Predominant usage of V beta 8.2 gene segments, encoding a T-cell receptor (TCR) beta chain variable region, has been reported for pathogenic Lewis rat T cells reactive to myelin basic protein (MBP). However, up to 75% of the alpha/beta T cells in a panel of MBP-specific T-cell lines did not display TCR V beta 8.2, V beta 8.5, V beta 10, or V beta 16 elements. To further investigate TCR usage, we sorted the T-cell lines for V beta 8.2- and V beta 10-positive T cells or depleted the lines of cells with these TCRs. V beta 8.2-positive T cells and one of the depleted T-cell lines strongly reacted against the MBP peptide MBP-(68-88). The depleted T-cell line caused marked experimental autoimmune encephalomyelitis (EAE) even in Lewis rats in which endogenous V beta 8.2-positive T cells had been eliminated by neonatal treatment with anti-V beta 8.2 monoclonal antibodies. T-cell hybridomas generated from this line predominantly used V beta 3 TCR genes coexpressed with TCR V alpha 2 transcripts, which were also used by V beta 8.2-positive T cells. Furthermore, V beta 10-positive T cells reactive to MBP-(44-67) were encephalitogenic when injected immediately after positive selection. After induction of EAE by sorted V beta 8.2- or V beta 10-positive T-cell lines, immunocytochemical analysis of the spinal cord tissue showed a predominance of the injected TCR or of nontypable alpha/beta T cells after injection of the depleted line. Our results demonstrate heterogeneity of TCR beta-chain usage even for a single autoantigen in an inbred strain. Moreover, V beta 8.2-positive T cells are not essential for the induction and progression of adoptive-transfer EAE.

Superantigens such as the staphylococcal enterotoxins can play an important role in exacerbation of autoimmune disorders such as experimental allergic encephalomyelitis (EAE) in mice. In fact, superantigens can reactivate EAE in PL/J mice that have been sensitized to rat myelin basic protein (MBP). The T-cell subset predominantly responsible for disease in PL/J mice bears the V beta 8+ T-cell antigen receptor (TCR). The question arises as to whether T cells bearing other V beta specificities are involved in induction or reactivation of EAE with superantigen. Thus, we have investigated the ability of a non-V beta 8-specific superantigen, staphylococcal enterotoxin A (SEA) (V beta specificities 1, 3, 10, 11, and 17), to induce EAE in PL/J mice that have been previously protected from disease by anergy and deletion of V beta 8+ T cells. PL/J mice were first pretreated with the V beta 8-specific superantigen staphylococcal enterotoxin B (SEB) and then immunized with MBP. These mice exhibited V beta 8-specific anergy and depletion and did not develop EAE, even when further treated with SEB. However, administration of SEA to these same mice induced an initial episode of EAE which was characterized by severe hindleg paralysis and accelerated onset of disease. In contrast to SEB pretreatment, PL/J mice pretreated with SEA did develop EAE when immunized with MBP, and after resolution of clinical signs of disease these mice were susceptible to relapse of EAE induced by SEB but not by SEA. Thus, superantigens can activate encephalitogenic MBP-specific non-V beta 8+ T cells to cause EAE in PL/J mice. These data suggest that superantigens can play a central role in autoimmune disorders and that they introduce a profound complexity to autoimmune diseases such as EAE, akin to the complexity seen in multiple sclerosis.

Experimental allergic encephalomyelitis (EAE) is an autoimmune disease of the central nervous system that serves as a model for the human disease multiple sclerosis. We evaluated rolipram, a type IV phosphodiesterase inhibitor, for its efficacy in preventing EAE in the common marmoset Callithrix jacchus. In a blinded experimental design, clinical signs of EAE developed within 17 days of immunization with human white matter in two placebo-treated animals but in none of three monkeys that received rolipram (10 mg/kg s.c. every other day) beginning 1 week after immunization. In controls, signs of EAE were associated with development of cerebrospinal fluid pleocytosis and cerebral MRI abnormalities. In the treatment group, there was sustained protection from clinical EAE, transient cerebrospinal fluid pleocytosis in only one of three animals, no MRI abnormality, and marked reduction in histopathologic findings. Rolipram-treated and control animals equally developed circulating antibodies to myelin basic protein. Thus, inhibition of type IV phosphodiesterase, initiated after sensitization to central nervous system antigens, protected against autoimmune demyelinating disease.

La présence d’Escherichia coli pathogènes en élevages porcins entraine des retards de croissance et la mortalité. La transmission des E. coli pathogènes entre les élevages et l'abattoir d’un même réseau de production n'est pas bien décrite. La détection des gènes de virulence des E. coli pathogènes pourrait permettre d’identifier un marqueur de contamination dans le réseau. L’objectif de cette étude a été d’identifier un marqueur de contamination E. coli dans un réseau de production porcine défini afin de décrire certains modes de transmission des E. coli pathogènes. Pour ce faire, une région géographique comprenant 10 fermes d’engraissement, un abattoir et un réseau de transport a été sélectionnée. Trois lots de production consécutifs par ferme ont été suivis pendant 12 mois. Des échantillons environnementaux ont été prélevés à l’intérieur et à l’extérieur des fermes (3 visites d’élevage), dans la cour de l’abattoir (2 visites lors de sorties de lot) et sur le camion de transport. La détection des gènes de virulence (eltB, estA, estB, faeG, stxA, stx2A, eae, cnf, papC, iucD, tsh, fedA) dans les échantillons a été réalisée par PCR multiplexe conventionnelle. La distribution temporelle et spatiale des gènes de virulence a permis d’identifier le marqueur de contamination ETEC/F4 défini par la détection d’au moins un gène d’entérotoxine ETEC (estB, estA et eltB) en combinaison avec le gène de l’adhésine fimbriaire (faeG). La distribution des échantillons positifs ETEC/F4 qualifie la cour de l’abattoir comme un réservoir de contamination fréquenté par les transporteurs, vecteurs de contamination entre les élevages. Ceci suggère le lien microbiologique entre l’élevage, les transporteurs et l’abattoir jouant chacun un rôle dans la dissémination des microorganismes pathogènes et potentiellement zoonotiques en production porcine.

BACKGROUND Intravenous immunoglobulin (IVIG) proved to be an efficient anti-inflammatory treatment for a growing number of neuroinflammatory diseases and protects against the development of experimental autoimmune encephalomyelitis (EAE), a widely used animal model for multiple sclerosis (MS). METHODS The clinical efficacy of IVIG and IVIG-derived F(ab')2 fragments, generated using the streptococcal cysteine proteinase Ide-S, was evaluated in EAE induced by active immunization and by adoptive transfer of myelin-specific T cells. Frequency, phenotype, and functional characteristics of T cell subsets and myeloid cells were determined by flow cytometry. Antibody binding to microbial antigen and cytokine production by innate immune cells was assessed by ELISA. RESULTS We report that the protective effect of IVIG is lost in the adoptive transfer model of EAE and requires prophylactic administration during disease induction. IVIG-derived Fc fragments are not required for protection against EAE, since administration of F(ab')2 fragments fully recapitulated the clinical efficacy of IVIG. F(ab')2-treated mice showed a substantial decrease in splenic effector T cell expansion and cytokine production (GM-CSF, IFN-γ, IL-17A) 9 days after immunization. Inhibition of effector T cell responses was not associated with an increase in total numbers of Tregs but with decreased activation of innate myeloid cells such as neutrophils, monocytes, and dendritic cells. Therapeutically effective IVIG-derived F(ab')2 fragments inhibited adjuvant-induced innate immune cell activation as determined by IL-12/23 p40 production and recognized mycobacterial antigens contained in Freund's complete adjuvant which is required for induction of active EAE. CONCLUSIONS Our data indicate that F(ab')2-mediated neutralization of adjuvant contributes to the therapeutic efficacy of anti-inflammatory IgG. These findings might partly explain the discrepancy of IVIG efficacy in EAE and MS.

Upon activation, T cells require energy for growth, proliferation, and function. Effector T (Teff) cells, such as Th1 and Th17 cells, utilize high levels of glycolytic metabolism to fuel proliferation and function. In contrast, Treg cells require oxidative metabolism to fuel suppressive function. It remains unknown how Teff/Treg-cell metabolism is altered when nutrients are limited and leptin levels are low. We therefore examined the role of malnutrition and associated hypoleptinemia on Teff versus Treg cells. We found that both malnutrition-associated hypoleptinemia and T cell-specific leptin receptor knockout suppressed Teff-cell number, function, and glucose metabolism, but did not alter Treg-cell metabolism or suppressive function. Using the autoimmune mouse model EAE, we confirmed that fasting-induced hypoleptinemia altered Teff-cell, but not Treg-cell, glucose metabolism, and function in vivo, leading to decreased disease severity. To explore potential mechanisms, we examined HIF-1α, a key regulator of Th17 differentiation and Teff-cell glucose metabolism, and found HIF-1α expression was decreased in T cell-specific leptin receptor knockout Th17 cells, and in Teff cells from fasted EAE mice, but was unchanged in Treg cells. Altogether, these data demonstrate a selective, cell-intrinsic requirement for leptin to upregulate glucose metabolism and maintain function in Teff, but not Treg cells.

L’inflammation du système nerveux central (SNC), appelée neuroinflammation, est un aspect inséparable des maladies neurodégénératives chroniques comme la sclérose en plaques (SEP) et la maladie d’Alzheimer (MA). La caractérisation de la signature moléculaire spécifique à chaque population cellulaire dans des pathologies distinctes va aboutir à la compréhension et donc au contrôle de la neuroinflammation. Le présent ouvrage a pour but de mieux comprendre les mécanismes d’action de deux types cellulaires myéloïdes, la microglie et les neutrophiles, au cours des affections neuroinflammatoires du SNC. Ainsi, le premier objectif a été de comprendre le rôle des cytokines IL-36 dans la neuroinflammation établie au cours de l’encéphalomyélite auto-immune expérimentale (EAE). Dans une seconde partie, l’objectif a été d’explorer l’action du GPR84, un récepteur couplé à la protéine G spécifique à la microglie dans le SNC, lors de l’altération des fonctions cérébrales dans un modèle de souris transgénique de la MA. Nos résultats démontrent que la voie de signalisation IL-36/IL36R est augmentée dans trois modèles différents de l’EAE, mais ne contribue pas au développement ni à la progression de la pathologie. En utilisant l’approche de cytométrie en flux nous identifions les neutrophiles comme la source majeure de l’IL-36γ. De plus, nous démontrons que la microglie exprime l’IL-36R et sa stimulation par l’IL-36γ conduit à la production de cytokines pro-inflammatoires. Dans un second temps, nous caractérisons l’augmentation de l’expression du GPR84 par la microglie dans le modèle murin de la MA APP/PS1. Ainsi, le croisement de ces souris avec des souris déficientes en GPR84 diminue l’activation et le recrutement de la microglie autour des plaques d’amyloïde-β et accélère le déclin cognitif. Nos études impliquent le GPR84 comme un acteur important dans le maintien de l’homéostasie neuronale puisque son absence favorise la dégénérescence des dendrites dans le cerveau. Les résultats obtenus dans cette thèse apportent de nouveaux éléments qui peuvent contribuer au développement des thérapies qui ciblent les cellules myéloïdes dans diverses pathologies du SNC. Ces données ouvrent de nouvelles pistes pour élucider le rôle de l’IL-36γ dans des maladies neurodégénératives. Enfin, pour une première fois, nous présentons un modèle murin permettant d’identifier le(s) ligand(s) endogène(s) du GPR84, une cible thérapeutique potentielle pour la prévention et/ou le traitement de la MA.

This thesis describes the application of multispectral imaging to several novel oximetry applications. Chapter 1 motivates optical microvascular oximetry, outlines oxygen transport in the body, describes the theory of oximetry, and describes the challenges associated with in vivo oximetry, in particular imaging through tissue. Chapter 2 reviews various imaging techniques for quantitative in vivo oximetry of the microvasculature, including multispectral and hyperspectral imaging, photoacoustic imaging, optical coherence tomography, and laser speckle techniques. Chapter 3 describes a two-wavelength oximetry study of two microvascular beds in the anterior segment of the eye: the bulbar conjunctival and episcleral microvasculature. This study reveals previously unseen oxygen diffusion from ambient air into the bulbar conjunctival microvasculature, altering the oxygen saturation of the bulbar conjunctiva. The response of the bulbar conjunctival and episcleral microvascular beds to acute mild hypoxia is quantified and the rate at which oxygen diffuses into bulbar conjunctival vessels is measured. Chapter 4 describes the development and application of a highly novel non-invasive retinal angiography technique: Oximetric Ratio Contrast Angiography (ORCA). ORCA requires only multispectral imaging and a small perturbation of blood oxygen saturation to produce angiographic sequences. A pilot study of ORCA in human subjects was conducted. This study demonstrates that ORCA can produce angiographic sequences with features such as sequential vessel filling and laminar flow. The application and challenges of ORCA are discussed, with emphasis on comparison with other angiography techniques, such as fluorescein angiography. Chapter 5 describes the development of a multispectral microscope for oximetry in the spinal cord dorsal vein of rats. Measurements of blood oxygen saturation are made in the dorsal vein of both healthy rats, and in rats with the Experimental autoimmune encephalomyelitis (EAE) disease model of multiple sclerosis. The venous blood oxygen saturation of EAE disease model rats was found to be significantly lower than that of healthy controls, indicating increased oxygen uptake from blood in the EAE disease model of multiple sclerosis. Chapter 6 describes the development of video-rate red eye oximetry; a technique which could enable stand-off oximetry of the blood-supply of the eye with high temporal resolution. The various challenges associated with video-rate red eye oximetry are investigated and their influence quantified. The eventual aim of this research is to track circulating deoxygenation perturbations as they arrive in both eyes, which could provide a screening method for carotid artery stenosis, which is major risk-factor for stroke. However, due to time constraints, it was not possible to thoroughly investigate if video-rate red eye can detect such perturbations. Directions and recommendations for future research are outlined.

Shiga toxin-producing Escherichia coli (STEC) and enteropathogenic E. coli (EPEC) strains may be responsible for food-borne infections in humans. Twenty-eight STEC and 75 EPEC strains previously isolated from French shellfish-harvesting areas and their watersheds and belonging to 68 distinguishable serotypes were characterized in this study. High-throughput real-time PCR was used to search for the presence of 75 E. coli virulence-associated gene targets, and genes encoding Shiga toxin (stx) and intimin (eae) were subtyped using PCR tests and DNA sequencing, respectively. The results showed a high level of diversity between strains, with 17 unique virulence gene profiles for STEC and 56 for EPEC. Seven STEC and 15 EPEC strains were found to display a large number or a particular combination of genetic markers of virulence and the presence of stx and/or eae variants, suggesting their potential pathogenicity for humans. Among these, an O26:H11 stx1a eae-β1 strain was associated with a large number of virulence-associated genes (n = 47), including genes carried on the locus of enterocyte effacement (LEE) or other pathogenicity islands, such as OI-122, OI-71, OI-43/48, OI-50, OI-57, and the high-pathogenicity island (HPI). One O91:H21 STEC strain containing 4 stx variants (stx1a, stx2a, stx2c, and stx2d) was found to possess genes associated with pathogenicity islands OI-122, OI-43/48, and OI-15. Among EPEC strains harboring a large number of virulence genes (n, 34 to 50), eight belonged to serotype O26:H11, O103:H2, O103:H25, O145:H28, O157:H7, or O153:H2.

We previously identified heme oxygenase 1 (HO-1) as a specific target of miR-155, and inhibition of HO-1 activity restored the capacity of miR-155-/- CD4+ T cells to promote antigendriven inflammation after adoptive transfer in antigen-expressing recipients. Protoporphyrins are molecules recognized for their modulatory effect on HO-1 expression and function. In the present study, we investigated the effect of protoporphyrin treatment on the development of autoimmunity in miR-155-deficient mice. MiR-155-mediated control of HO-1 expression in promoting T cell-driven chronic autoimmunity was confirmed since HO-1 inhibition restored susceptibility to experimental autoimmune encephalomyelitis (EAE) in miR-155- deficient mice. The increased severity of the disease was accompanied by an enhanced T cell infiltration into the brain. Taken together, these results underline the importance of miR- 155-mediated control of HO-1 expression in regulating the function of chronically-stimulated T cells in EAE.

El estrés laboral se presenta como una serie de reacciones físicas y emocionales que ocurren cuando las exigencias del trabajo no igualan las capacidades, los recursos o necesidades del profesional en anestesia y enfermería. Cuando éste se torna excesivo superando la tolerancia del organismo llevando consigo graves consecuencias como desgaste en la salud, actuación profesionalmente pobre, repercusión en la seguridad del paciente, en la vida familiar, deterioro psíquico-emocional. El objetivo es identificar el nivel estrés que presenta el personal de sala de operaciones que labora en el Hospital Nacional General "Dr. Jorge Arturo Mena" Santiago de María, y así conocer sintomatología psicosomática y nivel de estrés. La metodología utilizada en el estudio fue la hipotética deductiva. Para la recolección de la información se hizo uso de un cuestionario integrado inicialmente por datos generales y además un test psicológico dirigido al estrés laboral en profesionales de anestesiología y enfermería el cual consta: el cuestionario EAE está formado por 50 preguntas el cual midió el nivel de estrés laboral en el que se encontró el personal de anestesia y enfermería. La población fue constituida por 15 profesionales en anestesiología y 15 profesionales de enfermería que trabajan en el Hospital Nacional general "Dr. Jorge Arturo Mena" Santiago de María, Usulután, y tomando en cuenta es una población pequeña se incluyeron a todos los sujetos en el estudio. Los resultados fueron procesados mediante el Programa Estadístico Para las Ciencias Sociales (SPSS) versión 19, con el que se realizaron tablas de frecuencia y la prueba U de Mann Whitney, obteniendo que la mayoría de anestesistas tienen nivel de estrés bajo y en el personal de enfermería prevalece el nivel de estrés medio.

El interferón beta (IFNb) ha sido uno de los fármacos más utilizados en el tratamiento de la esclerosis múltiple (EM), gracias a su efecto inmunomodulador, antiproliferativo y antiviral. Sin embargo, existe un porcentaje de pacientes que responden de forma subóptima al tratamiento, sugiriendo la necesidad de buscar alternativas terapéuticas innovadoras. En este contexto se ha observado que las células madre mesenquimales derivadas del tejido adiposo (AdMSCs) presentan capacidad inmunomoduladora, neuroprotectora y regeneradora del tejido dañado en ensayos preclínicos con el modelo animal más frecuentemente usado en el estudio de la EM, la Encefalomielitis Autoinmune Experimental (EAE), lo que las hace buenas candidatas como terapia alternativa para la EM. Además, su capacidad de migración hacia el tejido dañado les confiere potencial para ser utilizadas como transportadoras de factores terapéuticos hacia las zonas lesionadas del SNC. Por ello, nos planteamos evaluar la eficacia terapéutica de las terapias celular con AdMSCs y génica con AdMSCs modificadas genéticamente para producir IFNb, en modelos de EAE. Para llevarlo a cabo se realizó la caracterización de la población de AdMSCs de la cepa de ratón SJL/jCrL (SJL-AdMSCs), usando como control las AdMSCs de la cepa C57BL/6, ampliamente caracterizadas en la literatura, la generación de líneas de AdMSCs secretoras de IFNb (AdMSCs-IFNb) mediante lentivirus y su posterior caracterización y comparación con las mismas células sin transducir, la evaluación de los efectos de las terapias celulares autóloga, alogénica y génica en los modelos de EAE crónico progresivo (EAE-CP) y remitente recurrente (EAE-RR) y el estudio de la migración de las AdMSCs administradas como terapia autóloga y de las AdMSCs-IFNb. Los resultados obtenidos en cada uno de los objetivos planteados nos condujeron a una serie de conclusiones: las SJL-AdMSCs aisladas, cultivadas y expandidas bajo nuestras condiciones experimentales, cumplen los criterios mínimos determinados para ser consideradas células madre mesenquimales. Además, estas células presentan eficacia clínica y efectos neuroinmunomoduladores al ser utilizadas como transplantes autólogos y alogénicos en animales con EAE-RR y EAE-CP respectivamente. Por otro lado, las SJL-AdMSCs constituyen una población apta para dar soporte al desarrollo de la terapia génica, ya que la alteración de su material genético por la inserción del IFNb no supone la modificación de sus propiedades biológicas ni funcionales en estudios preclínicos en modelos de EM. Estas AdMSCs-IFNb, constituyen una línea de células mesenquimales de crecimiento estable que produce elevados niveles de IFNb de forma constitutiva. Además, los transplantes con AdMSCs-IFNb son eficaces como tratamiento terapéutico en animales con EAE-RR y EAE-CP al modular tanto la sintomatología como los procesos inflamatorios y neurodegenerativos propios de la enfermedad. Sin embargo, los resultados no permiten discriminar si los efectos observados son debidos a las propiedades del inmunomodulador secretado, a las propias células mesenquimales o a la acción conjunta de ambos. En último lugar, la migración celular de las AdMSCs autólogas se potencia por los estados de inflamación activa en ambos modelos de EAE, mostrando una amplia biodistribución celular. La localización prioritaria fue inicialmente en pulmones y, posteriormente en zona de órganos linfáticos, como hígado y bazo, y del SNC a nivel de la médula espinal. La señal bioluminiscente de las AdMSCs-IFNb en el modelo EAE-CP es mayor que la emitida por las células de la terapia autóloga. Sin embargo, la migración de las células transfectadas no aparece fuertemente influenciada por los procesos proinflamatorios. En el modelo EAE-RR estas diferencias entre terapias son incluso más moderadas. Las áreas donde se registra señal son similares a las de las células autólogas, apareciendo principalmente en zonas correspondientes a pulmones, hígado, bazo y médula espinal a lo largo del tiempo experimental.

Ovine enzootic abortion, caused by Chlamydia abortus, leads to important economic losses worldwide. In addition to reproductive failures, infection may impact lamb growth during the first weeks after birth, yet this effect has not been well characterized. Vaccination can help to control the disease but variable efficacy values have been described, possibly related with factors associated with the host, the vaccine, the parameter used for efficacy determination and the challenge conditions. In this context, we evaluated the efficacy of an inactivated standard commercial vaccine and a 1/2 diluted dose in pregnant sheep challenged with C. abortus by examining multiple indicators ofvaccine effect (including incidence of reproductive failures, bacterial excretion, and evolution of weight gain of viable lambs during the first month of life). Three groups of ewes [control non-vaccinated, C (n = 18); vaccinated with standard dose, SV (n = 16) and vaccinated with 1/2 dose, DV (n = 17)], were challenged approximately 90 days post-mating and tested using direct PCR (tissue samples and vaginal swabs) and ELISA (serum) until 31 days post-reproductive outcome. There were not significant differences in the proportions of reproductive failures or bacterial shedding after birth/abortion regardless the vaccination protocol. However, a beneficial effect of vaccination on offspring growth was detected in both vaccinated groups compared with the controls, with a mean increase in weight measured at 30 days of life of 1.5 and 2.5 Kg (p = 0.056) and an increase in the geometric mean of the daily gain of 8.4 and 9.7% in lambs born from DV and SV ewes compared to controls, respectively. Our results demonstrate the effect of an inactivated vaccine in the development of the offspring of C. abortus-infected ewes at a standard and a diluted dose, an interesting finding given the difficulty in achieving sufficient antigen concentration in the production of EAE-commercial vaccines.

Etiological diagnosis of diarrheal diseases may be complicated by their multi-factorial nature. In addition, Escherichia coli strains present in the gut can occasionally harbor VGs without causing disease, which complicates the assessment of their clinical significance in particular. The aim of this study was to detect and quantify nine VGs (stx1, stx2, eae, aggR, ehxA, invA, est and elt) typically present in five E. coli enteric pathotypes (EHEC, ETEC, EPEC, EAEC and EIEC) in fecal samples collected from 49 patients with acute diarrhea and 32 healthy controls from Madrid, Spain. In addition, the presence of four serotype-related genes (wzxO104 and fliCH4, rbfO157 and fliCH7) was also determined. Presence of target genes was assessed using a quantitative real-time PCR assay previously developed, and the association of presence and burden of VGs with clinical disease and/or other risk factors was explored. Prevalence of ehxA (typically associated with STEC and EPEC), invA (EIEC) and the rbfO157+fliCH7 (STEC and/or STEC/EAEC) combination were significantly (p<0.02) higher in the diarrheic group, while the wzxO104+fliCH4 combination was significantly (p=0.014) more prevalent in the control group. On the other hand, eae was detected in more than 90% of the individuals in both patient and control populations, and it was not associated with bfpA, suggesting the absence of typical EPEC. No significant differences in the quantitative values were detected for any VG among study groups, but the difference in the load of aggR (EAEC) and invA in the patients with respect to the controls was close to the significance, suggesting a potential role of these VGs in the clinical signs observed when they are present at high levels.
