Ce mémoire porte sur le caractère heuristique et critique du discours utopique qu’il conviendra de mettre en lumière à travers les écrits du Sous-Commandant Marcos, écrits qui constituent un cas exemplaire des mécanismes que met en œuvre le discours utopique. Partant de la définition opératoire de l’utopie comme expression d’un imaginaire social qui relèverait d’une pratique utopique dont la fonction serait de critiquer le caractère idéologique des représentations spatio-temporelles induites par l’ordre dominant et, se questionnant par la suite sur la nature de l’ordre dominant contemporain et le caractère spatial de son hégémonie grâce aux travaux d’Henri Lefebvre et de David Harvey, il s’agira de montrer comment, au sein de ses textes, Marcos parvient à produire un espace représentationnel alternatif dans lequel opère un mécanisme de neutralisation des contradictions (Louis Marin), une véritable épochè, une suspension du jugement, seule condition de possibilité du dépassement des contraintes de l’ordre dominant dans le discours et les représentations.

Objectif: La caractérisation des facteurs immunitaires associés à la protection contre l’infection au VIH est cruciale pour le développement de stratégies de prévention. Cette étude évalue les IgGs dirigées contre l’enveloppe du VIH-1 dans le sérum et les liquides cervicovaginaux (LCV) de travailleuses du sexe béninoises. Méthode : Notre étude porte sur 23 travailleuses du sexe séropositives (TS+) et 20 travailleuses du sexe séronégatives (TS-). Le potentiel de neutralisation a été évalué par un essai de neutralisation. La détection d’IgGs a été effectuée par un ELISA sur base cellulaire. La capacité d’induire une réponse ADCC a été est évaluée par l’élimination de cellules cibles recouvertes de gp120 par le mécanisme d’ADCC. Résultats : Malgré que nous n’ayons pas détecté d’IgG dirigées contre l’enveloppe du VIH-1, ni d’activité de neutralisation ou d’élimination de cellules cibles par ADCC chez les TS-, nous avons facilement détecté ces activités de neutralisation et d’élimination de cellules cibles par ADCC chez les TS+ dans le sérum et les LCV qui reconnaissent mieux la forme de l’enveloppe liée à CD4. Ces IgGs pourraient être impliquées dans l’élimination par ADCC des cellules cibles présentant les glycoprotéines de l’enveloppe à leur surface, et ce à la muqueuse vaginale, le premier site de transmission virale. Conclusion : Ces résultats permettent pour la première fois de montrer la conformation de l’enveloppe préférentiellement reconnue par les IgGs présentes au site de transmission par voie sexuelle.

Au Canada, les discours politiques entourant la question des politiques pénales ont parfois été favorables à la répression, et parfois favorables à la réhabilitation et la littérature démontre qu’au cours des années, nous avons toujours nuancé notre approche à l’aide d’une philosophie pénale dichotomique, favorisant le juste équilibre entre ces deux rationalités pénales. Depuis l’arrivée du gouvernement conservateur en 2006 et malgré une baisse de l’activité criminelle enregistrée, l’activité législative en matière criminelle et pénale s’est intensifiée. En 2011, le gouvernement a ainsi déposé le projet de loi C-10, une loi omnibus comprenant 5 volets distincts, chacun mettant en place des changements important sur différentes dimensions du processus criminel ou pénal. L’objet de notre étude consiste, à partir de l’analyse des débats entourant l’adoption de ce projet de loi C-10, à comprendre les représentations du crime, du criminel et de la sanction pénale, et ce à partir de l’analyse documentaire thématique. Le corpus utilisé dans ce mémoire est formé de débats parlementaires ayant eu lieu à la Chambre des communes entre le 20 septembre 2011 et le 6 décembre 2011. Il ressort de notre analyse que la perspective dualiste d’autrefois a disparu au détriment d’une perspective pénale axée sur la protection en favorisant la rétribution, réprobation et la neutralisation. Nos résultats soulignent l’importance de l’émotion et du sensationnel dans les débats, ainsi que l’arrivée d’une pénalité politisée.

Les cellules myéloïdes incluant les monocytes, les macrophages et les cellules dendritiques (DCs, dendritic cells) contribuent à la pathogénèse de l’infection à VIH-1 en participant à la dissémination du virus, mais également en représentant des réservoirs viraux potentiels. Leurs fonctions sont exploitées par le VIH-1 afin d’assurer sa propagation à travers l’organisme. Notamment, une infection à VIH-1 est associée à une altération de la présentation antigénique et la perte de lymphocytes T CD4+ spécifiques à des antigènes. L’autophagie est un processus catabolique universel impliqué dans la régulation de la présentation antigénique subséquente à la neutralisation/destruction du pathogène. Des études récentes suggèrent que le VIH-1 altère le mécanisme d’autophagie afin d’assurer sa survie. Le premier volet de ce projet de maîtrise a visé la caractérisation des effets du VIH-1 sur l’autophagie dans les DCs dérivées de monocytes circulants (MDDC, monocyte-derived dendritic cells) et l’identification des stratégies thérapeutiques pour contrecarrer ces effets. Les objectifs spécifiques ont été de : (i) caractériser l’expression de marqueurs de maturation sur des MDDC exposées au VIH-1 in vitro, (ii) quantifier l’expression des protéines liées à la régulation positive (i.e., ATG5, LC3, p62) et négative (i.e., mTOR) de l’autophagie dans les MDDC exposées au VIH, (iii) déterminer le rôle de l’autophagie dans la trans infection du VIH-1 aux lymphocytes T CD4+ et (iv) explorer l’impact de l’autophagie sur la présentation antigénique par les MDDC infectées à VIH-1 in vitro. Nos résultats démontrent qu’une exposition des MDDC au VIH-1 in vitro altère dramatiquement leur maturation et leur habileté à induire la prolifération des cellules T autologues en réponse à Staphylococcus aureus et Cytomegalovirus (CMV) mais pas la réponse induite par Staphylococcal enterotoxin B (SEB). Nous démontrons que l’exposition des MDDC au VIH s’associe à une augmentation de l’expression de la protéine mTOR totale et de sa forme phosphorylée (phospho-mTOR) et de la protéine p62. Le traitement des MDDC à la rapamycine diminue l’expression de mTOR et réduit la capacité de trans infection du VIH-1 par les MDDC, sans toutefois restaurer leur potentiel immunogène. En effet, nous observons que la rapamycine réduit l’expression de CD83 par les MDDC et augmente l’expression de CCR7, indiquant ainsi que l’effet immunosuppresseur documenté de la rapamycine est associé à une défaillance de maturation des MDDC. Le second volet de ce projet de recherche s’est intéressé à la contribution des cellules myéloïdes à la persistance virale chez les sujets infectés par le VIH-1 sous thérapie antirétrovirale. Les objectifs spécifiques ont été : (i) d’évaluer la présence de différentes formes d’ADN viral dans les monocytes circulants de patients infectés par le VIH-1 et (ii) de mesurer l’intégration et la réplication virale dans des macrophages dérivés de monocytes (MDM) de patients infectés sous ART. Nos résultats indiquent que les monocytes portent des formes précoces de transcription virale inverse (ADN du VIH RU5) et que, malgré une charge virale plasmatique indétectable sous ART, les MDM supportent la réplication virale. Ces données très préliminaires apportent des évidences en faveur de la contribution des cellules myéloïdes à la persistance virale sous ART et représentent une ouverture pour un projet de recherche plus complexe dans le futur. En somme, nos résultats démontrent que le VIH-1 altère le potentiel immunogène des MDDC et que la rapamycine peut être employée pour limiter la trans infection des lymphocytes T CD4+ par les MDDC. Néanmoins, l’incapacité de la rapamycine à rétablir le potentiel immunogène des MDDC incite à identifier de nouvelles stratégies manipulant l’autophagie pour une restauration optimale de la compétence immunitaire chez les sujets infectés à VIH-1. Les cellules myéloïdes jouent un rôle primordial dans la dissémination et la persistance virale et doivent donc être ciblées par les stratégies actuelles d’éradication des réservoirs du VIH sous ART.

Liquid Crystalline DNA is emerging as an active area of research, due to its potential applications in diverse fields, ranging from nanoelectronics to therapeutics. Since, counter ion neutralization is an essential requirement for the expression of LC DNA, and the present level of understanding on the LC phase behavior of high molecular weight DNA is inadequate, a thorough investigation is required to understand the nature and stability of these phases under the influence of various cationic species. The present study is, therefore mainly focused on a comparative investigation of the effect of metal ions of varying charge, size, hydration and binding modes on the LC phase behavior of high molecular weight DNA. The main objectives of the works are investigations on the induction and stabilization of LC phases of high molecular weight DNA by alkali metal ions, investigations on the induction and stabilization of LC phases of high molecular weight DNA by alkaline earth metal ions, effects of multivalent, transition and heavy metal ions on the LC phase behavior of high molecular weight DNA and investigations on spermine induced LC behavior of high molecular weight DNA in the presence of alkali and alkaline earth metal ions. The critical DNA concentration (CD) required for the expression of LC phases, phase transitions and their stability varied considerably when the binding site of the metal ions changed from phosphate groups to the nitrogenous bases of DNA, with Li+ giving the highest stability. Multiple LC phases with different textures, sometimes diffused and unstable or otherwise mainly distinct and clear, were observed on mixing metal ions with DNA solutions, which in turn depended on the charge, size, hydration factor, binding modes, concentration of the metal ions and time. Cast Ai-Si alloys are widely used in the automotive, aerospace and general engineering industries due to their excellent combination of properties such as good castability, low coefficient of thermal expansion, high strength-to-weight ratio and good corrosion resistance. The present investigation is on the influence of alloying additions on the structure and properties of Ai-7Si-0.3Mg alloy. The primary objective of this present investigation is to study these beneficial effects of calcium on the structure and properties of Ai-7Si-0.3Mg-xFe alloys. The second objective of this work is to study the effects of Mn,Be and Sr addition as Fe neutralizers and also to study the interaction of Mn,Be,Sr and Ca in Ai-7Si-0.3Mg-xFe alloys. In this study the duel beneficial effects of Ca viz;modification and Fe-neutralization, comparison of the effects of Ca and Sr with common Fe neutralizers. The casting have been characterized with respect to their microstructure, %porosity and electrical conductivity, solidification behaviour and mechanical properties. One of the interesting observations in the present work is that a low level of calcium reduces the porosity compared to the untreated alloy. However higher level of calcium addition lead to higher porosity in the casting. An empirical analysis carried out for comparing the results of the present work with those of the other researchers on the effect of increasing iron content on UTS and % elongation of Ai-Si-Mg and Ai-Si-Cu alloys has shown a linear and an inverse first order polynomial relationships respectively.

Rare earth metal ion exchanged (La3+, Ce3+, RE3+) KFAU-Y zeolites were prepared by simple ion-exchange methods and have been characterized using different physico-chemical techniques. In this paper a novel application of solid acid catalysts in the dehydration/ Beckmann rearrangement of aldoximes; benzaldoxime and 4-methoxybenzaldoxime is reported. Dehydration/Beckmann rearrangement reactions of benzaldoxime and 4-methoxybenzaldoxime is carried out in a continuous down flow reactor at 473K. 4-Methoxybenzaldoxime gave both Beckmann rearrangement product (4-methoxyphenylformamide) and dehydration product (4-methoxybenzonitrile) in high overall yields. The difference in behavior of the aldoximes is explained in terms of electronic effects. The production of benzonitrile was near quantitative under heterogeneous reaction conditions. The optimal protocol allows nitriles to be synthesized in good yields through the dehydration of aldoximes. Time on stream studies show a fast decline in the activity of the catalyst due to neutralization of acid sites by the basic reactant and product molecules.

Rare earth exchanged Na–Y zeolites, H-mordenite, K-10 montmorillonite clay and amorphous silica-alumina were effectively employed for the continuous synthesis of nitriles. Dehydration of benzaldoxime and 4-methoxybenzaldoxime were carried out on these catalysts at 473 K. Benzonitrile (dehydration product) was obtained in near quantitative yield with benzaldoxime whereas; 4-methoxybenzaldoxime produces both Beckmann rearrangement (4-methoxyphenylformamide) as well as dehydration products (4-methoxy benzonitrile) in quantitative yields. The production of benzonitrile was near quantitative under heterogeneous reaction conditions. The optimal protocol allows nitriles to be synthesized in good yields through the dehydration of aldoximes. Time on stream (TOS) studies show decline in the activity of the catalysts due to neutralization of acid sites by the basic reactant and product molecules and water formed during the dehydration of aldoximes.

Novel thermo-reversible zinc sulphonated ionomers based on styrene butadiene rubber (SBR), and high styrene rubber (HSH) were synthesized by sulphonation followed by neutralization with zinc. The sulphonate content of the ionomer was estimated by using x-ray fluorescence spectroscopy. Presence of sulphonate groups has been confirmed by FTIR and FTNMR spectra. The TGA results show improvement in the thermo~oxidative stability of the modified rubber. Both DSC and DMTA studies show that the incorporation of the ionic groups affect the glass rubber transition of the base polymer. lntroduction ol ionic functionality in to the base material improved the physical properties. Retention of the improved physical properties of the novel ionomers even after three repeated cycles of mixing and molding may be considered as the evidence for the reprocessability of the ionomer. Effect of particulate fillers (HAF black, silica and zinc stearate) on the properties of the zinc sulphonated styrene butadiene rubber ionomer has been evaluated. Incorporation of tillers results in improvement in mechanical properties. Zinc stearate plays the dual role of reinforcement and plasticization. The evaluation of dielectric properties of zinc sulphonated styrene butadiene rubber iorpmers at microwave frequencies reveal that the materials show conductivity at semiconductor level. The real and imaginary parts of the complex permittivity increases with increase in ionic functionality. Use of the 38.5 ZnS-SBR ionomer as a compatibiliser for obtaining the technologically compatible blends from the immiscible SBR/NBR system has been discussed.

High styrene rubber ionomers were prepared by sulfonating styrene–butadiene rubber of high styrene content (high styrene rubber) in 1,2-dichloroethane using acetyl sulfate reagent, followed by neutralization of the precursor acids using methanolic zinc acetate. The ionomers were characterized using X-ray fluorescence spectroscopy, Fourier transform infrared spectroscopy (FTIR), nuclear magnetic resonance spectroscopy (NMR), dynamic mechanical analysis (DMA), and also by the evaluation of mechanical properties. The FTIR studies of the ionomer reveal that the sulfonate groups are attached to the benzene ring. The NMR spectra give credence to this observation. Results of DMA show an ionic transition (Ti) in addition to glass–rubber transition (Tg). Incorporation of ionic groups results in improved mechanical properties as well as retention of properties after three cycles of processing

A scheme to describe SDS−lysozyme complex formation has been proposed on the basis of isothermal titration calorimetry (ITC) and FTIR spectroscopy data. ITC isotherms are convoluted and reveal a marked effect of both SDS and lysozyme concentration on the stoichiometry of the SDS−lysozyme complex. The binding isotherms have been described with the aid of FTIR spectroscopy in terms of changes in the lysozyme structure and the nature of the SDS binding. At low SDS concentrations, ITC isotherms feature an exothermic region that corresponds to specific electrostatic binding of SDS to positively charged amino acid residues on the lysozyme surface. This leads to charge neutralization of the complex and precipitation. The number of SDS molecules that bind specifically to lysozyme is approximately 8, as determined from our ITC isotherms, and is independent of lysozyme solution concentration. At high SDS concentrations, hydrophobic cooperative association dominates the binding process. Saturated binding stoichiometries as a molar ratio of SDS per molecule of lysozyme range from 220:1 to 80:1, depending on the lysozyme solution concentration. A limiting value of 78:1 has been calculated for lysozyme solution concentrations above 0.25 mM.

The hemagglutinins (HAs) of human H1 and H3 influenza viruses and avian H5 influenza virus were produced as recombinant fusion proteins with the human immunoglobulin Fc domain. Recombinant HA-human immunoglobulin Fc domain (HA-HuFc) proteins were secreted from baculovirus-infected insect cells as glycosylated oligomer HAs of the anticipated molecular mass, agglutinated red blood cells, were purified on protein A, and were used to immunize mice in the absence of adjuvant. Immunogenicity was demonstrated for all subtypes, with the serum samples demonstrating subtype-specific hemagglutination inhibition, epitope specificity similar to that seen with virus infection, and neutralization. HuFc-tagged HAs are potential candidates for gene-to-vaccine approaches to influenza vaccination.

The aim of this study was to investigate the antimicrobial properties of fifteen selected strains belonging to the Lactobacillus, Bifidobacterium, Lactococcus, Streptococcus and Bacillus genera against Gram-positive and Gram-negative pathogenic bacteria. In vitro antibacterial activity was initially investigated by an agar spot method. Results from the agar spot test showed that most of the selected strains were able to produce active compounds on solid media with antagonistic properties against Salmonella Typhimurium, Escherichia coli, Enterococcus faecalis, Staphylococcus aureus and Clostridium difficile. These results were also confirmed when cell-free culture supernatants (CFCS) from the putative probiotics were used in an agar well diffusion assay. Neutralization of the culture supernatants with alkali reduced the antagonistic effects. These experiments are able to confirm the capacity of potential probiotics to inhibit selected pathogens. One of the main inhibitory mechanisms may result from the production of organic acids from glucose fermentation and consequent lowering of culture pH. This observation was confirmed when the profile of organic acids was analysed demonstrating that lactic and acetic acid were the principal end products of probiotic metabolism. Furthermore, the assessment of the haemolytic activity and the susceptibility of the strains to the most commonly used antimicrobials, considered as basic safety aspects, were also studied. The observed antimicrobial activity was mainly genus-specific, additionally significant differences could be observed among species.

The aims of the present study were to investigate in vitro the antimicrobial activity of Lactobacillus fermentum and Bifidobacterium longum, isolated from faeces of healthy elderly individuals, against enterohaemorrhagic Escherichia coli (E. coli O157:H7) and enteropathogenic E. coli (E. coli O86), to determine the capability of the selected strains to tolerate acid and bile in vitro, to select suitable carbohydrates in order to enhance the growth and maximise antimicrobial activity of the putative probiotic organisms and examine the adhesion properties of the synbiotics. Antimicrobial activity of the putative probiotics and synbiotics was investigated by a microtitre method using cell-free culture supernatants (CFCS). Results of the antimicrobial assay showed that both putative probiotic strains produced compounds at pH 5 that lead to higher lag phases of both E. coli O157:H7 and E. coli O86. When half the quantity of cell-free culture supernatants of both probiotic strains was used at pH 5, B. longum maintained the same antimicrobial effect against both strains of E. coli, whereas L. fermentum lead to a higher lag phase of E. coli O86 only. Neutralization of the culture supernatants with alkali reduced the antimicrobial effect with only cell-free supernatant of L. fermentum causing lower maximum growth rates of E. coli O157:H7 and E. coli O86. L. fermentum appeared to be acid tolerant whereas B. longum was more susceptible to acid and both isolates were bile tolerant. A short chain fructooligosaccharide (scFOS) and an isomalto-oligosaccharide (IMO) proved to be the most effective substrates, enhancing antimicrobial activity for L. fermentum and B. longum respectively. The adhesion of the synbiotic combinations showed that L. fermentum, exhibited higher percentage of adhesion when grown on glucose and as a synbiotic combination with scFOS whereas B. longum exhibited lowest percentage of adhesion when grown on both glucose and IMO.
