O oxigênio é fundamental para os vertebrados. No entanto, variações dos níveis de oxigênio na água podem provocar estresse oxidante em peixes porque privação de oxigênio seguida de reoxigenação forma espécies reativas de oxigênio (ERO) em células. Níveis intracelulares de ERO aumentados favorecem que moléculas de proteínas, fosfolipídios e ácidos nucleicos sofram alterações, vindo a prejudicar muitas funções celulares. No Pantanal, habitat do pacu, o nível de oxigênio varia circadianamente na água das lagoas rasas que acabam isoladas dos rios na seca. O pacu evoluiu sob a pressão contínua da exposição aos efeitos prejudiciais das ERO causados pelos pulsos de inundação. A melatonina, uma indolamina produzida na glândula pineal, influencia os níveis de atividade de enzimas antioxidantes que reduzem ERO, além de ser capaz de doar elétrons ou captar radicais livres de forma não enzimática. Os níveis de melatonina no pacu são mais altos no verão e menores no inverno. Isoenzimas de glutationa S-transferases que conjugam o tripetídeo glutationa com o 4-hidroxinonenal, aldeído derivado da peroxidação de ácidos graxos por ERO, são importantes para evitar alteração funcional de proteínas por ligação do 4-hidroxinonenal à sua estrutura. Neste trabalho procuramos relação entre estresse oxidante, níveis de atividades de glutationa S-transferase e melatonina, para estabelecer se a melatonina ajudaria pacus a superar os efeitos deletérios das espécies reativas de oxigênio. Ensaiamos atividades de isoenzimas de glutationa S-transferases no citosol de fígado de pacus mantidos em normoxia, hipoxia, reoxigenação e hiperoxia no inverno e no verão. Medimos o efeito da melatonina in vitro e in vivo sobre as atividades de isoenzimas de glutationa S-transferase. Medimos os efeitos do estresse oxidante sobre a ligação do 4-hidroxinonenal com proteínas nos fígados de pacus tratados com melatonina. Somente as isoenzimas que conjugam 4-hidroxinonenal com glutationa mostraram menor atividade no inverno em relação ao verão; outras isoenzimas de glutationa S-transferases não alteram suas atividades sazonalmente. In vitro a melatonina não alterou a atividade de isoenzimas de glutationa S-transferase que conjugam o 4-hidroxinonenal, mas inibiu outras isoenzimas de glutationa S-transferase. In vivo a melatonina aumentou a atividade encontrada no inverno das isoenzimas que conjugam o 4-hidroxinonenal para os níveis do verão. A ligação de 4-hidroxinonenal com proteínas foi menor em pacus inoculados com melatonina. Nossos resultados mostram que a melatonina pode influenciar os efeitos de ERO em fígado de pacus. Ficou claro que a melatonina do plasma mantém os níveis de atividade conjugadora de 4-hidroxinonenal do fígado em pacus e que a baixa produção de melatonina no inverno não é adequada para a conjugação do 4-hidroxinonenal em fígado de pacus.

A ação inibitória dos organofosforados sobre as esterases, por ser específica, pode ser empregada como um eficiente biomarcador da exposição de seres vivos aos organofosforados. A inibição da acetilcolinesterase (AChE; EC 3.1.1.7) provoca acúmulo do neurotransmissor acetilcolina nas fendas sinápticas colinérgicas, o que pode resultar na morte do indivíduo. Outra atividade também afetada por organofosforados é a da enzima carboxilesterase (CarbE; EC 3.1.1.1). CarbE estão envolvidas na fase I da biotransformação de xenobióticos e atuam como captadoras (scavengers) de organofosfatos, incluindo os formados pela biotransformação dos organofosforados. As CarbE estudadas até hoje se ligam com maior velocidade aos organofosfatos do que as colinesterases. Por isto se admite que CarbE possam diminuir, por captação estequiométrica, a ligação tóxica de moléculas de organofosfatos às acetilcolinesterases das sinapses colinérgicas e das placas motoras dos músculos. Experimentos realizados em nosso laboratório mostraram que a atividade da CarbE está aproximadamente 50% menor no soro e no fígado de pacus submetidos à hipoxia. Por causa disso, em razão de uma possível diminuição da capacidade captadora da CarbE, decidimos verificar se o pacu em hipoxia seria mais sensível aos agrotóxicos organofosforados. Para este propósito foram colocados seis pacus divididos em dois tanques. No primeiro tanque, os animais foram submetidos a 24 horas de hipoxia seguidos por mais 4 horas de exposição ao organofosforado metilparation em duas concentrações diferentes (0,02 ou 0,01 mg / L). No segundo tanque os animais permaneceram em normoxia durante o mesmo período de 24 horas e depois foram expostos ao metilparation como no primeiro tanque. As atividades da AChE ensaiada com acetiltiocolina, a da butirilcolinesterase (BChE) ensaiada com butiriltiocolina e a da CarbE ensaiada com p-nitrofenilacetato foram avaliadas no soro, fígado, cérebro, músculo e coração dos pacus. Houve redução de aproximadamente 35% da atividade de CarbE no soro dos pacus submetidos a 24 horas de hipoxia. Uma queda de 85% na atividade de CarbE do soro foi observada nos animais que sofreram hipoxia e subsequente exposição a 0,02 mg de metilparation por litro. Com metilparation a 0,01 mg/L a diminuição observada foi de 48,2%. No músculo dos pacus expostos a 0,02 mg/L, as atividades de AChE e BChE cairam pela metade quando os mesmos foram submetidos à hipoxia quando comparados a animais que permaneceram em normoxia. Nos diversos tecidos dos pacus expostos a 0,01 mg/L de metilparation não observamos diferenças significativas nas atividades de AChE, BChE ou CarbE. Concluímos que a duplicação da concentração de metilparation de 0,01 para 0,02 mg/L levou à atividade residual de CarbE do soro de 51,8% para 15%. A ausência de mudanças nas atividades das esterases dos tecidos de animais expostos a 0,01 mg/L entre os grupos hipoxia e normoxia deve ter ocorrido porque a concentração de organofosforado não foi suficiente para superar a primeira barreira de proteção das esterases séricas e atingir os tecidos. Mas, no experimento com 0,02 mg/L de metilparation, as inibições de AChE e de BChE no músculo dos animais em hipoxia podem ser explicadas pela diminuição da atividade de CarbE do soro dos pacus.

John Otterbein Snyder (1867–1943) was an early student of David Starr Jordan at Stanford University and subsequently rose to become an assistant professor there. During his 34 years with the university he taught a wide variety of courses in various branches of zoology and advised numerous students. He eventually mentored 8 M.A. and 4 Ph.D. students to completion at Stanford. He also assisted in the collection of tens of thousands of fish specimens from the western Pacific, central Pacific, and the West Coast of North America, part of the time while stationed as “Naturalist” aboard the U.S. Fish Commission’s Steamer Albatross (1902–06). Although his early publications dealt mainly with fish groups and descriptions (often as a junior author with Jordan), after 1910 he became more autonomous and eventually rose to become one of the Pacific salmon, Oncorhynchus spp., experts on the West Coast. Throughout his career, he was especially esteemed by colleagues as “a stimulating teacher,” “an excellent biologist,” and “a fine man.

Spencer Fullerton Baird (Fig. 1), a noted systematic zoologist and builder of scientific institutions in 19th century America, persuaded the U.S. Congress to establish the United States Commission of Fish and Fisheries1 in March 1871. At that time, Baird was Assistant Secretary of the Smithsonian Institution. Following the death of Joseph Henry in 1878, he became head of the institution, a position he held until his own demise in 1887. In addition to his many duties as a Smithsonian official, including his prominent role in developing the Smithsonian’s Federally funded National Museum as the repository for governmental scientific collections, Baird directed the Fish Commission from 1871 until 1887. The Fish Commission’s original mission was to determine the reasons and remedies for the apparent decline of American fisheries off southern New England as well as other parts of the United States. Charles Henry Gilbert (1859-1928) was a pioneering ichthyologist who made major contributions to the study of fishes of the American West. As chairman of the Department ofZoology at Leland Stanford Junior University in Palo Alto, Calif., during 1891-1925, Gilbert was extremely devoted to his work and showed little patience with those ofa different mindset. While serving as Naturalist-in-Charge of the U.S. Fish Commission Steamer Albatross during her exploratory expedition to the Hawaiian Islands in 1902, Gilbert engaged in an acrimonious feud with the ship's captain, Chauncey Thomas, Jr. (1850-1919), U.S.N., over what Gilbert perceived to be an inadequate effort by the captain. This essay focuses on the conflict between two strong figures, each operatingf rom different world views, and each vying for authority. Despite the difficulties these two men faced, the voyage of the Albatross in 1902 must be considered a success, as reflected by the extensive biological samples collected, the many new species of animals discovered, and the resulting publication of important scientific papers.

Fishery science pioneers often faced challenges in their field work that are mostly unknown to modern biologists. Some of the travails faced by ichthyologist and, later, fishery biologist Charles Henry Gilbert (1859-1928) during his service as Naturalist-in-Charge of the North Pacific cruise ofthe U.S. Bureau of Fisheries Steamer Albatross in 1906, are described here, as are accomplishments of the cruise. The vessel left San Francisco, Calif., on 3 May 1906, just after the great San Francisco earthquake, for scientific exploration of waters of the Aleutian islands, Bering Sea, Kamchatka, Sakhalin, and Japan, returning to San Francisco in December. Because the expedition occurred just after the war between Japan and Russia of 1904-05 floating derelict mines in Japanese waters were often a menace. Major storms caused havoc in the region, and the captain of the Albatross, Lieutenant Commander LeRoy Mason Garrett (1857-1906), U.S.N., was lost at sea, apparently thrown from the vessel during a sudden storm on the return leg of the cruise. Despite such obstacles, Gilbert and the Albatross successfully completed their assigned chores. They occupied 339 dredging and 48 hydrographic stations, and discovered over 180 new species of fishes and many new species of invertebrates. The expedition's extensive biological collections spawned over 30 descriptive publications, some of which remain today as standards of knowledge.

The dusky rockfish (Sebastes ciliatus) of the North Pacific Ocean has been considered a single variable species with light and dark forms distributed in deep and shallow water, respectively. These forms have been subjected to two distinct fisheries separately managed by federal and state agencies: the light deep form is captured in the offshore trawl fishery; the dark shallow form, in the nearshore jig fishery. The forms have been commonly recognized as the light dusky and dark dusky rockfishes. From morphological evidence correlated with color differences in some 400 specimens, we recognize two species corresponding with these color forms. Sebastes ciliatus (Tilesius) is the dark shallow-water species found in depths of 5−160 m in the western Aleutian Islands and eastern Bering Sea to British Columbia. The name Sebastes variabilis (Pallas) is resurrected from the synonymy of S. ciliatus to apply to the deeper water species known from depths of 12−675 m and ranging from Hokkaido, Japan, through the Aleutian Islands and eastern Bering Sea, to Oregon. Sebastes ciliatus is uniformly dark blue to black, gradually lightening on the ventrum, with a jet black peritoneum, a smaller symphyseal knob, and fewer lateral-line pores compared to S. variabilis. Sebastes variabilis is more variable in body color, ranging from light yellow to a more usual tan or greenish brown to a nearly uniform dark dorsum, but it invariably has a distinct red to white ventrum. Synonymies, diagnoses, descriptions, and geographic distributions are provided for each species.

Do hospitals experience safety tipping points as utilization increases, and if so, what are the implications for hospital operations management? We argue that safety tipping points occur when managerial escalation policies are exhausted and workload variability buffers are depleted. Front-line clinical staff is forced to ration resources and, at the same time, becomes more error prone as a result of elevated stress hormone levels. We confirm the existence of safety tipping points for in-hospital mortality using the discharge records of 82,280 patients across six high-mortality-risk conditions from 256 clinical departments of 83 German hospitals. Focusing on survival during the first seven days following admission, we estimate a mortality tipping point at an occupancy level of 92.5%. Among the 17% of patients in our sample who experienced occupancy above the tipping point during the first seven days of their hospital stay, high occupancy accounted for one in seven deaths. The existence of a safety tipping point has important implications for hospital management. First, flexible capacity expansion is more cost-effective for safety improvement than rigid capacity, because it will only be used when occupancy reaches the tipping point. In the context of our sample, flexible staffing saves more than 40% of the cost of a fully staffed capacity expansion, while achieving the same reduction in mortality. Second, reducing the variability of demand by pooling capacity in hospital clusters can greatly increase safety in a hospital system, because it reduces the likelihood that a patient will experience occupancy levels beyond the tipping point. Pooling the capacity of nearby hospitals in our sample reduces the number of deaths due to high occupancy by 34%.

According to the method of path integral quantization for the canonical constrained system in Becchi-Rouet-Stora-Tyutin scheme, the supersymmetric electromagnetic interaction system was quantized. Both the Hamiltonian of the supersymmetric electromagnetic interaction system in phase space and the quantization procedure were simplified. The BRST generator was constructed, and the BRST transformations of supersymmetric fields were gotten; the effective action was calculated, and the generating functional for the Green function was achieved; also, the gauge generator was constructed, and the gauge transformation of the system was obtained. Finally, the Ward-Takahashi identities based on the canonical Noether theorem were calculated, and two relations between proper vertices and propagators were obtained.
