Objetivou-se no presente trabalho verificar a degradação ruminal e a digestibilidade intestinal e total da matéria seca (MS) e da proteína bruta (PB) do farelo de soja, do grão de milho, do melaço em pó, da farinha de peixe, da farinha de penas e do feno de alfafa, por intermédio da técnica de degradabilidade ruminal in situ associada à técnica do saco de náilon móvel. As amostras dos alimentos foram moídas e colocadas em duplicata em sacos de náilon de 10x5 cm (48 micras) nas quantidades de 15 e 5 mg de MS/cm² para os alimentos concentrados e feno de alfafa, respectivamente. Os sacos de náilon permaneceram incubados no rúmen de bois holandeses por 0; 2; 6; 8; 24 e 48 h; e 0; 8; 12; 24; 48; 72 e 96 horas, respectivamente, sendo depois retirados e sua duplicata inserida no duodeno através de uma cânula. Posteriormente, os sacos foram coletados junto com as fezes. Os valores de degradabilidade efetiva da PB para uma velocidade de passagem de 5%/hora, para o melaço em pó, grão de milho, farelo de soja, farinha de peixe, farinha de penas e feno de alfafa, foram de 100,00; 62,50; 57,90; 39,30; 34,20 e 60,90%, respectivamente; a digestibilidade intestinal de 100,00; 96,05; 99,79; 98,19; 96,07 e 94,64%, respectivamente; e a digestibilidade total de 100,00; 97,86; 99,87; 98,88; 97,35 e 98,09%, respectivamente. Verificou-se que as proteínas do melaço foram totalmente solúveis no rúmen, sendo as do milho, feno e farelo de soja bastante degradadas, além de possuírem um aproveitamento quase total no intestino. As proteínas das farinhas de peixe e de penas apresentaram baixa solubilidade ruminal e alta digestibilidade intestinal, sendo a farinha de peixe levemente mais digerida no intestino do que a farinha de penas.

O equino é um herbívoro não ruminante capaz de suprir grande parte ou a totalidade da sua demanda nutricional pela ingestão de gramíneas. Apresenta a região ceco-cólica bastante desenvolvida, sendo este o principal sítio de fermentação. Este processo também ocorre na região aglandular do estômago, porém a produção de ácidos graxos voláteis é inferior, quando comparado ao intestino grosso. O conhecimento do sítio de aproveitamento de cada ingrediente é de suma importância para a combinação dos mesmos, favorecendo o ótimo aproveitamento de cada um deles e evitando excessos que podem ser prejudiciais ao metabolismo do equino. O estudo atual do fracionamento de carboidratos, por mais que não seja adaptado a fisiologia e metabolismo dos equinos, nos fornece informações sobre as diferentes frações que o compõem e com isso pode-se inferir sobre a produção de energia por cada fração gerada. A manutenção de equinos em pastejo ou sob o fornecimento de feno ou alimentos volumosos é essencial para a manutenção da atividade e saúde do seu trato digestório. O aporte mínimo de 12% de FDN garante tal situação. Atualmente, com o objetivo de aumentar a disponibilidade de energia, ingredientes como a polpa cítrica, polpa de beterraba e casca de soja, vem sendo utilizados nas formulações. Estes ingredientes de fermentação mais rápida e fácil, são uma alternativa segura para elevar a concentração energética da dieta de equinos, por proporcionar a diminuição do aporte de amido na dieta de equinos. Sabe-se entretanto, que com a elevação na qualidade do volumoso, maiores concentrações de carboidratos solúveis e de rápida fermentação são encontrados disponíveis ao longo do trato digestório dos equinos levando a alguns distúrbios metabólicos como a laminite e a sensibilidade a insulina. Com o correto manejo dos animais em pastejo e conhecimento do ciclo da planta é possível minimizar tais problemas. Objetivou-se revisão da ação fisiológica e metabólica das diferentes frações fibrosas, em cada compartimento do trato digestório, bem como destacar os ingredientes volumosos com suas diversas frações, os quais possibilitam sugerir manejo alimentar adequado para os equinos.

To evaluate the nutritional value of rations supplemented with different protein sources, an in vivo digestibility trial was conducted to determine the coefficients of digestibility for dry matter (DMDC) and crude protein (CPDC), nitrogen balance (NB) and the fecal non-protein nitrogen (NPN). Twenty - four 11-months - old lambs weighing 30.4 +/- 3.0 kg were blocked on the basis of their body weight and randomly allocated to three treatments groups; suplemented with cottonseed meal (CSM), starea (ST) or urea (UR). The DMDC was statistically superior for the Starea treatment (67.7%) over the others (64.3% and 64.1% for CSM and UR). The CSM, UR and ST rations had no influence on CPDC (63.9; 66.9 and 69.4%, respectively) and on fecal NPN (1,3; 1,2 e 1,3 g/day, respectively). The NB results were similar also among treatments (13,4; 9,0 e 10,5 g/day to CSM, UR and ST, respectively) although the ST treatment lambs excreted larger amounts of nitrogen in the urine (7.7 g/day) in relation to CSM (4.8g/day) and similar to UR treatment (6.5 g/day). Starea supplement in ruminant diets increased dry matter digestibility when compared to cottonseed meal and urea, but did not improve the efficiency of the nitrogen utilization by lambs.

The objectives of this study were to determine the technical viability of amniocentesis in sheep and to observe biochemical changes in the amniotic fluid components. Amniotic fluid samples were collected by puncture in the greatest curvature of the uterine hum at days 70, 100, and 145 of pregnancy. The surgical procedure for collection of amniotic fluid samples was safe and efficient. For three stages of pregnancy, the following results were obtained: pH values 8.36, 7.34 and 7.37; glucose concentrations, 16.06. 8.58, and 3.79 g/dl; urea values, 42.68, 33.53, and 25.49 mg/dl; creatinine, 0.85, 5.04, and 11.25 g/dl; Gama-GT enzyme, 12.58, 14.20, and 12.30 UI/l; sodium concentrations, 146.60, 129.42, and 103.8 mmol/l: potassium concentrations, 9.79, 6.15, and 8.65 mmol/l; chloride, 96.59, 85.28, and 65.35 mmol/l; total protein, 0.14, 0.23, and 0.24 g/dl, respectively. Protein is one of the limiting factors in animal production, and the knowledge of protein requirements by livestock is crucial for the success of a commercial animal raising enterprise. Thirty-four castrated lambs, 17 of them F1 Ideal x lie de France wool lambs and the remaining ones were Santa Ines hair lambs, with homogeneous initial BW, were used in the experiment. Five animals from each genotype were slaughtered in the beginning of the experimental period and used as reference. Diets (D) were composed of concentrate mix (C) and Cynodon sp. c.v. Tifton 85 hay (R), combined in three different ratios: D1 =60CAOR; D2=40C:60R and D3=20C:80R. Animals of each group of three lambs, that showed simultaneously an initial BW of 20 +/- 0.14 kg at the beginning of the dietary regimen, were slaughtered when one of them reached 35 kg, what always happened to be the one fed with D1. Net requirements for BW gain in wool lamb, fleece-free, ranged from 101 to 110 g of protein/kg BW, and for hair lamb ranged from 1 10 to 118 g of protein/kg BW. Net protein requirements for wool production ranged from 634 to 642 g/kg of produced wool. Hair lambs presented a 7.8-9.5% higher estimated net protein requirements than wool lambs, according to BW and daily weight gain (DG). Total net protein requirements for Santa Ines and wool lambs, with 30 kg of initial B W and an approximate 200 g mean DG, were 48.5 and 45.4 g/day, respectively. Metabolizable protein requirements for Santa Ines and wool lambs, with 20 kg of initial BW and an approximate 200 g mean DG were 59.4 g and 76.5 -/day, respectively. Net protein requirements for wool production was 64g/100g of produced wool. Thus, under the conditions of this experiment, it is concluded that hair lambs showed a higher concentration of protein in the body, more efficient use of the ingested protein and a consequent additional BW gain when fed isoproteic diets as compared to F1 Ideal x Ile de France wool lambs. The objective of this work was to evaluate the total endogenous N losses, and protein and energy net requirements for maintenance in growing lambs. Thirty-four castrated lambs, 17 F1 Ideal X Ile de France wool and 17 Santa Inas hair lambs, averaging 20 +/- 0.14 kg BW, were used in the experiment. Five animals from each genotype were slaughtered at the beginning of the experiment and taken as controls. Diets (D) were composed of concentrate mix (C) and Cynodon sp. c.v. Tifton 85 hay (R), combined in three different ratios: D1 = 60C:40R; D2 = 40C:60R and D3 = 20C:80R. Animals of each group of three lambs, that showed BW of 20 kg at the beginning of the dietary regimen, were slaughtered when one of them reached 35 kg, what always happened to be the one fed with D1. Total endogenous N losses estimated for wool lambs were 250 mg N/kg BW0.75. For hair lambs, total endogenous N losses reached 324 mg N/kg BW0.75 . Hair lambs showed higher (P < 0.01) (29.9%) net requirements of protein for maintenance than wool lambs. In contrast, net energy (NE) requirement for maintenance was similar (P > 0.05) for both genotypes (74.27 kcal/kg BW0.75 per day), the average of the antilog of the two intercept values obtained from the estimated regression equations of heat production for zero metabolizable energy (ME) consumption. Further studies should be done to check if this trend is also true for metabolizable energy and protein in animals exhibiting BW gains in tropical region. (C) 2003 Elsevier B.V. B.V. The present study aimed at determining the influence of condensed tannins present in the Brazilian legume species Mimosa hostilis, Mimosa caesalpinifolia and Bauhinia cheilantha on ruminal degradability, microbial colonization and enzymatic activity. Polyethylene glycol (PEG) was used to reduce the astringency and concentration of soluble condensed tannins. Four ruminally-cannulated Saanen goats (60 +/- 8 kg BW) were fed, in two experimental periods, with a hay diet based on the studied legumes treated or non-treated with PEG. Voluntary intake, microbial colonization, DM, CP, NDF, and ruminal degradability of PEG treated and non-treated forage leaves, as well as pH, ammonia and 1,4 P-endoglucanase activity of the rumen content were evaluated. Astringency and soluble tannin concentration of the studied legumes were reduced by approximately 70% and 50%, respectively, with PEG treatment. Average DM intake was higher for the treated diet (16.76 g DM/kg BW/day against 13.06 g DM/kg BW/day). Percentile values for degradation parameters and for potential and effective degradabilities of DM, CP and NDF were also affected by the tannins, but at different intensities. Electron microscopic observations of ruminally-incubated legume leaves showed a more effective microbial colonization of PEG-treated leaves for all legume species. A decrease in pH and an increase in ammonia concentration and in endoglucanase activity in the ruminal content was also observed for PEG-treated diets at all sampling periods. Condensed tannins of the studied legume species have influenced the adhesion conditions, colonization and enzymatic activity of the microbial ecosystem, and consequently the ruminal degradation of the different dietary fractions. For this reason, the reduction in condensed tannin would be of great importance to improve the nutrition of ruminant feeding of these species. (c) 2005 Elsevier B.V. All rights reserved.

Aiming at evaluating the use of those byproducts on ruminant feeding an experiment has been carried out in the Forage Research department (www.npf.ufc.br), in order to evaluate the effects of addition of increasing levels of mango processing byproducts (MB) in Elephant grass silages. Twenty experimental silos made of PVC pipe (100 x 340 mm), in a completely randomized design with 5 levels of addition (0; 5; 10; 15; and 20%) of MB and 4 replications. Upon 32 days of ensilage, samples were collected for determination of levels of dry Matter (DM), crude protein (CP), ethereal extract (EE), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicelluloses (HC), ammonia nitrogen (N-NH3), and pH values. Data were evaluated by analyses of variance and regression analyses using the F test at the 5% level. The levels of DM and CP increased of 0.50 and 0.03 percentage units, respectively, for every 1% of MB added. The levels of NDF and ADF decreased of 0.51 and 0.24 percentage units, respectively, for every 1% of MB added. The levels of N-NH3 decreased from 4 to 23 percentage units while pH values remained constant (p>0.05), with average values of 3.29. Addition of 20% of MB improved silages chemical composition by promoting an increase in the levels of MS and reduction in the levels of fiber.

In order to compare the in situ degradability of tanzania grass samples obtained as by extrusa or hand plucked, three ruminal fistulated cows were used in a completely randomized block design with split-plot scheme. Five grams of extrusa or hand harvested grasses were placed in nylon bags rumen incubated during 3, 6, 12, 24, 48, 96 and 120 hours. The degradability of DM, CP, NDF and ADF were, in this sequence, 62.59, 80.88, 50.73 and 46.65%, for hand-harvested grass; and 79.53, 90.97, 71.21 and 65.68%, for extrusa. In situ degradability data of hand harvested samples were not reliable.

Conjugated Linoleic Acids (CLAs) comprise a family of positional and geometric isomers of linoleic acid. The main form of CLA, cis-9, trans-11-C18:2 show positive effects in cancer prevention and treatment. The major dietary sources of these fatty acids are derived from ruminant animals, in particular dairy products. In these animals, the endogenous synthesis mainly occurs in mammary gland by the action of enzyme Stearoyl CoA Desaturase (SCD). Different levels of expression and activity of SCD in mammary gland can explain partially the variation of CLA levels in fat milk. Considering a great fat concentration in bubaline milk and the benefit of a high and positive correlation between fat milk and CLA production, this study was carried on with the intention of sequencing and characterizing part of the gene that codifies SCD in buffaloes. Genomic DNA was extracted from blood samples of lactating bubaline which begins to the breed Murrah. After the (acho que nao precisa desse the) extractions, PCR (Polymerase Chain Reaction) reactions were made by using primers Z (sic) (sic) D1 and E1 (sic) (sic) F1. The fragments obtained in PCR were cloned into T vectors and transformed in competent cells DH10B line. After this, three samples of each fragment were sequenced from 5' and 3' extremities using a BigDye kit in an automatic sequencer. Sequences were edited in a consensus of each fragment and were submitted to BLAST-n / NCBI for similarity comparisions among other species. The sequence obtained with Z (sic) (sic) D1 primers shows 938 bp enclosing exons 1 and 2 and intron 1. The primers E1 (sic) (sic) F1 show 70 bp corresponding to exon 3 of bubaline SCD gene. Similarities were obtained between 85% and 97% among bubaline sequences and sequences of SCD gene described in human, mouse, rat, swine, bovine, caprine and ovine species. This study has permitted the identification and partial characterization of SCD codifing region in Bubalus bubalis specie.

This work was conducted to evaluate food intake and digestive efficiency of temperate wool and tropic semi-arid hair lambs, according to different concentrate: forage ratios in diet. Twenty-four lambs, averaging 90 +/- 1.8 days old and a mean body weight of 20 +/- 0.69 kg, 12 of them wool lambs, F, from Ideal x Ile de France crossing, and 12 others pure Santa Ines hair lambs, were distributed into a four replication 3 x 2 factorial arrangement consisting of three diets and two genotypes. Experimental diets consisted of: D1 = 60% concentrate mix (C) and 40% Cynodon sp. cv. Tifton-85 hay (F), D2 = 40% C and 60% F, and D3 = 20% C and 80% F. D1 was formulated for a daily gain of 300g per animal. Increasing forage levels in diets resulted in linear reductions (P < 0.01) in DM, OM, CP, TCH and metabolizable energy (ME) intake, and a linear increase (P < 0.01) in NDF ingestion. Tropic semi-arid hair lambs had higher DM, OM, NDF, CP, and TCH intake than temperate wool lambs. Although there were no genotype effects in OM and GE coefficient of digestibility, hair lambs showed more efficient (P < 0.05) digestibility of DM, CP, NDF and TCH. Increases in forage levels of diets corresponded to a negative linear effect (P < 0.01) in the apparent digestibility of DM, OM, CP, TCH and GE, while apparent digestibility of NDF increased linearly (P < 0.01). Total endogenous nitrogen (fecal plus urinary N) for F(1) Ideal x Ilede France wool and Santa Ines hair lambs were, respectively, 182 and 312 mg/kg(0.75) per day. Thus, Santa Ines tropic semi-arid hair lambs showed to be more responsive than F(1) Ideal x Ile de France temperate wool lambs to low quality fibrous diets. (C) 2004 Elsevier B.V. A total of 116 goats originating from three heterozygous bucks for rob (5/15) were cytogenetically studied at Botucatu, Brazil. In the mating 59,XY,t(5/15) x 60,XX, the offspring karyotype proportion did not differ significantly from the expected ratio of 1 normal:1 heterozygous. In the mating 59,XY,t(5/15) x 59,XX,t(5/15) the observed proportion was 1 normal:2 heterozygous:1 homozygous, and in the mating 59,XY,t(5/15) X 58,XX,tt(5/15) the observed proportion was 1 heterozygous:1 homozygous. No animals had unbalanced karyotypes and no differences in the weight at birth of normal, heterozygous and homozygous kids were observed (P > 0.10).

The aim of this study was to compare the chemical composition, in vitro dry matter disappearance (IVDMD) and ruminal degradation of Panicum maximum, J. cv. Tanzania samples obtained by clipping (square method) or extrusa collection (animal selection). In the in situ trial, three ruminal fistulated dry crossbred cows, with 499 kg LW, were used in a completely randomized block design with split-plot arrangement design. Five grams of clipped (+/- 2 cm) grass or extrusa samples were placed in nylon bags (7 x 14 cm) and rumen incubated during 3, 6, 12, 24, 48, 96 and 120 hours. The IVDMD and the CP, NDF and ADF content were, respectively, 55.8, 7.6, 81.9 e 43.6%, for the clipped grass and .66.5, 12.1, 78.8 e 39.5%, for the extrusa samples. The potential degradability of DM, C P, NDF and ADF were 62.59, 80.88, 50.73 and 46.65%, for clipped grass; and 79.53, 90.97, 71.21 and 65.68%, for extrusa samples. The quality of the selected animal diet (extrusa) was better than the available forage in terms of IVDMD and chemical composition (high protein and low fiber content). In situ degradability trials carried out with clipped samples, and non selected by animal, could not supply reliable results closed to the animal diet.

Testicular biopsy has been a complementary technique for clinical and research purposes to evaluate reproductive function in males. However, hemorrhage, inflammation, degeneration, and adhesion are factors that might limit the use of this procedure. In order to minimize these potential problems, fibrin glue derived from snake venom, a tissue adhesive with sealing, hemostatic, and healing properties, was used in conjunction with bilateral testicular biopsy with the Tru-Cut needle and was compared with a more conventional technique that uses nylon suture. Thirty mature rams were randomly assigned to three groups of 10 animals each, as follows: nonsurgical control group (no scrotal surgery, or biopsy); biopsy + glue group (fibrin glue on puncture sites and skin incisions) and biopsy + suture group (compression with swab on puncture sites and suturing of skin incision). The surgeries of the rams in the biopsy groups were performed on the same day, which was designated Day 0 for all three groups. Data of scrotal circumference, number of spermatozoa per ejaculum, percentage of morphologically abnormal spermatozoa, spermatozoa motility, and serum testosterone concentrations from Days -7, 20, 40, 60, 80 and 100 were evaluated. There were no significant differences between groups within days for any of the parameters evaluated. In conclusion, the testicular biopsy procedure using the Tru-Cut needle in conjunction with conventional nylon suture or the more novel fibrin glue in rams did not affect any of the parameters of testicular function evaluated in this study and was shown to be relatively simple, safe and efficient.
