This paper studies the correlation properties of the speckles in the deep Fresnel diffraction region produced by the scattering of rough self-affine fractal surfaces. The autocorrelation function of the speckle intensities is formulated by the combination of the light scattering theory of Kirchhoff approximation and the principles of speckle statistics. We propose a method for extracting the three surface parameters, i.e. the roughness w, the lateral correlation length xi and the roughness exponent alpha, from the autocorrelation functions of speckles. This method is verified by simulating the speckle intensities and calculating the speckle autocorrelation function. We also find the phenomenon that for rough surfaces with alpha = 1, the structure of the speckles resembles that of the surface heights, which results from the effect of the peak and the valley parts of the surface, acting as micro-lenses converging and diverging the light waves.

More than thirty years after the discovery that Human Immunodeficiency Virus (HIV) was the causative agent of Acquired Immunodeficiency Syndrome (AIDS), the disease remains pandemic as long as no effective universal vaccine is found. Over 34 million individuals in the world are infected with the virus, and the vast majority of them have no access to the antiretroviral therapies that have largely reduced HIV to a chronic disease in the developed world. The first chapter of this thesis introduces the history of the virus. The key to the infectious mechanism of the virus lies in its envelope glycoprotein (Env), a trimeric spike on the viral surface that utilizes host T cell receptors for entry. Though HIV-1 Env is immunogenic, most infected patients do not mount an effective neutralizing antibody response against it. Broadly-neutralizing anti-Env antibodies (bNAbs) present in the serum of a minority of infected individuals are usually sufficient to prevent the progression to full blown AIDS. Thus, the molecular details of these bNAbs as well as the antibody-antigen interface are of prime interest for structural studies, as insight gained would contribute to the design of a more effective immunogen and potential vaccine candidate. The second chapter of this thesis describes the low-resolution crystal structure of one such antibody, 2G12 dimer, which targets a high mannose epitope on the surface of Env. Patients infected with HIV-2, a related virus with ~35% sequence identity in the Env region, can generally mount a robust antibody response sufficient for viral control for reasons still unknown. The final two chapters of this thesis focus on the first reported structural studies of HIV-2 Env, the molecular details of which may inform HIV-1 therapy and immunogen design.

O percurso histórico das representações da surdez, da educação de surdos e do estatuto da língua de sinais aponta para a necessidade de uma reflexão sobre as relações entre língua, cognição e cultura. Um estudo direcionado à identificação das estruturas conceptuais subjacentes à língua falada pelos surdos -a Libras- pode contribuir com algumas considerações pertinentes sobre a questão surdez/cultura, além de contribuir para desmistificar possíveis preconceitos relacionados à língua de sinais. A linguística cognitiva (LC), ciência que engloba os aspectos cognitivos envolvidos na significação, a influência do contexto para a compreensão/produção da linguagem e a forma como o mundo é experienciado individualmente e culturalmente, revela-se como um embasamento teórico adequado ao desenvolvimento de tal reflexão, uma vez que abarca dentre suas áreas de interesse o estudo dos mecanismos cognitivos de conceptualização e expressão da realidade, dentre os quais se inserem os modelos cognitivos e culturais, a metáfora e a metonímia conceptuais. Levando-se em conta que na LC a concepção de metáfora, estabelecida pela Teoria da Metáfora Conceptual (TMC), à luz de Lakoff e Johnson (2002[1980]) e Kövecses (2002, 2003, 2005), considera a metáfora como um mecanismo conceptual em que os seres humanos empregam um domínio experiencial mais concreto, estreitamente ligado à experiência com o próprio corpo e o mundo em que vivem, para compreender/conceptualizar um domínio mais abstrato; buscou-se, neste estudo, verificar a aplicabilidade de tal teoria na língua brasileira de sinais (Libras), hipotetizando-se que as metáforas conceptuais podem ser identificadas em qualquer língua, mesmo uma língua visuo-espacial, e que as manifestações metafóricas encontradas na Libras podem refletir as especificidades da cultura surda, bem como aspectos provenientes da cultura ouvinte devido à influência cultural gerada por sua inserção nesta cultura. A pesquisa realizada desenvolveu-se sob abordagem qualitativa/descritiva, com análise de um corpus heterogêneo da Libras, composto por sinais isolados, vídeos e transcrições de interações terapêuticas. Os resultados apontam não só para a manifestação da metáfora conceptual na Libras, como também para a manifestação de aspectos semânticos e fonológicos subjacentes à iconicidade cognitiva nos termos de (Wilcox, P. 2004) da Libras. Trata-se de um levantamento inicial, mas que fornece elementos para alguns questionamentos sobre o aspecto conceptual e cognitivo da iconicidade e sobre o alcance da TMC e sua relação com língua e cultura

Two kinds of nickel(II) and copper(II) P-diketone complexes derived from thenoyltrifluoroacetone ligand with blue-violet light absorption were synthesized by reacting free ligand and different metal(II) ions in sodium methoxide solution. Their structures were postulated based on elemental analysis, ESI-MS, FT-IR spectra and UV-vis electronic absorption spectra. Smooth films on K9 glass substrates were prepared using the spin-coating method. Two new hydrazone chelating ligands, 2-(2-(5-methylisoxazol-3-yl)hydrazono)-5,5-dimethylcyclohexane-1,3-dione (HL1) and 2-(2-(5-tert-butylisoxazol-3-yl)hydrazono)-5,5-dimethylcyclohexane- 1,3-dione (HL2), and their nickel(II) and copper(II) complexes were synthesized using the procedure of diazotization, coupling and metallization. Their structures were postulated based on elemental analysis, H-1 NMR, ESI-MS, FT-IR spectra and UV-vis electronic absorption spectra. Smooth films of these complexes on K9 glass substrates were prepared using spin-coating and their absorption properties were evaluated. Nanocrystalline Zn0.95-xNi0.05AlxO (x = 0.01, 0.02, 0.05 and 0.10) diluted magnetic semiconductors have been synthesized by an autocombustion method. X-ray absorption spectroscopy, high-resolution transmission electron microscopy, energy-dispersive spectrometry and Ni 2p core-level photoemission spectroscopy analyses revealed that some of the nickel ions were substituted for Zn2+ into the ZnO matrix while others gave birth to NiO nanoclusters embedded in the ZnO particles. The Zn0.95Ni0.05O sample showed no enhancement of room-temperature ferromagnetism after Al doping. (C) 2007 Acta Materialia Inc. Published by Elsevier Ltd. All rights reserved.
