Objetivou-se verificar qual o melhor estádio embrionário para o cultivo de embriões imaturos oriundos de frutos provenientes de hibridação entre 'Pêra Rio' x 'Poncã' , bem como o efeito de diferentes concentrações do meio de cultura MT. Os embriões em diferentes estádios de desenvolvimento (globulares, torpedo e cordiforme) foram excisados e inoculados em tubos de ensaio contendo 15 mL do meio MT com diferentes concentrações (0; 50; 100 e 150% da composição original e acrescido de 50 g.L-1 de sacarose). Após a inoculação, os embriões foram incubados à 27±1ºC, fotoperíodo de 16 horas e irradiância de 32 mmol.m-2.s-1. Após 90 dias, avaliou-se o comprimento da parte aérea e do sistema radicular, massa fresca e número de folhas das plântulas. Melhor desenvolvimento dos embriões imaturos foi obtido em estádio cotiledonar e com a concentração de 150% do meio MT.

O presente trabalho foi conduzido com o objetivo de avaliar o efeito da utilização de diferentes fontes de gonadotrofinas para maturação in vitro dos oócitos bovinos fecundados e desenvolvidos in vitro sobre as taxas de clivagem (TC) e de blastocistos (TBL). Oócitos imaturos provenientes de ovários de vacas de abatedouro foram submetidos a maturação in vitro sob diferentes condições: meio TCM 199, acrescido de 10% de soro de vaca em estro (SVE), aditivos, hepes, NaHCO3, piruvato de sódio, antibióticos (meio B-199), 20 UI/mL de PMSG e 10 UI/mL de hCG (PMSG/hCG) ou meio B-199, acrescido de 5 mig/mL de FSH e 5 mig/mL de LH (FSH/LH). Seguidos 24 h de cultura a 38,5ºC em atmosfera com 5% de CO2, os oócitos maturos foram incubados com sêmen descongelado durante 18 a 21 horas. Após esse período, os oócitos foram transferidos para placas contendo microgotas de meio Ménezo suplementado com 10% de SVE e células epiteliais do oviduto bovino em suspensão, cobertas com óleo de silicone, os quais permaneceram em cultura por mais 9 dias. Os dados foram analisados pelo teste do Qui-quadrado. A TC e a TBL, para PMSG/hCG e FSH/LH, foram 60 e 13,9% e 61,2 e 10,6%, respectivamente. Não houve diferença entre os tratamentos com relação a TC ou a TBL. Esses resultados sugerem que ambas as fontes de gonadotrofinas podem ser utilizadas para maturação in vitro dos oócitos fecundados e desenvolvidos in vitro.

The effect of four extracts from neem seeds (Azadirachta indica) containing 2000, 5000, 9000 and 10,000 ppm of azadirachtin A (AZA), quantified by high-performance liquid chromatography (HPLC) and diluted to 1.25%; 2.5%; 5.0%; 10.0% and 12.8% was verified by in vitro tests with engorged females and larvae of the cattle tick Rhipicephalus micro plus. The results from the bioassays with the engorged females showed that the main toxic effect of the extracts was reduction of the reproductive parameters, with a sharp drop in the number of eggs laid and the hatching rate, mainly when the extracts were diluted to 10.0% and 12.8%. The product effectiveness (PE) calculations for all the solutions tested showed that the AZA solution at 10,000 ppm (N10) was the most effective. However, statistical analysis of the PE data obtained for the proportional AZA concentrations in the different diluted extracts showed significance (P<0.05) of the effects included in the model (extract dilution, principle effect (classificatory) of the assay (extract) and the interaction between the two), indicating significant variations due to the dilution, the test and the interaction between the two factors in the tests with engorged females. For solutions N2, N5, and N9, it was not possible to estimate LC(90) values in the dilution range tested. The lowest LC(50) was observed for extract N5, and although extract N10 was the only extract for which the LC(90) could be estimated within the range tested, the LC(50) was higher than for N5 and N9. These results suggest that substances other than AZA present in the extracts influenced the efficacy, especially up to a certain LC range. In the tests with larvae, no mortality was observed, indicating zero effectiveness of all the extracts tested. The results of the tests with engorged females showed that the neem extracts had acaricide activity, inhibiting egg laying and the larval hatching rate. Complementary studies are necessary to develop new methods to isolate and/or identify other substances besides AZA contained in this plant, to enable using products made from it as acaricides. Avaliou-se o crescimento micelial in vitro das linhagens ABI-05/03, ABI-06/04, ABI-04/02, ABI-06/05 e ABI-01/01 de Agaricus bisporus em meios de cultura sólidos à base de composto. As avaliações foram realizadas por meio de medições de quatro diâmetros das colônias, a cada 48h, durante 12 dias de incubação, no escuro, a 20 e 25ºC. O delineamento experimental foi o de blocos casualizados, com uso do teste de Tukey para a comparação das médias. Co m base nos resultados obtidos, verificou-se que: o crescimento micelial de A. bisporus é influenciado pela temperatura de incubação; a temperatura de 25ºC foi mais favorável para o crescimento micelial de todas as linhagens de A. bisporus; na temperatura de 20ºC, o melhor crescimento foi obtido com as linhagens ABI-06/05 e ABI-01/01; na temperatura de 25ºC, a linhagem ABI-01/01 apresentou crescimento significativamente maior que todas as demais.

A mancha bacteriana do maracujá, causada pela bactéria Xanthomonas axonopodis pv. passiflorae, ocorre em todas as regiões produtoras do País, sendo responsável por grandes perdas econômicas na cultura do maracujazeiro-amarelo. O presente trabalho teve como objetivos testar a eficiência de argila silicatada na inibição da bactéria X. axonopodis pv. passiflorae in vitro e no controle preventivo e curativo da mancha bacteriana em mudas de maracujazeiro-amarelo. A argila silicatada foi adicionada ao meio de cultura batata-dextrose-ágar fundente, nas concentrações de 0,0; 0,5; 1,0; 1,5 e 2,0%; vertido em placas de Petri. Após resfriamento do meio, repicou-se a suspensão bacteriana (10(7) UFC.mL-1) com uma alça, incubando-se as placas a 28 °C por três dias, quando se avaliou o crescimento bacteriano. Posteriormente, o produto, nas mesmas concentrações citadas, foi pulverizado em mudas de maracujá 'Afruvec' de forma preventiva ou curativa. A inoculação da bactéria foi realizada através de pulverização foliar da suspensão bacteriana (10(7) UFC.mL-1), 24 h antes ou após os tratamentos curativo e preventivo, respectivamente. A severidade da doença foi avaliada com auxílio de uma escala diagramática nas quatro primeiras folhas verdadeiras contadas de baixo para cima. Nas concentrações avaliadas, a argila silicatada inibiu a bactéria in vitro e os sintomas da mancha bacteriana no tratamento curativo, enquanto no tratamento preventivo, controle significativo foi obtido a partir de 1,0% de argila silicatada. Com base nestes resultados, a argila silicada pode ser recomendada, na concentração de 1,0-2,0%, para o controle da mancha bacteriana do maracujazeiro em pulverizações foliares.

The bacterial spot in yellow passion fruit plants, caused by the bacteria Xanthomonas axonopodis pv. passiflorae, occurs in all producing areas of the country, and is responsible for great economic losses in the culture of passion fruit. This study aimed to test the efficiency of the silicate clay in the inhibition of the bacteria Xanthomonas axonopodis pv. passiflorae in vitro, and in both preventive and curative control of the bacterial spot in seedlings of yellow passion fruit plants. The silicate clay was added to the growth medium at concentrations of. 0.5, 1.0, 1.5 and 2.0%, placed in Petri dishes. After the culture medium was cooler, the bacterial suspension was inoculates (10(7) UFC.mL(-1)) with a handle, and left incubating at 28 degrees C for three days, and then the bacterial growth was evaluated. Subsequently, the product at the same concentrations above was sprayed on seedlings of 'Afruvec' passion fruit, as preventive or curative. The inoculation of the bacteria was made by foliar spraying of bacterial suspension (10(7) ufc.mL(-1)), 24 hours before or after the curative and preventive treatments, respectively. The severity of the disease was measured comparing each four true leaves from bottom up, with a diagrammatic scale. In the concentrations evaluated, the silicate clay inhibited both bacteria in vitro and symptoms of bacterial spot in the curative treatment. In preventive treatment, significant results were obtained using more than 1.0% of clay silicates. Based on these results, the clay silicate can be recommended, the concentration of 1.0-2.0% for the control of bacterial spot of passion fruit plants, in foliar sprays.

Cryopreservation is a process where cells or biological tissues are preserved by freezing at very low temperatures and aims to cease reversibly, in a controlled manner, all the biological functions of living tissues, i.e., maintain cell preservation so that it can recover with high degree of viability and functional integrity. This study aimed to evaluate the influence of cryopreservation on the mesenchymal stem cells originating from the periodontal ligament of human third molars by in vitro experiments. Six healthy teeth were removed and the periodontal cells grown in culture medium containing α-MEM supplemented with antibiotics and 15% FBS in a humidified atmosphere with 5% CO2 at 37° C. Cells isolated from each sample were divided into two groups: Group I - immediate cell culture (not fresh cryopreserved cells) and Group II - cell cryopreservation, during a period of 30 days. Analyses of rates of cell adhesion and proliferation in different groups were performed by counting the cells adhered to the wells, in intervals of 24, 48 and 72 hours after the start of cultivation. The number of cells in each well was obtained by counting viable cells with the use of hemocytometer and the method of exclusion of cells stained by trypan blue. The difference between groups for each of the times was analyzed by Wilcoxon test. Regarding the temporal evolution for each group, analysis was done by Friedman's test to verify the existence of differences between times and, when it existed, the Wilcoxon penalty was applied. The results showed no statistically significant difference between the two groups analyzed in this study. Therefore, we conclude that the cryopreservation process, after a period of 30 days, did not influence the cell type studied, and there was no difference in growth capacity in vitro between the groups

Photodynamic therapy (PDT) has been proposed as an alternative method for the treatment of biofilm-dependent oral diseases like dental caries. This therapy consists of simultaneous action of a visible light (L) and a photosensitizer (FS) in the presence of oxygen, which leads to production of different reactive oxygen species that can interact with the bacterial cell components, and promote cell death. This study aims to evaluate the antimicrobial action of PDT on oral bacteria in suspension, as well as the formation of mono and multi-species biofilms, in vitro, from a standard strain of Streptococcus mutans (ATCC 25175) and saliva samples, respectively. The dye methylene blue (MB) and toluidine blue (TB) were used at a concentration of 100 mg/ L and activated by halogen light (600 to 750 nm) from a modified hand held photopolymerizer (Ultralux ®, Dabi Atlante, Ribeirão Preto , São Paulo, Brazil.). Planktonic cultures were prepared and submitted to different experimental conditions: 1. PDT using TB 2. PDT using MB, 3. L+ FS- , 4. TB + L - ; 5. MB+ L-; 6. L- FS- (negative control) and 7. administration of 0.12% chlorhexidine digluconate (positive control) (Periogard ®, Colgate-Palmolive Company, New York, USA). The immediate and mediated action of PDT on bacterial suspensions, as well as its effect on biofilm formation were observed from the number of colony-forming units per milliliter (CFU/mL) and measures optical density (OD). The data were statistically analyzed using the Kruskal-Wallis and Mann-Whitney test for the significance level of 5%. According to the results, the PDT showed no antibacterial action on suspensions of S. mutans, regardless of the dye used. PDT with MB activated by halogen light was able to reduce 86.6% CFU/mL multi-species planktonic cultures, however, this reduction was not significant (p > 0.05). PDT showed antibacterial effect, mediate on multi-species planktonic cultures with TB (p < 0.001) and MB (p < 0.001), activated by halogen light. PDT was able to prevent the formation of multispecies biofilm, through the activation of TB by halogen light (p = 0.01). We conclude that activation of the dye toluidine blue and methylene blue, by halogen light (PDT) showed antimicrobial activity, compared to multi-species planktonic cultures prepared from saliva samples

The aim of this study was to comparatively evaluate the mechanical strength of squared and rectangular 2.0 mm system miniplates comparing them to the standard configuration with 2 straight miniplates in stabilizing fractures in the anterior mandible. Ninety synthetic polyurethane mandible replicas were used in mechanical test. The samples were divided into six groups of three different methods for fixation. Groups 1, 2 and 3 showed complete fractures in symphysis, characterized by a linear separation between the medial incisor, and groups 4, 5 and 6 showed complete fractures in parasymphysis with oblique design. Groups 1 and 4 were represented by the standard technique with two straight miniplates parallel to each other. Groups 2 and 5 were stabilized by squared miniplates and groups 3 and 6 were fixed by rectangular design. Each group was subjected to a mechanical test at a displacement speed of 10 mm/min on a universal testing machine, receiving linear vertical load on the region of the left first molar. The values of the maximum load and when displacements reached 5 mm were obtained and statistically analyzed by calculating the confidence interval of 95%. Fixation systems using squared (G2) and rectangular (G3) miniplates obtained similar results. No statistically significant differences with respect to the maximum load and the load at 5 mm displacement were found when compared to standard method in symphyseal fractures (G1). In parasymphysis the fixation method using squared miniplates (G5) obtained results without significant differences regarding the maximum load and the load at 5 mm when compared to the standard configuration (G4). The fixation method using rectangular miniplates (G6) showed inferior results which were statistically significant when compared to the standard configuration (G4) for parasymphysis fractures. The mechanical behavior of the fixation methods was similar, except when rectangular miniplates were used. The fixation methods showed better results with statistical significance in symphyseal fractures
