The goal of this work was to establish comparisons among environmental degradation in different areas from Southern Spain (Gulf of Cadiz) and Brazil (Santos and Sao Vicente estuary), by using principal component analyses (PCA) to integrate sediment toxicity (amphipods mortality) and chemical-physical data (Zn, Cd, Pb; Cu, Ni, Co, V, PCBs, PAHs concentrations, OC and fines contents). The results of PCA extraction of Spanish data showed that Bay of Cadiz, CA-1 did not present contamination or degradation; CA-2 exhibited contamination by PCBs, however it was not related to the amphipods mortality. Ria of Huelva was the most impacted site, showing contamination caused principally by hydrocarbons, in HV-1 and HV-2, but heavy metals were also important contaminants at HV-1, HV-2 and HV-3. Algeciras Bay was considered as not degraded in GR-3 and -4, but in GR-3' high contamination by PAHs was found. In the Brazilian area, the most degraded sediments were found in the stations situated at the inner parts of the estuary (SSV-2, SSV-3, and SSV-4), followed by SSV-6, which is close to the Submarine Sewage Outfall of Santos - SSOS. Sediments from SSV-1 and SSV-5 did not present chemical contamination, organic contamination or significant amphipod mortality. The results, of this investigation showed that both countries present environmental degradation related to PAHs: in Spain, at Ria of Huelva and Gudarranque river's estuary areas; and in Brasil, in the internal portion of the Santos and Sao Vicente estuary. The same situation is found for heavy metals, since all of the identified metals are related to toxicity in the studied areas, with few exceptions (V for both Brazil and Spain, and Cd and Co for Brazilian areas). The contamination by PCBs is more serious for Santos and Sao Vicente estuary than for the investigated areas in Gulf of Cadiz, where such compound did not relate to the toxicity. Ayahuasca is a hallucinogenic brew traditionally used by Northwestern Amazonian indigenous groups for therapeutic purposes. It is prepared by the decoction of Banisteriopsis caapi with the leaves of Psychotria viridis. Banisteriopsis caapi contains β-carbolines that are inhibitors of monoamine oxidase and P. viris is rich in dimethyltryptamine, a 5-HT(1A/2A/2C) agonist. Acute ayahuasca administration produces moderate cardiovascular effects in healthy volunteers, but information regarding long-term use is lacking. This study investigated the effects of ayahuasca (2-4 mL/kg) in the rat aorta after acute and chronic (14 days) administration. Ayahuasca caused flattening and stretching of vascular smooth muscle cells and changes in the arrangement and distribution of collagen and elastic fibers. Chronic treatment with the higher dose significantly increased media thickness and the ratio of media thickness to lumen diameter. More research is needed on the cardiovascular function of long-term ayahuasca consumers.

We aimed to develop site-specific sediment quality guidelines (SQGs) for two estuarine and port zones in Southeastern Brazil (Santos Estuarine System and Paranagua Estuarine System) and three in Southern Spain (Ria of Huelva, Bay of Cadiz, and Bay of Algeciras), and compare these values against national and traditionally used international benchmark values. Site-specific SQGs were derived based on sediment physical-chemical, toxicological, and benthic community data integrated through multivariate analysis. This technique allowed the identification of chemicals of concern and the establishment of effects range correlatively to individual concentrations of contaminants for each site of study. The results revealed that sediments from Santos channel, as well as inner portions of the SES, are considered highly polluted (exceeding SQGs-high) by metals, PAHs and PCBs. High pollution by PAHs and some metals was found in Sao Vicente channel. In PES, sediments from inner portions (proximities of the Ponta do Mix port`s terminal and the Port of Paranagua) are highly polluted by metals and PAHs, including one zone inside the limits of an environmental protection area. In Gulf of Cadiz, SQGs exceedences were found in Ria of Huelva (all analysed metals and PAHs), in the surroundings of the Port of CAdiz (Bay of CAdiz) (metals), and in Bay of Algeciras (Ni and PAHs). The site-specific SQGs derived in this study are more restricted than national SQGs applied in Brazil and Spain, as well as international guidelines. This finding confirms the importance of the development of site-specific SQGs to support the characterisation of sediments and dredged material. The use of the same methodology to derive SQGs in Brazilian and Spanish port zones confirmed the applicability of this technique with an international scope and provided a harmonised methodology for site-specific SQGs derivation. Sediment quality from Paranagua Estuarine System (PES), a highly important port and ecological zone, was evaluated by assessing three lines of evidence: (1) sediment physical-chemical characteristics; (2) sediment toxicity (elutriates, sediment-water interface, and whole sediment); and (3) benthic community structure. Results revealed a gradient of increasing degradation of sediments (i.e. higher concentrations of trace metals, higher toxicity, and impoverishment of benthic community structure) towards inner PES. Data integration by principal component analysis (PCA) showed positive correlation between some contaminants (mainly As, Cr, Ni, and Pb) and toxicity in samples collected from stations located in upper estuary and one station placed away from contamination sources. Benthic community structure seems to be affected by both pollution and natural fine characteristics of the sediments, which reinforces the importance of a weight-of-evidence approach to evaluate sediments of PES. L’obiettivo che l’elaborato si prefigge consiste nel dar forma ad un progetto palladiano rimasto sulla carta attraverso l’utilizzo delle tecnologie informatiche. Nello specifico si è ricostruita Villa Mocenigo in forma tridimensionale attraverso uno schizzo del progetto fatto su carta dallo stesso Palladio nel 1550. Lo schizzo della pianta, conservato al RIBA di Londra, anticiperebbe il successivo progetto finale pubblicato ne “I Quattro Libri dell’Architettura” ma rimasto comunque incompiuta. Il linguaggio di base utilizzato per la definizione dell’ipotesi progettuale del modello 3D è stato attinto dalle regole generali di proporzionamento “alla maniera degli antichi” che lo stesso Architetto illustra nel suo trattato. Al fine di rendere l’ipotesi ricostruttiva il più possibile in linea con il metodo di proporzionamento palladiano, come esercitazione, si è partiti dallo studio preparatorio di un altro progetto: Villa Sarego, edificata solo in parte. A partire quindi dalla ricostruzione 3D basata sui disegni dei rilievi eseguiti dal CISAAP è stata avanzata l’ipotesi di ricostruzione totale della villa, completa della parte incompiuta. Lo studio sintetizzato in questo elaborato ha anche riguardato: la composizione architettonica e lo studio delle regole proporzionali evolutesi nel tempo; il processo di digitalizzazione e archiviazione delle opere palladiane e i progetti attualmente in corso; escursus storico sulle ville romane; le opera di Palladio, costruite e progettate.

Tesi svolta presso l'azienda “Riba composites S.r.l.” con lo scopo di riprogettare una passerella nautica utilizzata in imbarcazioni a vela da competizione, attualmente realizzata mediante formatura in autoclave di tessuti pre-impregnati, per il processo produttivo di Vacuum Assisted Resin Injection (VARI). La formatura in autoclave di tessuti pre-­impregnati è una delle tecnologie più onerose, tra i vari processi produttivi nel settore dei materiali compositi, ma assicura proprietà meccaniche e livelli estetici superlativi. L’obiettivo della Riba Composites è ridurre i costi di produzione per offrire un prodotto dalle proprietà analoghe a un prezzo più competitivo. Nella fase di riprogettazione ci siamo affidati a un software di calcolo agli elementi finiti che simula il processo del VARI, l’applicativo PAM-­RTM, del gruppo ESI. Al fine di ottenere una simulazione quanto più precisa possibile del processo, abbiamo realizzato molteplici prove sperimentali per ricavare i valori di compressibilità e permeabilità dei rinforzi da inserire nel software FEM.

I componenti del carbonio in ambito strutturale (aeronautico, navale, automobilistico) sono soggetti a deterioramenti ambientali di difficile determinazione, in particolare alla temperatura e all'umidità. Scopo di questa tesi è determinare i danneggiamenti dei materiali compositi CFRP in funzione di un invecchiamento a diverse percentuali della temperatura di transizione vetrosa Tg. In particolare si vuole studiare e approfondire il processo di reazione della matrice e del carbonio. Per meglio descrivere il procedimento di deterioramento e reazione del materiale composito dovuto ad alti livelli di temperatura, mi sono avvalso del supporto pratico dell’azienda “Riba Composites” di Faenza che in particolare si occupa della prototipazione e produzione di componenti strutturali in materiali compositi avanzati e che si è dimostrata leader nel settore dei compositi CFRP. Pochi studi sono stati condotti su tale argomento. Da qui il mio interesse specifico nel volere studiare e dimostrare come questo processo possa ulteriormente apportare un aiuto agli studi in ambito strutturale già effettuati e pubblicati precedentemente. La dimostrazione pratica della seguente tesi è avvenuta, con l’aiuto dell’Ing. Paolo Proli, nel laboratorio di MaSTeR Lab dell’Università di Bologna, dove si è deciso di eseguire vari invecchiamenti termici a diverse temperature per constatare i livelli di deterioramento e influenza delle variazioni di temperatura sulla matrice del composito preso in analisi. Gli effetti di tale studi sono stati sostenuti anche grazie alla guida del Professore Lorenzo Donati, e verranno dettagliatamente evidenziati nello specifico con spiegazioni in ambito teorico e dimostrazioni pratiche affiancate da schemi dimostrativi e supporto grafico.

The antiviral potency of the cytokine IFN-α has been long appreciated but remains poorly understood. A number of studies have suggested that induction of the apolipoprotein B mRNA editing enzyme, catalytic polypeptide 3 (APOBEC3) and bone marrow stromal cell antigen 2 (BST-2/tetherin/CD317) retroviral restriction factors underlies the IFN-α-mediated suppression of HIV-1 replication in vitro. We sought to characterize the as-yet-undefined relationship between IFN-α treatment, retroviral restriction factors, and HIV-1 in vivo. APOBEC3G, APOBEC3F, and BST-2 expression levels were measured in HIV/hepatitis C virus (HCV)-coinfected, antiretroviral therapy-naïve individuals before, during, and after pegylated IFN-α/ribavirin (IFN-α/riba) combination therapy. IFN-α/riba therapy decreased HIV-1 viral load by -0.921 (±0.858) log(10) copies/mL in HIV/HCV-coinfected patients. APOBEC3G/3F and BST-2 mRNA expression was significantly elevated during IFN-α/riba treatment in patient-derived CD4+ T cells (P < 0.04 and P < 0.008, paired Wilcoxon), and extent of BST-2 induction was correlated with reduction in HIV-1 viral load during treatment (P < 0.05, Pearson's r). APOBEC3 induction during treatment was correlated with degree of viral hypermutation (P < 0.03, Spearman's ρ), and evolution of the HIV-1 accessory protein viral protein U (Vpu) during IFN-α/riba treatment was suggestive of increased BST-2-mediated selection pressure. These data suggest that host restriction factors play a critical role in the antiretroviral capacity of IFN-α in vivo, and warrant investigation into therapeutic strategies that specifically enhance the expression of these intrinsic immune factors in HIV-1-infected individuals.

Alpha interferon (IFN-α) suppresses human immunodeficiency virus type 1 (HIV-1) replication in vitro by inducing cell-intrinsic retroviral restriction mechanisms. We investigated the effects of IFN-α/ribavirin (IFN-α/riba) treatment on 34 anti-HIV-1 restriction factors in vivo. Expression of several anti-HIV-1 restriction factors was significantly induced by IFN-α/riba in HIV/hepatitis C virus (HCV)-coinfected individuals. Fold induction of cumulative restriction factor expression in CD4+ T cells was significantly correlated with viral load reduction during IFN-α/riba treatment (r2 = 0.649; P < 0.016). Exogenous IFN-α induces supraphysiologic restriction factor expression associated with a pronounced decrease in HIV-1 viremia.

Objective. This study was designed to determine the prevalence and incidence of HCV infection among non-sexual household contacts of HCV-infected women and to describe the association between HCV infection and potential household risk factors in order to examine whether non-sexual household contact is a route of transmission for HCV infection. ^ Methods. A baseline prevalence survey included 409 non-sexual household contacts of 241 HCV-infected index women in the Houston area from 1994 to 1997. A total of 470 non-sexual household contacts with no evidence of HCV infection at baseline investigation were re-assessed approximately three years after baseline enrollment. Information on potential risk factors was collected through face to face interviews and blood samples were tested for anti-HCV with ELISA-2 and Matrix/RIBA-2. The relationships between HCV infection and potential risk factors were examined by using univariate and multivariate logistic regression analyses. ^ Results. The overall prevalence of anti-HCV positivity among 409 non-sexual household contacts was 4.4%. The highest prevalence of anti-HCV was found in parents (19.5%), followed by siblings (8.1%) and other relatives (5.6%); the children had the lowest prevalence of anti-HCV (1.2%). The univariate analysis showed that IDU, blood transfusion, tattoos, sexual contact with injecting drug users, more than 3 sexual partners in a lifetime, history of a STD, incarceration, previous hepatitis, and contact with hepatitis patients were significantly associated with HCV infection, however, sharing razors, nail clippers, toothbrushes, gum, food or beds with HCV-infected women, and history of dialysis, health care job, body piercing, and homosexual activities were not. Multivariate analysis found that IDU (OR = 221.7 with 95% CI of 22.8 to 2155.7) and history of a STD (OR = 11.7 with 95% CI of 1.2 to 113.1) were the only variables significantly associated with HCV infection. No such associations remained for other risk factors. The three-year cumulative incidence of anti-HCV among 352 non-sexual household contacts of HCV-infected women was zero. ^ Conclusion. This study has provided no evidence that non-sexual household contact is a likely route of transmission for HCV infection. The risk of sharing razors, nail clippers, toothbrushes, gum, food and/or beds with HCV-infected women is not evident and has not been shown to be the likely mode for HCV spread among family members. This study does suggest that IDU is the likely route of transmission for most HCV infection. Association also has been shown independently with a history of STD. The prevalence of anti-HCV among non-sexual household contacts was low. Exposure to common parenteral risk factors and sexual transmission between sexual partners may account for HCV spread among household members of HCV-infected persons. ^
