This study evaluated levels of digestible lysine and organic zinc for male Ross strain broilers from 1 to 11 days of age. It was used 1,050 chicks distributed in randomized block design, in 5 × 2 factorial scheme, with seven repetitions of 15 birds per experimental unit. The dietary concentrations of digestible lysine were 0.90; 1.00; 1.10; 1.20; and 1.40% combined with 43 and 253 ppm zinc chelate. The diets contained 2,965 ± 18 kcal/kg of apparent metabolizable energy (AME) and 21.48 ± 0.18% of CP. It was determined chemical composition, protein, lipid, mineral and water depositions on carcass and empty body. There was no interaction among the factors digestible lysine and organic zinc. Effect of zinc concentration increase was observed on greatest deposition of body fat, indicating that there is interference in lipid metabolism of the birds in the studied phase. The most pronounced effects resulted from the dietary inclusion of lysine. There was a linear effect on reconstituted body weight as a response to the increase of dietary lysine, which suggests equal or superior requirement to the greatest studied level. However, the deposition of water and protein on the carcass had a quadratic increase, characterizing higher muscle mass accumulation up to the levels 1.25 and 1.27 of this amino acid in the diet. Considering the studied strain, broiler chickens from the first to the 11th days of age require 1.28 ± 0.01% of digestible lysine, according to the deposition of muscle mass.

This work was undertaken to provide further insight into the role of mammalian target of rapamycin complex 1 (mTORC1) in skeletal muscle regeneration, focusing on myofiber size recovery. Rats were treated or not with rapamycin, an mTORC1 inhibitor. Soleus muscles were then subjected to cryolesion and analyzed 1, 10, and 21 days later. A decrease in soleus myofiber cross-section area on post-cryolesion days 10 and 21 was accentuated by rapamycin, which was also effective in reducing protein synthesis in these freeze-injured muscles. The incidence of proliferating satellite cells during regeneration was unaltered by rapamycin, although immunolabeling for neonatal myosin heavy chain (MHC) was weaker in cryolesion+rapamycin muscles than in cryolesion-only muscles. In addition, the decline in tetanic contraction of freeze-injured muscles was accentuated by rapamycin. This study indicates that mTORC1 plays a key role in the recovery of muscle mass and the differentiation of regenerating myofibers, independently of necrosis and satellite cell proliferation mechanisms. Infection with GB virus C (GBV-C) or hepatitis G virus (HGV) is highly prevalent among HIV/AIDS patients. GBV-C/HGV viremia has not been associated with liver disease and seems to slow HIV disease progression. To study the GBV-C/HGV genotypes prevalence among HIV/AIDS patients and its association with HIV viral load (VL) and CD4+ lymphocyte counts. From February 2003 to February 2004, we analyzed 210 HIV-1-infected subjects who were on anti-retroviral therapy (ART). For 63 of them a PCR-nested to the non-coding 5` (5`NCR) region of the GBV-C/HGV was done, and for 49 a DNA direct sequencing was done. A phylogenetic analysis was performed by PHYLIP program. 63(30%) of the HIV-1-infected patients were co-infected with GBV-C/HGV. The phylogenetic analysis revealed the following genotypes (and respective relative frequencies): 1(10%), 2a (41%), 2b (43%), and 3 (6%). Co-infected patients presented lower HIV-1 VL and higher T CD4+ lymphocyte cells counts as compared with patients negative for GBV-C/HGV sequences (log = 4.52 vs. 4.71, p = 0.036), and T CD4+ lymphocyte counts (cells/mm(3) = 322.6 vs. 273.5, p = 0.081, respectively). T CD4+ cells counts equal to, or higher than, 200/mm(3) were significantly more common among co-infected patients than among HIV-infected-only patients (p = 0.042). The lowest T CD4+ cells counts were associated with genotype 1 and the highest with genotype 2b (p = 0.05). The GBV-C/HGV infection prevalence was 30% among HIV-1-infected subjects, and was associated with lower VL and higher CD4+ lymphocyte counts. GBV-C/HGV genotype 2b may be associated with better immunological response. Published by Elsevier B.V.

Background and purpose: Control of food intake is a complex behaviour which involves many interconnected brain structures. The present work assessed if the noradrenergic system in the lateral septum (LS) was involved in the feeding behaviour of rats. Experimental approach: In the first protocol, the food intake of rats was measured. Then non-food-deprived animals received either 100 nL of 21 nmol of noradrenaline or vehicle unilaterally in the LS 10 min after local 10 nmol of WB4101, an alpha(1)-adrenoceptor antagonist, or vehicle. In the second protocol, different doses of WB4101 (1, 10 or 20 nmol in 100 nL) were microinjected bilaterally into the LS of rats, deprived of food for 18 h and food intake was compared to that of satiated animals. Key results: One-sided microinjection of noradrenaline into the LS of normal-fed rats evoked food intake, compared with vehicle-injected control animals, which was significantly reduced by alpha(1)-adrenoceptor antagonism. In a further investigation, food intake was significantly higher in food-deprived animals, compared to satiated controls. Pretreatment of the LS with WB4101 reduced food intake in only food-deprived animals in a dose-related manner, suggesting that the LS noradrenergic system was involved in the control of food intake. Conclusion and implications: Activation by local microinjection of noradrenaline of alpha(1)-adrenoceptors in the LS evoked food intake behaviour in rats. In addition, blockade of the LS alpha(1)-adrenoceptors inhibited food intake in food-deprived animals, suggesting that the LS noradrenergic system modulated food intake behaviour and satiation.

Delta 9-THC is a component of Cannabis sativa that increases food intake in animals and humans, an effect prevented by selective CB1 receptor antagonists. Cannabidiol (CBD) is another constituent of this plant that promotes several opposite neuropharmacological effects compared to Delta 9-THC. CBD mechanisms of action are still not clear, but under specific experimental conditions it can antagonize the effects of cannabinoid agonists, block the reuptake of anandamide and act as an agonist of 5-HT1A receptors. Since both the cannabinoid and serotoninergic systems have been implicated in food intake control, the aim of the present work was to investigate the effects caused by CBD on hyperphagia induced by agonists of CB1 or 5-HT1A receptors. Fed or fasted Wistar rats received intraperitoneal (i.p.) injections of CBD (1, 10 and 20 mg/kg) and food intake was measured 30 min later for 1 h. Moreover, additional fed or fasted groups received, after pretreatment with CBD (20 mg/kg) or vehicle, i.p. administration of vehicle, a CBI receptor agonist WIN55,212-2 (2 mg/kg) or a 5-HT1A receptor agonist 8-OH-DPAT (1 mg/kg) and were submitted to the food intake test for 1 h. CBD by itself did not change food intake in fed or fasted rats. However, it prevented the hyperphagic effects induced by WIN55,212-2 or 8-OH-DPAT. These results show that CBD can interfere with food intake changes induced by a CB1 or 5-HT1A receptor agonist, suggesting that its role as a possible food intake regulator should be further investigate. Introduction. Endothelin-1 (ET-1), a potent vasoconstrictor peptide, acts mainly through the Gprotein-coupled ET(A) receptor (ET(A)R). Increased vascular ET-1 production and constrictor sensitivity have been observed in various cardiovascular diseases, including hypertension, as well as erectile dysfunction. The internal pudendal artery (IPA) supplies blood to the vagina and clitoris. Inadequate blood flow through the IPA may lead to insufficient vaginal engorgement and clitoral tumescence. Aim. Characterize the effects of ET-1 on the IPA and clitoral artery (CA). Methods. IPA and CA from female Sprague Dawley rats (225-250 g) were mounted in myograph chambers. Arterial segments were submitted to increasing concentrations of ET-1 (10-10-10-6 M). Segments were incubated with the ET(A)R antagonist, atrasentan (10-8 M) or the Rho-kinase inhibitor, Y-27632 (10-6 M) 30 minutes prior to agonist exposure. All E(max) values are expressed as % KCl-induced maximal contraction. ET(A)R, RhoA, and Rho-kinase expression from IPA was evaluated by Western blot. mRNA of preproET-1, ET(A)R, ET(B)R, RhoA, and Rho-kinase were measured by real time PCR. Main Outcome Measures. ET-1 constrictor sensitivity in IPA and CA, protein expression and messenger RNA levels of ET-1-mediated constriction components. Results. ET-1 concentration-dependently contracted IPA (% Contraction and pD2, respectively: 156 +/- 18, 8.2 +/- 0.1) and CA (163 +/- 12, 8.8 +/- 0.08), while ET(A)R antagonism reduced ET-1-mediated contraction (IPA: 104 +/- 23, 6.4 +/- 0.2; CA: 112 +/- 17, 6.6 +/- 0.08). Pretreatment with Y-27632 significantly shifted ET-1 pD2 in IPA (108 +/- 24, 7.9 +/- 0.1) and CA (147 +/- 58 and 8.0 +/- 0.25). Protein expression of ET(A)R, ET(B)R, RhoA, and Rho-kinase were detected in IPA. IPA and CA contained preproET-1, ET(A)R, ET(B)R, RhoA, and Rho-kinase message. Conclusion. We observed that the IPA and CA are sensitive to ET-1, signaling through the ET(A)R and Rho-kinase pathway. These data indicate that ET-1 may play a role in vaginal and clitoral blood flow and may be important in pathologies where ET-1 levels are elevated. Allahdadi KJ, Hannan JL, Tostes RC, and Webb RC. Cerebrospinal fluid amyloid-beta 1-42 (Aβ1-42) and phosphorylated Tau at position 181 (pTau181) are biomarkers of Alzheimer's disease (AD). We performed an analysis and meta-analysis of genome-wide association study data on Aβ1-42 and pTau181 in AD dementia patients followed by independent replication. An association was found between Aβ1-42 level and a single-nucleotide polymorphism in SUCLG2 (rs62256378) (P = 2.5×10(-12)). An interaction between APOE genotype and rs62256378 was detected (P = 9.5 × 10(-5)), with the strongest effect being observed in APOE-ε4 noncarriers. Clinically, rs62256378 was associated with rate of cognitive decline in AD dementia patients (P = 3.1 × 10(-3)). Functional microglia experiments showed that SUCLG2 was involved in clearance of Aβ1-42.

A Phase 1 double-blind placebo-controlled study was performed to evaluate a vaccine against American tegumentary leishmaniasis in 61 healthy male volunteers. Side effects and the immune response to the vaccine were evaluated, with 1- and 2- dose schemes, with intervals of 7 or 21 days, each dose containing 1440 mg of protein N antigen of a single strain of Leishmania amazonensis (PH 8) diluted in merthiolated saline (1:10,000). Merthiolated saline and an inert substance were used as placebos. No significant clinical alterations were found following the respective injections in the vaccinated individuals as compared to the placebos, except for local pain, which was associated significantly with injection of the vaccine. The laboratory alterations we observed bore no association with the clinical findings and were unimportant. We observed no differences between the groups with regard to seroconversion or the Montenegro skin test. However, the group that received a single dose of the vaccine and the one that received two doses with a 21-day interval displayed cutaneous induration significantly larger than in the control group, with 100%, 100%, and 66% conversion in the skin test, respectively. We concluded that the vaccine does not present any major side effect that would contraindicate its use in healthy individuals.

A sociedade brasileira vivencia, além da desnutrição e fome, problemas relacionados à obesidade - a transição nutricional. Este trabalho é um estudo transversal com amostra de 1322 crianças menores de 10 anos, residentes no município de Ferros, Minas Gerais, e cadastradas no SISVAN. Observou-se que 20,7% das crianças apresentaram alguma alteração nutricional (10,1% risco nutricional, 3,8% desnutrição e 6,7% sobrepeso). Fatores de risco para o desenvolvimento de desnutrição foram pesquisados: baixo peso ao nascer, RP=3,57, IC 95% (1,96-6,52); baixa estatura RP=19,36, IC 95% (11,53-32,52); aleitamento materno ausente RP=2,23, IC 95% (1,19-4,18); renda familiar de até R$ 95, RP = 2,39, IC 95% (1,10-5,16). Destaca-se maior prevalência de sobrepeso em relação à desnutrição. Enquanto a população rural apresentou maior prevalência de desnutrição e risco nutricional, a urbana destacou-se pelo sobrepeso. Esses resultados são importantes para mostrar que intervenções nutricionais são necessárias e devem considerar os problemas específicos apontados.

BACKGROUND & AIMS: In treatment-naive patients mono-infected with genotype 1 chronic HCV, treatments with telaprevir/boceprevir (TVR/BOC)-based triple therapy are standard-of-care. However, more efficacious direct-acting antivirals (IFN-based new DAAs) are available and interferon-free (IFN-free) regimens are imminent (2015). METHODS: A mathematical model estimated quality-adjusted life years, cost and incremental cost-effectiveness ratios of (i) IFN-based new DAAs vs. TVR/BOC-based triple therapy; and (ii) IFN-based new DAAs initiation strategies, given that IFN-free regimens are imminent. The sustained virological response in F3-4/F0-2 was 71/89% with IFN-based new DAAs, 85/95% with IFN-free regimens, vs. 64/80% with TVR/BOC-based triple therapy. Serious adverse events leading to discontinuation were taken as: 0-0.6% with IFN-based new DAAs, 0% with IFN-free regimens, vs. 1-10% with TVR/BOC-based triple therapy. Costs were euro60,000 for 12weeks of IFN-based new DAAs and two times higher for IFN-free regimens. RESULTS: Treatment with IFN-based new DAAs when fibrosis stage ⩾F2 is cost-effective compared to TVR/BOC-based triple therapy (euro37,900/QALY gained), but not at F0-1 (euro103,500/QALY gained). Awaiting the IFN-free regimens is more effective, except in F4 patients, but not cost-effective compared to IFN-based new DAAs. If we decrease the cost of IFN-free regimens close to that of IFN-based new DAAs, then awaiting the IFN-free regimen becomes cost-effective. CONCLUSIONS: Treatment with IFN-based new DAAs at stage ⩾F2 is both effective and cost-effective compared to TVR/BOC triple therapy. Awaiting IFN-free regimens and then treating regardless of fibrosis is more efficacious, except in F4 patients; however, the cost-effectiveness of this strategy is highly dependent on its cost.

Foram comparadas as concentrações de Na, K e P em extratos de solos, obtidas por um método de extração convencional, no qual é utilizada uma razão solo:extrator (Mehlich-1) de 1:10, com aquelas encontradas utilizando uma razão solo:extrator (Mehlich-1) de 1:5. Também, foram comparados os resultados obtidos por técnicas de quantificação convencionais, nas quais Na e K são quantificados por fotometria de chama e P por espectrofotometria de absorção molecular, com aqueles encontrados por espectrometria de emissão óptica com plasma indutivamente acoplado (ICP OES). Foram analisadas 15 amostras de solo brasileiro. No estudo de repetibilidade aplicado a todos os resultados, os maiores CVs foram encontrados para P e Na, principalmente quando as concentrações dos analitos foram menores (< 9 mg dm-3 para P e < 10 mg dm-3 para Na). Esse fato foi devido provavelmente à heterogeneidade dos extratos, que continham partículas coloidais. Filtração ou centrifugação em vez de decantação dos extratos provavelmente resultaria em menores CVs. No estudo de reprodutibilidade, realizado para três amostras, foram obtidos resultados não reprodutíveis somente para K em uma amostra. Todos os resultados obtidos por ICP OES foram semelhantes aos obtidos por espectrofotometria de absorção molecular UV-Vis. e fotometria de chama, indicando que a primeira técnica foi adequada para determinação de Na, K e P nos extratos de solos tropicais obtidos com solução de Mehlich-1. Os resultados de Na, K e P obtidos com razão solo:extrator de 1:5 foram estatisticamente diferentes daqueles em que se utilizou razão solo:extrator de 1:10. A maioria dos resultados obtidos com razão solo:extrator de 1:5 foi menor que os obtidos com razão solo:extrator 1:10, sobretudo nas amostras com concentrações de analitos mais elevadas. Portanto, para estudos comparativos envolvendo macronutrientes, deve-se utilizar o método convencional com razão solo:extrator de 1:10.

Nos laboratórios brasileiros, a determinação de micronutrientes para estudos de fertilidade de solos é realizada por extração com solução de Mehlich-1 (M-1), utilizando-se uma razão solo: extrator de 1:5 m/v e posterior quantificação por espectrometria de absorção atômica (AAS). No entanto, muitos laboratórios também empregam razão solo:extrator M-1 de 1:10 m/v para determinar macronutrientes e, ou, quantificar por espectrometria de emissão óptica com plasma indutivamente acoplado (ICP OES), em substituição ao AAS. Como estudos comparativos entre as concentrações de micronutrientes obtidos por esses métodos alternativos são escassos, o objetivo deste trabalho foi investigar se essas diferentes condições experimentais gerariam diferentes resultados. Os resultados foram estatisticamente tratados e revelaram que as concentrações de Fe e Mn, utilizando-se ambas as razões de solo:extrator M-1 (1:5 e 1:10 m/v) e de Zn, na razão solo:extrator M-1 de 1:10 m/v, foram estatisticamente iguais, enquanto as concentrações de Cu em ambas as razões solo:extrator M-1 e de Zn na razão solo:extrator de 1:5 m/v foram diferentes, quando as duas técnicas de quantificação (ICP OES e AAS) foram comparadas. Também, todas as concentrações de Cu, Fe, Mn e Zn obtidas utilizando razão solo:extrator M-1 de 1:5 m/v foram diferentes daquelas encontradas com a razão de 1:10 m/v, independentemente da técnica de quantificação. A maioria dessas concentrações foi maior quando se usou a razão de 1:5 m/v e alterou a classe de interpretação dos teores. Portanto, não recomenda-se o uso da razão solo:extrator de 1:5 para as extrações dos micronutrientes Cu, Fe, Mn e Zn com solução M-1 de amostras de solo.

Na execução de uma análise química de solo são empregados diversos procedimentos que, mesmo seguindo-se o protocolo preconizado pelo método de análise utilizado, estão sujeitos a variações nos resultados analíticos, causadas por manipulação das amostras ou dos materiais utilizados. Objetivou-se neste estudo avaliar a influência do grau de moagem da amostra, do tipo de frasco e do volume vazio no frasco na execução dos métodos Mehlich-1 e Mehlich-3 para determinar o P no solo. Para tanto, foram conduzidos três experimentos. No primeiro experimento, as amostras de solo foram moídas e tamisadas em peneiras com aberturas de 2,000; 1,700; 0,850; 0,600; e 0,300 mm. No segundo, foram utilizados dois modelos de frasco (erlenmeyer e snap-cap), ambos com volume de 50 mL. Já no terceiro, foi alterado o volume vazio no frasco, mantendo-se a relação solo:solução extratora utilizando-se as quantidades dentro do frasco de 1:10; 1,5:15; 2,5:25; 3:30; e 4:40 cm³ cm-3. O grau de moagem das amostras não influenciou a capacidade de extração do Mehlich-1; entretanto, a capacidade extrativa do Mehlich-3 foi influenciada, principalmente em solos argilosos. Tanto para o Mehlich-1 quanto para o Mehlich-3, os teores de P extraído foram significativamente mais elevados com o uso de frasco tipo snap-cap em relação ao erlenmeyer. O volume vazio no frasco alterou os teores de P extraído para o Mehlich-1 e Mehlich-3 em 100 e 64 % das amostras, respectivamente. Deve-se padronizar a intensidade da moagem das amostras de solo para extração do P pela solução de Mehlich-3. Um modelo único de frasco deve ser adotado pelos laboratórios de rotina para análise do P, independentemente do método de extração, mantendo-se sempre constante no frasco o volume da amostra (cm³) para o volume de solução extratora (cm³).
