The objective of this work was to evaluate the chemical composition and fatty acid contents of Amazonian and giant river prawns. After four-month farming, with the same diet for both species, palmitic and stearic acids were the main saturated fatty acids. Oleic acid was the main monounsatured fatty acid, and the eicosapentaenoic and docosahexaenoic acids were the most abundant polyunsaturated acids. Amazonian prawn has higher levels of protein and polyunsaturated fatty acids than those of the giant river prawn, which shows its potential for aquaculture.

The preference of predation on oocytes of Piaractus mesopotamicus, commonly known as 'Pacucaranha', by the prawn Macrobrachium amazonicum, under laboratory conditions, was evaluated regarding to other food items. The prawns were collected with sieves, placed below the underwater vegetation, and maintained in aquarium (12 L each) at a density of 10 individuals/aquarium for acclimatization. Oocytes (O) of P. mesopotamicus were obtained from the disposal of artisanal fishery. Artificial feed (R), fish muscle (M), organic matter accumulated in roots of Salvinia sp. (S) and plant organic matter in decomposition (MV) items were used to compare the preference by M. amazonicum. In all experiments the average of the prawns preying oocytes were higher; significant differences were detected only when comparing O/P and O/M. The preference for oocytes may be due to the presence of yolk, which has a high nutritional value and easy assimilation, in contrast to others items, which resent certain rigidity. This prawn species is exotic on site and the results of this study shows a possible environmental impact on P. mesopotamicus in the region.

The Amazon river prawn (Macrobrachium amazonicum) is a native species with great potential for aquaculture, based on promising results obtained from experimental culture trials in Brazil. The influence of different stocking densities on the development of prawns reared in cages in the nursery phase as well as on their growth when transferred to grow-out net pens at lower densities was evaluated. In the nursery phase, juveniles I (0.157 +/- 0.014 g, 47 days old) were stocked in 0.5 m(-2) cages at densities of 400, 800 and 1200 m(-2). After 71 days, prawns were transferred to grow-out net pens of 2.0 m(-2), at a density of 20 juveniles II m(-2). The treatments were determined by the mean weights registered for the prawns (118 days old) previously stocked at 400, 800 and 1200 juveniles I m(-2) in the nursery phase: 0.94 +/- 0.07 g (T1), 0.61 +/- 0.04 g (T2) and 0.48 +/- 0.07 g (T3), respectively. In the nursery phase, mean survival was above 96%, whereas mean weights were significantly higher (P<0.05) for the density of 400 prawns m(-2). The highest biomass (276.7 g) and productivity (1152 juveniles II m(-2)) were registered at the density of 1200 prawns m(-2), differing significantly (P<0.05) from the lower densities. One month after the transfer of the animals to the net pens, there was recovery in the specific growth rate (SGR) of prawns in all treatments which was significantly higher (P<0.05) in T3 (4.01 +/- 0.36% day(-1)) and T2 (3.60 +/- 0.18% day(-1)). The feed conversion efficiency (FCE) in the first month after the transfer was also significantly higher (P<0.05) in T3 (78.2 +/- 19.1%) when compared to T1 (39.8 +/- 9.5%). These results suggest the occurrence of a compensatory growth in M. amazonicum after transferring them to lower densities, which can point out high densities for nursery cages as a viable practice. After 277 days of grow-out phase in net pens in the cold season, survival, mean weight and biomass did not differ significantly among the treatments, indicating the viability of using net pens in stocking prawns during autumn and winter, since the minimum temperature does not drop below 17 degrees C. Influence of stocking density during the nursery phase on the grow-out of prawns was not observed. The population structure in prawns reared in net-pens was similar to that observed in earthen ponds. The implementation of a hypothetical aquaculture facility with hatchery, nursery and grow-out earthen ponds for raising the Amazon River Prawn Macrobrachium amazonicum in the Pantanal was considered. Eight larviculture cycles per year were projected: four to produce post-larvae for stocking in grow-out bait ponds, and four to stock nursery tanks to sell juveniles as seed to grow-out farms, which produce prawns for human consumption. Annual production would be 146,880 dozen bait prawns and 2,938 thousand juveniles. The assumed sale prices were US$ 1.38 per dozen baits and US$ 15.39 per thousand juveniles. The net present value was US$ 555,890.79, internal rate of return was 48% per year, payback period was 2.4 years and benefit-cost ratio was 3.90. The breakeven price to cover total costs per dozen baits was US$ 0.70 and per thousand juveniles was US$ 17.00, indicating that the selling price assumed for juveniles in base scenario is not realistic. Net return was US$ 84,773.80. The results indicate that this activity would be a lucrative and attractive investment in the Pantanal.

The paper contains a brief review of the studies on the life histories of Indian species of prawns chiefly belonging to the family Penaeidae. References to similar work carried out outside India are furnished where significant variations have been observed. The three main larval stages viz., Nauplius, Protozoea and Zoea (Mysis) and their important characteristics, including modes of locomotion, are described. The post-larval development of one species that has been studied in detail (Metapenaeus dobsoni) is indicated in outline. Some aspects of the bionomics of these prawns, especially breeding and migration, are also briefly dealt with in view of their relevance in their life cycle. An outline of the life histories of some Palaemonid prawns of both fresh water and marine habitats is added at the end and the need for well- planned investigations in regard to species of such economic value as Palaemon carcinus (Macrobrachium rosenbergii) is indicated.

An experiment was conducted for rearing of Meni, Nandus nandus in laboratory condition for seven months with the objective to select appropriate feed for the species and to develop a rearing technique of the species up to the stage of sexual maturation. Different trials were conducted using artificial feed (35.5% protein), dead fresh kachki (Carica soborna), dead fresh prawn (Macrobrachium lamarrei) and live prawn (Macrobrachium lamarrei). The provision of bottom sediment did not significantly influence the growth of fish. Between dead fresh kachki and dead fresh prawn, the fish preferred dead fresh prawn. The fish was found to be reluctant to take dead fresh kachki and prawn as food unless they became very hungry. The fish was found actively feeding on live prawn. The FCR of the prawn as food for N. nandus was found to be 2.5. From the study, it was observed that in laboratory rearing N. nandus preferred live prawn as food than artificial feed, dead fresh kachki and dead fresh prawn. The fish fed on live prawn became sexually matured (eggs or white milt extruded by gentle pressure on the abdomen of the fish) in the laboratory at the end of the experiment.

The effect of AC and DC electric stimulations on the heart-rate and the entire body of Heteropneustis fossillis, Tilapia mossambica and Macrobrachium rosenbergii were studied and presented in kymograph tracings. The reaction of spinal cord in Puntius ticto, Heteropneustis fossilis and Tilapia mossambica to D. C. field was observed to find out its role in electric shocks. A test-check of the electrical resistance of a few species was also conducted. The effect of D. C. and A.C. on the body muscle was found to be the same as that in the case of frog. Different degrees of cardiac slowing were observed in AC and DC. Unbalanced galvanotropic movements were also noticed in spinal fishes.

Decapsulated cysts of Artemia salina were experimentally fed to the larvae of Penaeus monodon, Metapenaeus ensis, M. endeavouri and Macrobrachium rosenbergii and were found to be comparable to freshly hatched brine shrimp nauplii.

对虾病害在世界范围内的广泛传播,给水产养殖和沿海农村经济造成了重大损失。深入开展对虾免疫机制研究并在此基础上寻找对虾疾病防治的有效方法已成为当务之急。研究表明,当对虾等甲壳动物受到外界病原刺激时,其体内的吞噬细胞在吞噬活动中会激活磷酸己糖支路的代谢,引起呼吸爆发,产生多种活性氧分子。另外,受到病原侵染的对虾还会产生其他多种免疫反应,这些免疫反应将消耗大量的能量(ATP),产能的呼吸链会加速运转,由此也会引发大量活性氧的产生。这些活性氧分子可以杀灭入侵的病原微生物,但同时由于活性氧分子反应的非特异性,它们也会对宿主的细胞、组织和器官造成严重伤害,进而导致对虾生理机能的损伤和免疫系统的破坏。所以,消除对虾体内因过度免疫反应产生的过量氧自由基将能够增强其抵御病原侵染的能力,提高免疫力。本论文从中国明对虾体内克隆了线粒体型超氧化物歧化酶(mMnSOD)、胞质型超氧化物歧化酶(cMnSOD)、过氧化氢酶(Catalase)和过氧化物还原酶(Peroxiredoxin)等四种与免疫系统相关的抗氧化酶基因,分析了它们的分子结构特征,组织分布及应答不同病原刺激的表达变化模式,并对其中的mMnSOD基因和Peroxiredoxin基因进行了体外重组表达、分离纯化和酶活性分析。 采用RACE技术从中国明对虾血细胞中克隆了两个超氧化物歧化酶(SOD)基因,通过序列比对分析发现,其中一个为mMnSOD基因,另一个为cMnSOD基因。mMnSOD基因的cDNA全长为1185个碱基,其中开放阅读框为660个碱基,编码220个氨基酸,其中推测的信号肽为20个氨基酸。多序列比对结果显示中国明对虾mMnSOD基因的推导氨基酸序列与罗氏沼虾、蓝蟹的推导氨基酸序列同源性分别为88%和82%。Northern blot结果表明,该基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。半定量RT-PCR结果显示,对虾感染病毒3 h时,该基因在血细胞和肝胰脏中的转录水平显著升高。此外,通过构建原核表达载体,本研究对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活分析。cMnSOD基因的cDNA全长为1284个碱基,其中开放阅读框为861个碱基,编码287个氨基酸。多序列比对结果显示中国明对虾cMnSOD基因的推导氨基酸序列与斑节对虾和凡纳滨对虾的同源性高达98%和94%。组织半定量结果显示,cMnSOD基因在对虾被检测的各个组织中均有表达。 另外,半定量RT-PCR结果表明,对虾感染病毒23h时,该基因在肝胰脏中的转录上升到正常水平的3.5倍;而感染后59 h时,该基因在血细胞中的转录上升到正常水平的2.5倍。 利用根据其他生物过氧化氢酶保守氨基酸序列设计的简并引物,结合RACE技术,从中国明对虾肝胰脏中克隆到了过氧化氢酶基因的部分片段,片段长1725个碱基。多序列比对结果发现目前所得中国明对虾Catalase基因部分片段的推导氨基酸序列与罗氏沼虾和皱纹盘鲍Catalase氨基酸序列的同源性分别达到95%和73%。通过实时荧光定量PCR技术对中国明对虾Catalase基因在各个组织中的分布情况及病毒感染后该基因在血细胞和肝胰脏中的转录变化进行了研究。结果发现,该基因在肝胰脏、鳃、肠和血细胞中表达水平较高,在卵巢、淋巴器官和肌肉中的表达水平相对较弱;感染病毒23 h和37 h时,对虾血细胞和肝胰脏中该基因mRNA的表达量分别出现显著性上升。 依据中国明对虾头胸部cDNA文库提供的部分片段信息,结合SMART-RACE技术,从中国明对虾肝胰脏中克隆到了过氧化物还原酶基因(Peroxiredoxin), 该基因的cDNA全长为942个碱基,其中开放阅读框为594个碱基,编码198个氨基酸。中国明对虾Peroxiredoxin基因的推断氨基酸序列与伊蚊、文昌鱼和果蝇等Peroxiredoxin基因的推断氨基酸序列同源性分别为77%、76%和73%。其蛋白理论分子量为22041.17 Da,pI为5.17。Northern blot结果表明,Peroxiredoxin基因在对虾的肝胰脏、血细胞、淋巴器官、肠、卵巢、肌肉和鳃等组织中均有表达。实时荧光定量PCR结果显示,弧菌感染后,该基因在对虾血细胞和肝胰脏中的转录水平都有明显变化并且表达模式不同。另外,对该基因进行了体外重组表达,并对纯化的重组蛋白进行了质谱鉴定和酶活性分析。酶活性分析表明,复性后的重组蛋白能在DTT存在的条件下还原H2O2。

In India scientific commercial farming of Macrobrachium is yet to get a start, the major constraint being the lack of seed. Seed collection from the natural sources alone may not be sufficient, and in such cases seed production in hatcheries is inevitable. This necessitates knowledge of the reproductive biology of the species technology development for brood stock and larval rearing, and the optimum conditions required for larval development to undertake large scale seed production.The present study was taken up with the major objectives of developing a proper, simple and feasible technology for the larval rearing of Macrobrachium, with special emphasis on. mass larval rearing suitable for commercial level operations.
