Although several alleles of susceptibility to Alzheimer’s disease (AD) have been studied in the last decades, few polymorphisms have been considered as risk factors for the disease. Among them, the APOE-e4 allele appears to be the major genetic risk factor for the onset of the disease. However, it is important to confirm the potential susceptibility of these genetic variants in different populations in order to establish a genetic profile for the disease in specific communities. This study analyzed the APOE polymorphisms regarding susceptibility to AD in a sample of 264 individuals (primarily Caucasians; 82 cases and 182 controls) in the population from Vitória, ES, Brazil, by PCR restriction fragment length polymorphism (PCR-RFLP) methods. The patients were selected according to clinical criteria for probable AD. Whereas the e4 allele showed statistically significant positive association with susceptibility to AD (OR = 3.01, 95%CI = 1.96-4.61; P < 0.0001), the e2 allele did not. The results of the e4 allele confirm the role of this polymorphism as a risk factor for AD in the sample studied as observed in other populations. Although the e3 allele has been considered neutral in several studies, our results suggest that it acts as a protective factor against AD in the population studied (OR = 0.46, 95%CI = 0.30-0.67; P < 0.0001). This study may provide a new insight into the role of the APOE-e3 allele in the etiology of AD and might help to estabilish a profile of risk for AD in the population from Vitória, ES.

Agmatine, an endogenous polyamine and putative neuromodulator, is known to have neuroprotective effects on various neurons in the central nervous system. We determined whether or not topically administered agmatine could reduce ischemic retinal injury. Transient ocular ischemia was achieved by intraluminal occlusion of the middle cerebral artery of ddY mice (30-35 g) for 2 h, which is known to also induce occlusion of the ophthalmic artery. In the agmatine group (N = 6), a 1.0 mM agmatine-containing ophthalmic solution was administered four times daily for 2 weeks before occlusion. In the control group (N = 6), a 0.1% hyaluronic acid ophthalmic solution was instilled at the same times. At 22 h after reperfusion, the eyeballs were enucleated and the retinal sections were stained by terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL). Transient ocular ischemia induced apoptosis of retinal cells in the entire retinal layer, and topically administered agmatine can significantly reduce this ischemic retinal injury. The proportion of apoptotic cells was definitely decreased (P < 0.001; Kruskal-Wallis test). Overall, we determined that topical agmatine application effectively decreases retinal damage in an in vivo ocular ischemic injury model. This implies that agmatine is a good candidate as a direct neuroprotective agent for eyes with ocular ischemic diseases.

We aimed to investigate the effects of an anti-tumor necrosis factor-α antibody (ATNF) on cartilage and subchondral bone in a rat model of osteoarthritis. Twenty-four rats were randomly divided into three groups: sham-operated group (n=8); anterior cruciate ligament transection (ACLT)+normal saline (NS) group (n=8); and ACLT+ATNF group (n=8). The rats in the ACLT+ATNF group received subcutaneous injections of ATNF (20 μg/kg) for 12 weeks, while those in the ACLT+NS group received NS at the same dose for 12 weeks. All rats were euthanized at 12 weeks after surgery and specimens from the affected knees were harvested. Hematoxylin and eosin staining, Masson's trichrome staining, and Mankin score assessment were carried out to evaluate the cartilage status and cartilage matrix degradation. Matrix metalloproteinase (MMP)-13 immunohistochemistry was performed to assess the cartilage molecular metabolism. Bone histomorphometry was used to observe the subchondral trabecular microstructure. Compared with the rats in the ACLT+NS group, histological and Mankin score analyses showed that ATNF treatment reduced the severity of the cartilage lesions and led to a lower Mankin score. Immunohistochemical and histomorphometric analyses revealed that ATNF treatment reduced the ACLT-induced destruction of the subchondral trabecular microstructure, and decreased MMP-13 expression. ATNF treatment may delay degradation of the extracellular matrix via a decrease in MMP-13 expression. ATNF treatment probably protects articular cartilage by improving the structure of the subchondral bone and reducing the degradation of the cartilage matrix.

In this work, through the use of thermal analysis techniques, the thermal stabilities of some antioxidants were investigated, in order to evaluate their resistance to thermal oxidation in oils, by heating canola vegetable oil, and to suggest that antioxidants would be more appropriate to increase the resistance of vegetable oils in the thermal degradation process in frying. The techniques used were: Thermal Gravimetric (TG) and Differential Scanning Calorimetry (DSC) analyses, as well as an allusion to a possible protective action of the vegetable oils, based on the thermal oxidation of canola vegetable oil in the laboratory under constant heating at 180 ºC/8 hours for 10 days. The studied antioxidants were: ascorbic acid, sorbic acid, citric acid, sodium erythorbate, BHT (3,5-di-tert-butyl-4-hydroxytoluene), BHA (2, 3-tert-butyl-4-methoxyphenol), TBHQ (tertiary butyl hydroquinone), PG (propyl gallate) - described as antioxidants by ANVISA and the FDA; and also the phytic acid antioxidant and the SAIB (sucrose acetate isobutyrate) additive, which is used in the food industry, in order to test its behavior as an antioxidant in vegetable oil. The following antioxidants: citric acid, sodium erythorbate, BHA, BHT, TBHQ and sorbic acid decompose at temperatures below 180 ºC, and therefore, have little protective action in vegetable oils undergoing frying processes. The antioxidants below: phytic acid, ascorbic acid and PG, are the most resistant and begin their decomposition processes at temperatures between 180 and 200 ºC. The thermal analytical techniques have also shown that the SAIB antioxidant is the most resistant to oxidative action, and it can be a useful choice in the thermal decomposition prevention of edible oils, improving stability regarding oxidative processes.

Covering the grapevine rows to delay the maturity and harvest date became widely practiced in 'Sultana Seedless' vineyards. The research work was conducted to test different cover materials (polypropylene cross-stitch, life pack, mogul and transparent polyethylene) in respect to their effects on grape quality and storability. Harvest was delayed for one month in covered plots. Harvested grapes were packed and transferred to storage rooms after pre-cooling. During packing, the grape clusters were sealed in PE bags with sulphur dioxide pads. The grapes were stored for 90 days in the first year and 120 days in the second year, at -0.5ºC and 90% RH. All the grape clusters were healthy and of marketable quality after 90 days of storage period. In the first year, at the end of the storage, only those grapes harvested from the rows covered with polypropylene cross-stitch showed fungal growth. The sensory quality scores revealed a lower level of preference after 120 days of storage. The effects of the covering materials tested were similar regarding grape quality and storage performance except the transparent polyethylene that damaged the grapevine leaves.

Resveratrol (RESV) is a polyphenolic compound found in various plants, including grapes, berries and peanuts, and its processed foods as red wine. RESV possesses a variety of bioactivities, including antioxidant, anti-inflammatory, cardioprotective, antidiabetic, anticancer, chemopreventive, neuroprotective, renal lipotoxicity preventative, and renal protective effects. Numerous studies have demonstrated that polyphenols promote cardiovascular health. Furthermore, RESV can ameliorate several types of renal injury in animal models, including diabetic nephropathy, hyperuricemic, drug-induced injury, aldosterone-induced injury, ischemia-reperfusion injury, sepsis-related injury, and endothelial dysfunction. In addition, RESV can prevent the increase in vasoconstrictors, such as angiotensin II (AII) and endothelin-1 (ET-1), as well as intracellular calcium, in mesangial cells. Together, these findings suggest a potential role for RESV as a supplemental therapy for the prevention of renal injury.

Older age increases the risk of developing a chronic atherosclerotic cardiovascular disease (CVD), such as coronary heart disease. Complications of CVDs, myocardial infarction or stroke often lead to loss of functional capacity or premature death. Dyslipidemia, high serum levels of total or low-density lipoprotein cholesterol (LDL-c) and low levels of high-density lipoprotein cholesterol (HDL-c), is among the most important modifiable risk factors for CVDs; it can be treated with lifestyle modifications, and with lipid-lowering drugs, primarily statins. In older persons, however, the association of cholesterol levels with cardiovascular and all-cause mortality has been inconsistent in previous studies. Furthermore, the beneficial effects of statins in older persons without previous CVD are still somewhat unclear, and older persons are more prone to adverse effects from statins. This thesis presents a prospective cohort study (TUVA), exploring associations of cholesterol levels with mortality and the changes in cholesterol levels of a 70-year-old population in long-term follow-up. Further, prevalence of CVDs, risk factors and preventive medication use in the TUVA cohort is compared with respective prevalences in another age-matched cohort (UTUVA) 20 years later in order to examine the changes in cardiovascular risk over time. Additionally, to evaluate statin use patterns among older persons, an observational register study was conducted covering the total Finnish population aged 70 and older during 2000-2008. Based on individual-level data retrieved from national health registries, the population was classified into low, moderate and high risk groups according to estimated CVD risk. The prevalence, incidence and persistence of statin use among the risk groups was then evaluated based upon yearly statin purchases tracked from the Prescription Register. The prospective cohort study demonstrated that low total cholesterol, LDL-c and HDL-c were associated with higher mortality in a cohort of home-dwelling 70-year-olds. However, after adjusting for traditional cardiovascular risk factors and cancer this association disappeared. Further, low total cholesterol seemed to be protective, whereas low HDL-c strongly predicted increased risk of CVD death. Cholesterol levels of those elderly who remained available for follow-up and were still home-dwelling at the age of 85 seemed to improve with advancing age. Compared to the TUVA cohort, the later born UTUVA cohort had less CVDs and their risk factors were better controlled, which was reflected in the higher use of preventive medications such as statins and antihypertensives. The register studies confirmed that statin use has increased significantly during 2000-2008 among older persons, especially among the oldest (80+) age groups and among those at high risk for cardiovascular events. Two-thirds of new statin users persisted with their use during the four years of follow-up; the most discontinuations were made during the first year of use. In conclusion, statins are commonly used among older age groups in Finland. Most of the older statin users had a high cardiovascular event risk, indicating that the treatment is well directed towards those who are likely to benefit from it the most. No age-limits should be put on the screening and treatment of dyslipidemia in older persons, but the benefits and adverse effects of statin treatment should be carefully weighed based on an individual assessment of the person’s general health status and functional capacity. Physicians should pay more attention to medication adherence, especially when prescribing preventive medications.

Le cancer du sein (CS) est la deuxième cause de décès liés au cancer parmi les femmes dans la plupart des pays industrialisés. Les personnes qui ont le CS peuvent ne pas hériter des mutations causant le cancer de leurs parents. Ainsi, certaines cellules subissent des mutations qui mènent au cancer. Dans le cas de cancer héréditaire, les cellules tumorales contiennent généralement des mutations qui ne sont pas trouvées ailleurs dans l'organisme, mais peuvent maintenir des mutations qui vont répartir dans toutes les cellules. La genèse du CS est le résultat des mutations de gènes qui assurent la régulation de la prolifération cellulaire et la réparation de l’ADN. Deux gènes semblent particulièrement concernés par les mutations. Les gènes ‘Breast Cancer 1’ (BRCA1) et ‘Breast Cancer 2’ (BRCA2), sont impliqués dans la prédisposition génétique de CS. On estime que 5-10% des cas de cancer du sein sont attribuables à une prédisposition génétique. La plupart de ces cancers sont liés à une anomalie du gène BRCA1 ou BRCA2. Plusieurs études ont été menées chez les femmes atteintes de CS sporadique et quelques études se sont concentrées sur celles qui sont porteuses de mutations de BRCA. Alors, notre recherche a été entreprise afin de vérifier l’hypothèse d’une association entre le CS, le mode vie et les habitudes alimentaires chez les Canadiennes-françaises non porteuses des 6 mutations de BRCA les plus fréquentes parmi cette population. Nous avons mené une étude cas-témoins dans cette population. Quelque 280 femmes atteintes du cancer du sein et non-porteuses de mutations de BRCA, ont été recrutées en tant que cas. Les témoins étaient recrutés parmi les membres de la famille des cas (n=15) ou à partir d'autres familles atteintes de CS (n=265). Les participantes étaient de tous âges, recrutées à partir d’une étude de cohorte qui est actuellement en cours, menée par une équipe de chercheurs au Centre Hospitalier Universitaire de Montréal (CHUM) Hôtel-Dieu à Montréal. Les apports alimentaires ont été recueillis par un questionnaire de fréquence semi-quantitatif validé et administré par une nutritionniste, qui portait sur la période avant les deux ans précédant le premier diagnostic de CS pour les cas et la période avant les deux ans précédant l’entrevue téléphonique pour les témoins. Un questionnaire de base était administré par l’infirmière de recherche aux participantes afin de colliger des renseignements sociodémographiques et sur les facteurs de risque du CS. Une association positive et significative a été détectée entre l’âge (plus de 50 ans) auquel les sujets avaient atteint leur Indice de Masse Corporel (IMC) le plus élevé et le CS rapport de cotes (OR) =2,83; intervalle de confiance à 95% (IC95%) (2,34-2,91). De plus, une association positive a été détectée entre un gain de poids de >34 lbs comparativement à un gain de poids de ≤15 lbs, dès l’âge de 20 ans OR=1,68; IC95% (1,10-2,58). Un gain de poids de >24 lbs comparativement à un gain de poids de ≤9 lbs, dès l’âge de 30 ans a aussi montré une augmentation de risque de CS OR=1,96; IC95% (1,46-3,06). Une association positive a aussi été détecté entre, un gain de poids de >12 lbs comparativement à un gain de poids de ≤1 lb, dès l’âge de 40 ans OR=1,91; IC95% (1,53-2,66). Concernant le tabagisme, nous avons observé une association positive et significative reliée à la consommation de plus de 9 paquets-années OR = 1,59; IC95% (1,57-2,87). Il fut suggéré que l’activité physique modéré confère une protection contre le CS: une pratique de > 24,8 (‘metabolic equivalent’) MET-hrs par semaine par rapport à ≤10,7 MET-hrs par semaine, diminue le risque du CS de 52% OR = 0,48 ; IC95% (0,31-0,74). L’activité physique totale (entre 16,2 et 33,2 MET-hrs par semaine), a aussi montré une réduction de risque de CS de 43% OR = 0,57 ; IC95% (0,37-0,87). Toutefois, il n'y avait aucune association entre une activité physique vigoureuse et le risque de CS. L’analyse portant sur les macro- et micro-nutriments et les groupes alimentaires a montré qu’un apport en énergie totale de plus de 2057 Kcal par jour augmentait le risque de CS de 2,5 fois OR = 2,54; IC95% (1,67-3,84). En ce qui concerne la consommation de café, les participantes qui buvaient plus de 8 tasses de café par jour avaient un risque de CS augmenté de 40% OR = 1,40; IC95% (1,09-2,24). Les sujets ayant une consommation dépassant 9 g d’alcool (éthanol) par jour avaient également un risque élevé de 55% OR = 1,55; IC95% (1,02-2,37). De plus, une association positive et significative a été détectée entre le CS et la consommation de plus de deux bouteilles de bière par semaine OR = 1,34; IC95% (1,28-2,11), 10 onces de vin par semaine OR = 1,16; IC95% (1,08-2,58) ou 6 onces de spiritueux par semaine OR = 1,09; IC95% (1,02-2,08), respectivement. En résumé, les résultats de cette recherche supportent l’hypothèse selon laquelle le mode de vie et les habitudes alimentaires jouent un rôle important dans l’étiologie de CS chez les Canadiennes-françaises non porteuses de mutations de BRCA. Les résultats nous permettent de constater que le gain de poids et le tabagisme sont liés à des risques élevés de CS, tandis que l'activité physique modérée aide à réduire ce risque. De plus, nos résultats suggèrent qu’un apport énergétique total relativement élevé et une consommation élevée de café et d'alcool peuvent accroître le risque de ce cancer. Ce travail a permis de mettre l’accent sur une nouvelle direction de recherche, jusqu'à présent non investiguée. Les résultats de ce travail de recherche pourraient contribuer à recueillir de nouvelles informations et des conseils pouvant influencer et aider la population à modifier son mode de vie et ses habitudes alimentaires afin de diminuer le risque de cancer du sein.

BACKGROUND: The secretory basic amino acid-specific proprotein convertases (PCs) have often been associated with cancer/metastasis. By controlling the cleavage of cancer-associated proteins, PCs play key roles in multiple steps of cancer development. Most analyses of the implication of PCs in cancer/metastasis relied on the use of in vitro overexpression systems or inhibitors that can affect more than one PC. Aside from the role of furin in salivary gland tumorigenesis, no other in vivo genetic model of PC-knockout was reported in relation to cancer development. RESULTS: Since PC5/6 is highly expressed in the small intestine, the present study examined its in vivo role in intestinal tumorigenesis. Analysis of human intestinal tumors at various stages showed a systematic down-regulation of PC5/6 expression. Since gene inactivation of PC5/6 leads to lethality at birth, we generated mice lacking PC5/6 in enterocytes and analyzed the impact of the presence or absence of this PC in the mouse ApcMin/+ model that develops numerous adenocarcinomas along the intestinal tract. This resulted in viable mice with almost no expression of PC5/6 in small intestine, but with no overt phenotype. The data showed that by themselves ApcMin/+ tumors express lower levels of PC5/6 mRNA, and that the lack of PC5/6 in enterocytes results in a significantly higher tumor number in the duodenum, with a similar trend in other intestinal segments. Finally, the absence of PC5/6 is also associated with a premature mortality of ApcMin/+ mice. CONCLUSION: Overall, these data suggest that intestinal PC5/6 is protective towards tumorigenesis, especially in mouse duodenum, and possibly in human colon.

This thesis follows and analyses the involvement of both social actors and the state in the industrial transformation of Mexico. While these actors have been key associates in the development of the Mexican economy for decades, with the 1982 debt crisis, and especially after the promotion of a structural reform in 1986, the patterns of their involvement changed. However, this change was more limited than expected, and the state did not abandon its participation in some key industrial sectors. This thesis will demonstrate that the role of the state remained important during the process of change towards an Export-Oriented developmental paradigm, although the state continued to participate in fashioning social relations, it did so in new ways that can be termed “protective liberalization.”

Le cancer du sein est une maladie multifactorielle, plusieurs facteurs socio-économiques, alimentaires ainsi que le style de vie ayant été incriminés dans son développement. Une mutation germinale sur un des gènes BRCA1 ou BRCA2 serait responsable d’une augmentation du risque de développer un cancer du sein de 50 à 80% chez les femmes porteuses d’une mutation sur BRCA1 ou BRCA2 par comparaison aux non- porteuses. Plusieurs études rapportant l’existence d’une association entre la fréquence des cancers du sein sporadiques, les habitudes alimentaires et le style de vie des femmes atteintes, ceci, nous a amené à nous interroger sur le rôle que pourrait jouer ces mêmes facteurs chez les femmes porteuses d’une mutation sur BRCA1 ou BRCA2. Pour répondre à cette question, nous avons effectué une étude cas-témoin. Nos quarante-quatre cas sont porteuses d’une mutation germinale parmi 6 des 14 mutations fondatrices sur BRCA1 et BRCA2 les plus fréquentes dans la population Canadienne Française. Elles sont issues d’une cohorte de plus de 1000 femmes Canadiennes Françaises atteintes de cancers du sein recrutées depuis 1994 et testées pour ces 6 mutations. Les témoins sont recrutés parmi des femmes de la population Canadienne Française, également porteuses d’une de ces six mutations fondatrices de BRCA1 ou BRCA2, mais indemnes de cancers. Quinze d’entre elles ont été recrutées dans les familles des cas de l’étude initiale. Vingt-neuf ont été recrutées à la clinique des cancers familiaux du CHUM, nous permettant ainsi de totaliser 44 témoins. Deux questionnaires ont été administrés aux cas et aux témoins. Le premier, dit Questionnaire de base, a servi à recenser les informations sociodémographiques et le style de vie, couvrant ainsi les deux années précédant la découverte du cancer du sein pour les cas et les deux années précédant la découverte de la mutation pour les témoins. Le deuxième questionnaire, Questionnaire de nutrition, a permis de colliger les informations sur les habitudes alimentaires durant la même période de recueil de données. Une association positive et significative entre le risque de cancer du sein et le niveau d’éducation a été observé parmi les sujets de niveau universitaire (>14 années d’étude) comparés aux sujets n’ayant pas dépassé le niveau d’études secondaires (<11 années d’études) [OR= 7,82; IC95% : (1,99-30,69); p=0,003]. Nous avons mis en évidence que le risque de cancer du sein augmentait lorsque les sujets atteignaient leur poids maximum à un âge avancé > 48 ans [OR = 4,27 ; IC 95% : (0,82-22,25)]. Nous avons montré que le risque du cancer du sein diminuait pour une durée d’allaitement supérieure à 7 mois par comparaison aux femmes n’ayant jamais allaité [OR= 0,35; IC 95% : (0,12-1,06)] mais cette association est non significative. Les porteuses qui pratiquent plus de 22,45 Met-h-sem d’activité physique modérée, comparativement à celles qui pratiquent moins de 11,45 Met-h-sem voient leur risque de cancer du sein diminué de 72% [OR=0,28- IC 95% : (0,08-0,95); p=0,04]. Celles qui pratiquent plus de 31,95 Met-h-sem d’activité physique totale comparativement à celles qui pratiquent moins de 16,40 Met-h-sem voient leur risque de cancer du sein réduit de 79 % [OR=0,21; IC 95% : (0,06-0,75); p= 0,02]. L’analyse des macro et micronutriments et des groupes alimentaires a démontré qu’une consommation de plus de 23,20 g/j d’acide gras monoinsaturés est responsable d’une augmentation du risque de cancer du sein de 6 fois par comparaison à une consommation inférieure à 17,08 g/j [OR=6,00; IC 95% : (0,97-37,02); p=0,05]. Une consommation de plus de 221,79 µg/j de vitamine K réduit le risque du cancer du sein de 83 % par comparaison à une consommation inférieure à 143,57 µg/j [OR= 0,17; IC95% : (0,05-0,61) ; p=0,007]. La consommation de fruits est associée à une réduction du risque de cancer du sein de 73% chez les porteuses de mutations qui en consomment plus de 563,31 g/j comparée à celles qui en consomment moins de 356,18 g/j [OR= 0,27; IC 95% : (0,07-1,01) ; p=0,05]. Nos résultats confortent l’hypothèse selon laquelle le style de vie et les habitudes alimentaires jouent un rôle dans le développement du cancer du sein chez les Canadiennes Françaises porteuses de mutations d’une des 6 mutations fondatrices de BRCA1 ou 2 étudiées. En effet, un niveau d’éducation élevé, un gain de poids sont associés à un risque élevé de développer un cancer du sein. De plus la pratique de l’allaitement et d’une activité physique modérée sont associées à une réduction de ce risque. Nous montrons aussi que la consommation d’acides gras monoinsaturés est responsable d’une augmentation du risque de ce cancer et que la consommation de vitamine K et de fruits permet de réduire ce risque. Nos résultats ouvrent une nouvelle voie de recherche par rapport au rôle de certains nutriments dans le développement du cancer du sein chez les porteuses de mutation d’un des gènes BRCA. Cette voie pourrait également être explorée chez les non porteuses.

The principal cause of mortality in patients with acute liver failure (ALF) is brain herniation resulting from intracranial hypertension caused by a progressive increase of brain water. In the present study, ex vivo high-resolution 1H-NMR spectroscopy was used to investigate the effects of ALF, with or without superimposed hypothermia, on brain organic osmolyte concentrations in relation to the severity of encephalopathy and brain edema in rats with ALF due to hepatic devascularization. In normothermic ALF rats, glutamine concentrations in frontal cortex increased more than fourfold at precoma stages, i.e. prior to the onset of severe encephalopathy, but showed no further increase at coma stages. In parallel with glutamine accumulation, the brain organic osmolytes myo-inositol and taurine were significantly decreased in frontal cortex to 63\% and 67\% of control values, respectively, at precoma stages (p<0.01), and to 58\% and 67\%, respectively, at coma stages of encephalopathy (p<0.01). Hypothermia, which prevented brain edema and encephalopathy in ALF rats, significantly attenuated the depletion of myo-inositol and taurine. Brain glutamine concentrations, on the other hand, did not respond to hypothermia. These findings demonstrate that experimental ALF results in selective changes in brain organic osmolytes as a function of the degree of encephalopathy which are associated with brain edema, and provides a further rationale for the continued use of hypothermia in the management of this condition.
