The paper describes the superiority of monoclonal antibodies (MAb) over conventional polyclonal antisera. Studies undertaken indicate that Aeromonas hydrophila isolates are highly heterogenous and variation exists even between isolates from a farm, requiring a large number of MAbs for classification and use of information in vaccine development. However, some of the MAbs could be used for detection of homologous isolates in fish kidney by immunodot assay and evaluation and standardization of biofilm of A. hydrophila for oral vaccination in carps.

Microbial biofilms have been found to increase fish production in ponds by increasing heterotrophic production through periphyton proliferation on available substrates. In this paper, the role of substrate based microbial biofilm in the production of Cyprinus carpio and Labeo rohita grown in ponds is investigated, using an easily available and biodegradable agricultural waste product (sugarcane bagasse) as substrate.

在植物中大多数功能基因是以基因家族的形式存在的,而基因重复则是基因家族的一种重要的进化方式。很多基因往往是由重复事件产生形成不同的拷贝,进而分化形成基因家族。谷胱甘肽转移酶(GSTs)是一类古老、庞大、行使解毒、抗逆、信号转导等多种功能的一个基因家族。本研究以栽培水稻(Oryza sativa ssp. japonica c.v. Nipponbare)为研究材料,以栽培水稻的Phi类GST的5个基因(OsGSTF3、OsGSTF6、OsGSTF14、OsGSTF15、OsGSTF16)为研究对象,分析了它们的系统发生和起源历史、不同组织的差异性表达、编码蛋白质的功能差异等问题,探讨了基因重复后5个基因的功能变化,主要结果如下: 1. OsGSTF3、OsGSTF14、OsGSTF15、OsGSTF16由串联重复产生,而OsGSTF6则由DNA转座产生;它们起源时间早在稻属(Oryza)分化之前。 2. 对水稻不同部位组织的RT-PCR结果表明这5个基因在水稻中的特异性表达组织部位有较大差异:OsGSTF3基因在叶、叶鞘、茎、根4个部位均有大量表达;OsGSTF6基因仅在叶中有表达;OsGSTF14基因在叶鞘、茎2个部位中有表达;OsGSTF15基因在茎、根2个部位中有表达;OsGSTF16则在叶、茎、根3个部位中有表达。 3. 将这5个基因连接原核表达载体PET30a并转化大肠杆菌BL21(DE3),获得了高表达菌株。将表达菌株进行大量表达,表达形式分析显示OsGSTF3蛋白是可溶性表达,而其余4个蛋白以包涵体的形式表达。通过亲和层析获得了纯化的OsGSTF3融合蛋白,OsGSTF3融合蛋白对底物CDNB和NBD-Cl具有高活性,酶动力学分析显示OsGSTF3融合蛋白对GSH与NBD-Cl有较高的亲和力,热力学分析显示该蛋白在40℃以下是热稳定的。通过对包涵体进行洗涤、亲和层析获得了纯化的OsGSTF6、OsGSTF14、OsGSTF15、OsGSTF16的融合蛋白,OsGSTF14融合蛋白对NBD-Cl有微弱活性,OsGSTF15融合蛋白对NBC较高的活性,而没有检测到OsGSTF6与OsGSTF16融合蛋白的活性。

Studies on genetic improvement of penaeid prawns for the character higher tail weight using methods of selective breeding were undertaken. Prior to the actual breeding experiments it was necessary to find out the quantum of available variability in the character tail weight amongst the natural populations of Penaeus merguiensis from the Indian waters. Thirteen morphometric variables were measured and various statistical analyses were carried out. The tail weight showed almost double values of coefficient of variation in the females than the males (C.V. 20.37 and 11.08 respectively). The combination of the characters viz. sixth segment length (SSL), sixth segment depth (SSD) and posterior abdominal circumference (PAC) gave the highest R super(2) values. These variables were easy to measure and gave maximum variation in the character tail weight without sacrificing the breeders in the brood stock. The quantitative character tail weight was influenced by both genetic and environmental factors was statistically ascertained by applying 2-Factor analysis.

Mahseers (Tor khudree), at one time considered to be of single species, are now represented by 6 valid species distributed all over India. Studies on their food habits, ecology of spawning grounds, eggs, larval development and especially methods of artificial propagation have advanced in recent years. Transport of eggs of Tor khudree by air in moist cotton has been possible for easy distribution. Breeding of T. khudree not only by hypophysation, but even without it in small ponds by manipulation of flow of water, exercise and feed has also been possible. Fry and fingerlings of T. khudree are being distributed to many of the states in the country to help conserve the species.

Studies conducted in the laboratory and field have shown that iced fish can be preserved for longer periods in fresh and edible condition in conventional bamboo baskets by providing additional insulated linings of double layer of gunny and polythene or bitumen coated kraft paper. The quality of fish is adjudged by chemical and organoleptic evaluations.

Majority of the prawn catch of the Kerala coast comes from the inshore regions within the 10 fathom line. A bottom deposit formed of fine mud rich in humus is found to be the ideal condition for the penaeid prawns. Similarly the lobster is observed in large numbers at localised areas very near the coast in the crevices of rocks. Though it has been observed that prawns and lobsters exist in deep water as well under different conditions, our knowledge about these forms along the Indian coasts is very scanty, being restricted to the works of Spence Bate, Alcock, De Man and Ramadan. Recently, during the cruises of the University Research Vessel Conch off the Kerala Coast (1958-1963) two species of deep water prawns and one species of lobster were collected from depths 100 - 180 fathoms. Of these, Penaeopsis philippi is found in large numbers occupying an almost continuous bed extending from Anjengo to Mangalore, while P. rectacutus has a restricted appearance between Cochin and Calicut. However, some sort of year to year variation has also been observed regarding the abundance of the species at various stations. Peurulus sewelli has a more restricted distribution, the maximum number being found between Puvar and Cochin. Attempt has been made to correlate the occurrence of the species with the hydrological conditions at the bottom and the nature of the substratum. It is observed that Peurulus sewelli occupies a more or less hard bed formed of sand with shell fragments or stones and small percentage of silt, the bottom temperature at the stations varying from 11°C to l4°C. P. sewelli is a more or less permanent inhabitant of the edge of the continental shell off the Kerala Coast worthy of attention for detailed investigation with a view to explore the possibilities of commercial exploitation.

Heating conditions have been standardised for measurement of moisture in dry cured fish using infrared irradiation source of 150w. Results obtained are comparable to those obtained from standard air oven method (drying to a constant weight at l02°c), the mean deviation being less than two units. The method works equally well for fresh fish muscle.

Shark livers are considered as an important raw material providing a quality fish oil. It has been reported to aid white — blood-cell production and act as an active ingredient in hemorrhoid treatments. It is also reported that liver oil as a good supplement of vitamin A and poly-unsaturated fatty acids which are important to the development of brain cells in human. Freshness of livers is very important to extract better quality oil. In Sri Lanka, the annual shark production amounts to 8000t, however the quality of livers collected from landing sites has not being measured yet. Present study was conducted to evaluate the quality of silky (Charcarninus fakiformis) shark livers available in Negombo and Beruwala landing sites in the West Coast of Sri Lanka and also to study the relationship between organoleptic and bio-chemical correlation on freshness of shark livers. Liver samples which were collected from landing sites in the West coast of Sri Lanka, were evaluated for external and internal colour, texture and odour. Total volatile nitrogen (TVN), pH value, free fatty acid (FFA%) and peroxide (PV) values of livers were also determined to assess quality. According to the organoleptic scoring system 4.3% of liver samples were categorized as best in quality while 30.4%, 56.5% and 8.7% rated as good, medium and poor in quality respectively at the Negombo and Beruwala landing sites. Bio-chemical analysis showed that the better quality livers had the highest score for sensory evaluation and low values for TVN, FFA and peroxide value while low quality livers gave low score for sensory evaluation and high TVN, FFA, peroxide values. Correlation coefficient of organoleptic scores against total volatile nitrogen value, pH value, free fatty acid % and peroxide value of shark livers were determined by statistical analysis.
