Type and rate of fertilizers influence the level of soil organic carbon (Corg) and total nitrogen (Nt) markedly, but the effect on C and N partitioning into different pools is open to question. The objectives of the present work were to: (i) quantify the impact of fertilizer type and rate on labile, intermediate and passive C and N pools by using a combination of biological, chemical and mathematical methods; (ii) explain previously reported differences in the soil organic matter (SOM) levels between soils receiving farmyard manure with or without biodynamic preparations by using Corg time series and information on SOM partitioning; and (iii) quantify the long-term and short-term dynamics of SOM in density fractions and microbial biomass as affected by fertilizer type and rate and determine the incorporation of crop residues into labile SOM fractions. Samples were taken from a sandy Cambisol from the long-term fertilization trial in Darmstadt, Germany, founded in 1980. The nine treatments (four field replicates) were: straw incorporation plus application of mineral fertilizer (MSI) and application of rotted farmyard manure with (DYN) or without (FYM) addition of biodynamic preparations, each at high (140 – 150 kg N ha-1 year-1; MSIH, DYNH, FYMH), medium (100 kg N ha-1 year-1; MSIM, DYNM, FYMM) and low (50 – 60 kg N ha-1 year-1; MSIL, DYNL, FYML) rates. The main findings were: (i) The stocks of Corg (t ha-1) were affected by fertilizer type and rate and increased in the order MSIL (23.6), MSIM (23.7), MSIH (24.2) < FYML (25.3) < FYMM (28.1), FYMH (28.1). Stocks of Nt were affected in the same way (C/N ratio: 11). Storage of C and N in the modelled labile pools (turnover times: 462 and 153 days for C and N, respectively) were not influenced by the type of fertilizer (FYM and MSI) but depended significantly (p ≤ 0.05) on the application rate and ranged from 1.8 to 3.2 t C ha 1 (7 – 13% of Corg) and from 90 to 140 kg N ha-1 (4-5% of Nt). In the calculated intermediate pool (C/N ratio 7), stocks of C were markedly higher in FYM treatments (15-18 t ha-1) compared to MSI treatments (12-14 t ha-1). This showed that differences in SOM stocks in the sandy Cambisol induced by fertilizer rate may be short-lived in case of changing management, but differences induced by fertilizer type may persist for decades. (ii) Crop yields, estimated C inputs (1.5 t ha-1 year-1) with crop residue, microbial bio¬mass C (Cmic, 118 – 150 mg kg-1), microbial biomass N (17 – 20 mg kg-1) and labile C and N pools did not differ significantly between FYM and DYN treatments. However, labile C increased linearly with application rate (R2 = 0.53) from 7 to 11% of Corg. This also applied for labile N (3.5 to 4.9% of Nt). The higher contents of Corg in DYN treatments existed since 1982, when the first sampling was conducted for all individual treatments. Contents of Corg between DYN and FYM treatments con-verged slightly since then. Furthermore, at least 30% of the difference in Corg was located in the passive pool where a treatment effect could be excluded. Therefore, the reported differences in Corg contents existed most likely since the beginning of the experiment and, as a single factor of biodynamic agriculture, application of bio-dynamic preparations had no effect on SOM stocks. (iii) Stocks of SOM, light fraction organic C (LFOC, ρ ≤ 2.0 g cm-3), light fraction organic N and Cmic decreased in the order FYMH > FYML > MSIH, MSIL for all sampling dates in 2008 (March, May, September, December). However, statistical significance of treatment effects differed between the dates, probably due to dif-ferences in the spatial variation throughout the year. The high proportion of LFOC on total Corg stocks (45 – 55%) highlighted the importance of selective preservation of OM as a stabilization mechanism in this sandy Cambisol. The apparent turnover time of LFOC was between 21 and 32 years, which agreed very well with studies with substantially longer vegetation change compared to our study. Overall, both approaches; (I) the combination of incubation, chemical fractionation and simple modelling and (II) the density fractionation; provided complementary information on the partitioning of SOM into pools of different stability. The density fractionation showed that differences in Corg stocks between FYM and MSI treatments were mainly located in the light fraction, i.e. induced by higher recalcitrance of the organic input in the FYM treatments. Moreover, the use of the combination of biological, chemical and mathematical methods indicated that effects of fertilizer rate on total Corg and Nt stocks may be short-lived, but that the effect of fertilizer type may persist for longer time spans in the sandy Cambisol.

Presentation given at the Al-Azhar Engineering First Conference, AEC’89, Dec. 9-12 1989, Cairo, Egypt. The paper presented at AEC'89 suggests an infinite storage scheme divided into one volume which is online and an arbitrary number of off-line volumes arranged into a linear chain which hold records which haven't been accessed recently. The online volume holds the records in sorted order (e.g. as a B-tree) and contains shortest prefixes of keys of records already pushed offline. As new records enter, older ones are retired to the first volume which is going offline next. Statistical arguments are given for the rate at which an off-line volume needs to be fetched to reload a record which had been retired before. The rate depends on the distribution of access probabilities as a function of time. Applications are medical records, production records or other data which need to be kept for a long time for legal reasons.

The main objective of this thesis was to determine the potential impact of heat stress (HS) on physiological traits of lactating cows and semen quality of bulls kept in a temperate climate. The thesis is comprised of three studies. An innovative statistical modeling aspect common to all three studies was the application of random regression methodology (RRM) to study the phenotypic and genetic trajectory of traits in dependency of a continuous temperature humidity index (THI). In the first study, semen quality and quantity traits of 562 Holstein sires kept on an AI station in northwestern Germany were analyzed in the course of THI calculated from data obtained from the nearest weather station. Heat stress was identified based on a decline in semen quality and quantity parameters. The identified general HS threshold (THI = 60) and the thermoneutal zone (THI in the range from 50 to 60) for semen production were lower than detected in studies conducted in tropical and subtropical climates. Even though adult bulls were characterized by higher semen productivity compared to younger bulls, they responded with a stronger semen production loss during harsh environments. Heritabilities (low to moderate range) and additive genetic variances of semen characteristics varied with different levels of THI. Also, based on genetic correlations genotype, by environment interactions were detected. Taken together, these findings suggest the application of specific selection strategies for specific climate conditions. In the second study, the effect of the continuous environmental descriptor THI as measured inside the barns on rectal temperatures (RT), skin temperatures (ST), vaginal temperatures (VT), respiration rates (RR), and pulse rate (PR) of lactating Holstein Friesian (HF) and dual-purpose German black pied cattle (DSN) was analyzed. Increasing HS from THI 65 (threshold) to THI 86 (maximal THI) resulted in an increase of RT by 0.6 °C (DSN) and 1 °C (HF), ST by 3.5 °C (HF) and 8 °C (DSN), VT by 0.3 °C (DSN), and RR by 47 breaths / minute (DSN), and decreased PR by 7 beats / minute (DSN). The undesired effects of rising THI on physiological traits were most pronounced for cows with high levels of milk yield and milk constituents, cows in early days in milk and later parities, and during summer seasons in the year 2014. In the third study of this dissertation, the genetic components of the cow’s physiological responses to HS were investigated. Heat stress was deduced from indoor THI measurements, and physiological traits were recorded on native DSN cows and their genetically upgraded crosses with Holstein Friesian sires in two experimental herds from pasture-based production systems reflecting a harsh environment of the northern part of Germany. Although heritabilities were in a low range (from 0.018 to 0.072), alterations of heritabilities, repeatabilities, and genetic components in the course of THI justify the implementation of genetic evaluations including heat stress components. However, low repeatabilities indicate the necessity of using repeated records for measuring physiological traits in German cattle. Moderate EBV correlations between different trait combinations indicate the potential of selection for one trait to simultaneously improve the other physiological attributes. In conclusion, bulls of AI centers and lactating cows suffer from HS during more extreme weather conditions also in the temperate climate of Northern Germany. Monitoring physiological traits during warm and humid conditions could provide precious information for detection of appropriate times for implementation of cooling systems and changes in feeding and management strategies. Subsequently, the inclusion of these physiological traits with THI specific breeding values into overall breeding goals could contribute to improving cattle adaptability by selecting the optimal animal for extreme hot and humid conditions. Furthermore, the recording of meteorological data in close distance to the cow and visualizing the surface body temperature by infrared thermography techniques might be helpful for recognizing heat tolerance and adaptability in cattle.

Almost 450 nuclear power plants are currently operating throughout the world and supplying about 17% of the world’s electricity. These plants perform safely, reliably, and have no free-release of byproducts to the environment. Given the current rate of growth in electricity demand and the ever growing concerns for the environment, the US consumer will favor energy sources that can satisfy the need for electricity and other energy-intensive products (1) on a sustainable basis with minimal environmental impact, (2) with enhanced reliability and safety and (3) competitive economics. Given that advances are made to fully apply the potential benefits of nuclear energy systems, the next generation of nuclear systems can provide a vital part of a long-term, diversified energy supply. The Department of Energy has begun research on such a new generation of nuclear energy systems that can be made available to the market by 2030 or earlier, and that can offer significant advances toward these challenging goals [1]. These future nuclear power systems will require advances in materials, reactor physics as well as heat transfer to realize their full potential. In this paper, a summary of these advanced nuclear power systems is presented along with a short synopsis of the important heat transfer issues. Given the nature of research and the dynamics of these conceptual designs, key aspects of the physics will be provided, with details left for the presentation.

The summary from Goodson’s group on their recent work on heat transfer issues in the microelectronics and data storage industries illustrate the critical role of heat transfer for some areas of information technology. In this article, we build on their work and discuss some directions worthy of further research.

La tesi s'ha estructurat en tres apartats que, en conjunt, han de permetre determinar les possibilitats d'aprofitament dins la mateixa indústria alimentària de la fracció plasmàtica de la sang de porc generada per escorxadors que utilitzen sistemes oberts de recollida higiènica. 1. En la primera part s'analitza la composició de la sang higiènica que s'està recollint actualment i s'estudien les característiques tant físico-químiques com microbiològiques que determinen la seva qualitat. La caracterització s'ha realitzat amb sang recollida en diferents escorxadors industrials de les comarques de Girona i s'ha centrat principalment en l'estudi de la contaminació microbiològica i el nivell d'hemòlisi de la sang. S'ha fet un disseny experimental que ha permès alhora valorar l'efecte d'alguns factors sobre la qualitat de la sang: possibles diferències relacionades amb (1) la climatologia del període de l'any en el qual es fa la recollida, (2) particularitats dels escorxadors (grandària, sistemes de dessagnat, tipus, dosi i sistema de dosificació de l'anticoagulant, condicions de processament, maneig i emmagatzematge després de la recollida, etc.). Els resultats obtinguts ens permeten constatar que, en les condicions actuals, la sang que s'està recollint en els escorxadors estudiats no es pot considerar adequada per a una matèria primera de productes destinats a alimentació humana. La major part de la microbiota contaminant s'adquireix en el propi sagnador. S'ha constatat que el sistema de dessagnat en posició horitzontal podria ser una mesura útil per minimitzar la contaminació d'origen fecal o provinent de la pell de l'animal sacrificat i que la separació immediata de les fraccions en el propi escorxador també pot contribuir a reduir la contaminació. Així doncs, en el benentès que l'efectivitat pot obtenir-se del conjunt de mesures preses, més que de l'aplicació d'una sola d'elles, es suggereix la introducció d'una sèrie d'actuacions que potser permetrien reduir els nivells de contaminació que s'obtenen actualment. El tractament mecànic de la sang, el sistema d'addició d'anticoagulant, el volum i concentració de la solució anticoagulant afegida i el període d'emmagatzematge són els factors responsables de l'hemòlisi; mentre que nivells elevats de contaminació microbiològica i el tipus d'anticoagulant utilitzat deterrminen la velocitat d'increment de l'hemòlisi de sang refrigerada. S'ha constatat que quan la sang no pot ser processada immediatament i s'ha d'emmagatzemar en refrigeració és millor utilitzar citrat sòdic enlloc de polifosfat com a anticoagulant ja que l'increment d'hemòlisi es dóna més lentament. 2. El segon apartat s'ha centrat en la fracció plasmàtica de la sang. S'ha utilitzat la deshidratació per atomització com a tecnologia de conservació del plasma i s'ha fet una caracterització del producte en pols resultant des del punt de vista de composició i qualitat. A més de la contaminació microbiològica, que determina la qualitat higiènico-sanitària del producte, s'ha realitzat un estudi de les propietats funcionals que podrien fer del plasma un producte útil en la formulació d'aliments (capacitat escumant, emulsionant, gelificant). S'ha fet especial incidència en (1) determinar l'efecte del procés tecnològic de deshidratació sobre la funcionalitat del producte i (2) estudiar l'estabilitat del plasma deshidratat durant el període d'emmagatzematge. En les condicions de deshidratació per atomització aplicades no es provoca desnaturalització de la fracció proteica i s'obté un producte suficientment deshidratat, amb una aw<0,4 per permetre suposar una bona estabilitat. Algunes mostres de plasma deshidratat analitzades presenten nivells detectables de determinats residus (sulfonamides i corticosteroides). La qualitat microbiològica del producte en pols reflecteix l'elevada contaminació que contenia la matèria primera utilitzada, tot i que la deshidratació per atomització ha comportat la reducció en una unitat logarítmica de la càrrega contaminant. Els recomptes generals de microorganismes són encara preocupants i més tenint en compte que s'ha evidenciat la presència de toxines estafilocòciques en algunes mostres. L'avaluació de les propietats funcionals del producte deshidratat en relació a les que presentava el plasma líquid ens ha permès comprovar que: (1) El procés de deshidratació no ha afectat la solubilitat de les proteïnes. Això, junt amb el fet que no s'obtinguin diferències significatives en l'anàlisi calorimètrica de mostres líquides o deshidratades, permet concloure que el procés no provoca desnaturalització proteica. (2) No s'observen efectes negatius del procés tecnològic sobre la capacitat escumant ni en l'activitat emulsionant de les proteïnes plasmàtiques, dues propietats funcionals que possibiliten l'aplicació del plasma amb aquestes finalitats en l'elaboració d'alguns aliments. (3) La deshidratació tampoc perjudica de manera important les característiques dels gels que s'obtenen per escalfament, ja que els gels obtinguts a partir del plasma líquid i del plasma deshidratat presenten la mateixa capacitat de retenció d'aigua i no s'observen diferències en la microestructura de la xarxa proteica d'ambdós tipus de gel. Tanmateix, els que s'obtenen a partir del producte en pols mostren una menor resistència a la penetració. L'estudi d'estabilitat ens ha permès comprovar que la mostra de plasma deshidratat per atomització perd algunes de les seves propietats funcionals (facilitat de rehidratació, capacitat de retenció d'aigua i fermesa dels gels) si s'emmagatzema a temperatura ambient, mentre que aquestes característiques es mantenen un mínim de sis mesos quan el producte en pols es conserva a temperatura de refrigeració. 3. En l'última part, tenint en compte les conclusions derivades dels resultats dels apartats anteriors, s'han assajat tres possibles sistemes de reducció de la contaminació aplicables a la fracció plasmàtica com a pas previ a la deshidratació, per tal de millorar les característiques de qualitat microbiològica i les perspectives d'estabilitat del producte durant l'emmagatzematge. S'ha determinat l'eficàcia, i l'efecte sobre les propietats del plasma deshidratat, que poden tenir tractaments d'higienització basats en la centrifugació, la microfiltració tangencial i l'aplicació d'altes pressions. Els tractaments de bactofugació aplicats permeten reduir entre el 96 i el 98% la contaminació microbiana del plasma. Aquesta reducció s'aconsegueix tant amb un sistema discontinu com amb un sistema continu treballant a una velocitat de 12 L/h, fet que permetria adaptar el tractament de bactofugació a un procés de producció industrial. Un sistema combinat de bactofugació en continu i microfiltració tangencial permet incrementar l'eficàcia fins a un 99,9 % de reducció. Cal tenir present, però, que aquest tractament provoca també una disminució de l'extracte sec que afecta negativament les propietats funcionals del plasma líquid. Malgrat suposar una pèrdua pel que fa al rendiment, aquest efecte negatiu sobre la funcionalitat no suposaria cap inconvenient si s'utilitzés la deshidratació com a tecnologia de conservació del plasma, ja que es podria corregir l'extracte sec durant la reconstitució del producte. Caldria avaluar si la millora en la qualitat higiènico-sanitària del producte compensa o no les pèrdues que suposa aquest sistema d'higienització combinat. Amb relació als tractaments d'alta pressió, de totes les condicions de tractament assajades, les pressions de fins 450 MPa permeten obtenir plasma sense modificacions importants que impedeixin la seva deshidratació per atomització. Així doncs, les condicions de procés que s'han aplicat són pressuritzacions a 450 MPa de 15 minuts de durada. La temperatura de tractament que s'ha mostrat més eficaç en la reducció dels recomptes de microorganismes ha estat de 40ºC. Els tractaments a aquesta temperatura permeten assolir reduccions del 99,97% i disminuir en un 80% la capacitat de creixement dels microorganismes supervivents a la pressurització en relació a la que presentava la població contaminant del plasma abans del tractament. L'estudi de l'efecte d'aquest tractament (450 MPa, 15 min i 40ºC) sobre les propietats funcionals del plasma ha permès observar que la pressurització comporta una disminució en la solubilitat del producte però una millora en les propietats de superfície -estabilitat de l'escuma i activitat emulsionant- i un increment de la capacitat de retenció d'aigua i de la duresa dels gels obtinguts per escalfament. Calen més estudis per confirmar i caracteritzar aquesta millora en la funcionalitat, així com per establir si el tractament de pressurització afecta també l'estabilitat del producte durant l'emmagatzematge. De totes les tecnologies d'higienització assajades, l'alta pressió és la que permet obtenir millors resultats en el sentit de poder garantir un producte de bona qualitat microbiològica i segur, des del punt de vista sanitari i tecnològic, per a la seva utilització com a ingredient alimentari.

In this paper, the available potential energy (APE) framework of Winters et al. (J. Fluid Mech., vol. 289, 1995, p. 115) is extended to the fully compressible Navier– Stokes equations, with the aims of clarifying (i) the nature of the energy conversions taking place in turbulent thermally stratified fluids; and (ii) the role of surface buoyancy fluxes in the Munk & Wunsch (Deep-Sea Res., vol. 45, 1998, p. 1977) constraint on the mechanical energy sources of stirring required to maintain diapycnal mixing in the oceans. The new framework reveals that the observed turbulent rate of increase in the background gravitational potential energy GPEr , commonly thought to occur at the expense of the diffusively dissipated APE, actually occurs at the expense of internal energy, as in the laminar case. The APE dissipated by molecular diffusion, on the other hand, is found to be converted into internal energy (IE), similar to the viscously dissipated kinetic energy KE. Turbulent stirring, therefore, does not introduce a new APE/GPEr mechanical-to-mechanical energy conversion, but simply enhances the existing IE/GPEr conversion rate, in addition to enhancing the viscous dissipation and the entropy production rates. This, in turn, implies that molecular diffusion contributes to the dissipation of the available mechanical energy ME =APE +KE, along with viscous dissipation. This result has important implications for the interpretation of the concepts of mixing efficiency γmixing and flux Richardson number Rf , for which new physically based definitions are proposed and contrasted with previous definitions. The new framework allows for a more rigorous and general re-derivation from the first principles of Munk & Wunsch (1998, hereafter MW98)’s constraint, also valid for a non-Boussinesq ocean: G(KE) ≈ 1 − ξ Rf ξ Rf Wr, forcing = 1 + (1 − ξ )γmixing ξ γmixing Wr, forcing , where G(KE) is the work rate done by the mechanical forcing, Wr, forcing is the rate of loss of GPEr due to high-latitude cooling and ξ is a nonlinearity parameter such that ξ =1 for a linear equation of state (as considered by MW98), but ξ <1 otherwise. The most important result is that G(APE), the work rate done by the surface buoyancy fluxes, must be numerically as large as Wr, forcing and, therefore, as important as the mechanical forcing in stirring and driving the oceans. As a consequence, the overall mixing efficiency of the oceans is likely to be larger than the value γmixing =0.2 presently used, thereby possibly eliminating the apparent shortfall in mechanical stirring energy that results from using γmixing =0.2 in the above formula.

The distribution of tracers in the ocean is often taken as an indication of the ventilation pathways for oceanic water masses. It has been suggested that under anthropogenic forcing heat will be taken up into the interior of the ocean along isopycnal ventilation pathways. This notion is investigated by examining distributions of potential temperature and a passive anomaly temperature tracer in a coupled climate experiment where CO2 is increased at a rate of 2% per year. We show that interior temperature changes cannot be explained solely by passive tracer transport along isopycnals. Heat uptake is strongly affected by changes in circulation and has a substantial diapycnal component.

This paper presents a new analysis of ocean heat content changes over the last 50 yr using isotherms by calculating the mean temperature above the 148C isotherm and the depth of the 148C isotherm as separate variables. A new quantity called the ‘‘relative heat content’’ (‘‘RHC’’) is introduced, which represents the minimum local heat content change over time, relative to a fixed isotherm. It is shown how mean temperature and isotherm depth changes make separable and additive contributions to changes in RHC. Maps of RHC change between 1970 and 2000 show similar spatial patterns to a traditional fixed-depth ocean heat content change to 220 m. However, the separate contributions to RHC suggest a more spatially uniform contribution from warming above the isotherm, while isotherm depth changes show wind-driven signals, of which some are identifiable as being related to the North Atlantic Oscillation. The time series show that the warming contribution to RHC dominates the global trend, while the depth contribution only dominates on the basin scale in the North Atlantic. The RHC shows minima associated with the major volcanic eruptions (particularly in the Indian Ocean), and these are entirely contributed by mean temperature changes rather than isotherm depth changes. The depth change contributions to RHC are strongly affected by the recently reported XBT fall-rate bias, whereas the mean temperature contributions are not. Therefore, only the isotherm depth change contributions toRHCwill need to be reassessed as fall-rate-corrected data become available.

This review summarizes the recent discovery of the cupin superfamily (from the Latin term "cupa," a small barrel) of functionally diverse proteins that initially were limited to several higher plant proteins such as seed storage proteins, germin (an oxalate oxidase), germin-like proteins, and auxin-binding protein. Knowledge of the three-dimensional structure of two vicilins, seed proteins with a characteristic beta-barrel core, led to the identification of a small number of conserved residues and thence to the discovery of several microbial proteins which share these key amino acids. In particular, there is a highly conserved pattern of two histidine-containing motifs with a varied intermotif spacing. This cupin signature is found as a central component of many microbial proteins including certain types of phosphomannose isomerase, polyketide synthase, epimerase, and dioxygenase. In addition, the signature has been identified within the N-terminal effector domain in a subgroup of bacterial AraC transcription factors. As well as these single-domain cupins, this survey has identified other classes of two-domain bicupins including bacterial gentisate 1, 2-dioxygenases and 1-hydroxy-2-naphthoate dioxygenases, fungal oxalate decarboxylases, and legume sucrose-binding proteins. Cupin evolution is discussed from the perspective of the structure-function relationships, using data from the genomes of several prokaryotes, especially Bacillus subtilis. Many of these functions involve aspects of sugar metabolism and cell wall synthesis and are concerned with responses to abiotic stress such as heat, desiccation, or starvation. Particular emphasis is also given to the oxalate-degrading enzymes from microbes, their biological significance, and their value in a range of medical and other applications.

Tolerance to high soil and air temperature during the reproductive phase is an important component of adaptation to and and semi-arid cropping environments in groundnut. Between 10 and 22 genotypes were screened for tolerance to high air and soil temperature in controlled environments. To assess tolerance to high soil temperature, 10 genotypes were grown from start of podding to harvest at ambient (28 degrees) and high (38 degreesC) soil temperatures, and crop growth rate (CGR), pod growth rate (PGR) and partitioning (ratio PGR:CGR) measured. To assess tolerance to high air temperature during two key stages-microsporogenesis (3-6 days before flowering, DBF) and flowering, fruit-set was measured in two experiments. In the first experiment, 12 genotypes were exposed to short (3-6 days) episodes of high (38 degreesC) day air temperature at 6 DBF and at flowering. In the second experiment, 22 genotypes were exposed to 40 degreesC day air temperature for I day at 6 DBF, 3 DBF or at flowering. Cellular membrane thermostability (relative injury, RI) was also measured in these 22 genotypes. There was considerable variation among genotypes in response to high temperature, whether assessed by growth rates, fruit-set or RI. Pod weight at high soil temperature was associated with variation in CGR rather than partitioning. Flowering was more sensitive to high air temperature than microsporogenesis. Genotypes tolerant to high air temperature at microsporogenesis were not necessarily tolerant at flowering, and nor was tolerance correlated with RI. Six genotypes (796, 55-437, ICG 1236, ICGV 86021, lCGV 87281 and ICGV 92121) were identified as heat tolerant based on their performance in all tests. These experiments have shown that groundnut genotypes can be easily screened for reproductive tolerance to high air and soil temperature and that several sources of heat tolerance are available in groundnut germplasm. Seeds of carrot, groundnut, lettuce, oilseed rape and onion were stored hermetically in laminated aluminium foil packets in four environments (dry or ultra-dry moisture contents combined factorially with temperatures of 20 degrees C or -20 degrees C), replicated at several sites. After ten years' hermetic storage, seed moisture content, equilibrium relative humidity, viability (assessed by ability to germinate normally in standard germination tests) and vigour were determined. After a decade, the change in seed moisture content of samples stored at -20 degrees C was small or nil. Except for groundnut and lettuce (where loss in viability was about 8 and 3%, respectively), no loss in viability was detected after 10 years' hermetic storage at -20 degrees C. In all cases, there was no difference in seed survival between moisture contents at this temperature (P > 0.25). Comparison of seed vigour (root length and rate of germination) also confirmed that drying to moisture contents in equilibrium with 10-12% r.h. had no detrimental effect to longevity when stored at -20 degrees C: the only significant (P < 0.05) differences detected were slightly greater root lengths for ultra-dry storage of four of the six seed lots. Seed moisture content had increased after a decade at 20 degrees C (generally to the level in equilibrium with ambient relative humidity). Hence, sub-zero temperature storage helped maintain the long-term integrity of the laminated aluminium foil packets, as well as that of the seeds within.

Cryopreservation using encapsulation-dehydration was developed for the long-term conservation of cocoa (Theobroma cacao L.) germplasm. Survival of individually encapsulated somatic embryos after desiccation and cryopreservation was achieved through optimization of cryoprotectants (abscisic acid (ABA) and sugar), duration of osmotic and evaporative dehydration, and embryo development stage. Up to 63% of the genotype SPA4 early-cotyledonary somatic embryos survived cryopreservation following 7 days preculture with 1 M sucrose and 4 h silica exposure (16% moisture content in bead). This optimized protocol was successfully applied to three other genotypes, e.g. EET272, IMC14 and AMAZ12, with recovery frequencies of 25, 40 and 72%, respectively (but the latter two genotypes using 0.75 M sucrose). Recovered SPA4 somatic embryos converted to plants at a rate of 33% and the regenerated plants were phenotypically comparable to non-cryopreserved somatic embryo-derived plants.

Experimental wind tunnel and smoke visualisation testing and CFD modelling were conducted to investigate the effect of air flow control mechanism and heat source inside rooms on wind catchers/towers performance. For this purpose, a full-scale wind catcher was connected to a test room and positioned centrally in an open boundary wind tunnel. Pressure coefficients (C-p's) around the wind catcher and air flow into the test room were established. The performance of the wind catcher depends greatly on the wind speed and direction. The incorporation of dampers and egg crate grille at ceiling level reduces and regulates the air flow rate with an average pressure loss coefficient of 0.01. The operation of the wind catcher in the presence of heat sources will potentially lower the internal temperatures in line with the external temperatures.

Tofu gels were rheologically examined to determine their storage or elastic (G′) and loss or viscous (G″) moduli as a function of frequency within their linear viscoelastic limits. The tofu gels were made using either glucono-δ-lactone (GDL) or calcium sulphate (CaSO4·2H2O), followed by either heat treatment (heated soymilk at 97 °C prior to coagulation and subsequently held at 70 °C for 60 min, HT) or high pressure treatment (400 MPa at 20 °C for 10 min, HP). The overall moduli values of the GDL gels and CaSO4·2H2O gels of both physical treatments were similar, each gave frequency profiles expected for weak viscoelastic materials. However, although both temperature and high pressure treatments could be used to produce tofu gels, the final products were not the same. Pressure formed gels, despite having a higher overall “consistency” (increasing values of their moduli), had a proportionately higher contribution from the loss modulus (increased tan δ). Differences could also be observed using confocal scanning laser microscopy. While such treatment may give rise to differing systems/structures, with new or modified organoleptic properties, the more “open” structures obtained by pressure treatment may well cause processing difficulties if subsequent reworking or moulding is required.
