Foram estudadas 100 progênies de meio-irmãos de uma sub-população de milho (Zea mays L.) Composto Flint com o objetivo de avaliar a resistência de genótipos à lagarta-da-espiga Helicoverpa zea (Bod.). Foram obtidos os valores de danos médios da lagarta-da-espiga de 1,14 cm de comprimento na espiga determinado pela escala de Widstrom e coeficiente de variação experimental (CVE) de 23,4%. Dos parâmetros genéticos avaliados, a estimativa de herdabilidade (h²) foi de 6%, variância genética (VG) de 0,0015 cm² e variância fenotÃpica (VF) de 0,025 cm² para danos de H. zea. No entanto, o comprimento da ponta da bráctea e compactação da bráctea alcançaram resultados de herdabilidade de 75% e 72% respectivamente. Essa sub-população de milho apresenta variabilidade genética suficiente para utilização em programas de melhoramento, sendo que a resistência à lagarta-da-espiga pode ser obtida através da melhoria dos caracteres morfológicos diretamente relacionados à praga, como a compactação e comprimento da bráctea.

L(+) Lactic acid fermentation was studied by Lactobacillus rhamnosus sp. under the effects of pH control and a lowcost nutritional medium (sugarcane juice and corn steep liquor-CSL). Central composite design (CCD) was employed to determine maximum lactic acid production at optimum values for process variables and a satisfactory fit model was realized. Statistical analysis of the results showed that the linear and quadratic terms of two variables (sugarcane juice and pH) had significant effects. The interactions between the three variables were found to contribute to the response at a significant level. A second-order polynomial regression model estimated that the maximum lactic acid production of 86.36 g/L was obtained when the optimum values of sucrose, CSL and pH were 112.65 g/L, 29.88 g/L and 6.2, respectively. Verification of the optimization showed that L(+) lactic acid production was of 85.06 g/L. Under these conditions, Y P/S and Q P values of 0.85 g/g and 1.77 g/Lh, respectively, were obtained after 48 h fermentation, with a maximal productivity of 2.2 g/L h at 30 h of process. © 2010 de Lima CJB, et al.

The objective of this work was to evaluate the effect of processing two corn hybrids conserved, dry and humid grains, the dry matter (DM) and crude protein (CP) degradability in situ. The particle size was determined and difference was verified in MGD (Medium Geometric Diameter) of processed ingredients. Three sheep were used with rumen canulated, in a completely randomized design, using a factorial outline 2 x 2 x 3, being two corn hybrid, two conservation methods and three processing forms (whole, coarsely and finely ground), with five times of incubation (3, 6, 12, 24 and 48 hours). The fraction A in SDC (silage of dent corn) of DM was superior to GDC (grain of dent corn) in all of the particles size. The ensiling process increased the DM solubility, reducing the fraction B in comparison to dry grain. The values regarding the fractions DP and DE the 5% per hour of the protein, were larger for SDC and GDC, it presents a decreasing when the incubation time advances. The fermentation rate was superior for SDC and GDC. The ensiling process has positive effect in the decreasing of DM and CP in comparison to GDC.

The genetic control of flowering time has been addressed by many quantitative trait locus (QTL) studies. A survey of the results from 29 independent studies reporting information on 441 QTLs led to the production of a QTL consensus map, which enabled the identification of 59 chromosome regions distributed on all chromosomes and shown to be frequently involved in the genetic control of flowering time and related traits. One of the major QTLs for flowering time, the Vegetative to generative transition 1 (Vgt1) locus , corresponds to an upstream (70 kb) non-coding regulatory element of ZmRap2.7, a repressor of flowering. A transposon (MITE) insertion was identified as a major allelic difference within Vgt1. One of the hypotheses is that Vgt1 might function by modifying ZmRap2.7 chromatin through an epigenetic mechanism. Therefore, the methylation state at Vgt1 was investigated using an approach that combines digestion with McrBc, an endonuclease that acts upon methylated DNA, and quantitative PCR. The analyses were performed on genomic DNA from leaves of six different maize lines at four stages of development. The results showed a trend of reduction of methylation from the first to the last stage with the exception of a short genomic region flanking the MITE insertion, which showed a constant and very dense methylation throughout leaf development and for both alleles. Preliminary results from bisulfite sequencing of a small portion of Vgt1 revealed differential methylation of a single cytosine residue between the two alleles. ZmRap2.7 expression was assayed in the four developmental stages afore mentioned for the six genotypes, in order to establish a link between methylation at Vgt1 and ZmRap2.7 transcription. To assess the role of Vgt1 as a transcriptional enhancer, two reporter vectors for stable transformation of plants have been developed.
Isolation and functional characterization of a high affinity urea transporter from roots of Zea mays

Background: Despite its extensive use as a nitrogen fertilizer, the role of urea as a directly accessible nitrogen source for crop plants is still poorly understood. So far, the physiological and molecular aspects of urea acquisition have been investigated only in few plant species highlighting the importance of a high-affinity transport system. With respect to maize, a worldwide-cultivated crop requiring high amounts of nitrogen fertilizer, the mechanisms involved in the transport of urea have not yet been identified. The aim of the present work was to characterize the high-affinity urea transport system in maize roots and to identify the high affinity urea transporter. Results: Kinetic characterization of urea uptake (<300 mu M) demonstrated the presence in maize roots of a high-affinity and saturable transport system; this system is inducible by urea itself showing higher Vmax and Km upon induction. At molecular level, the ORF sequence coding for the urea transporter, ZmDUR3, was isolated and functionally characterized using different heterologous systems: a dur3 yeast mutant strain, tobacco protoplasts and a dur3 Arabidopsis mutant. The expression of the isolated sequence, ZmDUR3-ORF, in dur3 yeast mutant demonstrated the ability of the encoded protein to mediate urea uptake into cells. The subcellular targeting of DUR3/GFP fusion proteins in tobacco protoplasts gave results comparable to the localization of the orthologous transporters of Arabidopsis and rice, suggesting a partial localization at the plasma membrane. Moreover, the overexpression of ZmDUR3 in the atdur3-3 Arabidopsis mutant showed to complement the phenotype, since different ZmDUR3-overexpressing lines showed either comparable or enhanced 15N]-urea influx than wild-type plants. These data provide a clear evidence in planta for a role of ZmDUR3 in urea acquisition from an extra-radical solution. Conclusions: This work highlights the capability of maize plants to take up urea via an inducible and high-affinity transport system. ZmDUR3 is a high-affinity urea transporter mediating the uptake of this molecule into roots. Data may provide a key to better understand the mechanisms involved in urea acquisition and contribute to deepen the knowledge on the overall nitrogen-use efficiency in crop plants.

Auxin (IAA) is an important regulator of plant development and root differentiation. Although recent studies indicate that salicylic acid (SA) may also be important in this context by interfering with IAA signaling, comparatively little is known about its impact on the plant’s physiology, metabolism, and growth characteristics. Using carbon-11, a short-lived radioisotope (t 1/2 = 20.4 min) administered as 11CO2 to maize plants (B73), we measured changes in these functions using SA and IAA treatments. IAA application decreased total root biomass, though it increased lateral root growth at the expense of primary root elongation. IAA-mediated inhibition of root growth was correlated with decreased 11CO2 fixation, photosystem II (PSII) efficiency, and total leaf carbon export of 11C-photoassimilates and their allocation belowground. Furthermore, IAA application increased leaf starch content. On the other hand, SA application increased total root biomass, 11CO2 fixation, PSII efficiency, and leaf carbon export of 11C-photoassimilates, but it decreased leaf starch content. IAA and SA induction patterns were also examined after root-herbivore attack by Diabrotica virgifera to place possible hormone crosstalk into a realistic environmental context. We found that 4 days after infestation, IAA was induced in the midzone and root tip, whereas SA was induced only in the upper proximal zone of damaged roots. We conclude that antagonistic crosstalk exists between IAA and SA which can affect the development of maize plants, particularly through alteration of the root system’s architecture, and we propose that the integration of both signals may shape the plant’s response to environmental stress.

Com o objetivo de estudar a influência da distribuição vertical de semente na variabilidade espacial da implantação de uma cultura de milho em diferentes sistemas de preparação do solo (mobilização convencional, mobilização mÃnima e sementeira direta), realizaram-se nas campanhas de 2011 e 2012 dois ensaios de campo na região do Alentejo. A avaliação da distribuição vertical de semente fez-se pela determinação da profundidade de sementeira medindo o comprimento do mesocótilo em plântulas de milho. Em sementeira direta foram ainda testadas duas velocidades de trabalho. Os resultados demonstraram que o melhor desempenho de distribuição vertical da semente foi conseguido em sementeira direta com o aumento de velocidade de trabalho de 4 para 6 km h-1 (com um coeficiente de variação, CV, de 10,1%). Uma ANOVA de duplo fator demonstrou que a resistência mecânica do solo e a velocidade de trabalho determinaram diferenças significativas na profundidade de sementeira; a profundidade de sementeira causou diferenças significativas no tempo médio (TME) e percentagem de emergência (PE) da cultura. Os elevados CV observados sugerem a necessidade de uma melhor calibração dos dispositivos de controlo de profundidade dos semeadores por parte dos operadores em campo ou o aperfeiçoamento dos mesmos.

Nessa pesquisa foi avaliada a utilização do sal de tetrazólio para determinar a maturidade fisiológica das sementes de milho. As sementes utilizadas foram dos hÃbridos Pioneer 4285 e Dow 2B587, semeadas em 03/10/2014 e 05/12/2014 respectivamente, e colhidas a partir dos 40 dias após o florescimento (DAF), com intervalos de 4 dias até os 68 DAF. As sementes colhidas foram avaliadas quanto à viabilidade e ao vigor (testes de germinação, de emergência da plântula, de condutividade elétrica, de envelhecimento acelerado e determinações do comprimento da plântula). Os parâmetros utilizados para determinar o ponto de maturidade fisiológica das sementes foram a camada preta, a linha de leite, a massa de matéria seca, o teor de água e a avaliação dos tecidos da semente utilizando o sal de tetrazólio, utilizando o método descrito para avaliar a viabilidade, complementado pela avaliação da atividade das células da chalaza e da zona de transferência do endosperma para o embrião. Para as sementes de milho dos dois hÃbridos a germinação foi superior a 95% e não houve diferença entre as épocas de colheita, somente nas últimas colheitas das sementes do hÃbrido Dow 2B587 houve redução da germinação e do vigor. O ponto de maturidade fisiológica (PM) foi identificado aos 56 DAF para as sementes de milho do hÃbrido P4285 e aos 48 DAF para as do hÃbrido Dow 2B587 e correspondeu ao estádio 4 da linha de leite e ao máximo de acúmulo da matéria seca. O máximo de vigor foi detectado por meio do resultado do teste de envelhecimento acelerado oito dias antes do (PM) para os dois hÃbridos. A atividade das células do endosperma está relacionada com os demais indicadores do PM (linha de leite, camada preta, massa de matéria seca e teor de água). O transporte de fotoassimilados da planta mãe para a semente cessa no ponto de maturidade fisiológica da semente, desativando o transporte no qual atuam as células da chalaza e da região basal do endosperma. A utilização do sal de tetrazólio possibilita identificar a morte das células da região basal do endosperma, uma vez que a partir desse momento não há mais a reação dessas células com o sal de tetrazólio, indicando que não têm atividade celular. Dessa forma, é possÃvel caracterizar o ponto de maturidade fisiológica da semente de milho, por meio da atividade do sal de tetrazólio; essa caracterização é confirmada pela expressão das enzimas CAT e MDH.

El presente trabajo se hizo con el objeto de evaluar la disponibilidad de fósforo en un suelo bajo en dicho elemento (8 ppm), el cual está ubicado en un área de la Estación Experimental y de Prácticas de la Facultad de Ciencias Agronómicas. Como planta indicadora de la fertilidad natural del suelo se utilizó el maÃz hÃbrido H-3. El diseño experimental utilizado fue un Factorial en Bloques al Azar, con los factores de variación siguientes: Niveles de fósforo y 4 secuencias de aplicación (estas se reúnen en 2 para 1976 y 1977). Se emplearon 6 niveles de fósforo: 0, 20, 40, 60, 80 y 100 Kg. De P205/ha. La fuente de fósforo utilizada fue el Superfosfato Simple (20%P205) y se hizo una aplicación general de Sulfato de Amonio (21% N). En un nivel de 100 Kg. N / Ha. Las épocas de aplicación se describe de la siguiente manera: para cada nivel de fósforo se distribuyen 4 parcelas que reciben su tratamiento respectivo de L1, L2, L3, L4. En el primer año se aplican los 4 tratamientos. En el 2º. año solo reciben fertilizante las parcelas bajo el sistema L1 y L3 (efecto acumulativo) y quedan L2 y L4 sin aplicación (efecto residual). En el 3er año parcelas con sistema L2 y L3 son fertilizados (efecto acumulativo) y L1 y L4 no reciben fertilizante. En el 4to. Año sólo L3 es fertilizado (efecto acumulativo) y el resto queda sin fertilización. En el presente estudio sólo se describen resultados de dos años. Se efectuaron dos muestreos de suelo en cada año de investigación: Uno a la siembra y el otro después de la cosecha, observándose en el análisis un aumento en el nivel de fósforo de acuerdo a las dosis aplicadas. Los datos que se tomaron fueron: Alturas de plantas y rendimiento de maÃz. Las alturas mostraron una diferencia significativa a las aplicaciones de Fósforo, siendo testigo menos a los niveles del elemento fertilizante, pero no existió variabilidad significativa en entre los niveles. El efecto de los niveles en el rendimiento fue significativo, al igual que las alturas el testigo absorbió toda la variabilidad del fósforo. Sin embargo en los promedios de producción existió un ligero incremento en el rendimiento al aumentar el nivel de fósforo, pero dicho incremento no fue significativo. La secuencia de aplicación de fósforo, hizo manifiesta su significación en el efecto acumulativo y residual, siendo el primer efecto superior al segundo. La interacción Fósforo x época de aplicación no manifestó variabilidad significativa. Sólo el desglose en sus grados de libertad el testigo absorbió el efecto de la interacción. Los análisis del suelo correlacionado con los rendimientos obtenidos demostraron que los suelos bajos en fósforo responden a las aplicaciones de fertilizantes fosforados.

The health-relevant functionality of 10 thermally processed Peruvian Andean grains (five cereals, three pseudocereals, and two legumes) was evaluated for potential type 2 diabetes-relevant antihyperglycemia and antihypertension activity using in vitro enzyme assays. Inhibition of enzymes relevant for managing early stages of type 2 diabetes such as hyperglycemia-relevant alpha-glucosidase and alpha-amylase and hypertension-relevant angiotensin I-converting enzyme (ACE) were assayed along with the total phenolic content, phenolic profiles, and antioxidant activity based on the 1,1-diphenyl-2-picrylhydrazyl radical assay. Purple corn (Zea mays L.) (cereal) exhibited high free radical scavenging-linked antioxidant activity (77%) and had the highest total phenolic content (8 +/- 1 mg of gallic acid equivalents/g of sample weight) and alpha-glucosidase inhibitory activity (51% at 5 mg of sample weight). The major phenolic compound in this cereal was protocatechuic acid (287 +/- 15 mu g/g of sample weight). Pseudocereals such as Quinoa (Chenopodium quinoa Willd) and Kaniwa (Chenopodium pallidicaule Aellen) were rich in quercetin derivatives (1,131 +/- 56 and 943 +/- 35 mu g [expressed as quercetin aglycone]/g of sample weight, respectively) and had the highest antioxidant activity (86% and 75%, respectively). Andean legumes (Lupinus mutabilis cultivars SLP-1 and H-6) inhibited significantly the hypertension-relevant ACE (52% at 5 mg of sample weight). No alpha-amylase inhibitory activity was found in any of the evaluated Andean grains. This in vitro study indicates the potential of combination of Andean whole grain cereals, pseudocereals, and legumes to develop effective dietary strategies for managing type 2 diabetes and associated hypertension and provides the rationale for animal and clinical studies.

Natural mycoflora and co-occurrence of fumonisins (FB(1), FB(2)) and aflatoxins (AFB(1), AFB(2), AFG(1) and AFG(2)) in freshly harvested corn grain samples from four regions of Brazil were investigated. Fusarium verticillioides was predominant in all samples. Analysis of fumonisins showed that 98% of the samples were contaminated with FB(1) and 74.5% with FB(1) + FB(2), with toxin levels ranging from 0.015 to 9.67 mu g/g for FB(1) and from 0.015 to 3.16 mu g/g for FB(2). Twenty-one (10.5%) samples were contaminated with AFB(1), seven (3.5%) with AFB(2) and only one (0.5%) with AFG(1) and AFG(2). Co-contamination with aflatoxins and fumonisins was observed in 7% of the samples. The highest contamination of fumonisins and aflatoxins was observed in Nova Odessa (SP) and Varzea Grande (MT), respectively. The lowest contamination of these mycotoxins was found in Varzea Grande and Nova Odessa, respectively.
