Polycomb Repressive Complex 2 (PRC2) represses the transcriptional activity of target genes through trimethylation of lysine 27 of histone H3. The functions of plant PRC2 have been chiefly described in Arabidopsis, but specific functions in other plant species, especially cereals, are still largely unknown. Here we characterize mutants in the rice EMF2B gene, an ortholog of the Arabidopsis EMBRYONIC FLOWER2 (EMF2) gene. Loss of EMF2B in rice results in complete sterility, and mutant flowers have severe floral organ defects and indeterminacy that resemble loss-of-function mutants in E-function floral organ specification genes. Transcriptome analysis identified the E-function genes OsMADS1, OsMADS6 and OsMADS34 as differentially expressed in the emf2b mutant compared with wild type. OsMADS1 and OsMADS6, known to be required for meristem determinacy in rice, have reduced expression in the emf2b mutant, whereas OsMADS34 which interacts genetically with OsMADS1 was ectopically expressed. Chromatin immunoprecipitation for H3K27me3 followed by quantitative (q)RT-PCR showed that all three genes are presumptive targets of PRC2 in the meristem. Therefore, in rice, and possibly other cereals, PRC2 appears to play a major role in floral meristem determinacy through modulation of the expression of E-function genes.

Small heat shock proteins (sHSPs) are a family of ATP-independent molecular chaperones which prevent cellular protein aggregation by binding to misfolded proteins. sHSPs form large oligomers that undergo drastic rearrangement/dissociation in order to execute their chaperone activity in protecting substrates from stress. Substrate-binding sites on sHSPs have been predominantly mapped on their intrinsically disordered N-terminal arms. This region is highly variable in sequence and length across species, and has been implicated in both oligomer formation and in mediating chaperone activity. Here, we present our results on the functional and structural characterization of five sHSPs in rice, each differing in their subcellular localisation, viz., cytoplasm, nucleus, chloroplast, mitochondria and peroxisome. We performed activity assays and dynamic light scattering studies to highlight differences in the chaperone activity and quaternary assembly of sHSPs targeted to various organelles. By cloning constructs that differ in the length and sequence of the tag in the N-terminal region, we have probed the sensitivity of sHSP oligomer assembly and chaperone activity to the length and amino acid composition of the N-terminus. In particular, we have shown that the incorporation of an N-terminal tag has significant consequences on sHSP quaternary structure.

Roles for the transcription factor RFL in rice axillary meristem development were studied. Its regulatory effects on LAX1, CUC1, and OsPIN3 reveal its functions in axillary meristem specification and outgrowth.Axillary meristems (AMs) are secondary shoot meristems whose outgrowth determines plant architecture. In rice, AMs form tillers, and tillering mutants reveal an interplay between transcription factors and the phytohormones auxin and strigolactone as some factors that underpin this developmental process. Previous studies showed that knockdown of the transcription factor gene RFL reduced tillering and caused a very large decrease in panicle branching. Here, the relationship between RFL, AM initiation, and outgrowth was examined. We show that RFL promotes AM specification through its effects on LAX1 and CUC genes, as their expression was modulated on RFL knockdown, on induction of RFL:GR fusion protein, and by a repressive RFL-EAR fusion protein. Further, we report reduced expression of auxin transporter genes OsPIN1 and OsPIN3 in the culm of RFL knockdown transgenic plants. Additionally, subtle change in the spatial pattern of IR4 DR5:GFP auxin reporter was observed, which hints at compromised auxin transport on RFL knockdown. The relationship between RFL, strigolactone signalling, and bud outgrowth was studied by transcript analyses and by the tillering phenotype of transgenic plants knocked down for both RFL and D3. These data suggest indirect RFL-strigolactone links that may affect tillering. Further, we show expression modulation of the auxin transporter gene OsPIN3 upon RFL:GR protein induction and by the repressive RFL-EAR protein. These modified forms of RFL had only indirect effects on OsPIN1. Together, we have found that RFL regulates the LAX1 and CUC genes during AM specification, and positively influences the outgrowth of AMs though its effects on auxin transport.

Two species of Pleurotus, Pleurotus florida and Pleurotus flabellatus were cultivated on two agro-residues (paddy straw; PS and coir pith; CP) singly as well as in combination with biogas digester residue (BDR, main feed leaf biomass). The biological efficiency, nutritional value, composition and nutrient balance (C, N and P) achieved with these substrates were studied. The most suitable substrate that produced higher yields and biological efficiency was PS mixed with BDR followed by coir pith with BDR. Addition of BDR with agro-residues could increase mushroom yield by 20-30%. The biological efficiency achieved was high for PS + BDR (231.93% for P. florida and 209.92% for P. flabellatus) and for CP + BDR (14831% for P. florida and 188.46% for P. flabellatus). The OC (organic carbon), TKN (nitrogen) and TP (phosphate) removal of the Pleurotus spp. under investigation suggests that PS with BDR is the best substrate for growing mushroom. Integrated agriculture-cum-fish farming has been practised profitably for ages in the Chinese small-scale farming system. There is a great potential for this system by utilizing the vast Nigerian flood plains (approx. 515,000 ha). Dogongari Bay in Lake Kainji Basin was identified as a suitable site for this system after some extensive fish culture trials. Polyculture of Clarias spp., Heterotis niloticus and Tilapia was proposed for integration with layers in the poultry house, 2-ha upland rain-fed rice farming and indirect cattle rearing in the 5-ha enclosure site. Cost benefit analysis showed that the system will consistently record profit as from the second year of operation. Various complex factors were identified to affect profitability of this mixed farming system. Concerted research approach is needed to fully understand the interrelationships of the various components of this integrated system. Generous funding of research activities is very crucial in this situation

This paper examines fish demand and supply and looks at on-going research works in rice-cum-fish culture in Nigeria. It examines all the pre-requisites for adopting this farming system. Economic and Financial analysis were made using experimental plots at two ecological zones of Nigeria. Details are given of a study carried out in Nigeria, to introduce the practice of fish-cum-rice culture, using Sarotherodon galilaeus. Two plots each measuring 360m super(2) were used for this study and were compared with the farmer's two plots measuring 300m super(2) and 350m super(2). The plots were modified and had two central canals. Rice seedlings were transplanted into the plots after 19 days using a planting distance of 20 x 20cm. Three rice seedlings were planted per hole, using rice variety FARO 40, and grown for 90 days. About 240 and 180 S. galilaeus fingerlings of mean weight of 30g and 26g were stocked in the two experimental plots, respectively. They were fed with pelleted feed of 25% C.P. and monitored for 100 days. A yield of 22.8kg was obtained in plot A while 15.66kg was obtained in plot B. A rice yield of 250kg (i.e 5 bags) was obtained in each of the plots. The results obtained were compared with plots with no fish

Representatives from the family of Lemnaceae (duckweed) make ideal experimental material for research into a succession of phytophysiological processes with regard to growth rate and vegetative reproduction. They are also easy to maintain in sterile cultures. Lemnaceae belong to the higher flowering plants (flowers are rarely produced), however they are distinguished by a much simplified morphological and anatomical structure. As water plants they possess the advantage, that they can be cultivated in synthetic media under laboratory conditions controlled by the application of both a known light intensity and temperature. This paper describes experimental research of growth of Lemnaceae in different conditions. Some of the variables were mineral media, illumination and aeration.

The objective of this short project progress report is to investigate the possible water quality implications of modern watercress growing practices. Chalk receiving watercourses are usually of high supply, amenity, game fishing and fish farming value. Any headwater pollution load, therefore, needs characterising and quantifying. Two sites of watercress farming were studied in 1986-87 and nutrient levels examined. Different approaches of watercress farmers in Dorset and Hampshire are summarised.

Several countries in Asia practice integrated rice-duck farming. On-farm resources such as duck manure and feed waste are not adequately used and recycled in the system. This indicates the potential for research to increase the productivity of the rice-duck system. The integration of fish and the nitrogen-fixing aquatic fern azolla show promise for increasing the production potential of the system. Fish, azolla and ducks integrated with rice farming can result in nutrient enhancement, pest control, feed supplementation and biological control. Some of the results of a case study on integrated rice-fish-azolla-duck farming systems conducted in the Philippines are presented in this paper.
