Mice infected with adult Schistosoma mansoni were dosed with a single oral dose of 125 or 250 mg/kg oltipraz, 50 or 100 mg/kg oxamniquine, or 200 or 400 mg/kg praziquantel. The mortality rate of worms and oogram changes were determined between 1 and 16 weeks after dosing. The time required between dosing and postmortem to obtain maximum effectiveness was 1 week for praziquantel, 2 weeks for oxamniquine and 8 weeks for oltipraz. Changes in oograms persisted throughout most of the experiment, although relapse has been observed at the 4th week on.

Oito grupos de camundongos albinos (Mus musculus), não isogênicos, foram infectados transcutaneamente com cerca de 450 cercárias (das cepas LE e SJ do S. mansoni), irradiadas com 3 Krad, 20 Krad e 40 Krad de radiação gama proveniente de cobalto-60, e não irradiados (grupos-controle). Os vermes provenientes de cercárias irradiadas com 20 e 40 Krad só foram encontrados em quantidades insignificantes no sistema porta. Verificou-se que os vermes irradiados com 3 Krad, que alcançom o sistema porta, mostram nítido retardo no desenvolvimento evolutivo quando comparados com os grupos-controles não irradiados. Os vermes da cepa SJ (irradiados ou não) têm evolução mais lenta do que os da cepa LE.

T-cell function was evaluated in 29 patients with either hepatointestinal or hepatosplenic schistosomiasis by intradermal tests to recall antigens. Immunodepression was detected in 26% of the subjects with hepatointestinal schistosomiasis and in 50% of those with the hepatosplenic form. Cellular immunodepression was related to worm load and spleen size. This non specific T-cell immunodepression may represent a serious constraint to the elimination of intracellular pathogens both in hepatosplenic or hepatointestinal schistosomiasis.

Protein nutritionalstatus indicators were studied in weanling albino Swiss mice infected with S. mansoni andfed the Regional Basic Diet (RBD)from Northeast Brazil, a multideficient diet of low-protein content. Each mouse was infected percutaneously with 80 cercariae. The experiment lasted 63 days. The growth curve, food consumption, protein intake, weight gain, Protein Efficiency Ratio (PER) and Net Protein Ratio (NPR) were the parameters investigated. RBD-fed mice showed a marked weight loss, a lower food and protein intake, a slower body weight gain and lower rates of food protein utilization when compared to casein-fed animals. Differences between infected and non-infected mice were not consistent. The present results suggest that the effects of RBD-induced malnutrition on health and nutritional conditions of the mice are more severe than those of Manson's schistosomiasis, in the initial phase of the disease.

Em duas alíquotas de 208 amostras de fezes foram feitos exames pelo método de Kato-Katz, sendo uma após o emprego de 0,2mg de azida sódica para 200mg e em outra sem o referido conservador. Os resultados percentuais com e sem conservador foram, respectivamente, para os Ancilostomídeos 12,5 e 25,9, para Ascaris lumbricoides 71,6 e 72,5, para Schistosoma mansoni 7,6 e 17,7 e para Trichuris trichiura 86 e 85. A contagem dos ovos com e sem o conservador foi, respectivamente, 264 e 539 para os Ancilostomídeos, 13816 e 33751 para A. lumbricoides, 55,5 e 63,5 para S. mansoni e 1345 e 2068 para T. trichiura. Os autores não confirmaram a vantagem do uso de azida sódica para estudo em áreas endêmicas. Sugerem que o ressecamento das fezes e a baixa intensidade das infecções possam explicar os resultados desfavoráveis do presente ensaio clínico.

Mice previously infected with Schistosoma mansoni, and cured by specific treatment (400mg/kg oxamniquine, p. o.) in the chronic phase of the disease, were reinfected 20 days after treatment to assess their capacityfor modulation ofthe granulomatous response. Histopathologic examination of the animals ' liver, at 60 days after reinfection, evidenced the presence of typical granulomas of the chronic phase in most animals. This infer that the capacity for modulation of the granulomatous response had been maintained, thus preventing a new acute phase of the disease. Conversely, a group of previously infected mice, untreated and submitted to reinfection, showed reactivation of the granulomatous response in 50% of the animals. The possible implications of these findings in human schistosomiasis mansoni are discussed.

In this study, which was undertaken in relation to the histopathologic behavior of two different strains (LE-Belo Horizonte, MG and SJ - São José dos Campos, SP) in infections and reinfections (homologous or heterologous) with Schistosoma mansoni, the authors confirmed a more accentuated pathogenicity of the SJ strain. All the reinfections showed the presence of typical granulomas of the acute phase, when performed either with the same strain (homologous) or with a different strain (heterologous) of the parasite of the primo infection. The possible mechanisms responsible for reactivation of the immunopathologic response in reinfections are discussed.

In this study, four compounds were utilized at the dose of 12.5mg/kg body weight, p.o., to treat Cebus monkeys experimentally infected with about 200 cercariae of Schistosoma mansoni (SJ strain), via transcutaneous route. The oograms performed with rectal snips, as well as stool examinations carried out periodically, showed no viable eggs of the parasite, from day 29 to 226post-treatment. The perfusion undertaken after killing the animals showed absence of worms in the treated monkeys, whereas 83 worms were recovered from the control, thus corroborating the results obtained by means of oograms and coproscopy. These results confirm the efficacy of 9-acridanone- hydrazones previously tested against the LE strain of S. mansoni. The low curative dose and apparent absence of toxicity render these dmgs an important therapeutic reserve, taking into consideration the reports on the resistance of S. mansoni to the modern drugs oxamniquine and praziquantel.

Os níveis de infecciosidade de Schistosoma mansoni para as três espécies de Biomphalaria, hospedeiras intermediárias do parasita no Brasil, foram pesquisados após exposições conjuntas dos moluscos a miracídios, no laboratório e no campo. Foram utilizadas as cepas LE e SJ de S. mansoni, mantidas no laboratório e WVS e RFS obtidas de fezes de escolares de Belo Horizonte, MG. Os resultados mostraram a infecciosidade maior de S. mansoni para B. glabrata, com taxas de infecção de 4, 7 a 85,5%. A B. straminea foi suscetível às cepas LE, SJ e WVS, com taxas de infecção de 11,0 a 24,6%. A B. tenagophila foi suscetível a LE e Sj com taxas de infecção de 2,5 a 6,5%. As médias de cercárias da cepa WVS, eliminadas por dia por B. straminea e B. glabrata variaram de 93 ± 59 e 782 ± 1.120, respectivamente.
