Redox imbalance generates multiple cellular damages leading to oxidative stress-mediated pathological conditions such as neurodegenerative diseases and cancer progression. Therefore, maintenance of reactive oxygen species (ROS) homeostasis is most important that involves well-defined antioxidant machinery. In the present study, we have identified for the first time a component of mammalian protein translocation machinery Magmas to perform a critical ROS regulatory function. Magmas overexpression has been reported in highly metabolically active tissues and cancer cells that are prone to oxidative damage. We found that Magmas regulates cellular ROS levels by controlling its production as well as scavenging. Magmas promotes cellular tolerance toward oxidative stress by enhancing antioxidant enzyme activity, thus preventing induction of apoptosis and damage to cellular components. Magmas enhances the activity of electron transport chain (ETC) complexes, causing reduced ROS production. Our results suggest that J-like domain of Magmas is essential for maintenance of redox balance. The function of Magmas as a ROS sensor was found to be independent of its role in protein import. The unique ROS modulatory role of Magmas is highlighted by its ability to increase cell tolerance to oxidative stress even in yeast model organism. The cytoprotective capability of Magmas against oxidative damage makes it an important candidate for future investigation in therapeutics of oxidative stress-related diseases.

We have synthesized a series of 4'-aryl substituted 2,2':6',2 `'-terpyridine (terpy) derivatives, namely 4'-(4-methylphenyl)-2,2':6',2 `'-terpyridine (C-1), 4'-(2-furyl)-2,2':6'2 `'-terpyridine (C-2), and 4'-(3,4,5-trimethoxyphenyl)-2,2':6',2 `'-terpyridine (C-3). The synthesized terpy compounds were characterized by elemental analyses, FTIR, NMR (H-1 and C-13), and ESI-Mass spectrometry. Photophysical, electrochemical and thermal properties of terpy compounds were systematically studied. Maximum excitation band was observed between 240 and 330 nm using UV-visible spectra, and maximum emission peaks from PL spectra were observed at 385, 405 and 440 nm for C-1, C-2 and C-3 respectively. Fluorescence lifetime (tau) of the fluorophores was found to be 035 and 1.55 ns at the excitation wavelength of 406 nm for C-1 and C-2 respectively, and tau value for C-3 was found to be 0.29 ns at the excitation wavelength of 468 nm. We noticed that the calculated values of HOMO energy levels were increased from 5.96 (C-1) to 6.08 (C-3) eV, which confirms that C-3 derivative is more electrons donating in nature. The calculated electrochemical band gaps were 2.95, 2.82 and 3.02 eV for C-1, C-2 and C-3 respectively. These blue fluorescent emitter derivatives can be used as an electron transport and electroluminescent material to design the blue fluorescent organic light emitting diode (OLED) applications. Biogenesis of the iron-sulfur (Fe-S) cluster is an indispensable process in living cells. In mammalian mitochondria, the initial step of the Fe-S cluster assembly process is assisted by the NFS1-ISD11 complex, which delivers sulfur to scaffold protein ISCU during Fe-S cluster synthesis. Although ISD11 is an essential protein, its cellular role in Fe-S cluster biogenesis is still not defined. Our study maps the important ISD11 amino acid residues belonging to putative helix 1 (Phe-40), helix 3 (Leu-63, Arg-68, Gln-69, Ile-72, Tyr-76), and C-terminal segment (Leu-81, Glu-84) are critical for in vivo Fe-S cluster biogenesis. Importantly, mutation of these conserved ISD11 residues into alanine leads to its compromised interaction with NFS1, resulting in reduced stability and enhanced aggregation of NFS1 in the mitochondria. Due to altered interaction with ISD11 mutants, the levels of NFS1 and Isu1 were significantly depleted, which affects Fe-S cluster biosynthesis, leading to reduced electron transport chain complex (ETC) activity and mitochondrial respiration. In humans, a clinically relevant ISD11 mutation (R68L) has been associated in the development of a mitochondrial genetic disorder, COXPD19. Our findings highlight that the ISD11 R68A/R68L mutation display reduced affinity to form a stable subcomplex with NFS1, and thereby fails to prevent NFS1 aggregation resulting in impairment of the Fe-S cluster biogenesis. The prime affected machinery is the ETC complex, which showed compromised redox properties, causing diminished mitochondrial respiration. Furthermore, the R68L ISD11 mutant displayed accumulation of mitochondrial iron and reactive oxygen species, leading to mitochondrial dysfunction, which correlates with the phenotype observed in COXPD19 patients.

Functional linkage between reef habitat quality and fish growth and production has remained elusive. Most current research is focused on correlative relationships between a general habitat type and presence/absence of a species, an index of species abundance, or species diversity. Such descriptive information largely ignores how reef attributes regulate reef fish abundance (density-dependent habitat selection), trophic interactions, and physiological performance (growth and condition). To determine the functional relationship between habitat quality, fish abundance, trophic interactions, and physiological performance, we are using an experimental reef system in the northeastern Gulf of Mexico where we apply advanced sensor and biochemical technologies. Our study site controls for reef attributes (size, cavity space, and reef mosaics) and focuses on the processes that regulate gag grouper (Mycteroperca microlepis) abundance, behavior and performance (growth and condition), and the availability of their pelagic prey. We combine mobile and fixed-active (fisheries) acoustics, passive acoustics, video cameras, and advanced biochemical techniques. Fisheries acoustics quantifies the abundance of pelagic prey fishes associated with the reefs and their behavior. Passive acoustics and video allow direct observation of gag and prey fish behavior and the acoustic environment, and provide a direct visual for the interpretation of fixed fisheries acoustics measurements. New application of biochemical techniques, such as Electron Transport System (ETS) assay, allow the in situ measurement of metabolic expenditure of gag and relates this back to reef attributes, gag behavior, and prey fish availability. Here, we provide an overview of our integrated technological approach for understanding and quantifying the functional relationship between reef habitat quality and one element of production – gag grouper growth on shallow coastal reefs.

This thesis summarizes the application of conventional and modern electron paramagnetic resonance (EPR) techniques to establish proximity relationships between paramagnetic metal centers in metalloproteins and between metal centers and magnetic ligand nuclei in two important and timely membrane proteins: succinate:ubiquinone oxidoreductase (SQR) from Paracoccus denitrificans and particulate methane monooxygenase (pMMO) from Methylococcus capsulatus. Such proximity relationships are thought to be critical to the biological function and the associated biochemistry mediated by the metal centers in these proteins. A mechanistic understanding of biological function relies heavily on structure-function relationships and the knowledge of how molecular structure and electronic properties of the metal centers influence the reactivity in metalloenzymes. EPR spectroscopy has proven to be one of the most powerful techniques towards obtaining information about interactions between metal centers as well as defining ligand structures. SQR is an electron transport enzyme wherein the substrates, organic and metallic cofactors are held relatively far apart. Here, the proximity relationships of the metallic cofactors were studied through their weak spin-spin interactions by means of EPR power saturation and electron spin-lattice (T_1) measurements, when the enzyme was poised at designated reduction levels. Analysis of the electron T_1 measurements for the S-3 center when the b-heme is paramagnetic led to a detailed analysis of the dipolar interactions and distance determination between two interacting metal centers. Studies of ligand environment of the metal centers by electron spin echo envelope modulation (ESEEM) spectroscopy resulted in the identication of peptide nitrogens as coupled nuclei in the environment of the S-1 and S-3 centers.
Finally, an EPR model was developed to describe the ferromagnetically coupled trinuclear copper clusters in pMMO when the enzyme is oxidized. The Cu(II) ions in these clusters appear to be strongly exchange coupled, and the EPR is consistent with equilateral triangular arrangements of type 2 copper ions. These results offer the first glimpse of the magneto-structural correlations for a trinuclear copper cluster of this type, which, until the work on pMMO, has had no precedent in the metalloprotein literature. Such trinuclear copper clusters are even rare in synthetic models.

In LiNbO3:Fe, anomalous behaviour of grating erasure is observed with different wavelenghts, i.e. rapid grating erasure in the short wavelength range, which deviates from the results predicted by the electron transport band model. The deviation is related to the coexistance of electrons and holes in photorefraction, and charge-transfer process including electrons and hole has been proposed. The electron and hole contributions to photo-excitation coefficient S of the Fe centre on the wavelength.

Part I: The mobilities of photo-generated electrons and holes in orthorhombic sulfur are determined by drift mobility techniques. At room temperature electron mobilities between 0.4 cm2/V-sec and 4.8 cm2/V-sec and hole mobilities of about 5.0 cm2/V-sec are reported. The temperature dependence of the electron mobility is attributed to a level of traps whose effective depth is about 0.12 eV. This value is further supported by both the voltage dependence of the space-charge-limited, D.C. photocurrents and the photocurrent versus photon energy measurements.
As the field is increased from 10 kV/cm to 30 kV/cm a second mechanism for electron transport becomes appreciable and eventually dominates. Evidence that this is due to impurity band conduction at an appreciably lower mobility (4.10-4 cm2/V-sec) is presented. No low mobility hole current could be detected. When fields exceeding 30 kV/cm for electron transport and 35 kV/cm for hole transport are applied, avalanche phenomena are observed. The results obtained are consistent with recent energy gap studies in sulfur.
The theory of the transport of photo-generated carriers is modified to include the case of appreciable thermos-regeneration from the traps in one transit time.
Part II: An explicit formula for the electric field E necessary to accelerate an electron to a steady-state velocity v in a polarizable crystal at arbitrary temperature is determined via two methods utilizing Feynman Path Integrals. No approximation is made regarding the magnitude of the velocity or the strength of the field. However, the actual electron-lattice Coulombic interaction is approximated by a distribution of harmonic oscillator potentials. One may be able to find the “best possible” distribution of oscillators using a variational principle, but we have not been able to find the expected criterion. However, our result is relatively insensitive to the actual distribution of oscillators used, and our E-v relationship exhibits the physical behavior expected for the polaron. Threshold fields for ejecting the electron for the polaron state are calculated for several substances using numerical results for a simple oscillator distribution.

Indivíduos obesos apresentam maior risco de morbidade e mortalidade atribuída às doenças cardiovasculares. A composição da dieta é um fator que prediz o fenótipo cardíaco em resposta a obesidade e, o tipo de ácido graxo pode afetar de forma diferencial a estrutura e a função do miocárdio. Estudos têm mostrado que a disfunção mitocondrial exerce um papel chave na patogênese da insuficiência e hipertrofia cardíaca, e as alterações mitocondriais observadas em falhas cardíacas apontam para defeitos em sítios específicos da cadeia transportadora de elétrons. Desta forma, o objetivo deste estudo foi avaliar a função contrátil ventricular em camundongos, alimentados com dieta hiperlipídica, rica em ácidos graxos poliinsaturados, buscando elucidações através da bioenergética mitocondrial. Após desmame, camundongos machos C57Bl/6 passaram a receber dieta manipulada contendo 7% (C) ou 19% (HF) de óleo de soja, até os 135 dias de idade. A ingestão alimentar e a massa corporal foram monitoradas e foi realizado teste de tolerância à glicose. No final do período experimental, os animais foram anestesiados e submetidos à avaliação da composição corporal por Absortimetria de Raios X de Dupla Energia (DXA), e em seguida, sacrificados por exsanguinação. No plasma foram determinados o perfil lipídico e a insulina. O coração, o tecido adiposo intra-abdominal e o subcutâneo foram coletados, pesados, processados para análise histomorfológica. Fibras cardíacas do ventrículo esquerdo foram utilizadas para análise da respiração mitocondrial através de oxígrafo. O coração também foi utilizado para a técnica de perfusão de coração isolado de Langendorff, e para análise da expressão de proteínas relacionadas à bioenergética de cardiomiócitos, através de Western Blotting. O índice de HOMA e de adiposidade foram calculados. O grupo HF apresentou maior adiposidade, sem alteração na ingestão alimentar. Foi observada intolerância a glicose, hiperinsulinemia e resistência à insulina, além de alterações desfavoráveis no perfil lipídico. Foi observado alteração na morfologia cardíaca e quadro de cardiomiopatia hipertrófica, refletindo em alteração hemodinâmica, determinando maior contratilidade, maior pressão ventricular e função diastólica prejudicada. Em relação à atividade mitocondrial dos cardiomiócitos foi observada menor oxidação de carboidratos (-47%) e de ácidos graxos (-60%). Porém, sem alteração na expressão de proteínas relacionadas à bioenergética de cardiomiócitos, CPT1, UCP2, GLUT1, GLUT4, AMPK e pAMPK. A partir desses resultados, concluímos que o tipo e a quantidade de ácidos graxos predizem o fenótipo cardíaco na obesidade, promovendo alteração na capacidade oxidativa mitocondrial, na morfologia e na hemodinâmica cardíaca

本部分研究以菠菜和水稻为材料,比较系统的研究了高温对类囊体膜、PSII颗粒、PSII外周捕光天线LHCII、PSII核心复合物和PSII反应中心等不同层次膜蛋白结构与功能的影响,以探讨高温对光合膜蛋白的伤害机理。其主要结果如下: 1.类囊体膜结构与功能的完整性对于维持PSII的结构与功能在高温胁迫下的稳定性具有重要作用。当类囊体膜的完整性受到破坏,当与PSI有关的一系列保护机制失去作用时,PSII对高温胁迫的敏感性会大大加强。 2.虽然PSI的功能在高温下保持相对稳定,但PSI的结构在高温胁迫下并不稳定。本文的研究发现LHCI对高温非常敏感,在中度高温胁迫下就开始降解,但PSI的核心在高温下比较稳定,所以PSI介导的电子传递活性仍然维持在较高水平。 3.高温胁迫会对PSII的结构和功能产生多重破坏。这个过程首先应该是放氧复合体的失活:其次是反应中心的可逆失活;接下来可能是核心天线CP43和CP47的失活导致捕光天线同反应中心的能量传递受阻;再下来是QA到QR电子传递的受阻、反应中心的不可逆失活、捕光效率下降等过程;最后是大范围色素蛋白的变性和失活,PSII的结构和功能遭到彻底破坏。 4.高湿胁迫下Fo显著升高,Fo的升高的原因可能源于少量捕光天线同反应中心的分离和反应中心的失活。 5.高等植物体的类囊体膜中存在多种Chla和Chlb的光谱吸收形式。这些代表不同的光谱吸收形式的组分在高温胁迫下表现出不同程度的降解,其中C678 和C684组分降解最快。这些不同的光谱吸收形式组分可能以不同的比例存在于每一种色素蛋白复合物中。 6.LHCII的结构与功能对于维持PSII结构与功能的热稳定性具有重要作用,LHCII完全缺失的水稻突变体VG28及分离纯化的PSII核心复介物都人大增强了对高温的敏感性。但一种LHCII减少的水稻突变体249-Mutant,反而增加了PSII的热稳定性,进一步的研究表明,类囊体膜中 LHCII本身含量的多少对PSII热稳定性的影响不足决定性的,关键性因素可能主要取决于 LHCII含量改变而引起的膜脂组成和膜脂不饱和程度的改变,以及由膜脂变化引起的PSII放氧复合体结构与功能的变化。 7.本研究首次发现,高温可以促使分离纯化的LHCII的红区吸收光谱发生显著红移,而680nm处的荧光发射降低,长波长荧光组分大大增强。绿胶电泳表明中度高温胁迫能够诱导LHCII产生寡聚体,这种寡聚体可能在调节能量耗散方面具有重要生理意义:而严重高温胁迫下LHCII倾向于聚合产生大分子的非活性聚集体。 8.分离纯化的反应中心对高温非常敏感,各种色素的结构和功能在轻度高温胁迫下就开始受到破坏和抑制,各种色素变性和降解的顺序由快到慢是:P680>Pheo>Chla>β-Car。反应中心的多肽组分在高温胁迫下显著减少,Dl和D2减少的原因可能归因于高温胁迫导致大分子聚合物的产生,D2的减少显著快于D1的减少。

本文主要研究了在非离子表面活性剂-辛基硫代葡萄糖苷(OTG)处理条件下菠菜叶绿体中光系统I(PSI)颗粒的光合特性,主要的研究结果如下: 1. 在所研究的OTG浓度范围内(0.01~13%,w/v),OTG对PSI的电子传递活性有显著的促进作用。而与此相对照,阴离子表面活性剂-十二烷基硫酸钠(SDS)对PSI的活性具有抑制作用。 2. OTG对PSI在SDS和高温(70℃)处理后失去的电子传递活性具有恢复作用。 3. OTG对PSI的色素结合状态和能量传递都有很大的影响,影响程度和影响方式与OTG的浓度有关。在其临界胶束浓度(cmc)以上的一定浓度范围内,OTG处理会导致PSI捕光天线色素蛋白复合体(LHCI)的解离;而更大浓度的OTG会使PSI中产生自由色素。

为了阐明蕨类植物铁芒萁(Dicranopteris dichotoma Bernh.)体内稀土元素的分布及其光合特性,采用电感耦合等离子质谱分析了中国江西省龙南县轻、重稀土矿区和非矿区铁芒萁植物体内的稀土元素含量,并采用透射电子显微镜对其叶片细胞内的稀土元素进行精确定位。还比较系统的研究了自然条件下的铁芒萁与高浓度稀土元素处理条件下的非稀土元素富集植物黄瓜(Cucumis sativus Linn)的光合特性。结果表明: 1、0.5 mmol•L-1 LaCl3处理黄瓜后,可以诱导激发能向PS II分配。1和2 mmol•L-1 LaCl3处理黄瓜后,对黄瓜幼苗抑制作用表现在对其生长率,光合放氧活性和叶绿体完整率的抑制。这是由于LaCl3对黄瓜细胞结构和叶绿体膜结构的破坏所致。其表现为对类囊体膜结构的破坏,而导致PS II光合活性下降,并最终抑制黄瓜生长。 2、铁芒萁可以富集稀土元素,轻、重稀土矿区铁芒萁植物稀土元素的分布规律为叶片>根>土壤>茎>叶柄,非矿区铁芒萁植物稀土元素的分布规律为叶片>根>茎>叶柄。稀土元素在铁芒萁体内的运输和迁移过程中,发生了明显的分异作用,茎、叶柄、叶片中的重稀土相对贫乏,叶片中可以富集高浓度的轻稀土元素。 3、稀土元素可以进入完整的铁芒萁表皮细胞和叶肉细胞中,但多以沉淀的形式聚集在一起。非矿区铁芒萁叶绿体中的稀土元素含量约占其叶片中含量的5%。轻稀土矿区铁芒萁叶绿体中的稀土元素含量约占其叶片中含量的10%。部分稀土元素定位于富含PS II的基粒片层上。 4、铁芒萁富集稀土元素受环境和遗传特性的双重影响,但主要由其自身的生理、生化特性决定。其富集稀土元素的机制是隔离稀土元素在细胞壁、液泡中和分泌结合物质使稀土元素成为沉淀沉积下来,从而避免对光合活性的破坏。 5、与非矿区铁芒萁相比,轻稀土矿区植物叶绿体膜的全链电子传递速率增加了34.9%,PS II的电子传递活性增高了252.9%,PS I的电子传递活性增加了16.8%。轻稀土矿区铁芒萁全链电子传递活性的增加主要来自PS II电子传递活性的大幅提高,这可能与其调节激发能更多向PS II分配,提高PS II反应中心色素蛋白复合体(67.0%)和捕光色素蛋白复合体的含量相关。 6、与非矿区铁芒萁相比,重稀土矿区植物叶绿体膜的全链电子传递速率增加了46.3%,PS II的电子传递活性增高了23.8%,PS I的电子传递活性增加了60.4%。重稀土矿区铁芒萁电子传递活性的提高主要来自PS I电子传递活性的大量增加,这可能与其PS I反应中心蛋白复合体含量的提高(60.0%)有关。 铁芒萁富集并吸收稀土元素主要是由自身的理化特性决定的。它能够将稀土元素以沉淀的形式固定在细胞内部,并通过改变生理代谢来避免高浓度稀土元素对其光合作用的影响。可以在治理稀土元素污染的环保工程中用作植物修复材料。

A systematic study of the Cu-catalyzed chemical vapor deposition of graphene under extremely low partial pressure is carried out. A carbon precursor supply of just P CH4∼ 0.009 mbar during the deposition favors the formation of large-area uniform monolayer graphene verified by Raman spectra. A diluted HNO 3 solution is used to remove Cu before transferring graphene onto SiO 2/Si substrates or carbon grids. The graphene can be made suspended over a ∼12 μm distance, indicating its good mechanical properties. Electron transport measurements show the graphene sheet resistance of ∼0.6 kΩ/□ at zero gate voltage. The mobilities of electrons and holes are ∼1800 cm 2/Vs at 4.2 K and ∼1200 cm 2/Vs at room temperature. © 2011 IEEE.

Amyloid fibres displaying cytochrome b562 were probed using scanning tunnelling microscopy (STM) in vacuo. The cytochromes are electron transfer proteins containing a haem cofactor and could, in principle, mediate electron transfer between the tip and the gold substrate. If the core fibres were insulating and electron transfer within the 3D haem network was detected, then the electron transport properties of the fibre could be controlled by genetic engineering. Three kinds of STM images were obtained. At a low bias (<1.5 V) the fibres appeared as regions of low conductivity with no evidence of cytochrome mediated electron transfer. At a high bias, stable peaks in tunnelling current were observed for all three fibre species containing haem and one species of fibre that did not contain haem. In images of this kind, some of the current peaks were collinear and spaced around 10 nm apart over ranges longer than 100 nm, but background monomers complicate interpretation. Images of the third kind were rare (1 in 150 fibres); in these, fully conducting structures with the approximate dimensions of fibres were observed, suggesting the possibility of an intermittent conduction mechanism, for which a precedent exists in DNA. To test the conductivity, some fibres were immobilized with sputtered gold, and no evidence of conduction between the grains of gold was seen. In control experiments, a variation of monomeric cytochrome b562 was not detected by STM, which was attributed to low adhesion, whereas a monomeric multi-haem protein, GSU1996, was readily imaged. We conclude that the fibre superstructure may be intermittently conducting, that the cytochromes have been seen within the fibres and that they are too far apart for detectable current flow between sites to occur. We predict that GSU1996, being 10 nm long, is more likely to mediate successful electron transfer along the fibre as well as being more readily detectable when displayed from amyloid.

It is widely reported that threshold voltage and on-state current of amorphous indium-gallium-zinc-oxide bottom-gate thin-film transistors are strongly influenced by the choice of source/drain contact metal. Electrical characterisation of thin-film transistors indicates that the electrical properties depend on the type and thickness of the metal(s) used. Electron transport mechanisms and possibilities for control of the defect state density are discussed. Pilling-Bedworth theory for metal oxidation explains the interaction between contact metal and amorphous indium-gallium-zinc-oxide, which leads to significant trap formation. Charge trapping within these states leads to variable capacitance diode-like behavior and is shown to explain the thin-film transistor operation. © 2013 AIP Publishing LLC.

The physiological differences for three bloom-forming cyanobacteria (Cylindrospermopsis raciborskii, Microcystis aeruginosa, and Aphanizomenon flos-aquae) were investigated. In comparison with M. aeruginosa and A. flos-aquae, C. raciborskii exhibited a significantly higher concentration of carotenoids, higher values in maximum photosynthesis rate (P-m), apparent photosynthetic efficieny (a), and maximum electron transport rate (ETRmax) during the growth period. In addition, higher extracellular alkaline phosphatase activities and lower light compensation point (I-c) were also detected in C raciborskii (p < 0.05, ANOVA). Therefore, it is suggested that the higher photosynthetic activities, more effective uptake and utilization to phosphate, and low light requirements might play important roles in the occurrence and invasive behavior of C. raciborskii.
