The lanthanide binuclear helicate [Eu(2)(L(C2(CO(2)H)))(3)] is coupled to avidin to yield a luminescent bioconjugate EuB1 (Q = 9.3%, tau((5)D(0)) = 2.17 ms). MALDI/TOF mass spectrometry confirms the covalent binding of the Eu chelate and UV-visible spectroscopy allows one to determine a luminophore/protein ratio equal to 3.2. Bio-affinity assays involving the recognition of a mucin-like protein expressed on human breast cancer MCF-7 cells by a biotinylated monoclonal antibody 5D10 to which EuB1 is attached via avidin-biotin coupling demonstrate that (i) avidin activity is little affected by the coupling reaction and (ii) detection limits obtained by time-resolved (TR) luminescence with EuB1 and a commercial Eu-avidin conjugate are one order of magnitude lower than those of an organic conjugate (FITC-streptavidin). In the second part of the paper, conditions for growing MCF-7 cells in 100-200 microm wide microchannels engraved in PDMS are established; we demonstrate that EuB1 can be applied as effectively on this lab-on-a-chip device for the detection of tumour-associated antigens as on MCF-7 cells grown in normal culture vials. In order to exploit the versatility of the ligand used for self-assembling [Ln(2)(L(C2(CO(2)H)))(3)] helicates, which sensitizes the luminescence of both Eu(III) and Tb(III) ions, a dual on-chip assay is proposed in which estrogen receptors (ERs) and human epidermal growth factor receptors (Her2/neu) can be simultaneously detected on human breast cancer tissue sections. The Ln helicates are coupled to two secondary antibodies: ERs are visualized by red-emitting EuB4 using goat anti-mouse IgG and Her2/neu receptors by green-emitting TbB5 using goat anti-rabbit IgG. The fact that the assay is more than 6 times faster and requires 5 times less reactants than conventional immunohistochemical assays provides essential advantages over conventional immunohistochemistry for future clinical biomarker detection.

La deformación plástica puede inducir a la transformación de la austenita a martensita en los aceros inoxidables austeníticos metaestables. Para analizar este hecho, el inoxidable austenítico metaestable grado AISI 301 LN fue estudiado en dos condiciones diferentes: recocido y laminado en frío. En el primer caso, el acero era completamente austenítico, mientras que después de la laminación presentaba un importante porcentaje de α’-martensita. Se evaluó el cambio de fase cuando el acero es sometido a ensayos monotónicos y cíclicos, así como cuando ha sido modificada la superficie mediante el granallado o se han realizado tratamientos térmicos de reversión. Se utilizaron diferentes técnicas de caracterización microestructural para detectar y cuantificar la martensita, como microscopía óptica, difracción de rayos-X (DRX) y difracción de electrones retrodispersados (EBSD); como también de caracterización mecánica para evaluar el comportamiento de los aceros, trabajo esencial de fractura (TEF), conformabilidad, fatiga de alto número de ciclos (HCF) y nanoindentación. Los resultados mostraron un incremento en la resistencia mecánica del acero laminado en comparación al acero recocido; este hecho está relacionado con la presencia de martensita originada por la laminación. Sin embargo, en términos de deformación y endurecimiento el acero recocido presenta un mejor desempeño como consecuencia del elevado porcentaje de fase austenítica. Así mismo, revertir la martensita de laminación a austenita y refinar la austenita presente permite obtener un acero con una propiedades mecánicas similares a cuando esta en la condición laminado.

The sentinel or tumor-draining lymph node (tdLN) serves as a metastatic niche for many solid tumors and is altered via tumor-derived factors that support tumor progression and metastasis. tdLNs are often removed surgically, and therapeutic vaccines against tumor antigens are typically administered systemically or in non-tumor-associated sites. Although the tdLN is immune-suppressed, it is also antigen experienced through drainage of tumor-associated antigens (TAA), so we asked whether therapeutic vaccines targeting the tdLN would be more or less effective than those targeting the non-tdLN. Using LN-targeting nanoparticle (NP)-conjugate vaccines consisting of TAA-NP and CpG-NP, we compared delivery to the tdLN versus non-tdLN in two different cancer models, E.G7-OVA lymphoma (expressing the nonendogenous TAA ovalbumin) and B16-F10 melanoma. Surprisingly, despite the immune-suppressed state of the tdLN, tdLN-targeting vaccination induced substantially stronger cytotoxic CD8+ T-cell responses, both locally and systemically, than non-tdLN-targeting vaccination, leading to enhanced tumor regression and host survival. This improved tumor regression correlated with a shift in the tumor-infiltrating leukocyte repertoire toward a less suppressive and more immunogenic balance. Nanoparticle coupling of adjuvant and antigen was required for effective tdLN targeting, as nanoparticle coupling dramatically increased the delivery of antigen and adjuvant to LN-resident antigen-presenting cells, thereby increasing therapeutic efficacy. This work highlights the tdLN as a target for cancer immunotherapy and shows how its antigen-experienced but immune-suppressed state can be reprogrammed with a targeted vaccine yielding antitumor immunity.

Mouse models of Leishmania major infection have shown that a predominant CD4(+) T helper type 1 cell (Th1) response leads to protection, while T helper type 2 cell (Th2) predominance confers susceptibility. Dendritic cells (DCs) are antigen-presenting cells that orchestrate the T cell response. The immune response to L. major involves direct antigen presentation by migrating DCs or transfer of antigens to resident DCs to prime T cells. In this review, we discuss the timing and consequences of antigen presentation by DC subsets and how this affects Leishmania susceptibility. We propose a model where dermal DCs and Langerhans cells play a role early in infection, followed by inflammatory monocyte-derived DC and lymph node (LN)-resident DCs at later time points of infection to establish the resistant Th1 response.

Mouse mammary tumor virus (MMTV) encodes a superantigen (SAg) that promotes stable infection and virus transmission. Upon subcutaneous MMTV injection, infected B cells present SAg to SAg-reactive T cells leading to a strong local immune response in the draining lymph node (LN) that peaks after 6 d. We have used the reverse transcriptase inhibitor 3'-azido-3'-deoxythymidine (AZT) to dissect in more detail the mechanism of SAg-dependent enhancement of MMTV infection in this system. Our data show that no detectable B or T cell response to SAg occurs in AZT pretreated mice. However, if AZT treatment is delayed 1-2 d after MMTV injection, a normal SAg-dependent local immune response is observed on day 6. Quantitation of viral DNA in draining LN of these infected mice indicates that a 4,000-fold increase in the absolute numbers of infected cells occurs between days 2 and 6 despite the presence of AZT. Furthermore MMTV DNA was found preferentially in surface IgG+ B cells of infected mice and was not detectable in SAg-reactive T cells. Collectively our data suggest that MMTV infection occurs preferentially in B cells without SAg involvement and is completed 1-2 d after virus challenge. Subsequent amplification of MMTV infection between days 2 and 6 requires SAg expression and occurs in the absence of any further requirement for reverse transcription. We therefore conclude that clonal expansion of infected B cells via cognate interaction with SAg-reactive T cells is the predominant mechanism for increasing the level of MMTV infection. Since infected B cells display a memory (surface IgG+) phenotype, both clonal expansion and possibly longevity of the virus carrier cells may contribute to stable MMTV infection.

PURPOSE: Local delivery of therapeutic molecules encapsulated within liposomes is a promising method to treat ocular inflammation. The purpose of the present study was to define the biodistribution of rhodamine-conjugated liposomes loaded with vasoactive intestinal peptide (VIP), an immunosuppressive neuropeptide, following their intravitreal (IVT) injection in normal rats. METHODS: Healthy seven- to eight-week-old Lewis male rats were injected into the vitreous with empty rhodamine-conjugated liposomes (Rh-Lip) or with VIP-loaded Rh-Lip (VIP-Rh-Lip; 50 mM of lipids with an encapsulation efficiency of 3.0+/-0.4 mmol VIP/mol lipids). Twenty-four h after IVT injection, the eyes, the cervical, mesenteric, and inguinal lymph nodes (LN), and spleen were collected. The phenotype and distribution of cells internalizing Rh-Lip and VIP-Rh-Lip were studied. Determination of VIP expression in ocular tissues and lymphoid organs and interactions with T cells in cervical LN was performed on whole mounted tissues and frozen tissue sections by immunofluorescence and confocal microscopy. RESULTS: In the eye, 24 h following IVT injection, fluorescent liposomes (Rh-Lip and VIP-Rh-Lip) were detected mainly in the posterior segment of the eye (vitreous, inner layer of the retina) and to a lesser extent at the level of the iris root and ciliary body. Liposomes were internalized by activated retinal Müller glial cells, ocular tissue resident macrophages, and rare infiltrating activated macrophages. In addition, fluorescent liposomes were found in the episclera and conjunctiva where free VIP expression was also detected. In lymphoid organs, Rh-Lip and VIP-Rh-Lip were distributed almost exclusively in the cervical lymph nodes (LN) with only a few Rh-Lip-positive cells detected in the spleen and mesenteric LN and none in the inguinal LN. In the cervical LN, Rh-Lip were internalized by resident ED3-positive macrophages adjacent to CD4 and CD8-positive T lymphocytes. Some of these T lymphocytes in close contact with macrophages containing VIP-Rh-Lip expressed VIP. CONCLUSIONS: Liposomes are specifically internalized by retinal Müller glial cells and resident macrophages in the eye. A limited passage of fluorescent liposomes from the vitreous to the spleen via the conventional outflow pathway and the venous circulation was detected. The majority of fluorescent liposomes deposited in the conjunctiva following IVT injection reached the subcapsular sinus of the cervical LN via conjuntival lymphatics. In the cervical LN, Rh-Lip were internalized by resident subcapsular sinus macrophages adjacent to T lymphocytes. Detection of VIP in both macrophages and T cells in cervical LN suggests that IVT injection of VIP-Rh-Lip may increase ocular immune privilege by modulating the loco-regional immune environment. In conclusion, our observations suggest that IVT injection of VIP-loaded liposomes is a promising therapeutic strategy to dampen ocular inflammation by modulating macrophage and T cell activation mainly in the loco-regional immune system.

La réflexion sur la biographie a connu un renouvellement important ces vingt dernières années dans plusieurs disciplines. Florissant sur le marché ditorial, ce genre ancien trouve aujourd'hui des formes nouvelles, nourries de la galaxie des sciences humaines contemporaines, comme en témoigne l'ouvrage de François Dosse, Le Pari biographique (La Découverte, 2005). Genre fondateur en histoire de l'art, la biographie est pratiquée peu ou prou dans la plupart des disciplines des lettres. Bonne raison pour se pencher de manière interdisciplinaire sur le genre et sa pratique. La vogue des éloges académiques au XVIIIe siècle, et la profusion du discours biographique sur les artistes à l'orée du Romantisme ont ouvert la voie à la fameuse «méthode biographique» de Sainte-Beuve, et à sa reconversion dans les programmes d'enseignement, par le biais de Gustave Lanson et de ses émules. Inculqué à des générations d'élèves, devenu une structure invisible de l'entendement universitaire dans les disciplines littéraires, le couple « la vie et l'oeuvre » a été sévèrement reconsidéré, au XXe siècle, par les approches formalistes, structuralistes ou sociologiques. Qu'en est-il de ce genre actuellement? Comment les travaux de littéraires, d'historiens, d'historiens de l'art, de philosophes envisagent-ils les données biographiques?

Le but de cet article est de montrer à l'aide d'un exemple concret comment les catégories « biographie » et « hagiographie » posent problème dans la discipline d'histoire comparée des religions. Il ne s'agit pas de traiter de la biographie ou de l'hagiographie en tant que genres littéraires, mais bien de voir comment se différencient, dans plusieurs types de récits sur la vie et l'oeuvre, les éléments hagiographiques et les éléments biographiques. Pour ce faire, je me concentrerai sur un personnage : la poétesse mystique Mīrābāī, qui a vécu au XVIe siècle de notre ère en Inde du Nord. Ce cas me paraît être intéressant puisque nous n'avons aucun matériel qui permette de rendre des conclusions définitives sur la vie de ce personnage qu'on postule comme historique. Mīrābāī n'a pas laissé de journal intime, de chroniques, ni d'autobiographie. Elle était une femme dévouée au dieu Krishna, une poétesse visant à vivre pleinement sa relation à une entité supra- humaine postulée culturellement, le dieu masculin, Krishna. Ce type de dévotion amoureuse, impliquant une relation d'amour total et d'abandon pour une divinité de prédilection, est appelé bhakti.

Se ha analizado la variación oceanográfica y las respuestas de los ensambles de microfitoplancton, mesozooplancton, ictioplancton y macrobentos en las áreas costeras (<20 mn) frente a Paita (05°S) y a San José (06°45’S) durante el período 1994 a 2002. La variación oceanográfica presentó componentes a varias escalas temporales, moduladas por el ciclo ENOS, la propagación de ondas atrapadas hacia la costa y la intensificación estacional del afloramiento costero. La sucesión de los eventos El Niño (EN) 1997-98 y La Niña (LN) 1998-99 presentó características bien diferenciadas en las condiciones físicas superficiales y en la estructura vertical de la columna de agua. El evento EN 1997-98 fue antecedido por el impacto de una onda Kelvin en febrero de 1997, provocando anomalías positivas de temperatura, profundización de la estructura vertical y presencia de algunos indicadores de masas de agua cálida en el plancton, entre febrero y abril de 1997. Estas condiciones se mantuvieron, o se acentuaron, hasta el final del evento. El evento LN 1998-99 se caracterizó por la ausencia de masas de agua cálidas cerca de la costa y la dominancia de aguas costeras frías, la no propagación de ondas Kelvin, la posición somera de aguas frías y pobres en oxígeno, así como la hegemonía de indicadores planctónicos de aguas costeras frías. Estacionalmente, durante otoño-invierno tendieron a desarrollarse condiciones subsuperficiales más oxigenadas (una oxiclina más profunda), mientras que durante el verano las condiciones tendieron a ser menos oxigenadas (hipóxicas, con una oxiclina más somera). Tal patrón no responde a la estacionalidad del afloramiento costero y más bien coincide con la dinámica esperada de la Extensión Sur de la Corriente de Cromwell (ESCC). En general, se determinó un muy buen ajuste de los rangos de tolerancia de algunos organismos planctónicos a las características de las masas de agua dominantes en la capa superficial: Aguas Costeras Frías (ACF), Aguas Ecuatoriales Superficiales (AES) y Aguas Subtropicales Superficiales (ASS), validando la utilidad de estas especies como eficaces indicadores biológicos de masas de agua. Se determinaron los rangos de tolerancia en temperatura y salinidad de los dinoflagelados Protoperidinium obtusum (ACF), Ceratium breve (AES) y Ceratium praelongum (ASS), así como de los copépodos Centropages brachiatus (ACF), Eucalanus inermis (ACF), Centropages furcatus (AES) y Mecynocera clausi (ASS), entre otras. Los indicadores presentaron variaciones en su distribución a lo largo del período estudiado. Los indicadores de ACF fueron detectados durante la mayor parte del estudio, pero ocurrieron hechos sobresalientes durante EN 1997-98: (a) entre los dinoflagelados, Goniodoma polyedricum alcanzó su mayor frecuencia en San José y Pyrocystis lunula frente a Paita; (b) el copépodo Centropages furcatus (AES) incrementó su abundancia frente a Paita y fue hallado frente a San José; (c) se evidenciaron cambios en la composición específica del plancton, detectándose el ingreso de especies no residentes y aumento de la riqueza de especies; (d) las biomasas fitoplanctónica y zooplanctónica tendieron a mostrar una relación directa bajo condiciones neutras del ENOS; sin embargo, al ocurrir variaciones ambientales (EN y LN) presentaron una tendencia contraria; (e) las comunidades del macrobentos en estas dos áreas, mostraron disminuciones significativas en los parámetros comunitarios, contrastando con la respuesta de la macrofauna bentónica frente a la costa central. Este comportamiento frente a Paita pudo obedecer a la alteración del ambiente sedimentario por las muy altas des- cargas del río Chira; y frente a San José, pudo resultar de la disminución local de la producción primaria y del flujo de alimento particulado al bentos. Se sugiere que la disponibilidad de alimento, influenciada por los procesos erosivos sobre el fondo, marca una diferencia clave en la dinámica de estas comunidades en relación a las registradas frente a la costa central, ya que estas últimas habitan en áreas donde pre- dominan los procesos deposicionales y la acumulación de materia orgánica en los sedimentos. Los anfípodos gamáridos, especialmente de la familia Ampeliscidae, mostraron ser más sensibles a los cambios ambientales en el fondo (interfase sedimento-agua) al disminuir significativamente sus poblaciones, en ambas áreas costeras.

Background/Introduction: There is little information regarding intergenerational trends in obesity levels in Switzerland. We aimed at assessing generational differences in obesity levels. Methods: Data from MONICA (1984-1986) and CoLaus-(2003-2006) surveys. Analyses were stratified by gender and age groups (35-44, 45-54, 55-64 and 65-74 years). Results: No changes were found for body mass index (BMI) between surveys (26.2±3.4 vs. 26.6±4.0 kg/m2 in men and 24.8±4.3 vs. 25.1±4.8 kg/m2 in women, for MONICA and CoLaus, respectively). ln men, the prevalence of overweight decreased from 48.6% to 46.0% and the prevalence of obesity increased slightly from 12.4% to 16.7% (p=NS). ln women, the prevalence of overweight decreased from 29.4% to 28.4% and the prevalence of obesity increased slightly from 12.9% to 14.5% (p=NS). After multivariate adjustment on age, education and smoking levels, the odds ratio (OR) and (95% confidence interval) of being obese in 2003-6 relative to 1984-6 was 1.36 (1.01-1.83) in men and 1.44 (1.07-1.93) in women, while no significant trend was found for overweight. After stratifying for age, no increase in BMI levels was found for bath genders. Obesity levels increased in participants aged 35-44 years (from 6.2% to 11.5% in men and from 4.9% to 10.0% in women, p<0.001) and 45-54 years (from 6.2% to 14.5% in men and 4.9% to 14.5% in women, p<0.001 ). After multivariate adjustment on age, education and smoking levels, the increase in obesity levels was signifiant in women aged 35-44, OR=2.10 (1.02-4.30), while a similar, albeit nonsignificant trend was obser11ed in men: 1.85 (0.97-3.51 ). Conclusion: BMI levels appear to have levelled off in Switzerland, but the prevalence of obesity is still on the rise. The increase in obesity levels among the youngest generations is of particular concern.

A capacidade de troca de cátions (CTC) e a proporção dos principais cátions retidos nas cargas negativas do solo são de grande importância para a caracterização química e para o diagnóstico da fertilidade do solo. Essas características, juntamente com o Al, o Ca e o Mg trocáveis, foram incluídas na análise de solo realizada pelos laboratórios oficiais do Rio Grande do Sul e de Santa Catarina, a partir de 1994. Este trabalho avaliou a relação entre o valor de H + Al e o pH SMP, visando estabelecer uma equação para a estimativa do valor de H + Al (acidez potencial), o qual pode ser utilizado no cálculo da CTC (método da soma de cátions trocáveis). O trabalho foi realizado nos laboratórios de solos da Universidade de Passo Fundo (UPF) e da Universidade Federal do Rio Grande do Sul (UFRGS), em 1995, constando da determinação do teor de H + Al, extraído com acetato de cálcio 1,0 mol L-1, pH 7,0, e do pH SMP em um grupo de 113 amostras de solo. Os resultados obtidos foram reunidos com os de outros pesquisadores, procedendo-se à análise de regressão para um conjunto de 250 amostras representativas de solos dos dois estados. Observou-se relação significativa entre o valor de H + Al e o pH SMP. Concluiu-se que as equações log (H + Al) = 3,9014 - 0,391 SMP (R² = 0,90) ou ln (H + Al) = 8,9832 - 0,9004 SMP (R² = 0,90), com os valores de H + Al expressos em mmol c dm-3, podem ser utilizadas para a estimativa do teor de H + Al pelo uso do pH SMP.

Through a rational design approach, we generated a panel of HLA-A*0201/NY-ESO-1(157-165)-specific T cell receptors (TCR) with increasing affinities of up to 150-fold from the wild-type TCR. Using these TCR variants which extend just beyond the natural affinity range, along with an extreme supraphysiologic one having 1400-fold enhanced affinity, and a low-binding one, we sought to determine the effect of TCR binding properties along with cognate peptide concentration on CD8(+) T cell responsiveness. Major histocompatibility complexes (MHC) expressed on the surface of various antigen presenting cells were peptide-pulsed and used to stimulate human CD8(+) T cells expressing the different TCR via lentiviral transduction. At intermediate peptide concentration we measured maximum cytokine/chemokine secretion, cytotoxicity, and Ca(2+) flux for CD8(+) T cells expressing TCR within a dissociation constant (K(D)) range of ∼1-5 μM. Under these same conditions there was a gradual attenuation in activity for supraphysiologic affinity TCR with K(D) < ∼1 μM, irrespective of CD8 co-engagement and of half-life (t(1/2) = ln 2/k(off)) values. With increased peptide concentration, however, the activity levels of CD8(+) T cells expressing supraphysiologic affinity TCR were gradually restored. Together our data support the productive hit rate model of T cell activation arguing that it is not the absolute number of TCR/pMHC complexes formed at equilibrium, but rather their productive turnover, that controls levels of biological activity. Our findings have important implications for various immunotherapies under development such as adoptive cell transfer of TCR-engineered CD8(+) T cells, as well as for peptide vaccination strategies.
