Transcript patterns elicited in response to attack reveal, at the molecular level, how plants respond to aggressors. These patterns are fashioned both by inflicted physical damage as well as by biological components displayed or released by the attacker. Different types of attacking organisms might therefore be expected to elicit different transcription programs in the host. Using a large-scale DNA microarray, we characterized gene expression in damaged as well as in distal Arabidopsis thaliana leaves in response to the specialist insect, Pieris rapae. More than 100 insect-responsive genes potentially involved in defense were identified, including genes involved in pathogenesis, indole glucosinolate metabolism, detoxification and cell survival, and signal transduction. Of these 114 genes, 111 were induced in Pieris feeding, and only three were repressed. Expression patterns in distal leaves were markedly similar to those of local leaves. Analysis of wild-type and jasmonate mutant plants, coupled with jasmonate treatment, showed that between 67 and 84% of Pieris-regulated gene expression was controlled, totally or in part, by the jasmonate pathway. This was correlated with increased larval performance on the coronatine insensitive1 glabrous1 (coi1-1 gl1) mutant. Independent mutations in COI1 and GL1 led to a faster larval weight gain, but the gl1 mutation had relatively little effect on the expression of the insect-responsive genes examined. Finally, we compared transcript patterns in Arabidopis in response to larvae of the specialist P. rapae and to a generalist insect, Spodoptera littoralis. Surprisingly, given the complex nature of insect salivary components and reported differences between species, almost identical transcript profiles were observed. This study also provides a robustly characterized gene set for the further investigation of plant-insect interaction.

Recht spielt auf allen Stufen der Verwaltungsführung und in allen Bereichen der Verwaltung eine zentrale Rolle. Dieser Bedeutung von Recht für die Verwaltungsführung muss mit der Schaffung geeigneter Strukturen begegnet werden, die es der Verwaltung ermöglichen, rechtliche Aspekte und Vorgaben jederzeit und umfassend zu berücksichtigen. Rechtsdiensten kommt dabei eine Scharnierfunktion zu. Im Zentrum der vorliegenden Studie stehen die Fragen, die sich im Rahmen der Organisationsgestaltung von Rechtsdiensten in der Bundesverwaltung stellen. Sie geht dabei von der Leitmaxime aus, wonach Rechtsdiensten eine Querschnittfunktion innerhalb von Verwaltungseinheiten zukommt, die sich in deren Organisation widerspiegeln muss. Aus der Tatsache, dass das Recht für die Verwaltungseinheit in all ihren Tätigkeitsbereichen von Bedeutung ist, ergibt sich die Notwendigkeit, rechtliche Aspekte bereits bei der Festlegung der Struktur einer Verwaltungseinheit sowie bei der Gestaltung der Prozesse zu berücksichtigen und auch tatsächlich in die Abläufe der Verwaltungseinheit zu integrieren. Le droit intervient à tous les niveaux de la gestion publique et dans tous les domaines de l'administration. Cette importance du droit pour la gestion publique doit se traduire par la mise en place de structures appropriées, qui permettent à l'administration de respecter les aspects et les exigences juridiques à tout moment et de manière globale. Les services juridiques jouent un rôle charnière à cet égard. La présente étude se consacre avant tout aux questions qui se posent en matière d'organisation des services juridiques à l'administration fédérale. Le fil conducteur de ce travail repose sur le principe selon lequel les questions juridiques occupent une fonction transversale, qui doit se refléter dans l'organisation des services juridiques. Le fait que le droit joue un rôle dans tous les domaines d'activité d'une unité administrative implique la nécessité d'en tenir compte lors de l'établissement de la structure de celle-ci et de la conception des processus, en l'intégrant effectivement dans les différents processus de ladite unité administrative.

BACKGROUND: In a previous randomised EORTC study on adjuvant dibromodulcitol (DBD) and bichloroethylnitrosourea (BCNU) in adults with glioblastoma multiforme (GBM) and anaplastic astrocytoma (AA), a clinically significant trend towards a longer overall survival (OS) and a progression-free survival (PFS) was observed in the subgroup of AA. The aim of the present study was to test this adjuvant regimen in a larger number of AA patients. METHODS: Continuation of the previous phase III trial for newly diagnosed AA according to the local pathologist. Patients were randomised to either radiotherapy only or to radiotherapy in combination with BCNU on day 2 and weekly DBD, followed by adjuvant DBD and BCNU in cycles of six weeks for a maximum total treatment duration of one year. OS was the primary end-point. RESULTS: Patients (193 ) with newly diagnosed AA according to local pathological assessment were randomised to radiotherapy (RT) alone (n=99), or to RT plus DBD/BCNU (n=94); 12 patients were considered not eligible. At central pathology review, over half (53%) of the locally diagnosed AA cases could not be confirmed. On intent-to-treat analysis, no statistically significant differences in OS (p=0.111) and PFS (p=0.087) were observed, median OS after RT was only 23.9 months 95% confidence interval (CI), [18.4-34.0] after RT plus DBD/BCNU 27.3 months 95% CI [21.4-46.8]. CONCLUSION: No statistically significant improvement in survival was observed after BCNU/DBD adjuvant chemotherapy in AA patients. The trend towards improved survival is consistent with previous reports. Central pathology review of grade 3 tumours remains crucial.

PURPOSE: To assess the feasibility and activity of radio-chemotherapy with mitomycin C (MMC) and cisplatin (CDDP) in locally advanced squamous cell anal carcinoma with reference to radiotherapy (RT) combined with MMC and fluorouracil (5-FU). PATIENTS AND METHODS: Patients with measurable disease >4 cmN0 or N+ received RT (36Gy+2 week gap+23.4Gy) with either MMC/CDDP or MMC/5-FU (MMC 10mg/m(2) d1 of each sequence; 5-FU 200mg/m(2)/day c.i.v. daily; CDDP 25mg/m(2) weekly). Forty patients/arm were needed to exclude a RECIST objective response rate (ORR), 8 weeks after treatment, of <75% (Fleming 1, alpha=10%, beta=10%). RESULTS: The ORR was 79.5% (31/39) (lower bound confidence interval [CI]: 68.8%) with MMC/5-FU versus 91.9% (34/ 37) (lower bound CI: 82.8%) with MMC/CDDP. In the MMC/5-FU group, two patients (5.1%) discontinued treatment due to toxicity versus 11 (29.7%) in the MMC/CDDP group. Nine grade 3 haematological events occurred with MMC/CDDP versus none with 5-FU/MMC. The rate of other toxicities did not differ. There was no toxic death. Thirty-one patients in the MMC/5-FU arm (79.5%) and 18 in the MMC/CDDP arm (48.6%) were fully compliant with the protocol treatment (p=0.005). CONCLUSIONS: Radio-chemotherapy with MMC/CDDP seems promising as only MMC/CDDP demonstrated enough activity (RECIST ORR >75%) to be tested further in phase III trials; MMC/5-FU did not. MMC/CDDP also had an overall acceptable toxicity profile.
