Vários trabalhos vem sendo desenvolvidos sobre o cultivo in vitro de cacau (T.cacao), mas são raros para a maioria das outras espécies do gênero, como o cupuaçu (T. grandiflorum), cuja a área plantada vem aumentando expressivamente, e outras que poderiam servir de fonte de genes para as espécies economicamente já reconhecidas. Protocolos para obtenção de embriões somáticos in vitro para as espécies T. cacao,T. grandiflorum,T. speciosum e o híbrido T. grandiflorum x T. obovatum foram avaliados a partir de duas fontes de explantes, estaminódios e pétalas (formadas por lígulas e cógulas) cultivados em meio de crescimento primário de calo, consistindo de sais DKW, suplementado com 20 g l-1 de sacarose, 250 mg l-1de glutamina, 200 mg l-1 de mio-inositol, 0,2 mg l-1 de tiamina-HCl, 0,1 mg l-1 de ácido nicotínico, 0,2 mg l-1 de glicina, 2 mg l-1 de 2,4-D, 2,2 g l-1 de Gelrite® e pH 5,8. A este meio foram adicionadas diferentes concentrações de tidiazuron (0, 5 e 10 µg l-1). As culturas foram mantidas no escuro por 14 dias, à temperatura de 25 ± 2 ºC, e então transferidas para meio de crescimento secundário de calo, constituído de sais WPM, vitaminas de Gamborg, 20 g l-1 de sacarose, 2 mg l-1 de 2,4 D, 0,3 mg l-1 de cinetina, 50 ml l-1 de água de côco, 2,2 g l-1 de Gelrite® e pH 5,8. A formação de calos ocorreu em todas as espécies. Embriões somáticos foram obtidos somente para T. cacao. A calogênese mostrou-se influenciada pelo genótipo e foi maior nos estaminódios.

Chronic osteomyelitis with proliferative periostitis is a kind of osteomyelitis that is characterized by a great periosteal reaction resulting of low grade stimulation, usually a caries lesion associated a periapical pathology. It affects especially children and young adults, with the mandible as the most common site, usually in the inferior aspect. Clinically presents as a painless hard swelling intra and/or extra-oral. Radiographic aspects include bone layers new formed, parallel to each other and to the underlying cortical surface, giving the characteristic appearance of "onion skin". Treatment includes elimination of the cause, with or without antibiotics, being restored the facial symmetry of patient. The present article is a clinical case report of chronic osteomyelitis with proliferative periostitis affecting a female 11 year old patient, caused by a caries lesion in the left permanent first molar, that was submissed to a exodontia and the facial symmetry of the patient was restored with management.

Objectives: The aim of this study was to evaluate the association between serum levels of 25(OH) vitamin D-3 with midupper arm muscle circumference (MUAMC), handgrip strength and length of hospital stay (LOS) after hip fracture.Methods: In total, 102 consecutive patients with hip fracture over the age of 65 were admitted to the orthopedic unit and prospectively evaluated. All of the patients were treated according to specific protocols depending on the type of fracture. Anthropometric measurements and handgrip strength were performed, and blood samples were taken for serum biochemistry and 25(OH) vitamin D-3 analysis within the first 72 h of admission. All of the patients were followed during their hospital stay, and the length of stay was recorded.Results: Of the patients, two were excluded because of pathologic fractures. In total, 100 patients with a mean age of 80 +/- 7 y were included in the analysis. Among these patients, 73% were female, and 37% had vitamin D deficiency. The median LOS was 7 (5-11) d. Patients with vitamin D deficiency had lower handgrip strength in univariate analysis. In the multiple linear regression analysis with robust standard error, serum vitamin D levels adjusted by age and sex were associated with handgrip strength but not with MUAMC and LOS after hip fracture.Conclusions: In conclusion, vitamin D serum levels were associated with handgrip strength but not with muscle mass or length of hospital stay after hip fracture. Tephrocybella (Basidiomycetes) represents a novel genus from wood (Italy). Morphological and culture characteristics along with ITS DNA barcodes are provided for all taxa.

Investigou-se o período mínimo de colheitas sucessivas necessário para avaliação do desempenho de cultivares de cacau. Os cultivares locais não-melhorados 'Maranhão', 'Pará' e 'Parazinho', juntamente com os cultivares melhorados 'ICS 1' polinização livre e híbrido comercial, foram avaliados em quadrado latino 5 x 5 com parcelas de 196 plantas, durante dez anos (1984-93). O número de frutos sadios por planta (NFSP) e o peso de sementes úmidas por hectare (PSUH) e por fruto (PSUF) foram os componentes de produção avaliados. A análise dos coeficientes de correlação entre produções anuais e acumuladas no decênio mostrou a necessidade da combinação dos primeiros seis e oito anos de colheitas sucessivas para NFSP e PSUH, respectivamente, para avaliação dos cultivares. A ampla variabilidade e o desempenho superior no pós-clímax (1989-93) e a tendência de os cultivares interagirem com os anos no pré-clímax indicaram que a avaliação deve ser conduzida em cacaueiros cultivados em Linhares somente após o oitavo ano de cultivo.

The causal agent of witches' broom disease, Moniliophthora perniciosa is a hemibiotrophic and endemic fungus of the Amazon basin and the most important cocoa disease in Brazil. The purpose of this study was to analyze the genetic diversity of polysporic isolates of M. perniciosa to evaluate the adaptation of the pathogen from different Brazilian regions and its association with different hosts. Polysporic isolates obtained previously in potato dextrose agar cultures of M. perniciosa from different Brazilian states and different hosts (Theobroma cacao, Solanum cernuum, S. paniculatum, S. lycocarpum, Solanum sp, and others) were analyzed by somatic compatibility grouping where the mycelium interactions were distinguished after 4-8 weeks of confrontation between the different isolates of M. perniciosa based on the precipitation line in the transition zone and by protein electrophoresis through SDS-PAGE. The diversity of polysporic isolates of M. perniciosa was grouped according to geographic proximity and respective hosts. The great genetic diversity of M. perniciosa strains from different Brazilian states and hosts favored adaptation in unusual environments and dissemination at long distances generating new biotypes.

We have characterized in vitro and in vivo effects of trypsin inhibitors from Theobroma seeds on the activity of trypsin- and chymotrypsin-like proteins from Lepidopteran pest insects. The action of semipurified trypsin inhibitors from Theobroma was evaluated by the inhibition of bovine trypsin and chymotrypsin activities determined by the hydrolysis of N-Benzoyl-DL-Arginine-p-Nitroanilide (BAPA) and N-Succinyl-Ala-Ala-Pho-Phe p-Nitroanilide (S-(Ala)2ProPhe-pNA). Proteinase inhibitor activities from Theobroma cacao and T. obovatum seeds were the most effective in inhibiting trypsin-like proteins, whereas those from T. obovatum and T. sylvestre were the most efficient against chymotrypsin-like proteins. All larvae midgut extracts showed trypsin-like proteolytic activities, and the putative trypsin inhibitors from Theobroma seeds significantly inhibited purified bovine trypsin. With respect to the influence of Theobroma trypsin inhibitors on intact insects, the inclusion of T. cacao extracts in artificial diets of velvet bean caterpillars (Anticarsia gemmatalis) and sugarcane borer (Diatraea saccharalis) produced a significant increase in the percentage of adult deformation, which is directly related to both the survival rate of the insects and oviposition.

The forest-like characteristics of agroforestry systems create a unique opportunity to combine agricultural production with biodiversity conservation in human-modified tropical landscapes. The cacao-growing region in southern Bahia, Brazil, encompasses Atlantic forest remnants and large extensions of agroforests, locally known as cabrucas, and harbors several endemic large mammals. Based on the differences between cabrucas and forests, we hypothesized that: (1) non-native and non-arboreal mammals are more frequent, whereas exclusively arboreal and hunted mammals are less frequent in cabrucas than forests; (2) the two systems differ in mammal assemblage structure, but not in species richness; and (3) mammal assemblage structure is more variable among cabrucas than forests. We used camera-traps to sample mammals in nine pairs of cabruca-forest sites. The high conservation value of agroforests was supported by the presence of species of conservation concern in cabrucas, and similar species richness and composition between forests and cabrucas. Arboreal species were less frequently recorded, however, and a non-native and a terrestrial species adapted to open environments (Cerdocyon thous) were more frequently recorded in cabrucas. Factors that may overestimate the conservation value of cabrucas are: the high proportion of total forest cover in the study landscape, the impoverishment of large mammal fauna in forest, and uncertainty about the long-term maintenance of agroforestry systems. Our results highlight the importance of agroforests and forest remnants for providing connectivity in human-modified tropical forest landscapes, and the importance of controlling hunting and dogs to increase the value of agroforestry mosaics.

The enzyme chitinase from Moniliophthora perniciosa the causative agent of the witches' broom disease in Theobroma cacao, was partially purified with ammonium sulfate and filtration by Sephacryl S-200 using sodium phosphate as an extraction buffer. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes were obtained: ChitMp I, ChitMp II, ChitMp III and ChitMp IV. ChitMp I had an optimum temperature at 44-73ºC and an optimum pH at 7.0-8.4. ChitMp II had an optimum temperature at 45-73ºC and an optimum pH at 7.0-8.4. ChitMp III had an optimum temperature at 54-67ºC and an optimum pH at 7.3-8.8. ChitMp IV had an optimum temperature at 60ºC and an optimum pH at 7.0. For the computational biology, the primary sequence was determined in silico from the database of the Genome/Proteome Project of M. perniciosa, yielding a sequence with 564 bp and 188 amino acids that was used for the three-dimensional design in a comparative modeling methodology. The generated models were submitted to validation using Procheck 3.0 and ANOLEA. The model proposed for the chitinase was subjected to a dynamic analysis over a 1 ns interval, resulting in a model with 91.7% of the residues occupying favorable places on the Ramachandran plot and an RMS of 2.68.

[ES] La vida de las provincias de San Salvador y Sonsonate, que conformaron el actual El Salvador, en América Central, estuvo caracterizada por la dependencia de la capital y el monocultivo. Para la segunda mitad del siglo XVIII aconteció el despunte añilero (sustituyendo al cacao sonsonateco tras su declive) que tanta relevancia tuvo para la economía del reino y que repercutió de manera particular en las economías locales, «tironeando» el desarrollo de otros productos. El más sobresaliente de ellos fue la caña de azúcar, con una producción tan considerable que llegó a hacerse de una importante porción del mercado capitalino guatemalteco. El presente trabajo sostiene que esto sólo fue posible gracias al empleo del apante o regadío, ya presente en los limitados espacios tecnológicos y comerciales de aquella sociedad rural. Rompiendo los paradigmas del sistema productivo cañero, las elevadas posibilidades productivas del apante, los antecedentes de su uso y el mismo sistema productivo añilero, aseguraron el éxito de la empresa, en un ejemplo poco conocido del aporte de la tecnología indígena en las sociedades y economías del interior colonial.

The macroscopic properties of oily food dispersions, such as rheology, mechanical strength, sensory attributes (e.g. mouth feel, texture and even flavour release) and as well as engineering properties are strongly determined by their microstructure, that is considered a key parameter in the understanding of the foods behaviour . In particular the rheological properties of these matrices are largely influenced by their processing techniques, particle size distribution and composition of ingredients. During chocolate manufacturing, mixtures of sugar, cocoa and fat are heated, cooled, pressurized and refined. These steps not only affect particle size reduction, but also break agglomerates and distribute lipid and lecithin-coated particles through the continuous phase, this considerably modify the microstructure of final chocolate. The interactions between the suspended particles and the continuous phase provide information about the existing network and consequently can be associated to the properties and characteristics of the final dispersions. Moreover since the macroscopic properties of food materials, are strongly determined by their microstructure, the evaluation and study of the microstructural characteristics, can be very important for a through understanding of the food matrices characteristics and to get detailed information on their complexity. The aim of this study was investigate the influence of formulation and each process step on the microstructural properties of: chocolate type model systems, dark milk and white chocolate types, and cocoa creams. At the same time the relationships between microstructural changes and the resulting physico-chemical properties of: chocolate type dispersions model systems dark milk and white chocolate were investigated.
