Epicuticular wax production was evaluated along the length of expanding leek (Allium porrum L.) leaves to gain insight into the regulation of wax production. Leaf segments from the bottom to the top were analyzed for (a) wax composition and load; (b) microsomal fatty acid elongase, plastidial fatty acid synthase, and acyl-acyl carrier protein (ACP) thioesterase activities; and (c) tissue and cellular morphological changes. The level of total wax, which was low at the bottom, increased 23-fold along the length of the leaf, whereas accumulation of the hentriacontan-16-one increased more than 1000-fold. The onset of wax accumulation was not linked to cell elongation but, rather, occurred several centimeters above the leaf base. Peak microsomal fatty acid elongation activity preceded the onset of wax accumulation, and the maximum fatty acid synthase activity was coincident with the onset. The C16:0- and C18:0-ACP-hydrolyzing activities changed relatively little along the leaf, whereas C18:1-ACP-hydrolyzing activity increased slightly prior to the peak elongase activity. Electron micrographic analyses revealed that wax crystal formation was asynchronous among cells in the initial stages of wax deposition, and morphological changes in the cuticle and cell wall preceded the appearance of wax crystals. These studies demonstrated that wax production and microsomal fatty acid elongation activities were induced within a defined and identifiable region of the expanding leek leaf and provide the foundation for future molecular studies.

In both animal and plant acyl elongation systems, it has been proposed that fatty acids are first activated to acyl-coenzyme A (CoA) before their elongation, and that the ATP dependence of fatty acid elongation is evidence of acyl-CoA synthetase involvement. However, because CoA is not supplied in standard fatty acid elongation assays, it is not clear if CoA-dependent acyl-CoA synthetase activity can provide levels of acyl-CoAs necessary to support typical rates of fatty acid elongation. Therefore, we examined the role of acyl-CoA synthetase in providing the primer for acyl elongation in leek (Allium porrum L.) epidermal microsomes and Brassica napus L. cv Reston oil bodies. As presented here, fatty acid elongation was independent of CoA and proceeded at maximum rates with CoA-free preparations of malonyl-CoA. We also showed that stearic acid ([1-14C]18:0)-CoA was synthesized from [1-14C]18:0 in the presence of CoA-free malonyl-CoA or acetyl-CoA, and that [1-14C]18:0-CoA synthesis under these conditions was ATP dependent. Furthermore, the appearance of [1-14C]18:0 in the acyl-CoA fraction was simultaneous with its appearance in phosphatidylcholine. These data, together with the s of a previous study (A. Hlousek-Radojcic, H. Imai, J.G. Jaworski [1995] Plant J 8: 803–809) showing that exogenous [14C]acyl-CoAs are diluted by a relatively large endogenous pool before they are elongated, strongly indicated that acyl-CoA synthetase did not play a direct role in fatty acid elongation, and that phosphatidylcholine or another glycerolipid was a more likely source of elongation primers than acyl-CoAs.

BACKGROUND Gastrointestinal and respiratory diseases in calves and piglets lead to significant economic losses in livestock husbandry. A high morbidity has been reported for diarrhea (calves ≤ 35 %; piglets ≤ 50 %) and for respiratory diseases (calves ≤ 80 %; piglets ≤ 40 %). Despite a highly diverse etiology and pathophysiology of these diseases, treatment with antimicrobials is often the first-line therapy. Multi-antimicrobial resistance in pathogens results in international accordance to strengthen the research in novel treatment options. Medicinal plants bear a potential as alternative or additional treatment. Based on the versatile effects of their plant specific multi-component-compositions, medicinal plants can potentially act as 'multi-target drugs'. Regarding the plurality of medicinal plants, the aim of this systematic review was to identify potential medicinal plant species for prevention and treatment of gastrointestinal and respiratory diseases and for modulation of the immune system and inflammation in calves and piglets. RESULTS Based on nine initial sources including standard textbooks and European ethnoveterinary studies, a total of 223 medicinal plant species related to the treatment of gastrointestinal and respiratory diseases was identified. A defined search strategy was established using the PRISMA statement to evaluate 30 medicinal plant species starting from 20'000 peer-reviewed articles published in the last 20 years (1994-2014). This strategy led to 418 references (257 in vitro, 84 in vivo and 77 clinical trials, thereof 48 clinical trials in veterinary medicine) to evaluate effects of medicinal plants and their efficacy in detail. The findings indicate that the most promising candidates for gastrointestinal diseases are Allium sativum L., Mentha x piperita L. and Salvia officinalis L.; for diseases of the respiratory tract Echinacea purpurea (L.) MOENCH, Thymus vulgaris L. and Althea officinalis L. were found most promising, and Echinacea purpurea (L.) MOENCH, Camellia sinensis (L.) KUNTZE, Glycyrrhiza glabra L. and Origanum vulgare L. were identified as best candidates for modulation of the immune system and inflammation. CONCLUSIONS Several medicinal plants bear a potential for novel treatment strategies for young livestock. There is a need for further research focused on gastrointestinal and respiratory diseases in calves and piglets, and the findings of this review provide a basis on plant selection for future studies.

In vitro selection is one of the most effective and efficient techniques for plant improvement. This is due to its ability to isolate plants with the desired character(s), either by applying a selection agent on the culture media to drive the selection of somaclones with the required character(s), or by establishing particular conditions that change in the genomes of somaclones toward the required character. The objective of this study was to identify a suitable protocol for in vitro selection of Allium white rot disease ( Sclerotium cepivorum ) tolerance in commercial Egyptian onion varieties, namely Giza 20, Giza 6 and Beheri Red. Oxalic acid (OA), the phytotoxin produced by Sclerotium cepivorum, was used as the selective agent. Seeds of the three Egyptian varieties were germinated on four concentrations (0.0, 0.02, 0.2, 2 and 20 mM) of Oxalic acid. Among the tested cultivars, Beheri Red had the highest germination frequency (52%) at all concentrations tested, followed by Giza 20 (42.6%), and Giza 6 at (32%). Cotyledon explants from the varieties were cultured on toxic MSBDK medium, supplemented with 0, 3, 6 and 12 mM OA. The survival of calli on MSBDK free toxic medium was 70.7% for all tested cultivars; however, MSBDK-stressed medium, with 3 mM OA reduced the viable calli to 42.1%. The highest OA concentration (12 mM) completely inhibited calli induction from cotyledons explants. A medium supplement with 3 mM OA retarded 80% of calli growth. Among 156 tested calli of Beheri Red, only 23 calli (14.7%) survived on toxic medium for 45 days. Similarly, there was 15.6% survival for Giza 20 calli, while 40.1% of the Giza 6 calli survived. Plantlets were regenerated from surviving calli and transplanted onto ex vitro, and formed bulb after acclimatisation.

Este artículo informa sobre la valoración de la fitotóxicidad del Material Particulado Sedimentable (MPS) en la zona urbana del cantón Cuenca. El MPS se recolectó en tres sitios de la zona urbana, mediante el método descrito en el Texto Unificado de Legislación Ambiental Secundaria del Ministerio del Ambiente (TULASMA). Por el material particulado se realizó la determinación de la concentración de los componentes utilizando el análisis gravimétrico, la de los metales pesados por espectroscopía de absorción atómica, y la de aniones por método adaptado de referencia EPA 300.1. Posteriormente se evaluó su fitotóxicidad en cuatro especies, mediante el bioensayo de germinación y elongación de raíz e hipocótilo, en semillas de Lactuca sativa, Brassica oleracea itálica, Allium cepa y Raphanus sativus. En el bioensayo las semillas se expusieron a diferentes diluciones de la suspensión acuosa de MPS. Los resultados muestran que el efecto de los MPS dependió de la concentración y de la especie analizada, evidenciando diferentes grados de fitotóxicidad.

Background: Agricultural products and by products provide the primary materials for a variety of technological applications in diverse industrial sectors. Agro-industrial wastes, such as cotton and curaua fibers, are used to prepare nanofibers for use in thermoplastic films, where they are combined with polymeric matrices, and in biomedical applications such as tissue engineering, amongst other applications. The development of products containing nanofibers offers a promising alternative for the use of agricultural products, adding value to the chains of production. However, the emergence of new nanotechnological products demands that their risks to human health and the environment be evaluated. This has resulted in the creation of the new area of nanotoxicology, which addresses the toxicological aspects of these materials.Purpose and methods: Contributing to these developments, the present work involved a genotoxicological study of different nanofibers, employing chromosomal aberration and comet assays, as well as cytogenetic and molecular analyses, to obtain preliminary information concerning nanofiber safety. The methodology consisted of exposure of Allium cepa roots, and animal cell cultures (lymphocytes and fibroblasts), to different types of nanofibers. Negative controls, without nanofibers present in the medium, were used for comparison.Results: The nanofibers induced different responses according to the cell type used. In plant cells, the most genotoxic nanofibers were those derived from green, white, and brown cotton, and curaua, while genotoxicity in animal cells was observed using nanofibers from brown cotton and curaua. An important finding was that ruby cotton nanofibers did not cause any significant DNA breaks in the cell types employed.Conclusion: This work demonstrates the feasibility of determining the genotoxic potential of nanofibers derived from plant cellulose to obtain information vital both for the future usage of these materials in agribusiness and for an understanding of their environmental impacts.

Esta pesquisa estudou a ação inseticida de extrato alcoólico de alho (Allium sativum L.) e pimenta-do-reino (Piper nigrum L.), já utilizados no controle de pragas em sistemas agroflorestais no nordeste paraense, contra larvas de 4º instar de Tenebrio molitor (Col., Tenebrionidae) em laboratório. Os experimentos foram desenvolvidos no Laboratório de Entomologia da Embrapa Amazônia Oriental, em Belém, Pará. Os extratos alcoólicos de A. sativum e P. nigrum foram obtidos de 50g dos produtos vegetais, colocados em repouso em 200 mL de álcool etílico hidratado (92,8º INPM) por 5 dias. As formulações à base de 50 mL de extrato alcoólico para cada extrato isoladamente e 25 mL de ambos na mistura, com 25mL de sabão líquido cada, foram aplicadas nas concentrações de 1% e 10%, além de um tratamento testemunha com água destilada, em um delineamento experimental inteiramente casualizado, com 20 repetições. Os insetos foram expostos ao tratamento por aplicação tópica com auxílio de micropipetas com pressão e precisão de 0,2 mL e mantidos em placas de Petri com discos de papel de filtro (9,0 cm Ø), providas de pedaços de chuchu para alimentação e armazenadas em câmaras climatizadas, tipo B.O.D, à temperatura de 27 ± 2oC, umidade relativa de 70 ± 5% e fotofase de 12 horas. A mortalidade foi avaliada por 5 dias após a exposição dos mesmos aos tratamentos. Extratos alcoólicos de A. sativum e P. nigrum apresentaram ação inseticida contra larvas de T. molitor, com taxa de mortalidade variando de 60% (1º dia) a 80% (5º dia) na maior dosagem de resposta biológica (10%) do experimento

Bryla Keywords: Allium cepa, crop growth, nitrogen uptake, soil nitrate Abstract: The effect of nitrogen application on growth, nitrogen (N) uptake, yield, and quality of intermediate-day onion (Allium cepa 'Guimar') was evaluated in the field in southern Portugal. Plants were fertilized with 30 kg ha-1 N at transplanting, 10 kg ha-1 N at 29 days after transplanting (DAT) during early leaf growth, and with 0, 20, 40 and 60 kg ha-1 N at 51 DAT at the initiation of bulbing. The root system of plants in each treatment were concentrated in the top 0.1 m of soil and limited to 0.3 m depth but neither root length density nor rooting depth were affected by N application during later stages of bulb development. Leaf and bulb dry matter, on the other hand, increased linearly with N rate during bulb growth (85 DAT) and at harvest (114 DAT), respectively. Soil nitrate-N (NO3-N) at 0-0.3 m depth likewise increased linearly with N rate during bulb growth but declined from 15-30 mg kg-1 at bulbing to >10 mg kg-1 in each treatment by harvest. A substantial amount of N in the plants, which ranged from 302-525 mg, was taken up from the soil. Application of 60 kg ha-1 N resulted in luxury consumption. Yield (fresh bulb weight) increased from 0.19 kg plant-1 with no N at bulbing to as much as 0.28 kg plant-1 with 60 kg ha-1 N. Bulbs harvested from plants fertilized 40-60 kg ha-1 N averaged 8.2-8.5 cm in diameter, while those from plants with no N at bulbing averaged only 7.2 cm in diameter. Application of N fertilizer is thus recommended at bulbing to increase N uptake, yield, and bulb size of intermediate-day onions, particularly in dry Mediterranean climates where many onions are produced. Other components of quality, including neck diameter, bulb water content, total soluble solids, and juice pH, were not affect by N applied at bulbing.

Os elementos mais absorvidos em termos de porcentagem na matéria seca da cebola são o potássio e nitrogênio. Como o objetivo de avaliar a produtividade e classificação de bulbos de cultivares de cebola em função de doses de nitrogênio foi conduzido um experimento no período de junho a setembro de 2013, em Petrolina-PE. O delineamento experimental utilizado foi de blocos ao acaso no esquema fatorial 5x2, compreendendo cinco doses de nitrogênio (0; 60; 120, 180 e 240 kg ha-1) e duas cultivares (Brisa IPA-12 e Vale Ouro IPA-11), com três repetições. A cultivar Brisa IPA-12 (80,7 t ha-1) foi superior a cultivar Vale Ouro IPA-11 (77,0 t ha-1). A dose de 183,2 kg ha-1 de N promoveu a maior produtividade comercial (94,7 t ha-1). A cultivar Brisa IPA-12 apresentou maior produção de refugos (2,4 t ha-1) comparativamente a cultivar Vale Ouro IPA-11 (1,1 t ha-1). Verificou-se com o aumento das doses uma redução gradativa da produção de bulbos considerados não comerciais, sendo a menor produção de refugos estimada na dose de 167,7 kg ha-1 de N. Maior massa fresca e bulbos de diâmetro superior foram obtidos com o incremento das doses de nitrogênio

Este trabalho teve como objetivo avaliar o efeito da temperatura na infecção de Colletotrichum gloeosporioides em cebola. Mudas de cebola, da variedade Alfa São Francisco, foram inoculadas numa suspensão de esporos, na concentração de 105 esporos/mL até o ponto de escorrimento. Após a inoculação, as mudas foram cobertas com saco plástico preto umedecido, vedado próximo à base dos vasos, por 24 horas, para simular o período de molhamento do campo, e mantidas em estufas sob as temperaturas de 15 oC, 20 oC, 25 oC, 30 oC, 35 oC e 40 oC. Foi determinada a severidade da doença considerando-se a porcentagem da área foliar com sintomas. As variáveis significativas no teste F da análise de variância foram submetidas à análise de regressão. A elevação da temperatura propiciou aumento na severidade da antracnose da cebola.

Beta Cristal e na germinação de conídios de Colletotrichum gloeosporioides f. sp. cepae.
