Objective: To evaluate, through the application of different dentin bonding systems, the influence of wetness on shear bond strength in enamel. Methods: This study evaluated three etch-and-rinse adhesive systems (Scotchbond MP [used with and without primer]; Singlebond; and Prime&Bond 2.1) and two self-etching adhesive systems (Clearfil SE Bond; and Xeno IV). Flat bovine enamel surfaces were either air-dried for 30 seconds or blotted with absorbent paper after acid-etching for the conventional bonding agents or before the application of self-etching bonding agents. The resin composite EsthetX was bonded to flat surfaces that had been treated with one of the adhesives, following the manufacturer`s instructions. After being stored in water at 37 degrees C for one week, bonded specimens were broken in shear. Data were evaluated with two-way analysis of variance (ANOVA) and Student-Newman-Keuls tests (alpha=0.05). For comparing each condition individually, regardless of the adhesive or wetness condition, a one-way ANOVA and a Student-Newman-Keuls test (alpha=0.05) were applied. Results: The two-way ANOVA showed significant differences among adhesive systems. An interaction effect was also observed (p < 0.05), but wetness did not influence shear bond strength (p=0.98). The one-way ANOVA showed that the all-in-one adhesive was the only material influenced by the presence of water at the enamel`s surface. Conclusion: The all-in-one adhesive behaved differently depending on whether the enamel surface was dry or wet.

Ethylene is a plant hormone that is of fundamental importance to in vitro morphogenesis, but in many species, it has not been thoroughly studied. Its relationship with polyamines has been studied mainly because the two classes of hormones share a common biosynthetic precursor, S-adenosylmethionine (SAM). In order to clarify whether competition between polyamines and ethylene influences in vitro morphogenetic responses of Passiflora cincinnata Mast., a climacteric species, different compounds were used that act on ethylene biosynthesis and action, or as ethylene scavengers. Treatment with the ethylene inhibitor, aminoethoxyvinylglycine (AVG) caused a greater regeneration frequency in P. cincinnata, whereas treatment with the ethylene precursor, 1-aminocyclopropane-1-carboxylic-acid (ACC) lessened regeneration frequencies. The data suggested that levels of polyamines and ethylene are not correlated with morphogenic responses in P. cincinnata. It was ascertained that neither the absolute ethylene and polyamine levels, nor competition between the compounds, correlated to the obtained morphogenic responses. However, sensitivity to, and signaling by, ethylene appears to play an important role in differentiation. This study reinforces previous reports regarding the requirement of critical concentrations and temporal regulation of ethylene levels for morphogenic responses. Temporal regulation also appeared to be a key factor in competition between the two biosynthetic pathways, without having any effects on morphogenesis. Further studies investigating the silencing or overexpression of genes related to ethylene perception, under the influence of polyamines in cell differentiation are extremely important for the complete understanding of this process.

Arthropods display different mechanisms to protect themselves against infections, among which antimicrobial peptides (AMPs) play an important role, acting directly against invader pathogens. We have detected several factors with inhibitory activity against Candida albicans and Micrococcus luteus on the surface and in homogenate of eggs of the tick Rhipicephalus (Boophilus) microplus. One of the anti-M. luteus factors of the egg homogenate was isolated to homogeneity. Analysis by electrospray mass spectrometry (ESI-MS) revealed that it corresponds to microplusin, an AMP previously isolated from the cell-free hemolymph of X (B.) microplus. Reverse transcription (RT) quantitative polymerase chain reactions (qPCR) showed that the levels of microplusin mRNA gradually increase along ovary development, reaching an impressive highest value three days after the adult females have dropped from the calf and start oviposition. Interestingly, the level of microplusin mRNA is very low in recently laid eggs. An enhance of microplusin gene expression in eggs is observed only nine days after the onset of oviposition, achieving the highest level just before the larva hatching, when the level of expression decreases once again. Fluorescence microscopy analysis using an anti-microplusin serum revealed that microplusin is present among yolk granules of oocytes as well as in the connecting tube of ovaries. These results, together to our previous data. suggest that microplusin may be involved not only in protection of adult female hemocele, but also in protection of the female reproductive tract and embryos, what points this AMP as a considerable target for development of new methods to control R. The anomalous alternating magnetoresistivity in HgTe quantum wells with thicknesses of 5.8 and 8.3 nm, i.e., near the transition from the direct band spectrum to an inverted spectrum, has been revealed and analyzed. It has been shown that the revealed anomalous alternating magnetoresistivity in wells with an inverted spectrum is well described by the theory developed by S.V. Iordanskii et al. [JETP Lett. 60, 206 (1994)] and W. Knap et al. [Phys. Rev. B 53, 3912 (1996)]. A detailed comparison of the experimental data with the theory indicates the presence of only the cubic term in the spin splitting of the electronic spectrum. The applicability conditions of the mentioned theory are not satisfied in a well with a direct gap and, for this reason, such a certain conclusion is impossible. The results indicate the existence of a strong spin-orbit interaction in symmetric HgTe quantum wells near the topological transition.

Serum amyloid A (SAA) levels are elevated highly in acute phase response and elevated slightly and persistently in chronic diseases such as rheumatoid arthritis and diabetes. Given that fibroblasts exert profound effects on progression of inflammatory chronic diseases, the aim of this study was to investigate the response of fibroblasts to SAA. A dose-dependent increase in O(2)(-) levels was observed by treatment of fibroblasts with SAA (r = 0.99 and P <= 0.001). In addition, the expression of p47-phox was up-regulated by SAA (P < 0.001) and diphenyliodonium (DPI), a nicotinamide adenine dinucleotide phosphate (NADPH) oxidase inhibitor, reduced the release of O(2)(-) by 50%. Also, SAA raised fibroblast proliferation (P < 0.001) and this effect was completely abolished by the addition of anti-oxidants (P < 0.001). These findings support the notion that, in chronic inflammatory sites, SAA activated fibroblast proliferation and ROS production.

Schistosomiasis affects more than 200 million people worldwide; another 600 million are at risk of infection. The schistosomulum stage is believed to be the target of protective immunity in the attenuated cercaria vaccine model. In an attempt to identify genes up-regulated in the schistosomulum stage in relation to cercaria, we explored the Schistosoma mansoni transcriptome by looking at the relative frequency of reads in EST libraries from both stages. The 400 genes potentially up-regulated in schistosomula were analyzed as to their Gene Ontology categorization, and we have focused on those encoding-predicted proteins with no similarity to proteins of other organisms, assuming they could be parasite-specific proteins important for survival in the host. Up-regulation in schistosomulum relative to cercaria was validated with real-time reverse transcription polymerase chain reaction (RT-PCR) for five out of nine selected genes (56%). We tested their protective potential in mice through immunization with DNA vaccines followed by a parasite challenge. Worm burden reductions of 16-17% were observed for one of them, indicating its protective potential. Our results demonstrate the value and caveats of using stage-associated frequency of ESTs as an indication of differential expression coupled to DNA vaccine screening in the identification of novel proteins to be further investigated as potential vaccine candidates.

A pesquisa objetivou compreender a inserção de organizações de catadores em programas empresariais de logística reversa para reciclagem de embalagens pós-consumo. O estudo multi-casos englobou programas empresariais de quatro setores de embalagens: vidro, papelão, embalagem longa vida e plástico PET. As unidades de análise foram as relações empresa/cooperativas de catadores. Utilizou-se como requisito para seleção dos programas o avanço das cooperativas na cadeia reversa através da sua atuação como fornecedoras de primeira camada, ou seja, sem “intermediários”. Foram realizadas entrevistas semiestruturadas com quatro empresas e quatro cooperativas de catadores, sendo uma delas analisada como explanação rival. De maneira geral observou-se que as características dos programas são função das motivações para estruturação da logística reversa, seja pela responsabilidade socioambiental, recuperação de materiais, competitividade da embalagem, imagem corporativa ou compras de materiais secundários. Além da obtenção de escala e regularização, o avanço das cooperativas de catadores nessas cadeias de suprimentos foi viabilizado através da aprendizagem prática quanto à correta identificação e seleção de materiais recicláveis e desenvolvimento de processos de pré-processamento para garantia de qualidade conforme, exigências das empresas. Vale destacar também a infraestrutura obtida através de convênios com a Prefeitura Municipal de São Paulo e de financiamentos pelo Banco Nacional do Desenvolvimento - BNDES. Em dois casos analisados constatou-se a atuação de empresas de bens de consumo na gestão de fluxos de informações, permitindo uma gestão eficiente das operações de logística reversa. Em todos os programas estudados, apesar da comercialização direta, faz-se necessária a realização de etapas de beneficiamento pelas empresas com emprego de tecnologias apropriadas. Para as cooperativas de catadores a comercialização direta tem proporcionado melhores preços dos materiais recicláveis e perspectivas de vendas em longo prazo e, consequentemente, perspectivas de vida e reconhecimento profissional dos catadores.

Esta tese analisa três lacunas que ainda não foram discutidas na literatura. As lacunas dizem respeito aos efeitos da internacionalização na estrutura de capital e de propriedade de empresas multinacionais. Para tanto, três ensaios sobre os efeitos da internacionalização foram elaborados. O primeiro analisou os efeitos dos modos de entrada sobre o endividamento; o segundo os efeitos da internacionalização nas formas de endividamento e o terceiro os efeitos da internacionalização na estrutura de propriedade. A partir de dados de empresas multinacionais e domésticas de capital aberto latino-americanas de 2007 a 2011, foram montados painéis de dados com variáveis de teste e controle para cada ensaio. Os resultados do primeiro ensaio apontam que os modos de entrada são relevantes para determinar o nível de endividamento das multinacionais e complementam os resultados de trabalhos baseados na hipótese upstream-downstream. Evidenciou-se também que empresas com modos de entrada patrimoniais tendem a ser mais endividadas no longo prazo e total que empresas com modos de entrada não-patrimoniais, reforçando explicações dadas pela teoria do static trade-off; e menos endividadas no curto prazo, reforçando as explicações das teorias da agência e pecking order. Os resultados do segundo ensaio apontam que o grau de internacionalização: (i) aumenta o nível de endividamento da quase totalidade das dívidas providas por agentes financeiros (bancos), conforme a hipótese upstream-downstream e diminuem as dívidas providas por agentes não-financeiros (trade-credit), conforme teoria da restrição financeira; (ii) não produz efeitos sobre a maturidade das dívidas (iii) diferente do que esperava, não aumenta endividamento via banco nacional de desenvolvimento (empréstimos incentivados), (iv) foi relevante para alterar a composição do endividamento das multinacionais se comparada às empresas domésticas e (v) a forma de entrada na internacionalização não afeta a composição de endividamento. No terceiro ensaio o principal resultado encontrado foi que as empresas mais internacionalizadas e com modo de entrada patrimonial têm menores níveis de concentração de propriedade. Outro importante resultado do terceiro ensaio foi de que há uma simultaneidade ainda não explorada em estudos anteriores na determinação do nível de internacionalização e da concentração de propriedade. Ambos os resultados do terceiro ensaio estão amparados pela Visão Baseada em Recursos contrapondo a visão tradicional da teoria da agência.

A indústria de transformação é considerada de suma importância para o desenvolvimento do país tanto em termos econômicos como sociais, visto que é responsável por encadeamentos produtivos e efeitos multiplicadores no emprego e na renda do país. Entre os anos de 2000 e 2011 ainda que haja indícios de desindustrialização da economia brasileira, devido à queda da participação do valor adicionado da indústria de transformação no Produto Interno Bruto e à deterioração da indústria de transformação frente ao comércio exterior, no que concerne ao emprego, em termos absolutos e relativos, não há suspeitas de que esteja em curso no Brasil um processo de desindustrialização. Paralelamente, ressalta-se que embora se façam presentes sintomas da chamada “doença holandesa” no Brasil, ou seja, balança comercial superavitária em commodities acompanhada de déficits da indústria de transformação e de taxa de câmbio valorizada; o Brasil não parece sofrer da “doença”

Background: Over the last century the incidence of cutaneous melanoma has increased worldwide, a trend that has also been observed in Brazil. The identified risk factors for melanoma include the pattern of sun exposure, family history, and certain phenotypic features. In addition, the incidence of melanoma might be influenced by ethnicity. Like many countries, Brazil has high immigration rates and consequently a heterogenous population. However, Brazil is unique among such countries in that the ethnic heterogeneity of its population is primarily attributable to admixture. This study aimed to evaluate the contribution of European ethnicity to the risk of cutaneous melanoma in Brazil. Methodology/Principal Findings: We carried out a hospital-based case-control study in the metropolitan area of Sao Paulo, Brazil. We evaluated 424 hospitalized patients (202 melanoma patients and 222 control patients) regarding phenotypic features, sun exposure, and number of grandparents born in Europe. Through multivariate logistic regression analysis, we found the following variables to be independently associated with melanoma: grandparents born in Europe-Spain (OR = 3.01, 95% CI: 1.03-8.77), Italy (OR = 3.47, 95% CI: 1.41-8.57), a Germanic/Slavic country (OR = 3.06, 95% CI: 1.05-8.93), or >= 2 European countries (OR = 2.82, 95% CI: 1.06-7.47); eye color-light brown (OR = 1.99, 95% CI: 1.14-3.84) and green/blue (OR = 4.62; 95% CI 2.22-9.58); pigmented lesion removal (OR = 3.78; 95% CI: 2.21-6.49); no lifetime sunscreen use (OR = 3.08; 95% CI: 1.03-9.22); and lifetime severe sunburn (OR = 1.81; 95% CI: 1.03-3.19). Conclusions: Our results indicate that European ancestry is a risk factor for cutaneous melanoma. Such risk appears to be related not only to skin type, eye color, and tanning capacity but also to others specific characteristics of European populations introduced in the New World by European immigrants.

Correlations between GABA(A) receptor (GABA(A)-R) activity and molecular organization of synaptosomal membranes (SM) were studied along the protocol for cholesterol (Cho) extraction with beta-cyclodextrin (beta-CD). The mere pre-incubation (PI) at 37A degrees C accompanying the beta-CD treatment was an underlying source of perturbations increasing [H-3]-FNZ maximal binding (70%) and K (d) (38%), plus a stiffening of SMs' hydrocarbon core region. The latter was inferred from an increased compressibility modulus (K) of SM-derived Langmuir films, a blue-shifted DPH fluorescence emission spectrum and the hysteresis in DPH fluorescence anisotropy (A (DPH)) in SMs submitted to a heating-cooling cycle (4-37-4A degrees C) with A (DPH,heating) < A (DPH,cooling). Compared with PI samples, the beta-CD treatment reduced B (max) by 5% which correlated with a 45%-decrement in the relative Cho content of SM, a decrease in K and in the order parameter in the EPR spectrum of a lipid spin probe labeled at C5 (5-SASL), and significantly increased A (TMA-DPH). PI, but not beta-CD treatment, could affect the binding affinity. EPR spectra of 5-SASL complexes with beta-CD-, SM-partitioned, and free in solution showed that, contrary to what is usually assumed, beta-CD is not completely eliminated from the system through centrifugation washings. It was concluded that beta-CD treatment involves effects of at least three different types of events affecting membrane organization: (a) effect of PI on membrane annealing, (b) effect of residual beta-CD on SM organization, and (c) Cho depletion. Consequently, molecular stiffness increases within the membrane core and decreases near the polar head groups, leading to a net increase in GABA(A)-R density, relative to untreated samples.
