Ce mémoire a pour but de comparer deux romans policiers contemporains et ayant eu un grand succès, l’un écrit par Henning Mankell dont la traduction française a paru sous le titre Les Morts de la Saint-Jean et le second est Pars vite et reviens tard de Fred Vargas. L’objectif de la candidate est d’essayer de trouver les éléments réalistes dans le décor, les personnages et l’enquête. Pour ce qui est du premier paramètre, la candidate montre que Mankell n’essaie jamais de cacher la vérité derrière une surface idéale ; il en va de même pour Vargas qui accentue les points de vue divergents des Français sur la réalité de leur pays. En ce qui a trait aux personnages, on distingue dans le portrait qu’en fait Mankell un combat compliqué et non manichéen, proche de la réalité psychologique; idem pour ceux de Vargas, mais le réalisme est quelque peu atténué par un recours au mystère. Enfin, l’enquête menée par Wallander aboutit un peu par hasard grâce à son intuition alors qu’Adamsberg résout l’énigme grâce à sa faculté de voir, de s’imaginer et d’aller au-delà de la capacité des autres. La conclusion générale du mémoire est que ces deux œuvres sont tout à fait réalistes, mais à un degré un peu différent. Le roman de Mankell l’est sans doute davantage dans la mesure où le recours au mystère est écarté et que l’enquête est menée de façon logique alors que le roman de Vargas, tout en suivant les règles de la logique, a tendance à faire appel au merveilleux. En tout état de cause, il s’agit de deux œuvres réalistes comportant un côté romantique plus ou moins bien exprimé.

We generalize a procedure proposed by Mancera and Hunt [P.F.A. Mancera, R. Hunt, Some experiments with high order compact methods using a computer algebra software-Part 1, Appl. Math. Comput., in press, doi: 10.1016/j.amc.2005.05.015] for obtaining a compact fourth-order method to the steady 2D Navier-Stokes equations in the streamfunction formulation-vorticity using the computer algebra system Maple, which includes conformal mappings and non-uniform grids. To analyse the procedure we have solved a constricted stepped channel problem, where a fine grid is placed near the re-entrant corner by transformation of the independent variables. In this work, a non-linear Boundary Element Method (BEM) formulation with damage model is extended for numerical simulation of structural masonry walls in 2D stress analysis. The formulation is reoriented to analyse structural masonry, the component materials of which, clay bricks and mortar, are considered as damaged materials. Also considered are the internal variables and cell discretization of the domain. A damage model is used to represent the material behaviour and the domain discretization is also proposed and discussed. The paper presents the numerical parameters of the damage model for the material properties of the masonry components, clay bricks and mortar. Some examples are shown to validate the formulation.

Insulin resistance is a common risk factor in chronic kidney disease patients contributing to the high cardiovascular burden, even in the absence of diabetes. Glucose-based peritoneal dialysis (PD) solutions are thought to intensify insulin resistance due to the continuous glucose absorption from the peritoneal cavity. The aim of our study was to analyse the effect of the substitution of glucose for icodextrin on insulin resistance in non-diabetic PD patients in a multicentric randomized clinical trial. This was a multicenter, open-label study with balanced randomization (1:1) and two parallel-groups. Inclusion criteria were non-diabetic adult patients on automated peritoneal dialysis (APD) for at least 3 months on therapy prior to randomization. Patients assigned to the intervention group were treated with 2L of icodextrin 7.5%, and the control group with glucose 2.5% during the long dwell and, at night in the cycler, with a prescription of standard glucose-based PD solution only in both groups. The primary end-point was the change in insulin resistance measured by homeostatic model assessment (HOMA) index at 90 days. Sixty patients were included in the intervention (n = 33) or the control (n = 27) groups. There was no difference between groups at baseline. After adjustment for pre-intervention HOMA index levels, the group treated with icodextrin had the lower post-intervention levels at 90 days in both intention to treat [1.49 (95% CI: 1.23-1.74) versus 1.89 (95% CI: 1.62-2.17)], (F = 4.643, P = 0.03, partial η(2) = 0.078); and the treated analysis [1.47 (95% CI: 1.01-1.84) versus 2.18 (95% CI: 1.81-2.55)], (F = 7.488, P = 0.01, partial η(2) = 0.195). The substitution of glucose for icodextrin for the long dwell improved insulin resistance measured by HOMA index in non-diabetic APD patients.

The aim of this study was to analyse the characteristics of the asymmetries in the dominant and non-dominant limbs when kicking stationary and rolling balls. Ten experienced Brazilian amateur futsal players participated in this study. Each participant performed kicks under two conditions (stationary ball vs. rolling ball) with the dominant and non-dominant limbs (five kicks per condition per limb). We analysed the kicking accuracy, ball and foot velocities, angular joint displacement and velocity. The asymmetry between the dominant and non-dominant limbs was analysed by symmetry index and two-way repeated measures ANOVA. The results did not reveal any interaction between the condition and limb for ball velocity, foot velocity and accuracy. However, kicking with the dominant limb in both kicks showed higher ball velocity (stationary ball: dominant - 24.27 ± 2.21 m · s(-1) and non-dominant - 21.62 ± 2.26 m · s(-1); rolling ball: dominant - 23.88 ± 2.71 m · s(-1) and non-dominant - 21.42 ± 2.25 m · s(-1)), foot velocity (stationary ball: dominant - 17.61 ± 1.87 m · s(-1) and non-dominant - 15.58 ± 2.69 m · s(-1); rolling ball: dominant - 17.25 ± 2.26 m · s(-1) and non-dominant - 14.77 ± 2.35 m · s(-1)) and accuracy (stationary ball: dominant - 1.17 ± 0.84 m and non-dominant - 1.56 ± 1.30 m; rolling ball: dominant - 1.31 ± 0.91 m and non-dominant - 1.97 ± 1.44 m). In addition, the angular joint adjustments were dependent on the limb in both kicks (the kicks with non-dominant limb showed lower hip external rotation than the kicks with the dominant limb), indicating that the hip joint is important in kick performance. In conclusion, the kicks with the non-dominant limb showed different angular adjustments in comparison to kicks with the dominant limb. In addition, kicking a rolling ball with the non-dominant limb showed higher asymmetry for accuracy, indicating that complex kicks are more asymmetric.

Recent studies of the large cheilostome bryozoan genus Scrupocellaria have shown a greater degree of taxonomically informative morphological variation in zooids, opesia, and polymorphic structures than previously recognized. Only one subgenus has been named within the genus, Retiscrupocellaria d'Hondt, 1988, erected for Scrupocellaria jolloisii. In this work we further analyse S. jolloisii and its related species, resurrecting an earlier genus name, Licornia van Beneden, 1850 for Licornia jolloisii, and nine relatives, L. annectens, L. cervicornis, L. cyclostoma, L. diadema, L. ferox, L. gaspari, L. longispinosa, L. macropora, and L. prolata. Licornia jolloisii was originally described from the Red Sea, and most species of the genus occur in the Indo-Pacific region. The species, however, has now been found in the Western Atlantic, in the Florida Keys, US, and in Bahia de Todos Santos, Brazil.

Human reasoning is a fascinating and complex cognitive process that can be applied in different research areas such as philosophy, psychology, laws and financial. Unfortunately, developing supporting software (to those different areas) able to cope such as complex reasoning it’s difficult and requires a suitable logic abstract formalism. In this thesis we aim to develop a program, that has the job to evaluate a theory (a set of rules) w.r.t. a Goal, and provide some results such as “The Goal is derivable from the KB5 (of the theory)”. In order to achieve this goal we need to analyse different logics and choose the one that best meets our needs. In logic, usually, we try to determine if a given conclusion is logically implied by a set of assumptions T (theory). However, when we deal with programming logic we need an efficient algorithm in order to find such implications. In this work we use a logic rather similar to human logic. Indeed, human reasoning requires an extension of the first order logic able to reach a conclusion depending on not definitely true6 premises belonging to a incomplete set of knowledge. Thus, we implemented a defeasible logic7 framework able to manipulate defeasible rules. Defeasible logic is a non-monotonic logic designed for efficient defeasible reasoning by Nute (see Chapter 2). Those kind of applications are useful in laws area especially if they offer an implementation of an argumentation framework that provides a formal modelling of game. Roughly speaking, let the theory is the set of laws, a keyclaim is the conclusion that one of the party wants to prove (and the other one wants to defeat) and adding dynamic assertion of rules, namely, facts putted forward by the parties, then, we can play an argumentative challenge between two players and decide if the conclusion is provable or not depending on the different strategies performed by the players. Implementing a game model requires one more meta-interpreter able to evaluate the defeasible logic framework; indeed, according to Göedel theorem (see on page 127), we cannot evaluate the meaning of a language using the tools provided by the language itself, but we need a meta-language able to manipulate the object language8. Thus, rather than a simple meta-interpreter, we propose a Meta-level containing different Meta-evaluators. The former has been explained above, the second one is needed to perform the game model, and the last one will be used to change game execution and tree derivation strategies.

The thesis studies the economic and financial conditions of Italian households, by using microeconomic data of the Survey on Household Income and Wealth (SHIW) over the period 1998-2006. It develops along two lines of enquiry. First it studies the determinants of households holdings of assets and liabilities and estimates their correlation degree. After a review of the literature, it estimates two non-linear multivariate models on the interactions between assets and liabilities with repeated cross-sections. Second, it analyses households financial difficulties. It defines a quantitative measure of financial distress and tests, by means of non-linear dynamic probit models, whether the probability of experiencing financial difficulties is persistent over time. Chapter 1 provides a critical review of the theoretical and empirical literature on the estimation of assets and liabilities holdings, on their interactions and on households net wealth. The review stresses the fact that a large part of the literature explain households debt holdings as a function, among others, of net wealth, an assumption that runs into possible endogeneity problems. Chapter 2 defines two non-linear multivariate models to study the interactions between assets and liabilities held by Italian households. Estimation refers to a pooling of cross-sections of SHIW. The first model is a bivariate tobit that estimates factors affecting assets and liabilities and their degree of correlation with results coherent with theoretical expectations. To tackle the presence of non normality and heteroskedasticity in the error term, generating non consistent tobit estimators, semi-parametric estimates are provided that confirm the results of the tobit model. The second model is a quadrivariate probit on three different assets (safe, risky and real) and total liabilities; the results show the expected patterns of interdependence suggested by theoretical considerations. Chapter 3 reviews the methodologies for estimating non-linear dynamic panel data models, drawing attention to the problems to be dealt with to obtain consistent estimators. Specific attention is given to the initial condition problem raised by the inclusion of the lagged dependent variable in the set of explanatory variables. The advantage of using dynamic panel data models lies in the fact that they allow to simultaneously account for true state dependence, via the lagged variable, and unobserved heterogeneity via individual effects specification. Chapter 4 applies the models reviewed in Chapter 3 to analyse financial difficulties of Italian households, by using information on net wealth as provided in the panel component of the SHIW. The aim is to test whether households persistently experience financial difficulties over time. A thorough discussion is provided of the alternative approaches proposed by the literature (subjective/qualitative indicators versus quantitative indexes) to identify households in financial distress. Households in financial difficulties are identified as those holding amounts of net wealth lower than the value corresponding to the first quartile of net wealth distribution. Estimation is conducted via four different methods: the pooled probit model, the random effects probit model with exogenous initial conditions, the Heckman model and the recently developed Wooldridge model. Results obtained from all estimators accept the null hypothesis of true state dependence and show that, according with the literature, less sophisticated models, namely the pooled and exogenous models, over-estimate such persistence.

Das Humane Cytomegalovirus (HCMV) ist ein Erreger von großer klinischer Relevanz. Die HCMV-Infektion, die insbesondere bei immunsupprimierten Patienten mit hoher Morbidität und Mortalität assoziiert ist, wird vorwiegend durch CD8+-zytotoxische T-Lymphozyten (CTL) kontrolliert. Das Tegumentprotein pp65 und das immediate early 1-Protein (IE1) waren als die dominanten CTL-Antigene bekannt. Ziel dieser Arbeit war es, die zur Immundominanz des pp65 führenden molekularen Mechanismen aufzuklären und die Grundlagen für die Analyse der IE1-spezifischen Immunantwort zu erarbeiten. Durch Peptidimmunisierung HLA-A2-transgener Mäuse wurden hochaffine pp65-spezifische CTL-Klone generiert. Für die Generierung ähnlicher CTL-Klone gegen IE1 konnte erstmals ein konserviertes HLA-A2-bindendes Peptid identifiziert werden. Mit Hilfe der pp65-spezifischen CTL-Klone konnte gezeigt werden, dass das durch Viruspartikel in die Zelle eingebrachte pp65 die Erkennung infizierter Zellen durch CD8+-CTL vermittelt. Durch den Nachweis der außergewöhnlichen Stabilität von pp65 in der Zelle gelang es, eine hohe metabolische Umsatzrate als eine Ursache von Immundominanz auszuschließen. Dagegen hob die Blockierung des CRM1-vermittelten nukleären Exportweges durch Zugabe von Hemmstoffen oder Zutransfektion kompetitiver Inhibitoren die Erkennung des pp65 nahezu auf. Hiermit wurde erstmalig eine Abhängigkeit der Präsentation eines immundominanten nukleären Proteins vom nukleozytoplasmatischen Transport nachgewiesen. Die Erkenntnisse dieser Arbeit stellen die Grundlage für die detaillierte Analyse der Zusammenhänge zwischen nukleärem Export und Antigenpräsentation dar.

Die Aufklärung von Biosynthesewegen erfolgt häufig mit Hilfe von Fütterungsexperimenten mit radioaktiven oder stabilen Isotopen markierten Präkusoren oder auf der Basis der Enzymreinigung mit anschließender molekularbiologischer Charakterisierung. Die erstgenannte Methode verlangt die Isolierung der Produkte. Jedoch besteht bei Aufarbeitung und Extraktion immer die Gefahr, daß sich der Metabolit teilweise oder vollständig chemisch verändert. Ein weiterer Nachteil der genannten Methoden ist, daß diese generell mühsam und zeitaufwendig sind. Mit Hilfe der in vivo NMR-Spektroskopie können diese Nachteile umgangen werden. In der vorliegenden Arbeit wurden Biotransformationen und Biosynthesesequenzen des Ajmalin-Biosyntheseweges mit Hilfe der in vivo NMR-Spektroskopie in Pflanzenzellkulturen von Rauvolfia serpentina und Rauvolfia serpentina x Rhazya stricta anhand der natürlichen 13C-Häufigkeit untersucht. Dafür wurden ein 700 MHz, 800 MHz und ein 500 MHz CryoProbe Spektrometer eingesetzt, um die Biotransformationen von Isatin-3-oxim und Isatin sowie die Metabolisierungen der Alkaloide Vellosimin, Vinorin, Vomilenin, Ajmalin, Nß-Methyl-dihydrochano-ajmalin und Perakin mit der 1H-13C invers korrelierten NMR-Spektroskopie zu verfolgen.

Alpha- und Beta-Dystroglycan, die zentralen Komponenten eines multimeren Dystrophin-assoziierten Proteinkomplexes wurden bislang im Wesentlichen in der Skelettmuskulatur charakterisiert. Dort stellt der DAG eine molekulare Verbindung zwischen dem Aktin-Zytoskelett der Muskelfaser und einer Basalmembran her, die die einzelne Muskelfaser umhüllt. Dystroglycan vermittelt auf diese Weise die mechanische Festigkeit der Muskelfasern während der Kontraktion. Außerdem dient der DAG als Gerüst für die Anlagerung von Proteinen. Mutationen in den strukturgebenden oder signaltransduzierenden Proteinen des DAG verursachen Muskeldystrophie. Besonders schwere Muskeldystrophien werden durch Mutationen hervorgerufen, die eine veränderte Glykosylierung von Dystroglycan und damit eine verminderte Bindung von alpha-Dystroglycan an Matrixproteine verursachen. Dies führt zu einer Beeinträchtigung der Basalmembranbiosynthese sowie sich daraus ergebende Störungen in der Migration, Schichtung und Differenzierung von Nervenzellen im ZNS. Welche Rolle Dystroglycan im sich entwickelnden ZNS spielt, sollte in dieser Arbeit an der Hühnerretina untersucht werden. Durch Anwendung der in ovo Elektroporation wurden zwei modifizierte Dystroglycankonstrukte in Neuroepithelzellen transfiziert. Die Überexpression eines verkürtzten Dystroglycanproteins, verursachte eine Abrundung der Neuroepithelzellen. Dies führte zur Hyperproliferation der Zellen deren Folge die Bildung von Verdickungen in der Retina war sowie eine verstärkte Bildung postmitotischer Neurone. Die Elektroporation eines nicht-spaltbaren Dystroglycans, führte im Gegensatz dazu zu einer Abnahme der Anzahl proliferierender und differenzierender Nervenzellen. Als Konsequenz veränderte sich die Orientierung der Axone von retinalen Ganglienzellen. Nach der Überexpression des verkürzten Dystroglycans verloren die Axone ihre zentripetale Orientierung auf den optischen Nerv, während die Elektroporation von Wt-Dystroglycan und nicht-spaltbarem Dystroglycan nur einen gelegentlichen Richtungswechsel der Axone verursachte. Die Daten zeigen, dass Dystroglycan einen entscheidenden Einfluss auf die Proliferation, Differenzierung und Polarität der Neuroepithelzellen ausübt. Dies geschieht vermutlich durch die Vermittlung der Adhäsion des Endfußes von Neuroepithelzellen an die Basalmembran. Die Veränderungen nach der Überexpression der modifizierten Dystroglycankonstrukte liefern möglicherweise eine Erklärung für den ZNS-Phänotyp der sich bei verschiedenen Formen von Muskeldystrophie zeigt.
