Background: The main goal of the present study was to analyse the genetic architecture of mRNA expression in muscle, a tissue with an outmost economic importance for pig breeders. Previous studies have used F2 crosses to detect porcine expression QTL (eQTL), so they contributed with data that mostly represents the between-breed component of eQTL variation. Herewith, we have analysed eQTL segregation in an outbred Duroc population using two groups of animals with divergent fatness profiles. This approach is particularly suitable to analyse the within-breed component of eQTL variation, with a special emphasis on loci involved in lipid metabolism. Methodology/Principal Findings: GeneChip Porcine Genome arrays (Affymetrix) were used to determine the mRNA expression levels of gluteus medius samples from 105 Duroc barrows. A whole-genome eQTL scan was carried out with a panel of 116 microsatellites. Results allowed us to detect 613 genome-wide significant eQTL unevenly distributed across the pig genome. A clear predominance of trans- over cis-eQTL, was observed. Moreover, 11 trans-regulatory hotspots affecting the expression levels of four to 16 genes were identified. A Gene Ontology study showed that regulatory polymorphisms affected the expression of muscle development and lipid metabolism genes. A number of positional concordances between eQTL and lipid trait QTL were also found, whereas limited evidence of a linear relationship between muscle fat deposition and mRNA levels of eQTL regulated genes was obtained. Conclusions/Significance: Our data provide substantial evidence that there is a remarkable amount of within-breed genetic variation affecting muscle mRNA expression. Most of this variation acts in trans and influences biological processes related with muscle development, lipid deposition and energy balance. The identification of the underlying causal mutations and the ascertainment of their effects on phenotypes would allow gaining a fundamental perspective about how complex traits are built at the molecular level.

La evaluación de la sostenibilidad en modelos de producción porcina en Cataluña (España), mediante el análisis multicriterio, se estructura en dos partes. Una primera de establecimiento del marco de análisis donde, de forma participativa, se fijan los criterios de evaluación, sus indicadores y las escalas de evaluación. En la segunda se implementa la metología expuesta para evaluar la sostenibilidad de tres modelos productivos y detectar posibles conflictos entre los diferentes grupos de interés. Los resultados obtenidos demuestran que desde el punto de vista de la sostenbilidad, en base a los criterios considerados y bajo distintas opciones de robustez, el modelo productivo más sostenible es el ecológico, seguido del familiar y, en último puesto, el del modelo denominado de integración. En cuanto a la formación de coaliciones se observa que hay un importante grupo de actores que apuestan, en primer lugar, por el modelo de integración. Los anteriores resultados avalan la necesidad de potenciar modelos intermedios entre el ecológico y el de integración. Una opción conciliadora, entre las existentes, podría ser el tipo familiar, aplicando algunas mejoras en el manejo si se quieren satisfacer plenamente los requerimientos de sostenibilidad.

There is growing public concern about reducing saturated fat intake. Stearoyl-CoA desaturase (SCD) is the lipogenic enzyme responsible for the biosynthesis of oleic acid (18:1) by desaturating stearic acid (18:0). Here we describe a total of 18 mutations in the promoter and 3′ non-coding region of the pig SCD gene and provide evidence that allele T at AY487830:g.2228T>C in the promoter region enhances fat desaturation (the ratio 18:1/18:0 in muscle increases from 3.78 to 4.43 in opposite homozygotes) without affecting fat content (18:0+18:1, intramuscular fat content, and backfat thickness). No mutations that could affect the functionality of the protein were found in the coding region. First, we proved in a purebred Duroc line that the C-T-A haplotype of the 3 single nucleotide polymorphisms (SNPs) (g.2108C>T; g.2228T>C; g.2281A>G) of the promoter region was additively associated to enhanced 18:1/18:0 both in muscle and subcutaneous fat, but not in liver. We show that this association was consistent over a 10-year period of overlapping generations and, in line with these results, that the C-T-A haplotype displayed greater SCD mRNA expression in muscle. The effect of this haplotype was validated both internally, by comparing opposite homozygote siblings, and externally, by using experimental Duroc-based crossbreds. Second, the g.2281A>G and the g.2108C>T SNPs were excluded as causative mutations using new and previously published data, restricting the causality to g.2228T>C SNP, the last source of genetic variation within the haplotype. This mutation is positioned in the core sequence of several putative transcription factor binding sites, so that there are several plausible mechanisms by which allele T enhances 18:1/18:0 and, consequently, the proportion of monounsaturated to saturated fat.

Reproductive traits play a key role in pig production in order to reduce costs and increase economic returns. Among others, gene expression analyses represent a useful approach to study genetic mechanisms underlying reproductive traits in pigs. The application of reverse-transcription quantitative PCR requires the selection of appropriate reference genes, whose expression levels should not be affected by the experimental conditions, especially when comparing gene expression across different physiological stages.

Aplicació de càlcul nutricional que permet les següents funcionalitats: Gestió d’Individus/Pacients permet organitzar els pacients en carpetes i subcarpetes, podent incorporar un històric de dades antropomètriques i dietètic i calcular els següents dades: IMC (Índex de Massa Corporal), Índex Cintura / Maluc, Estima el pes ideal orientatiu, Estimació de Despesa Energètica per: Harris-Benedict, FAO-OMS, Mifflin-St. Jeor. En base a la informació introduïda a la pestanya Activitat Física estima també la despesa energètica tenint en compte l’activitat física realitzada per l’individu (permet estimar la despesa energètica per 605 activitats).Estimació de necessitats energètiques en malalts per Long et al. i Ireton-Jones amb la base del càlcul del Metabolisme Basal de Harris-Benedict. Introduint els plecs (Plec Pectoral, Plec Bíceps, Plec Abdominal, Plec Supraespinal, Plec Cuixa anterior, Plec Cama medial, Plec Subescapular, Plec Tricipital), els diàmetres (Diàmetre Antero-Posterior Tòrax, Diàmetre Sagital, Diàmetre Húmer, Diàmetre Fèmur, Diàmetre Biacromial, Diàmetre Transversal Tòrax, Diàmetre Biileocrestal) i els perímetres (Perímetre Braç flexionat, Perímetre Braç, Perímetre Canell, Perímetre Cuixa, Perímetre Cama, Perímetre Tòrax) el programa és capaç de calcular el % de greix, massa òssia, massa muscular i massa residual per diferents fórmules (% Gas Yuhasz, % Gras Faulkner, % Gras Siri, % Gras Brozek, Distribució corporal de Drinkwater). També calcula el perímetre muscular del braç i la cama. També calcula el Pes Objectiu en relació a un % Greix objectiu, calculant quin pes hauria de tenir el pacient per un % de Greix donat.

Objectives: The objective of this study was to evaluate the oral health status of professional soccer players of F.C. Barcelona and its relation to the incidence of sport lesions. Methods: Thirty professional soccer players were consecutively evaluated in the seasons 2003/4, 2004/5 and 2005/6. A research protocol to assess their oral health was developed. DMFT, Quigley & Hein plaque index (PI), Löe & Silness gingival index (GI), World Health Organization malocclusion index, Ramfjord teeth probing pocket depth (PPD), TMJ examination and history of dental trauma were recorded. All physical injuries sustained by players during the season were documented from F.C. Barcelona medical services. Results: Mean DMFT score was 5.7 (SD 4.1), Quigley & Hein plaque index score was 2.3 (SD 1.1), Löe & Silness gingival index was 1.1 (SD 0.8), and periodontal pocket depth was 1.9 mm (SD 0.3). Pearson"s analysis showed a significant correlation between PI and GI (p<0.01). Nine players (30%) presented bruxism - the same proportion of those with severe malocclusion. Seven (23.3%) players had suffered uncomplicated crown fractures. The mean incidence of physical injuries was 8 (SD 3.4) per player. PI and PPD showed a statistically significant correlation to muscle injuries (p<0.05). Conclusions: Soccer players, despite intensive medical follow-up, have significant oral health problems such as untreated caries, gingivitis or malocclusion, and suffer dental trauma as a result of sports activities. Their physical condition could also be associated to oral health

Brain oxidative processes play a major role in age-related cognitive decline, thus consumption of antioxidant-rich foods might help preserve cognition. Our aim was to assess whether consumption of antioxidant-rich foods in the Mediterranean diet relates to cognitive function in the elderly. In asymptomatic subjects at high cardiovascular risk (n = 447; 52% women; age 55-80 y) enrolled in the PREDIMED study, a primary prevention dietary-intervention trial, we assessed food intake and cardiovascular risk profile, determined apolipoprotein E genotype, and used neuropsychological tests to evaluate cognitive function.We also measured urinary polyphenols as an objective biomarker of intake. Associations between energy-adjusted food consumption, urinary polyphenols, and cognitive scores were assessed by multiple linear regression models adjusted for potential confounders. Consumption of some foods was independently related to better cognitive function. The specific associations [regression coefficients (95% confidence intervals)] were: total olive oil with immediate verbal memory [0.755 (0.151-1.358)]; virgin olive oil and coffee with delayed verbal memory [0.163 (0.010-0.316) and 0.294 (0.055-0.534), respectively];walnuts with working memory [1.191 (0.061-2.322)]; and wine with Mini-Mental State Examination scores [0.252 (0.006-0.496)]. Urinary polyphenols were associated with better scores in immediate verbal memory [1.208 (0.236-2.180)]. Increased consumption of antioxidant-rich foods in general and of polyphenols in particular is associated with better cognitive performance in elderly subjects at high cardiovascular risk. The results reinforce the notion that Mediterranean diet components might counteract age-related cognitive decline.

Adipose tissue (AT) is distributed as large differentiated masses, and smaller depots covering vessels, and organs, as well as interspersed within them. The differences between types and size of cells makes AT one of the most disperse and complex organs. Lipid storage is partly shared by other tissues such as muscle and liver. We intended to obtain an approximate estimation of the size of lipid reserves stored outside the main fat depots. Both male and female rats were made overweight by 4-weeks feeding of a cafeteria diet. Total lipid content was analyzed in brain, liver, gastrocnemius muscle, four white AT sites: subcutaneous, perigonadal, retroperitoneal and mesenteric, two brown AT sites (interscapular and perirenal) and in a pool of the rest of organs and tissues (after discarding gut contents). Organ lipid content was estimated and tabulated for each individual rat. Food intake was measured daily. There was a surprisingly high proportion of lipid not accounted for by the main macroscopic AT sites, even when brain, liver and BAT main sites were discounted. Muscle contained about 8% of body lipids, liver 1-1.4%, four white AT sites lipid 28-63% of body lipid, and the rest of the body (including muscle) 38-44%. There was a good correlation between AT lipid and body lipid, but lipid in"other organs" was highly correlated too with body lipid. Brain lipid was not. Irrespective of dietary intake, accumulation of body fat was uniform both for the main lipid storage and handling organs: large masses of AT (but also liver, muscle), as well as in the"rest" of tissues. These storage sites, in specialized (adipose) or not-specialized (liver, muscle) tissues reacted in parallel against a hyperlipidic diet challenge. We postulate that body lipid stores are handled and regulated coordinately, with a more centralized and overall mechanisms than usually assumed.

La situación económica actual ha dado pie al tema de esta conferencia que se desarrollará en dos grandes apartados: primero, trataremos de ideas generales sobre la racionalización versus el racionamiento en la sanidad, y, posteriormente, del racionamiento en la práctica de la nutrición clínica. Racionalizar, según la Real Academia Española de la Lengua, es organizar la producción o el trabajo de manera que aumente los ren - dimientos o reduzca los costos con el mínimo esfuerzo. Mientras que por racionamiento se entiende la acción y efecto de racionar o limitar el consumo de algo para evitar consecuencias negativas. En Europa, el porcentaje del Producto Interior Bruto destinado a Sanidad cae progresivamente mientras el coste de la Sanidad no para de aumentar. Desde el punto de vista económico, éste sería el principal motivo por el que las autoridades sanitarias parece que no tienen más alternativa que racionar. ¿Hasta qué punto el principio ético de justicia es compatible con el racionamiento? Éticamente, parece que para aceptar el racionamiento debería cumplirse no sólo una distribución justa de los recursos limitados sino también el uso racional de los mismos. Si se acepta que el recorte en prestaciones sanitarias es necesario, deberíamos responder a diversas preguntas: ¿qué es lo ético no recortar?, ¿quién decide lo que es médicamente necesario?, ¿cómo se decide? Sin respuestas coherentes a estas preguntas resulta díficil, éticamente, aceptar el racionamiento a nivel sanitario. Al tratar el racionamiento en la práctica de la nutrición clínica, debemos centrarnos en cómo el racionamiento afecta a la desnutrición, y más concretamente de la desnutrición relacionada con la enfermedad, ya que ello centra el origen de la Nutrición Clínica. Por su importancia e implicaciones se están llevando a cabo, en diversos países entre ellos en España, acciones integradas en la estrategia de la Unión Europea: “Together for health: a Strategic Approach for the EU 2008-2013” encaminadas a tomar medidas dirigidas a prevenir y tratar la desnutrición. Pero a pesar de ello, las restricciones persisten, situándonos en la necesidad imperativa de utilizar todas las herramientas a nuestro alcance para prevenir el desarrollo de la desnutrición en los pacientes en riesgo, para detectar precozmente los pacientes con desnutrición o riesgo de desarrollarla y para establecer las medidas de actuación más adecuadas.

El objetivo de este trabajo es proponer cuáles son los portadores de coste de las explotaciones ganaderas porcinas intensivas y su valoración. Para ello en primer lugar hemos descrito el proceso productivo para poder determinar los portadores de coste y finalmente la valoración de los mismos así como la producción en curso. Al tratarse de seres biológicos una de las problemáticas que se plantea para la valoración es la existencia de animales fallecidos durante el proceso productivo. A partir de este aspecto proponemos un sistema de valoración de los portadores de coste. Para comprobar que este modelo es factible, desarrollamos en el último apartado una aplicación práctica a los datos de una explotación ganadera porcina. En la elaboración del producto, en cada una de sus fases de transformación, se obtendrán los portadores de coste del proceso de producción y, eslabón a eslabón, se irá completando de forma minuciosa todo el proceso de valoración.

Les lesions musculars són molt freqüents en el futbol, superant el 30% de totes les lesions; provocant una interrupció en la pràctica esportiva normal del jugador durant dies o setmanes. Així doncs, la prevenció és un factor molt important. L’objectiu d’aquest estudi és el disseny d’un pla integral preventiu de fisioteràpia englobant diferent àmbits: nutrició, salut muscular i preparació física, sempre dins la perspectiva de la salut; per poder amb l’avaluació d’aquest pla disminuir la incidència de lesions musculars en el futbol amateur. Aquesta pràctica de prevenció es durà a terme a un dels dos equips sèniors masculins de futbol aficionat que formaran part de l’estudi (grup d’intervenció), mentre que l’altre equip de característiques el més similars possibles seguirà la seva preparació habitual (grup control). Després d’una temporada es compararà la incidència de lesions musculars entre els dos equips. Si les dades demostren evidències, aquest pla es facilitarà al servei interessat corresponent, perquè el puguin distribuir als equips associats, podent disminuir el número de lesions i així reduir els costos en tractaments.

La caries del biberón es la aparición brusca de caries en los incisivos y molares temporales, relacionadas con el hábito de succionar el chupete o el biberón, impregnados de sustancias cariogénicas. Afecta a los niños en las primeras etapas de la vida. Los dientes de leche son fundamentales para diferentes funciones como la masticación y el habla. Este trabajo revisa cuáles son sus causas más comunes y las medidas preventivas más indicadas.

The finding that tissue δ15N values increase with protein catabolism has led researchers to apply this value to gauge nutritive condition in vertebrates. However, its application to marine mammals has in most occasions failed. We investigated the relationship between δ15N values and the fattening/fasting cycle in a model species, the fin whale, a migratory capital breeder that experiences severe seasonal variation in body condition. We analyzed two tissues providing complementary insights: one with isotopic turnover (muscle) and one that keeps a permanent record of variations in isotopic values (baleen plates). In both tissues δ15N values increased with intensive feeding but decreased with fasting, thus contradicting the pattern previously anticipated. The apparent inconsistency during fasting is explained by the fact that a) individuals migrate between different isotopic isoscapes, b) starvation may not trigger significant negative nitrogen balance, and c) excretion drops and elimination of 15N-depleted urine is minimized. Conversely, when intensive feeding is resumed in the northern grounds, protein anabolism and excretion start again, triggering 15N enrichment. It can be concluded that in whales and other mammals that accrue massive depots of lipids as energetic reserves and which have limited access to drinking water, the δ15N value is not affected by fasting and therefore cannot be used as an indicatior of nutritive condition.
