Coccidiosis is an economically important parasitic disease of chickens that, in Australia, is caused by seven species of the genus Eimeria.1 The disease has traditionally been controlled by prophylactic drugs, but vaccination with attenuated lines of the parasites2–4 is rapidly gaining acceptance world wide. Live Eimeria vaccines are produced in batches which are not frozen and have a limited shelf life. The per cent infectivity of vaccine seed stocks and the vaccines produced from them must therefore be accurately monitored using standardised dose dependant assays to ensure that shelf life, quality control and vaccine release specifications are met. Infectivity for the chicken host cannot readily be determined by microscopic observation of oocysts or sporocyst hatching.5 Dose dependent parameters such as body weight gain, feed conversion ratio, visual lesion scores, mortality, oocysts production, clinical symptoms and microscopic lesion counts could be used as measures of infectivity.6–11 These parameters show significant dose dependant effects with field strains, but lines of vaccine parasites that have been selected for precocious development with associated reduced virulence and reproductive capability may not have the same effect.3,4 The aim of this trial was to determine which parameters provide the most effective measures of infective dose in birds inoculated with a precocious vaccine strain.

Methane emissions from ruminant livestock represent a loss of carbon during feed conversion, which has implications for both animal productivity and the environment because this gas is considered to be one of the more potent forms of greenhouses gases contributing to global warming. Many strategies to reduce emissions are targeting the methanogens that inhabit the rumen, but such an approach can only be successful if it targets all the major groups of ruminant methanogens. Therefore, a thorough knowledge of the diversity of these microbes in different breeds of cattle and sheep, as well as in response to different diets, is required. A study was undertaken using the molecular techniques denaturing gradient gel electrophoresis, DNA cloning and DNA sequence analysis to define the extent of diversity among methanogens in ruminants, particularly Bos indicus cross cattle, on differing forages in Queensland. It was found that the diversity of methanogens in forage-fed cattle in Queensland was greater than in grain-fed cattle but there was little variability in methanogen community composition between cattle fed different forages. The species that dominate the rumen microbial communities of B. indicus cross cattle are from the genus Methanobrevibacter, although rumen-fluid inoculated digestors fed Leucaena leucocephala leaf were populated with Methanosphaera-like strains, with the Methanobrevibacter-like strains displaced. If ruminant methane emissions are to be reduced, then antimethanogen bioactives that target both broad groups of ruminant methanogens are most likely to be needed, and as a part of an integrated suite of approaches that redirect rumen fermentation towards other more useful end products.

In the European Union, conventional cages for laying hens will be faded out at the beginning of 2012. The rationale behind this is a public concern over animal welfare in egg production. As alternatives to conventional cages, the European Union Council Directive 1999/74/EC allows non-cage systems and enriched (furnished) cages. Layer performance, behavior, and welfare in differently sized furnished cages have been investigated quite widely during recent decades, but nutrition of hens in this production system has received less attention. This thesis aims to compare production and feed intake of laying hens in furnished and conventional cages and to study the effects of different dietary treatments in these production systems, thus contributing to the general knowledge of furnished cages as an egg production system. A furnished cage model for 8 hens was compared with a 3-hen conventional cage. Three consecutive experiments each studied one aspect of layer diet: The first experiment investigated the effects of dietary protein/energy ratio, the second dietary energy levels, and the third the effects of extra limestone supplementation. In addition, a fourth experiment evaluated the effects of perches on feed consumption and behavior of hens in furnished cages. The dietary treatments in experiments 1 3 generally had similar effects in the two cage types. Thus, there was no evidence supporting a change in nutrient requirements for laying hens when conventional cages are replaced with small-group furnished cages. Moreover, the results from nutritional experiments conducted in conventional cages can be applied to small-group furnished cage systems. These results support the view that production performance comparable with conventional cages can be achieved in furnished cages. All of the advantages of cages for bird welfare are sustained in the small-group furnished cages used here. In addition, frequent use of perches and nests implies a wider behavioral repertoire in furnished cages than in conventional cages. The increase observed in bone ash content may improve bird welfare in furnished cages. The presence of perches diminished feed consumption during the prelaying period and enhanced the feed conversion ratio during the early laying period in furnished cages. However, as the presence or absence of perches in furnished cages had no significant effect on feed consumption after the prelaying period, the lower feed consumption observed in furnished cages than in conventional cages could be attributed to other factors, such as the presence of wood shavings or a nest box. The wider feed trough space per hen in conventional than in furnished cages may partly explain the higher feed consumption observed in conventional cages.

The development of a more efficient in vitro regeneration system for somatic embryos (SEs) of avocado (Persea americana) would facilitate the development of new superior cultivars for this valuable horticultural crop. In this study, we report a new and efficient method for maintenance and regeneration of avocado SEs. Avocado SEs of four cultivars remained healthy and viable in vitro for 11 months on a medium used for mango somatic embryogenesis, compared with 3-4 months on Murashige and Skoog medium. Various supplements and media modifications were investigated to improve the low conversion rate of regenerated plants from avocado SEs reported previously. The one-step system for regeneration of white-opaque somatic embryos (WOSEs) used solid medium only over a period of 12-14 weeks (sub-culturing every 6 weeks). Addition of praline and glutamine improved the total regeneration from 0 to 17.5% and 10.5%, and plant/shoot recovery from 0 to 12.5% and 5%, respectively. A two-step culture system involving the transfer of WOSEs of cultivar 'Reed' after 6 weeks on solid to liquid medium for 12-15 days as an intermediate step, followed by subculturing again onto solid medium for 6 weeks improved total regeneration to 29% and plant/shoot recovery to 18.3 from 0% when regenerated by subculturing on solid medium only. Supplementation with proline in the solid as well as liquid medium in the two-step culture system at 0.4 g/L increased total regeneration to 35% and plant/shoot recovery to 20%. We were able to achieve highest regeneration using glutamine at 1 g/L in the two-step culture system in terms of both total regeneration (58.3%, including 43.3% bipolar regeneration) and plant/shoot recovery (36.7%) rates, which were significantly higher than in any other treatment investigated. This study examined the physical and chemical properties of a novel, fully-recirculated prawn and polychaete production system that incorporated polychaete-assisted sand filters (PASF). The aims were to assess and demonstrate the potential of this system for industrialisation, and to provide optimisations for wastewater treatment by PASF. Two successive seasons were studied at commercially-relevant scales in a prototype system constructed at the Bribie Island Research Centre in Southeast Queensland. The project produced over 5.4 tonnes of high quality black tiger prawns at rates up to 9.9 tonnes per hectare, with feed conversion of up to 1.1. Additionally, the project produced about 930 kg of high value polychaete biomass at rates up to 1.5 kg per square metre of PASF, with the worms feeding predominantly on waste nutrients. Importantly, this closed production system demonstrated rapid growth of healthy prawns at commercially relevant production levels, using methods that appear feasible for application at large scale. Deeper (23 cm) PASF beds provided similar but more reliable wastewater treatment efficacies compared with shallower (13 cm) beds, but did not demonstrate significantly greater polychaete productivity than (easier to harvest) shallow beds. The nutrient dynamics associated with seasonal and tidal operations of the system were studied in detail, providing technical and practical insights into how PASF could be optimised for the mitigation of nutrient discharge. The study also highlighted some of the other important advantages of this integrated system, including low sludge production, no water discharge during the culture phase, high ecosystem health, good prospects for biosecurity controls, and the sustainable production of a fishery-limited resource (polychaetes) that may be essential for the expansion of prawn farming industries throughout the world. Regarding nutrient discharge from this prototype mariculture system, when PASF was operating correctly it proved feasible to have no water (or nutrient) discharge during the entire prawn growing season. However, the final drain harvest and emptying of ponds that is necessary at the end of the prawn farming season released 58.4 kg ha-1 of nitrogen and 6 kg ha-1 of phosphorus (in Season 2). Whilst this is well below (i.e., one-third to one-half of) the current load-based licencing conditions for many prawn farms in Australia, the levels of nitrogen and chlorophyll a in the ponds remained higher than the more-stringent maximum limits at the Bribie Island study site. Zero-net-nutrient discharge was not achieved, but waste nutrients were low where 5.91 kg of nitrogen and 0.61 kg of phosphorus was discharged per tonne of prawns produced. This was from a system that deployed PASF at 14.4% of total ponded farm area which treated an average of 5.8% of pond water daily and did not use settlement ponds or other natural or artificial water remediation systems. Four supplemental appendices complement this research by studying several additional aspects that are central to the industrialisation of PASF. The first details an economic model and decision tool which allows potential users to interactively assess construction and operational variables of PASF at different scales. The second provides the qualitative results of a prawn maturation trial conducted collaboratively with the Commonwealth Scientific and Industrial Research Organisation (CSIRO) to assess dietary inclusions of PASF-produced worms. The third provides the reproductive results from industry-based assessments of prawn broodstock produced using PASF. And the fourth appendix provides detailed elemental and nutritional analyses of bacterial biofilm produced by PASF and assesses its potential to improve the growth of prawns in recirculated culture systems.

Metal-free CNTs exhibit high activity (conversion rate 99.6%, 6 h) towards the synthesis of chiral hydrobenzoin from benzaldehyde under near-UV light irradiation (320–400 nm). The CNT structure before and after the reaction, the interaction between the molecule and the CNT surface, the intermediate products, the substitution effect and the influence of light on the reaction were examined using various techniques. A photo-excited conduction electron transfer (PECET) mechanism for the photocatalytic reduction using CNTs has been proposed. This finding provides a green photocatalytic route for the production of hydrobenzoin and highlights a potential photocatalytic application of CNTs.

Nanocrystalline Ce1-xFexO2-delta (0 <= x <= 0.45) and Ce0.65Fe0.33Pd0.02O2-delta of similar to 4 nm sizes were synthesized by a sonochemical method using diethyletriamine (DETA) as a complexing agent. Compounds were characterized by powder X-ray diffraction (XRD), X-ray photo-electron spectroscopy (XPS) and transmission electron microscopy (TEM). Ce1-xFexO2-delta (0 <= x <= 0.45) and Ce0.65Fe0.33Pd0.02O2-delta crystallize in fluorite structure where Fe is in +3, Ce is in +4 and Pd is in +2 oxidation state. Due to substitution of smaller Fe3+ ion in CeO2, lattice oxygen is activated and 33% Fe substituted CeO2 i.e. Ce0.67Fe0.33O1.835 reversibly releases 0.31O] up to 600 degrees C which is higher or comparable to the oxygen storage capacity of CeO2-ZrO2 based solid solutions (Catal. Today 2002, 74, 225-234). Due to interaction of redox potentials of Pd2+/0(0.89 V) and Fe3+/2+ (0.77 V) with Ce4+/3+ (1.61 V), Pd ion accelerates the electron transfer from Fe2+ to Ce4+ in Ce0.65Fe0.33Pd0.02O1.815, making it a high oxygen storage material as well as a highly active catalyst for CO oxidation and water gas shift reaction. The activation energy for CO oxidation with Ce0.65Fe0.33Pd0.02O1.815 is found to be as low as 38 kJ mol(-1). Ce0.67Fe0.33O1.835 and Ce0.65Fe0.33Pd0.02O1.815 have also shown high activity for the water gas shift reaction. CO conversion to CO2 is 100% H-2 specific with these catalysts and conversion rate was found to be as high 27.2 mu moles g(-1) s(-1) and the activation energy was found to be 46.4 kJ mol(-1) for Ce0.65Fe0.33Pd0.02O1.815.

Hydrogen is a clean energy carrier and highest energy density fuel. Water gas shift (WGS) reaction is an important reaction to generate hydrogen from steam reforming of CO. A new WGS catalyst, Ce(1-x)Ru(x)O(2-delta) (0 <= x <= 0.1) was prepared by hydrothermal method using melamine as a complexing agent. The Catalyst does not require any pre-treatment. Among the several compositions prepared and tested, Ce(0.95)Ru(0.05)O(2-delta) (5% Ru(4+) ion substituted in CeO(2)) showed very high WGS activity in terms of high conversion rate (20.5 mu mol.g(-1).s(-1) at 275 degrees C) and low activation energy (12.1 kcal/mol). Over 99% conversion of CO to CO(2) by H(2)O is observed with 100% H(2) selectivity at >= 275 degrees C. In presence of externally fed CO(2) and H(2) also, complete conversion of CO to CO(2) was observed with 100% H(2) selectivity in the temperature range of 305-385 degrees C. Catalyst does not deactivate in long duration on/off WGS reaction cycle due to absence of surface carbon and carbonate formation and sintering of Ru. Due to highly acidic nature of Ru(4+) ion, surface carbonate formation is also inhibited. Sintering of noble metal (Ru) is avoided in this catalyst because Ru remains in Ru(4+) ionic state in the Ce(1-x)Ru(x)O(2-delta) catalyst.

The present fishing effort regulation for bottom trawls in the North Sea (EU 40/2008) reduces the fishing effort with larger co-dend mesh sizes (> 100 mm mesh opening) to 86 fishing days per year. The aim of this technical measure is the protection of the weak cod stock usually harvested with this mesh opening. By combining the results of our selectivity investigations on plaice during a commercial fishing trip and the samples taken within the frame of the EU data collection regulation (DCR) we could draw conclusions on commercial and non-commercial fish species. When using codend meshes with larger mesh opening the discard was reduced remarkably. In the reference codend with 80 mm mesh opening the undersized bycatch was 47 % of the total catch of plaice, in the experimental codends with 120 mm mesh opening it was only 7 % and with 130 mm mesh opening just 3 %. On the other hand however, the applied mesh openings in the experimental codends let escape marketable plaice to some extent. The loss of catch was assessed on the basis of fish numbers per length class using a length-weight conversion rate from the DCR. The loss by weight of marketable fish with 120 mm codend mesh opening was 18 %, and 28 % with 130 mm codend mesh opening. To assure the protection of young round and flatfish with one general mesh size, a new regulation should prescribe a minimum mesh opening of 120 mm, accompanied by an expansion of the fishing effort.

The growth responses and feed utilization of Sarotherodon niloticus held in metal cages in a pond and fed diets containing fish-meal, cow blood-meal or sorghum was studied. Results indicate that the best growth, feed conversion and protein efficiency ratio were obtained with the diet containing 60% fish-meal. The growth performance of fish on 40% fish-meal, and 40% and 60% blood meal were not significantly different, and were quite close to the performance with 60% fish-meal. The growth and food utilization of fish on 84% sorghum diet was significantly lower than the rest. The Caged fish without supplemental feeding had a light gain in weight. All fish with supplemental feeding appeared healthy. It is concluded that cow blood meal at 40% or 60% inclusion in diet can adequately replace fish-meal in S. niloticus supplemental diet in pond culture

The effect of inclusion of grasshopper meal on the growth, feed conversion ratio and survival of Clarias gariepinus fingerlings of mean weight 9.71g was investigated. The results show that the best growth and food utilization indexes were recorded in the fingerling fed with 10% grasshopper meal and 30% fish meal, while the poorest growth and food utilization indexes were recorded with the diet containing 25% grasshopper meal and 15% fish meal. The best survival of 100% was observed in the diet containing 30% grasshopper meal and 10% fish meal while the worst survival of 73.3% was observed in the diet containing 25% grasshopper meal and 15% fish meal

Os principais constituintes do ar, nitrogênio, oxigênio e argônio, estão cada vez mais presentes nas indústrias, onde são empregados nos processos químicos, para o transporte de alimentos e processamento de resíduos. As duas principais tecnologias para a separação dos componentes do ar são a adsorção e a destilação criogênica. Entretanto, para ambos os processos é necessário que os contaminantes do ar, como o gás carbônico, o vapor dágua e hidrocarbonetos, sejam removidos para evitar problemas operacionais e de segurança. Desta forma, o presente trabalho trata do estudo do processo de pré-purificação de ar utilizando adsorção. Neste sistema a corrente de ar flui alternadamente entre dois leitos adsorvedores para produzir ar purificado continuamente. Mais especificamente, o foco da dissertação corresponde à investigação do comportamento de unidades de pré-purificação tipo PSA (pressure swing adsorption), onde a etapa de dessorção é realizada pela redução da pressão. A análise da unidade de pré-purificação parte da modelagem dos leitos de adsorção através de um sistema de equações diferenciais parciais de balanço de massa na corrente gasosa e no leito. Neste modelo, a relação de equilíbrio relativa à adsorção é descrita pela isoterma de Dubinin-Astakhov estendida para misturas multicomponentes. Para a simulação do modelo, as derivadas espaciais são discretizadas via diferenças finitas e o sistema de equações diferenciais ordinárias resultante é resolvido por um solver apropriado (método das linhas). Para a simulação da unidade em operação, este modelo é acoplado a um algoritmo de convergência relativo às quatro etapas do ciclo de operação: adsorção, despressurização, purga e dessorção. O algoritmo em questão deve garantir que as condições finais da última etapa são equivalentes às condições iniciais da primeira etapa (estado estacionário cíclico). Desta forma, a simulação foi implementada na forma de um código computacional baseado no ambiente de programação Scilab (Scilab 5.3.0, 2010), que é um programa de distribuição gratuita. Os algoritmos de simulação de cada etapa individual e do ciclo completo são finalmente utilizados para analisar o comportamento da unidade de pré-purificação, verificando como o seu desempenho é afetado por alterações nas variáveis de projeto ou operacionais. Por exemplo, foi investigado o sistema de carregamento do leito que mostrou que a configuração ideal do leito é de 50% de alumina seguido de 50% de zeólita. Variáveis do processo foram também analisadas, a pressão de adsorção, a vazão de alimentação e o tempo do ciclo de adsorção, mostrando que o aumento da vazão de alimentação leva a perda da especificação que pode ser retomada reduzindo-se o tempo do ciclo de adsorção. Mostrou-se também que uma pressão de adsorção maior leva a uma maior remoção de contaminantes.

Esta dissertação avaliou a reação de amidação de poli(metacrilato de metila) (PMMA) comercial de massa molecular de 90000 g/mol (determinado por viscosimetria) com alilamina. A reação foi estudada a temperatura ambiente e sob refluxo. A reação apresentou baixos índices de conversão, 0,5 %, tanto após 24 dias, a temperatura ambiente, quanto sob refluxo por 2 horas. Experimentos executados sob refluxo por períodos maiores apresentaram diminuição no índice de conversão. Todos os experimentos foram acompanhados pela análise elementar e FT-IR e RMN-1H. O material derivatizado foi epoxidado pelo tratamento com ácido m-cloro-perbenzóico (AMCPB) seguido de tratamento com ácido periódico gerado in situ (NaIO4/HCl). Todos os produtos preparados foram submetidos à análise térmica apresentando, em sua maioria, um aumento na Tg relativa ao PMMA

Great advances have been, and are being made in our knowledge of the genetics and molecular biology (including genomics, proteomics and structural biology). Global molecular profiling technologies such as microassays using DNA or oligonucleotide chip, and protein and lipid chips are being developed. The application of such biotechnological advances are inevitable in aquaculture in the areas of improvement of aquaculture stocks where many molecular markers such as RFLPs, AFLDs and RAPD are now available for genome analysis, finger printing and genetic linkage mapping. Transgenic technology has been developed in a number of fish species and research is being pursed to produce transgenic fish carrying genes that encode antimicrobial peptides such as lysozyme thereby achieving disease resistance in fish. Also it is a short cut to achieving genetic change for fast growth and other desirable traits like early sexual maturity, temperature tolerance and feed conversion efficiency. Three different stocking rates in a semi-intensive pilot shrimp project was adopted in duplicates of three treatments designated as T1, T2 and T3 having initial per meter square stocking density of shrimp of 40, 44 and 51 respectively of 0.025g size post larva. The study was conducted for 84 days. Commercial pelleted diets designated as starter - 1, 2, 3 and grower were fed at a satiation level during the study period with a feeding frequency of 4 to 5 times per day. Feed rationing was based on the survivability, body weight and tray checking. 5-25% of the pond water was exchanged daily. Sampling was done for growth after every 2nd week. Monthly sampling was done for mortality in the ponds. Mean weight gain of the shrimp in treatments T1, T2 and T3 were 16.96 ± 1.14, 16.04 ± 1.38 and 14.08 ± 1.17g respectively and T1 with a low stocking density showed a significantly best growth among the treatments. Total mortality in treatments T1, T2 and T3 were as 30.00, 39.77 and 31.37% respectively. Significantly higher feed conversion ratio (FCR) of 1.87 was obtained with shrimp in treatment T3 followed by shrimp in T1 and T2 with FCR values of 1.70 and 1.41 respectively. A positive correlation of growth and salinity was observed during the study. Total production per unit area was the highest in the treatment T3 (4928 kg/ha) and followed by T1 (4747 kg/ha) and T2 (4251 kg/ha). The result show significantly negative correlation between individual growth and density.
