In this paper an unprecedent thermo-reversible sol-gel transition for titania nanoparticles dispersed in a solution of p-toluene sulfonic acid (PTSH) in isopropanol is reported. The sol formed by the thermo-hydrolysis at 60 degrees C of titanium tetraisopropoxide (Ti((OPr)-Pr-i)(4)) reversibly changes into a turbid gel upon cooling to room temperature. Turbidimetric measurements performed for samples containing different nominal acidity ratios (A = [PTSH]/[Ti]) have evidenced that the gel transformation temperature increases from 20 to 35 degrees C as the [PTSH]/[Ti] ratio increases from 0.2 to 2.0. SAXS results indicate that the thermo-reversible gelation is associated to a reversible aggregation of a monodisperse set of titania nanoparticles with average gyration radius of approximate to 2 nm. From the different PTSH species evidenced by Raman spectroscopy and TG/DTA of dried gels we proposed that the then-no-reversible gelation in this systems is induced by the formation of a supramolecular network, in which the protonated surface of nanoparticles is interconnected through cooperative hydrogen bonds between -SO3 groups of p-toluene sulfonic acid. We study the existence of periodic solutions in the neighbourhood of symmetric (partially) elliptic equilibria in purely reversible Hamiltonian vector fields. These are Hamiltonian vector fields with an involutory reversing symmetry R. We contrast the cases where R acts symplectically and anti-symplectically. In case R acts anti-symplectically, generically purely imaginary eigenvalues are isolated, and the equilibrium is contained in a local two-dimensional invariant manifold containing symmetric periodic solutions encircling the equilibrium point. In case R acts symplectically, generically purely imaginary eigenvalues are doubly degenerate, and the equilibrium is contained in two two-dimensional invariant manifolds containing nonsymmetric periodic solutions encircling the equilibrium point. In addition, there exists a three-dimensional invariant surface containing a two-parameter family of symmetric periodic solutions.

Antimony based glasses have been investigated for the first time regarding the possibility of holographic data storage using visible lasers sources. Changes in both refractive index and the absorption coefficient were measured using a holographic setup. The modulation of the optical constants is reversible by heat treatment. Bragg gratings were written under visible light of an Ar laser and erased thermally.

Human respiratory syncytial virus (HRSV) is the major cause of lower respiratory tract infections in children under 5 years of age and the elderly, causing annual disease outbreaks during the fall and winter. Multiple lineages of the HRSVA and HRSVB serotypes co-circulate within a single outbreak and display a strongly temporal pattern of genetic variation, with a replacement of dominant genotypes occurring during consecutive years. In the present study we utilized phylogenetic methods to detect and map sites subject to adaptive evolution in the G protein of HRSVA and HRSVB. A total of 29 and 23 amino acid sites were found to be putatively positively selected in HRSVA and HRSVB, respectively. Several of these sites defined genotypes and lineages within genotypes in both groups, and correlated well with epitopes previously described in group A. Remarkably, 18 of these positively selected tended to revert in time to a previous codon state, producing a flipflop phylogenetic pattern. Such frequent evolutionary reversals in HRSV are indicative of a combination of frequent positive selection, reflecting the changing immune status of the human population, and a limited repertoire of functionally viable amino acids at specific amino acid sites.

A difração múltipla de raios-X utilizando radiação Síncrotron foi aplicada para o estudo de cátions de metais de transição Mn3+ e Ni2+ incorporados a rede cristalina do Fosfato de Amônio Monobásico (ADP) e Fosfato de Potássio Monobásico (KDP). Em todos os diagramas Renninger obtidos para as diferentes amostras e diferentes comprimentos de onda podemos observar que as posições angulares e o número de picos não sofrem alteração. Este fato nos diz que os parâmetros da célula unitária e a simetria do cristal são praticamente os mesmos, independentemente da incorporação de cátíons Mn3+ e Ni2+. Cálculos precisos dos parâmetros da célula unitária revelam que há expansão dos parâmetros de rede a = b e contração do parâmetro de rede c do cristal de ADP dopado com Ni2+ e Mn3+. Nas medidas com ambos os comprimentos de onda no ADP:Mn o digrama Renninger apresenta picos com perfis semelhantes aos perfis dos picos nos diagrama Renninger do cristal de ADP puro. Nenhum pico extra aparece no diagrama Renninger do cristal dopado. A partir dos diagramas resultantes das medidas no cristal de ADP:Ni pode-se observar claramente: (i) alguns picos que tinham um perfil assimétrico no diagrama do cristal de ADP puro apresentam perfis quase totalmente simétricos no diagrama do cristal dopado com Ni

This work quantifies, using ADP and rating curve techniques, the instantaneous outflows at estuarine interfaces: higher to middle estuary and middle to lower estuary, in two medium-sized watersheds (72 000 and 66 000 km(2) of area, respectively), the Jaguaribe and Contas Rivers located in the northeastern (semi-arid) and eastern (tropical humid) Brazilian coasts, respectively. Results from ADP showed that the net water balances show the Contas River as a net water exporter, whereas the Jaguaribe River Estuary is a net water importer. At the Jaguaribe Estuary, water retention during flood tide contributes to 58% of the total volume transferred during the ebb tide from the middle to lower estuary. However, 42% of the total water volume (452 m(3) s(-1)) that entered during flood tide is retained in the middle estuary. In the Contas River, 90% of the total water is retained during the flood tide contributing to the volume transported in the ebb tide from the middle to the lower estuary. Outflows obtained with the rating curve method for the Contas and Jaguaribe Rivers were uniform through time due to river flow normalization by dams in both basins. Estimated outflows with this method are about 65% (Contas) and 95% (Jaguaribe) lower compared to outflows obtained with ADP. This suggests that the outflows obtained with the rating curve method underestimate the net water balance in both systems, particularly in the Jaguaribe River under a semi-arid climate. This work is concerned with dynamical systems in presence of symmetries and reversing symmetries. We describe a construction process of subspaces that are invariant by linear Gamma-reversible-equivariant mappings, where Gamma is the compact Lie group of all the symmetries and reversing symmetries of such systems. These subspaces are the sigma-isotypic components, first introduced by Lamb and Roberts in (1999) [10] and that correspond to the isotypic components for purely equivariant systems. In addition, by representation theory methods derived from the topological structure of the group Gamma, two algebraic formulae are established for the computation of the sigma-index of a closed subgroup of Gamma. The results obtained here are to be applied to general reversible-equivariant systems, but are of particular interest for the more subtle of the two possible cases, namely the non-self-dual case. A gene encoding a-L-arabinofuranosidase (abfA) from Aspergillus niveus was identified, cloned, and successfully expressed in Aspergillus nidulans. Based on amino acid sequence comparison, the 88.6 kDa enzyme could be assigned to the GH family 51. The characterization of the purified recombinant AbfA revealed that the enzyme was active at a limited pH range (pH 4.0-5.0) and an optimum temperature of 70 degrees C. The AbfA was able to hydrolyze arabinoxylan, xylan from birchwood, debranched arabinan, and 4-nitrophenyl arabinofuranoside. Synergistic reactions using both AbfA and endoxylanase were also assessed. The highest degree of synergy was obtained after the sequential treatment of the substrate with endoxylanase, followed by AbfA, which was observed to release noticeably more reducing sugars than that of either enzyme acting individually. The immobilization of AbfA was performed via ionic adsorption onto various supports: agarose activated by polyethyleneimine polymers, cyanogen bromide activated Sepharose, DEAE-Sepharose, and Sepharose-Q The Sepharose-Q derivative remained fully active at pH 5 after 360 min at 60 degrees C, whereas the free AbfA was inactivated after 60 min. A synergistic effect of arabinoxylan hydrolysis by AbfA immobilized in Sepharose-Q and endoxylanase immobilized in glyoxyl agarose was also observed. The stabilization of arabinofuranosidases using immobilization tools is a novel and interesting topic. (C) 2012 Elsevier Ltd. All rights reserved.

Monomers based on plant oil derivatives bearing furan heterocycles appended through thiol-ene click chemistry were prepared and, subsequently, polymerized via a second type of click reaction, i. e. the Diels-Alder (DA) polycondensation between furan and maleimide complementary moieties. Two basic approaches were considered for these DA polymerizations, namely (i) the use of monomers with two terminal furan rings in conjunction with bismaleimides (AA + BB systems) and (ii) the use of a protected AB monomer incorporating both furan and maleimide end groups. This study clearly showed that both strategies were successful, albeit with different outcomes, in terms of the nature of the ensuing products. The application of the retro-DA reaction to these polymers confirmed their thermoreversible character, i. e. the clean-cut return to their respective starting monomers, opening the way to original macromolecular materials with interesting applications, like mendability and recyclability.

Air conditioning and lighting costs can be reduced substantially by changing the optical properties of "intelligent windows." The electrochromic devices studied to date have used copper as an additive. Copper, used here as an electrochromic material, was dissolved in an aqueous animal protein-derived gel electrolyte. This combination constitutes the electrochromic system for reversible electrodeposition. Cyclic voltammetry, chronoamperometric and chromogenic analyses indicated that were obtained good conditions of transparency (initial transmittance of 70%), optical reversibility, small potential window (2.1 V), variation of transmittance in visible light (63.6%) and near infrared (20%) spectral regions. Permanence in the darkened state was achieved by maintaining a lower pulse potential (-0.16 V) than the deposition potential (-1.0 V). Increasing the number of deposition and dissolution cycles favored the transmittance and photoelectrochemical reversibility of the device. The conductivity of the electrolyte (10(-3) S/cm) at several concentrations of CuCl2 was determined by electrochemical impedance spectroscopy. A thermogravimetric analysis confirmed the good thermal stability of the electrolyte, since the mass loss detected up to 100 degrees C corresponded to water evaporation and decomposition of the gel started only at 200 degrees C. Micrographic and small angle X-ray scattering analyses indicated the formation of a persistent deposit of copper particles on the ITO. (C) 2012 Elsevier B.V. All rights reserved.

We study a probabilistic model of interacting spins indexed by elements of a finite subset of the d-dimensional integer lattice, da parts per thousand yen1. Conditions of time reversibility are examined. It is shown that the model equilibrium distribution converges to a limit distribution as the indexing set expands to the whole lattice. The occupied site percolation problem is solved for the limit distribution. Two models with similar dynamics are also discussed.

Nel presente lavoro di tesi sono state messe a confronto le ATP sintasi wild-type e γM23-K in cromatofori del batterio fotosintetico Rhodobacter capsulatus sotto gli aspetti funzionale e regolatorio. Si pensava inizialmente che la mutazione, in base a studi riportati in letteratura condotti sull’omologa mutazione in E. coli, avrebbe indotto disaccoppiamento intrinseco nell’enzima. Il presente lavoro ha chiarito che il principale effetto della mutazione è un significativo aumento dell’affinità dell’enzima per l’ADP inibitorio, che ne determina il ridotto livello di ATP idrolisi e la rapidissima reinattivazione in seguito ad attivazione da forza protonmotiva. Il residuo 23 della subunità γ si trova posizionato in prossimità della regione conservata DEELSED carica negativamente della subunità β, e l’introduzione nel mutante di una ulteriore carica positiva potrebbe determinare una maggiore richiesta di energia per indurre l’apertura del sito catalitico. Un’analisi quantitativa dei dati di proton pumping condotta mediante inibizione parziale dell’idrolisi del wildtype ha inoltre mostrato come il grado di accoppiamento del mutante in condizioni standard non differisca sostanzialmente da quello del wild-type. D’altro canto, è stato recentemente osservato come un disaccoppiamento intrinseco possa venire osservato in condizioni opportune anche nel wild-type, e cioè a basse concentrazioni di ADP e Pi. Nel presente lavoro di tesi si è dimostrato come nel mutante l’osservazione del fenomeno del disaccoppiamento intrinseco sia facilitata rispetto al wild-type. È stato proprio nell’ambito delle misure condotte sul mutante che è stato possibile dimostrare per la prima volta il ruolo fondamentale della componente elettrica della forza protonmotiva nel mantenere lo stato enzimatico ad elevato accoppiamento. Tale ruolo è stato successivamente messo in luce anche nel wild-type, in parte anche grazie all’uso di inibitori specifici di F1 e di FO. Il disaccoppiamento intrinseco nel wild-type è stato ulteriormente esaminato anche nella sua dipendenza dalla rimozione di ADP e Pi; in particolare, oltre all’amina fluorescente ACMA, è stata utilizzata come sonda di ΔpH anche la 9-aminoacridina e come sonda di Δψ l’Oxonolo VI. In entrambi i casi il ruolo accoppiante di questi due ligandi è stato confermato, inoltre utilizzando la 9-aminoacridina è stato possibile calibrare il segnale di fluorescenza con salti acido-base, dando quindi una base quantitativa ai dati ottenuti. Noi riteniamo che il più probabile candidato strutturale coinvolto in questi cambiamenti di stato enzimatici sia la subunità ε, di cui è noto il coinvolgimento in processi di regolazione e in cambiamenti strutturali indotti da nucleotidi e dalla forza protonmotiva. In collaborazione con il Dipartimento di Chimica Fisica dell’Università di Friburgo è in atto un progetto per studiare i cambiamenti strutturali presumibilmente associati al disaccoppiamento intrinseco tramite FRET in singola molecola di complessi ATP-sintasici marcati con fluorofori sia sulla subunità ε che sulla subunità γ. Nell’ambito di questa tesi sono stati creati a questo fine alcuni doppi mutanti cisteinici ed è stato messo a punto un protocollo per la loro marcatura con sonde fluorescenti.

Gegenstand dieser Arbeit war die Untersuchung der Bedeutung der Poly(ADP-Ribose)-Polymerase 1 (PARP 1), der AP Endonuklease 1 (Ape 1) und des Xeroderma pigmentosum A (XPA) Proteins für die DNA-Reparatur in Säugerzellen.Zunächst wurde der Einfluss der PARP 1-Aktivität auf die Reparatur verschiedener DNA-Modifikationen untersucht. Die Ergebnisse zeigen erstmalig, dass eine Hemmung der PARP-Aktivität nicht nur eine deutliche Verlangsamung der Reparatur von Einzelstrangbrüchen, sondern auch von oxidativen Purinmodifikationen und Pyrimidindimeren zur Folge hat. Interessanterweise erfolgte diese Verlangsamung der DNA-Reparatur nicht in Csb-defizienten Zellen. Diese Ergebnisse deuten darauf hin, dass die Aktivierung der PARP 1 und das Csb-Protein zusammen an einem neuartigen Mechanismus beteiligt sind, der die globale Reparatur verschiedener DNA-Modifikationen beschleunigt.Weiterhin wurde die Bedeutung der Nukleotidexcisionsreparatur als back-up Reparatur von 8 Hydroxyguanin untersucht. Dazu wurden normale und XPA-defiziente Fibroblasten des Menschen mit einem hOgg1-anitsense Konstrukt transfiziert und dann in diesen Zellen die Reparaturkinetiken oxidativer Basenmodifikationen bestimmt. Dadurch konnte eine Beteiligung des XPA-Proteins an diesem Reparaturweg ausgeschlossen werden.Außerdem wurden die Auswirkungen einer AP Endonuklease-1-Überexpression in XRCC1-defizienten Zellen auf die Reparatur von Einzelstrangbrüchen untersucht. Die Reparatur der induzierten Einzelstrangbrüche war in XRCC1-defizienten Zellen erwartungsgemäß deutlich langsamer als in XRCC1-profizienten Zellen. Die Überexpression der AP Endonuklease 1 in XRCC1-defizienten Zellen führte zu einer teilweisen Beschleunigung der Einzelstrangbruchreparatur.
