority of taxa are held in a small number of collections, dominated by non-wild origin accessions, and are not adequately documented. The majority of specimens in botanic gardens are cultivated out of the range country and not contributing to a specific conservation project. We review the genetic representation and documentation of origin in collections. Existing plant collections contain representatives of populations, now lost in the wild and maintain samples of at least nine European plant taxa identified as 'Extinct in the Wild'. However, inadequate standards of record keeping has compromised the conservation value of many collections. We highlight the dangers of hybridisation and disease in ex situ collections. The results suggest that botanic garden collections are skewed towards horticulturally robust and ornamental species and do not fully reflect priorities as defined by the Bern Convention. Recognising the limitations of traditional botanic garden collections we propose that botanic gardens more effectively utilise their two core competencies, namely scientific horticulture and public display and interpretation. The unique horticultural skills resident in European botanic gardens could be more effectively utilised through the application of horticulture to the management of wild populations.

Losses of cultivated cocoa (Theobroma cacao L.) due to diseases and continued depletion of forests that harbour the wild progenitors of the crop make ex situ conservation of cocoa germplasm of paramount importance. In order to enhance security of in situ germplasm collections, 2-3 mm floral-derived secondary somatic embryos were cryopreserved by vitrification. This work demonstrates the most uncomplicated clonal cocoa cryopreservation. Optimal post-cryostorage survival (74.5%) was achieved by 5 d preculture of SSEs on 0.5 M sucrose medium followed by 60 min dehydration in cold PVS2. To minimise free radical related cryo-injury, cation sources were removed from the embryo development solution and/or the recovery medium, the former treatment resulting in a significant benefit. After optimisation with cocoa genotype AMAZ 15, the same protocol was effective across all five additional cocoa genotypes tested. For the multiplication of clones, embryos regenerated following cryopreservation were used as explant sources, and vitrification was found to maintain their embryogenic potential.

General principles of climate change adaptation for biodiversity have been formulated, but do not help prioritize actions. This is inhibiting their integration into conservation planning. We address this need with a decision framework that identifies and prioritizes actions to increase the adaptive capacity of species. The framework classifies species according to their current distribution and projected future climate space, as a basis for selecting appropriate decision trees. Decisions rely primarily on expert opinion, with additional information from quantitative models, where data are available. The framework considers in-situ management, followed by interventions at the landscape scale and finally translocation or ex-situ conservation. Synthesis and applications: From eight case studies, the key interventions identified for integrating climate change adaptation into conservation planning were local management and expansion of sites. We anticipate that, in combination with consideration of socio-economic and local factors, the decision framework will be a useful tool for conservation and natural resource managers to integrate adaptation measures into conservation plans.

In the present study Tradescantia pallida micronucleus (Trad-MCN) bioassay was used to assess the genotoxicity of particulate matter with a mass median aerodynamic diameter less than 10 pm (PM(10)) in Tangara da Serra (MT), a Brazilian Amazon region that suffers the impact of biomass burning. The levels of PM (coarse and fine size fractions) and black carbon (BC) collected were also measured. Furthermore, the alkanes and polycyclic aromatic hydrocarbons (PAHs) were identified and quantified in the samples taken during the burning period by gas chromatography with flame ionization detection (GC-FID). The PM and BC results for both fractions indicate a strong correlation (p < 0.001). The analysis of alkanes indicates an anthropic influence. Retene was the most abundant PAH found, an indicator of biomass burning, and 12 other PAHs considered to be potentially mutagenic and/or carcinogenic were identified in this sample. The Trad-MCN bioassay showed a significant increase in micronucleus frequency during the period of most intense burning, possibly related to the mutagenic PAHs that were found in such extracts. This study demonstrated that Trad-MCN was sensitive and efficient in evaluating the genotoxicity of organic compounds from biomass burning. It further emphasizes the importance of performing chemical analysis, because changes in chemical composition generally have a negative effect on many living organisms. This bioassay (ex situ), using T. pallida with chemical analysis, is thus recommended for characterizing the genotoxicity of air pollution. Crown Copyright (C) 2011 Published by Elsevier Inc. CuO/CeO(2), CuO/Al(2)O(3) and CuO/CeO(2)-Al(2)O(3) catalysts, with CuO loading varying from 1 to 5 wt.%, were prepared by the citrate method and applied to the preferential oxidation of carbon monoxide in a reaction medium containing large amounts of hydrogen (PROX-CO). The compounds were characterized ex situ by X-ray diffraction, specific surface area measurements, temperature-programmed reduction and temperature-programmed reduction of oxidized surfaces; XANES-PROX in situ experiments were also carried out to study the copper oxidation state under PROX-CO conditions. These analyses showed that in the reaction medium the Cu(0) is present as dispersed particles. On the ceria, these metallic particles are smaller and more finely dispersed, resulting in a stronger metal-support interaction than in CuO/Al(2)O(3) or CuO/CeO(2)-Al(2)O(3) catalysts, providing higher PROX-CO activity and better selectivity in the conversion of CO to CO(2) despite the greater BET area presented by samples supported on alumina. It is also shown that the lower CuO content, the higher metal dispersion and consequently the catalytic activity. The redox properties of the ceria support also contributed to catalytic performance. This study presents the results of the degradation of the pesticide atrazine using electrochemical and photo-assisted electrochemical degradation techniques using SnO(2)-containing electrode of nominal composition electrodes of composition Ti/Ru(x)Sni-(x)O(2) (where X = 0.10, 0.15, 0.20, 0.25 and 0.30). The materials were characterized ex situ and in situ in order to correlate the observed atrazine removal rates with electrode morphology/composition. The results obtained demonstrate the effectiveness of the photo-assisted electrochemical degradation. Using purely electrochemical methods the rate of atrazine removal is almost zero at all the electrodes studied. However, the application of photo-assisted degradation results in almost complete atrazine removal in 1 h of electrolysis. The efficiency of atrazine degradation does not seem to be greatly affected by the electrode material or by SnO(2) content, but the overall COD removal is dependent on the SnO(2) content. Overall, the SnO(2)-containing electrodes do not reach the level of COD removal (maximum similar to 21%) seen for the Ti/Ru(0.3)Ti(0.2)O(2) electrode. Cu(II) ions previously coordinated with typical electroplating organic additives were investigated as an alternative source of metal for plating bath. The coordination complexes were isolated from reaction between CuSO(4) and organic additives as ligands (oxalate ion, ethylenediamine or imidazole). Deposits over 1010 steel were successfully obtained from electroplated baths using the complexes without any addition of free additives, at pH = 4.5 (H(2)SO(4)/Na(2)SO(4)). These deposits showed better morphologies than deposits obtained from CuSO(4) solution either in the absence or presence of oxalate ion as additive (40 mmol L(-1)), at pH = 4.5 (H(2)SO(4)/Na(2)SO(4))It is suggestive that the starting metal plating coordinated with additives influences the electrode position processes, providing deposits with corrosion potentials shifted over + 200 mV in 0.5 mol L(-1) NaCl (1 mV s(-1)). The resistance against corrosion is sensitive to the type of additive-complex used as precursor. The complex with ethylenediamine presented the best deposit results with the lowest pitting potential (-0.27 V vs 3.0 mol L(-1) CE). It was concluded that the addition of free additives to the electrodeposition baths is not necessary when working with previously coordinated additives. Thus, the complexes generated in ex-situ are good alternatives as plating precursors for electrodeposition bath. (C) 2009 Elsevier B.V. All rights reserved.

Introdução: A solução de UW (University of Wisconsin) tem sido utilizada como padrão para preservação de enxertos hepáticos para transplante ortotópico de fígado (TOF) desde 1990. Seu custo é alto, e não existem, em nosso meio, estudos clínicos que comparem a sua efetividade com a de outras soluções. A solução de HTK (histidina-triptofano-cetoglutarato) foi desenvolvida inicialmente para cardioplegia, porém estudos experimentais e alguns estudos clínicos retrospectivos demonstraram sua eficácia na preservação hepática, mesmo com tempos de isquemia prolongados. Objetivos: Comparar a efetividade das soluções de preservação de órgãos HTK e UW com relação as variáveis: disfunção primária do enxerto (DPE), tempo de isquemia fria (TIF), complicações de via biliar (CVB), alterações de provas funcionais hepáticas (PFH) e sobrevida do enxerto e dos pacientes. Método: Foram estudados os fígados de doadores de múltiplos órgãos, implantados nos receptores pela técnica de piggyback, segundo ordem cronológica de ingresso em lista de espera no RS no período janeiro de 2003 a agosto de 2004. As soluções de preservação HTK e UW foram utilizadas de forma randomizada em blocos. A perfusão na aorta foi feita com 4 litros de HTK ou 2 litros de UW e a perfusão venosa portal com 1 litro de ambas as soluções, utilizando-se 500 ml para perfusão venosa e arterial adicional ex-situ e armazenagem do enxerto. Realizou-se biópsia hepática em cunha do lobo esquerdo quando a esteatose macroscópica estava presente. A análise bioquímica sérica foi diária na primeira semana e, em 15 e 30 dias pós-operatórios. Resultados: Foram estudados 102 pacientes submetidos ao TOF, sendo 65 no grupo UW (63,7%) e 37 no grupo HTK (36,3%). As frequências de sexo, raça, estado hemodinâmico, o uso de vasopressores e a presença de esteatose nos doadores foram igual nos dois grupos (pα>,05). A idade média dos doadores foi de 38,1 anos (DP +-14,4) no grupo UW e de 44,6 anos (DP +-14,2) no HTK (pα=,036). A distribuição de sexo, raça, idade, etiologia da cirrose, re-transplante, hepatite fulminante, trombose portal e escore de Child-Pugh dos receptores foi igual nos dois grupos (pα>,05). O grupo HTK teve 8 casos (25,8%) de CVB (4 estenose, 2 fístulas e 2 lesões do tipo isquêmica) contra 5 casos (8,6%) do grupo UW (pα=,033) em 89 pacientes que completaram 4 meses de seguimento (OR=2,0; IC 95%=1,2 a 3,5). A média do TIF nos dois grupos foi semelhante (UW= 579,2 min.; HTK= 527,9 min. pα>,05) e não houve diferenças nas incidências de CVB, DPE e óbito com relação a TIF estratificados entre os grupos. Não houve variação nas medianas das PFH (pα>,05 para BT, AST, ALT, FA, GGT, LDH, Fator 5 e TP). A incidência de óbito foi similar em ambos os grupos: UW= 6 (9,4%) e HTK= 4 (11,1%). A incidências de DPE foi de 2,8% no grupo HTK (1 caso) e 9,4% no grupo UW (6 casos) (pα=0,15), dos quais 5 (71,4%) evoluíram para o óbito por não funcionamento primário do enxerto, com ou sem outras morbidades. Conclusão: As soluções de UW e HTK foram igualmente efetivas na preservação dos enxertos hepáticos de doadores cadavéricos na amostra analisada, considerando-se aspectos clínicos, laboratoriais e sobrevida dos pacientes e dos enxertos. A utilização rotineira da solução de HTK poderá diminuir os custos do TOF. A média de idade maior dos doadores e a utilização de um volume reduzido de solução podem ter contribuído para uma incidência maior de CVB no grupo HTK.

About 40% of the earth is occupied by tropical and subtropical forests, including 42% of dry forests, where there is Caatinga Bioma, contemplating tree forests and shrubs, with xerophytic characteristics. Study and conservations of Caatinga biologic diversity is one of the greatest challenges of Brazilian science because those are, proportionally, the less studied among natural areas, with most of the scientific effort centered in very few points around the main cities in the area and also because it is the less protected natural Brazilian area. The environmental degradation is constantly increasing and has its rhythm accelerated by the men appropriation to meet or not their own needs. Therefore, species conservation should be based in three principles: the use of natural resources by present generation, waste prevention and use of the natural resources to benefit the majority of the citizens. Among the strategies to species conservation, we can mention the ex situ conservation , in which the conservation of genetic resources may be realized outside of the natural environment in which the species occur, and in situ conservation , or, in other words, in the places where the species occur. In ex situ conservation, the germplasm collections are maintained in the field and/or in laboratories (conservation chambers), and this mainly conserves intraspecific diversity (genetic variance), the ex situ collections are continuously enriched by collection activities, introduction and germplasm interchange; the in situ conservation preserving ecosystems and habitats, maintaining and recovering native population of species of interest. So, the objective of this paper is the search for strategies to the conservation of Mimosa caesalpiniifolia B. (sabiá) using instruments of environmental perception and plant biotechnology, as mechanisms of in situ and ex situ conservation. To environmental perception, were realized open, semi-structured and qualitative interviews. The questions included socioeconomic data and knowledge of Sabiá specie. To plant biotechnology, Sabiá seed collection were realized in different location to formation of a germplasm bank. The specie micropropagation was made from nodal segment of plants from the matrizeiro. About the knowledge of rural populations and the use of Sabiá plant, some preferences occurred from speeches that the plant possesses a firm wood, not attacked by termites, legalized for exploration by the Brazilian environmental organ (IBAMA), and is a native specie. This research found the rural population has knowledge about Sabiá specie and the natural resources are exhausting. The proposal that the rural community brought was the donation of the Sabiá specie seeding initiating on the rain season, in which the seeding would be plated between the lots, in individual plantations. To the formation of a matrix bank, plant biothecnology brought answers favorable to Sabiá specie seeding, with the formation of multiple shoots
